The purpose of this study was to investigate genotoxicity and heat shock protein 70 (Hsp70) in human airway smooth muscle cells (HASMCs) exposed to cigarette smoke extract (CSE). HASMCs was exposed to CSE with different doses for 24 h. The level of 8-hydroxydeoxyguanosine (8-OHdG) was determined by using HPLC-ECD, the DNA damage was ana- lyzed by using comet assay, and apoptosis was examined by using Annexin-FITC/PI staining. The pro- duction of Hsp70 after CSE stimulation was tested. Results indicated that CSE significantly increased the level of 8-OHdG, DNA damage and cell apoptosis, and reduced the production of Hsp70. In par- ticular, levels of Hsp70 were inversely correlated with 8-OHdG, DNA damage and cell apoptosis. It was concluded that cigarette smoke induced genotoxicity and decreased the production of cell protective protein Hsp70, which may contribute to the development of some airway diseases.展开更多
BACKGROUND Cigarette smoking(CS)is the most common method of consuming tobacco.Deleterious effects on bone integrity,increased incidence of fractures,and delayed fracture healing are all associated with CS.Over 150 of...BACKGROUND Cigarette smoking(CS)is the most common method of consuming tobacco.Deleterious effects on bone integrity,increased incidence of fractures,and delayed fracture healing are all associated with CS.Over 150 of the 6500 molecular species contained in cigarette smoke and identified as toxic compounds are inhaled by CS and,via the bloodstream,reach the skeletal system.New technologies designed to develop a reduced-risk alternative for smokers are based on electronic nicotine delivery systems,such as e-cigarettes and tobacco heating systems(THS).THS are designed to heat tobacco instead of burning it,thereby reducing the levels of harmful toxic compounds released.AIM To examine the effects of THS on osteoprogenitor cell viability and function compared to conventional CS.METHODS Human immortalized mesenchymal stem cells(n=3)and primary human preosteoblasts isolated from cancellous bone samples from BG Unfall Klinik Tübingen(n=5)were osteogenically differentiated in vitro with aqueous extracts generated from either the THS 2.4“IQOS”or conventional“Marlboro”cigarettes for up to 21 d.Cell viability was analyzed using resazurin conversion assay(mitochondrial activity)and calcein-AM staining(esterase activity).Osteogenic differentiation and bone cell function were evaluated using alkaline phosphatase(AP)activity,while matrix formation was analyzed through alizarin red staining.Primary cilia structure was examined by acetylatedα-tubulin immunofluorescent staining.Free radical production was evaluated with 2’,7’-dichlorofluoresceindiacetate assay.RESULTS Our data clearly show that THS is significantly less toxic to bone cells than CS when analyzed by mitochondrial and esterase activity(P<0.001).No significant differences in cytotoxicity between the diverse flavors of THS were observed.Harmful effects from THS on bone cell function were observed only at very high,non-physiological concentrations.In contrast,extracts from conventional cigarettes significantly reduced the AP activity(by two-fold)and matrix mineralization(four-fold)at low concentrations.Additionally,morphologic analysis of primary cilia revealed no significant changes in the length of the organelle involved in osteogenesis of osteoprogenitor cells,nor in the number of ciliated cells following THS treatment.Assessment of free radical production demonstrated that THS induced significantly less oxidative stress than conventional CS in osteoprogenitor cells.CONCLUSIONTHS was significantly less harmful to osteoprogenitor cells during osteogenesisthan conventional CS. Additional studies are required to confirm whether THS isa better alternative for smokers to improve delays in bone healing followingfracture.展开更多

It is necessary to develop burn-resistant and thermal barrier complex functional coatings on the titanium alloys surface due to the poor high temperature performance for titanium materials and the problem of“titanium...It is necessary to develop burn-resistant and thermal barrier complex functional coatings on the titanium alloys surface due to the poor high temperature performance for titanium materials and the problem of“titanium fire”which is easily happened.MTU Aero Engines GmbH has developed a complex functional coating which has great performance of burn resistant and sealing,the coatings has already been applied on compressor blades and casing for aero-engines and showed great performance.In this study,the complex functional coating which is composed of an electro-spark deposited amorphous Ti40Zr25Ni3 burn-resistant layer and a high-energy plasma spraying prepared YSZ thermal barrier layer was prepared on titanium alloys surface.Then the heat insulation ability,burn resistant performance and the bonding strength were investigated.The experimental results showed that the interface between the coating and the matrix was typical metallurgical bonded,and the average bonding strength was 36.335 MPa;when the temperature of the flame on the one side of the specimen reached 600℃,the average insulated temperature of the coating samples was 70.67℃;when the flame temperature was 350℃,the titanium samples without the complex functional coatings were burned,while the samples with the coating showed great performance of burn resistant even when the temperature was 750℃.This indicates that the new functional coating has good heat-insulating and burn-resistant performance.展开更多

