A Study on the Integration of Chinese Culture in High School English Teaching

The inseparable relationship between language and culture indicates that culture teaching is indispensable in English teaching.The ultimate goal of culture teaching in English teaching is to cultivate learners’cross-cultural communication skills.Cross-cultural communication is a two-way process,which requires not only knowledge of English culture but also the use of English as a medium for disseminating Chinese culture to the outside world.However,in actual English teaching,the teaching of Chinese culture is a mere formality,and the teaching effect is not obvious.Therefore,it is necessary to conduct a survey and research on Chinese culture teaching in high school English classrooms to understand its current situation,existing problems,and their causes,so as to provide suggestions for Chinese culture teaching in high school.

Establishment of a Highly Efficient Regeneration System for the Mature Embryo Culture of Wheat

Establishment of a highly efficient regeneration system for the mature embryo of wheat will provide a convenient tool for wheat tissue culture and transformation, thereby facilitating the transformation of foreign genes into wheat. By using the mature embryos derived from 20 different wheat lines including Shi 4185, Yumai 66, Lunxuan 987, CB037, Yangmai 6, Xinchun 9, Bobwhite, Han 6172, Zheng 9023, Jimai 20, Ningchun 4, and Jing 411, the effects of some factors including inoculation methods, initiating culture media, organic additives, antioxidants, and auxins on the regeneration from the explants were evaluated. The results indicated that the scraping embryo culture was better than the whole embryo culture, the Aa medium was better than the SD2 medium and dicamba was better than 2,4-D in increasing the regeneration frequency. An Adi medium was established in this study by adding silver nitrate, cysteine, ascorbic acid, dicamba, glutamine into the Aa medium at the concentration of 4,40, 100, 2, and 5 mg L^-1, respectively. By using the Adi medium and the scraping technique, the regeneration frequencies of the mature embryos of CB037, Lunxuan 987, Hart 6172, Yangmai 6, Bobwhite, Zheng 9023, Shi 4 185, and Jimai 20 became 85.6, 60,1, 46.0, 42.1,42.0, 34.0, 33.0, and 32.0%, respectively, which were about 5-8 times higher than that obtained from the conventional culture mediums and techniques. This novel regeneration system could be helpful in wheat transformation.

On the Cultural Conflicts in The Wedding Banquet Based on Hall's High-Low Context Culture Theory

The Wedding Banquet is a movie about a gay Taiwan Residents immigrant man who marries a mainland Chinese woman to placate his parents and get her a green card. His plan backfires and things get out of hand when his parents arrive in the United States to plan his wedding banquet. Based on Hall's theory of high-context and low-context culture, this paper analyzes the cultural conflicts in the movie from six perspectives, and offers suggestions to the harmonious development of China and America. It is hoped that the paper can help readers get an in-depth comprehension of Hall's theory of high-context and low-context culture and discover some invisible conflicts between China and the West and finally raise people's consciousness of cultural conflicts and ways of handling the conflicts. With the advancement of globalization, intercultural communicative competence is becoming more and more important, and social communication between China and the West will become more relaxed and fruitful if people acquire more knowledge in intercultural communication.

FATTY ACIDS PROFILE IN A HIGH CELL DENSITY CULTURE OF ARACHIDONIC ACID-RICH PARIETOCHLORIS INCISA (TREBOUXIOPHYCEAE,CHLOROPHYTA) EXPOSED TO HIGH PFD

The changes in arachidonic acid (AA) and fatty acids profiles along the growth curve of Parietochloris incisa, a coccoid snow green alga, were studied in a 2.8 cm light-path flat photobioreactor, exposed to strong photon flux density [PFD, 2400 μEmol/(m 2·s)]. Sixteen fatty acids were identified by gas chromatography showing that AA was the dominant fatty acid (33%-41%) followed by linoleic acid (17%-21%). AA content was closely investigated with respect to total fatty acids (TFA), ash free dry weight (AFDW) of cell mass as well as total culture content. These parameters were influenced significantly in a similar manner by culture growth phase, i.e., slightly decreasing in the lag period, gradually increasing in the logarithmic phase, becoming maximal at the early stationary phase, starting to decrease at the late stationary phase, sharply dropping at the decline phase. The increase in AA per culture volume during the logarithmic phase was not only associated with the increase in AFDW but also connected with a corresponding increase in AA/TFA, TFA/AFDW as well as AA/AFDW. The sharp decrease in AA content of the culture during the decline phase was mainly due to the decrease in AA/TFA, TFA/AFDW and AA/AFDW, although AFDW declined only a small extent. Maximal AA concentration, obtained at the early stationary phase, was 900 mg/L culture volume, and the average daily net increase of AA during 9 days logarithmic growth was 1.7 g/(m 2·day). Therefore, harvesting prior to the decline phase in a batch culture, or at steady state in continuous culture mode seems best for high AA production. The latter possibility was also further confirmed by continuous culture with 5 gradients of harvesting rate.