Human bronchi smooth muscle cells were cultured with CSE at the different concentrations. The expression of Hsp70 mRNA and Hsp70 was detected by reverse translation-polymerase chain reaction (RT-PCR) and Western blotting respectively. Levels of Hsp70 mRNA and Hsp70 in lymphocytes from 20 patients with COPD and 20 healthy smoking control subjects were measured by RT-PCR and Western blotting. The results showed the expression of both Hsp70 mRNA and Hsp70 was decreased conformably in human bronchi smooth muscle cells treated with CSE at certain concentration in vitro. The A values of the Hsp70 mRNA expression were 0.24±0.11 and 0.42±0.13 respectively in COPD patients and healthy smoking controls with the difference being significant (P<0.01). There was also significant difference in the A values of the Hsp70 expression between COPD patients and healthy smoking controls (20.9±9.9 vs 44.8±15.3, P<0.01). The levels of Hsp70 mRNA had strongly positive correlation with Hsp70 protein (r = 0.85, P<0 01). It was suggested that the expression of Hsp70 mRNA was in concordance with the expression of Hsp70, which could provide a basis on the study of Hsp70 gene regulation and Hsp70 gene in the development of COPD.展开更多

The assumed flame structure consists of a broad preheat-vaporization zone where the rate of gas-phase chemical reaction is small, a thin reaction zone composed of three regions: gas, tar and char combustion where convection and the vaporization rate of the fuel particles are small, and a broad convection zone. The analysis is performed in the asymptotic limit, where the value of the characteristic Zeldovich number is large and the equivalence ratio is larger than unity(i.e.u≥1). The principal attention is made on the determination of a non-linear burning velocity correlation. Consequently, the impacts of radiation, heat loss and particle size as the determining factors on the flame temperature and burning velocity of biomass particles are declared in this research.展开更多

It is found that the amount ...The effect of heat treatment and thermal exposure on the microstructure and mechanical properties of non burning β titanium alloy Ti 25V 15Cr 2Al 0.2C (mass fraction, %) was investigated. It is found that the amount of α precipitation in samples after solution treatment followed by air cooling increases with increasing solution temperature. After solution treatment, the alloy was subjected to two different heat treatments to stabilize the material. The results show that the single ageing treatment(700 ℃, 4 h, AC) leads to a large amount of α precipitation, while the triplex treatment(850 ℃, 2 h, FC+700 ℃, 6 h, FC+540 ℃, 6 h, AC) significantly suppresses the formation of α precipitates. It is more important that after long term exposure at 540 ℃(the expected application temperature) samples heat treated by the triplex treatment have markedly higher ductility than those given the single aging treatment. The intermetallic compound TiCr 2 is observed in samples after long term exposure, which further degrades the ductility of the alloy.展开更多

Onsite mine fire generates large volumes of heat-affected coal in Jharia coalfields,India.Direct utilization of such heat-affected coal in thermal utilities is not feasible as such coal does not have the desirable vol...Onsite mine fire generates large volumes of heat-affected coal in Jharia coalfields,India.Direct utilization of such heat-affected coal in thermal utilities is not feasible as such coal does not have the desirable volatile matter required for combustion.In the present work,experimental studies have been carried out to investigate the possible utilization of such heat-affected coal in thermal utilities by blending with other coal.Heat-affected coal(31%ash and 5300 kcal/kg GCV)collected from Jharia coalfield were blended with thermal coal(28%ash and 5650 kcal/kg GCV)in different ratios of 90:10,80:20,70:30 and 60:40 to identify the desirable blend ratio for burning of blended coal in thermal utilities.Burning characteristics of all the coals were carried out using TGA.Various combustion parameters such as ignition temperature,peak temperature,burnout temperature,ignition index,burnout index,combustion performance index,rate and heat intensity index of the combustion process and activation energy were evaluated to analyse the combustion process.Experimental and theoretical analysis shows the blend ratio of 90:10 can be used in place of only thermal coal in utilities to reduce the fuel cost.展开更多

In general, rice husks burnt outdoors at a lower temperature range of 300-400 ℃, which cannot assure complete combustion, thus generating a large volume of toxic air pollutants. A heat flux of 40 kW/m^2, with a cone calorimeter, is the equivalent to the 700 ℃ of an incinerator. The test result shows that the mass reduction rate of the sample at this or at a higher temperature condition was 99.5% or higher, meaning that the sample was almost completely combusted. In this study using rice husks, the amount of carbon dioxide, which is a greenhouse gas, discharged were 1.57-3.61 kg/kg. This is as high as 10 times, than produced in other studies. When the rice husks are burnt outdoors, they are not completely combusted as the combustion temperature remains low, and the rice husk residuals are continuously being combusted in a smoldering phase which creates a large volume of carbon dioxide and carbon monoxide. Therefore, it is recommended to burn rice husks at 700 ℃ or higher to minimize the carbon dioxide and carbon monoxide emissions.展开更多

The microstructures and mechanical properties of the alloy after the five heat treatments and thermal expo...Five new heat treatment processes were designed,which were divided into three groups by their characteristics. The microstructures and mechanical properties of the alloy after the five heat treatments and thermal exposure at 500,550 ℃ for 100 h were tested. The results indicate that a little differences exist in the performance of mechanical properties at room-temperature after the five heat treatments,and the thermal stability is the key factor for determining heat treatment process. Among the three groups of heat treatment processes,the best thermal stability is achieved after the first group of heat treatment. After annealing treatment at intermediate temperature,some defects and uneven grain boundaries are remained,which leads to the reduction fractions of precipitations on unit grain boundary and the harmful effect of precipitations on grain boundary is weakened. The process of annealing at 650 ℃ for 4 h is recommended the best heat treatment process for Ti40 alloy.展开更多

文摘Onsite mine fire generates large volumes of heat-affected coal in Jharia coalfields,India.Direct utilization of such heat-affected coal in thermal utilities is not feasible as such coal does not have the desirable volatile matter required for combustion.In the present work,experimental studies have been carried out to investigate the possible utilization of such heat-affected coal in thermal utilities by blending with other coal.Heat-affected coal(31%ash and 5300 kcal/kg GCV)collected from Jharia coalfield were blended with thermal coal(28%ash and 5650 kcal/kg GCV)in different ratios of 90:10,80:20,70:30 and 60:40 to identify the desirable blend ratio for burning of blended coal in thermal utilities.Burning characteristics of all the coals were carried out using TGA.Various combustion parameters such as ignition temperature,peak temperature,burnout temperature,ignition index,burnout index,combustion performance index,rate and heat intensity index of the combustion process and activation energy were evaluated to analyse the combustion process.Experimental and theoretical analysis shows the blend ratio of 90:10 can be used in place of only thermal coal in utilities to reduce the fuel cost.
文摘This paper analyzes the combustion characteristics and greenhouse gas emissions from varied heat fluxes with rice husks. In general, rice husks burnt outdoors at a lower temperature range of 300-400 ℃, which cannot assure complete combustion, thus generating a large volume of toxic air pollutants. A heat flux of 40 kW/m^2, with a cone calorimeter, is the equivalent to the 700 ℃ of an incinerator. The test result shows that the mass reduction rate of the sample at this or at a higher temperature condition was 99.5% or higher, meaning that the sample was almost completely combusted. In this study using rice husks, the amount of carbon dioxide, which is a greenhouse gas, discharged were 1.57-3.61 kg/kg. This is as high as 10 times, than produced in other studies. When the rice husks are burnt outdoors, they are not completely combusted as the combustion temperature remains low, and the rice husk residuals are continuously being combusted in a smoldering phase which creates a large volume of carbon dioxide and carbon monoxide. Therefore, it is recommended to burn rice husks at 700 ℃ or higher to minimize the carbon dioxide and carbon monoxide emissions.

文摘Five new heat treatment processes were designed,which were divided into three groups by their characteristics. The microstructures and mechanical properties of the alloy after the five heat treatments and thermal exposure at 500,550 ℃ for 100 h were tested. The results indicate that a little differences exist in the performance of mechanical properties at room-temperature after the five heat treatments,and the thermal stability is the key factor for determining heat treatment process. Among the three groups of heat treatment processes,the best thermal stability is achieved after the first group of heat treatment. After annealing treatment at intermediate temperature,some defects and uneven grain boundaries are remained,which leads to the reduction fractions of precipitations on unit grain boundary and the harmful effect of precipitations on grain boundary is weakened. The process of annealing at 650 ℃ for 4 h is recommended the best heat treatment process for Ti40 alloy.