Effects of High Ammonium Concentration on Growth and Nutrient Uptake of Lettuce Plants with Solution Culture

A nutrition solution experiment was conducted over two months to investigate the response of vegetable crops to high concentrations of ammonium, using lettuce （Lactuca sativa L. cv. Angustana Irish） as a test crop. Ammonium concentrations were designed in 5 levels, ranging from 12 mmol N L^-1 to 22 mmol N L^-1 and local tap water was used as water source. At the first culture stage （0-9 days）, lettuce plants maintained normal growth while the lettuce roots were increasingly impaired. During the subsequent three stages the root structure was greatly damaged, and roots became brown or black through continuous supply of high concentration of ammonium. However, there was no obvious reduction of the aboveground biomass of the plants in the high ammonium treatments compared to those supplied with nitrate alone. In contrast to results obtained in another experiment from us with distilled water, the detrimental effect of high ammonium concentration on lettuce growth was greatly alleviated. Based on the results, it was postulated that the small amount of nitrate and the higher amount of bicarbonate existed in the tap water might mitigate the adverse effects of high ammonium N. The higher bicarbonate content in water and soil has usually been regarded as a major constraint factor limiting plant growth in calcareous soil areas. However, the reaction of bicarbonate to ammonium might produce positively interactive effect on reduction of both damages. The lettuce plants grown in ammonium solutions took up less P, K, Fe, Mn and Cu and more Ca than those grown in the nitrate nutrient solution. In conclusion, the results indicated that the N form imposed an obvious influence on absorption of cations and anions. Supplying ammonium-N stimulated transport of Ca, Mg and Mn to shoots of lettuce.

pFTAA: a high affinity oligothiophene probe that detects filamentous tau in vivo and in cultured neurons

Tauopathies describe a group of neurodegenerative diseases in which the protein tau,encoded by the gene MAPT,is aberrantly misfolded,leading to tau aggregation,neural dysfunction,and cell death（Spillantini and Goedert,2013）.In Alzheimer＇s disease（AD）,tau forms the characteristic intracellular neurofibrillary tangles（NFTs）,which are thought to be the major cause of neurodegeneration（Bloom,2014）.In other tauopathies,including frontotemporal dementia with Parkinsonism linked to chromo- some 17 （FTDP-17T）, corticobasal degeneration and progressive supranuclear palsy, there are specific forms of tau aggregates and filaments without any amyloid pathology, demonstrating tau＇s po- tent disease-causing potential （Spillantini and Goedert, 2013）. Tau is a microtubule （MT） binding protein, which becomes abnormally hyperphosphorylated on several residues prior/during the process of aggregation, thereby causing loss of its MT binding activity （Mandelkow and Mandelkow, 2012）.

Microfluidic three-dimensional cell culture of stem cells for high-throughput analysis

Although the recent advances in stem cell engineering have gained a great deal of attention due to their high potential in clinical research,the applicability of stem cells for preclinical screening in the drug discovery process is still challenging due to difficulties in controlling the stem cell microenvironment and the limited availability of high-throughput systems.Recently,researchers have been actively developing and evaluating three-dimensional(3D)cell culture-based platforms using microfluidic technologies,such as organ-on-a-chip and organoid-on-a-chip platforms,and they have achieved promising breakthroughs in stem cell engineering.In this review,we start with a comprehensive discussion on the importance of microfluidic 3D cell culture techniques in stem cell research and their technical strategies in the field of drug discovery.In a subsequent section,we discuss microfluidic 3D cell culture techniques for high-throughput analysis for use in stem cell research.In addition,some potential and practical applications of organ-on-a-chip or organoid-on-a-chip platforms using stem cells as drug screening and disease models are highlighted.

Assessment of AMBR^TMas a model for high-throughput cell culture process development strategy

The development and delivery of high quality therapeutic products necessitates the need for highthrough-put (HTP) process development tools. Traditionally, these works requires a combination of shake flask and small-scale stirred tank bioreactor (STR) which are labor and resource intensive and time-consuming. Here we demonstrate a strategy for rapid and robust cell culture process development by evaluating and implementing the use of a new HTP disposable micro bioreactor (MBR) called AMBRTM system (Advanced Microscale Bioreactor) that has the capabilities for automated sampling, feed addition, pH, dissolved oxygen (DO), gassing and agitation controls. In these studies the performance of two monoclonal antibody (MAb) producing cell lines (MAb1 and MAb2) was evaluated both in the AMBR system and 3-L STR. We demonstrated that cell culture performance (growth and viability, production titer and product quality) were similar in both vessel systems. Furthermore, process control and feed optimization were demonstrated in an additional cell line (MAb3) in the disposable MBR and its performance confirmed at STR scale. The results indicate that the AMBR system can be used to streamline the process development effort and facilitate a rapid and robust cell culture process development effort for MAb programs in a HTP manner.

Swab culture monitoring of automated endoscope reprocessors after high-level disinfection

AIM:To conduct a bacterial culture study for monitoring decontamination of automated endoscope reprocessors(AERs) after high-level disinfection(HLD).METHODS:From February 2006 to January 2011,authors conducted randomized consecutive sampling each month for 7 AERs.Authors collected a total of 420 swab cultures,including 300 cultures from 5 gastroscope AERs,and 120 cultures from 2 colonoscope AERs.Swab cultures were obtained from the residual water from the AERs after a full reprocessing cycle.Samples were cultured to test for aerobic bacteria,anaerobic bacteria,and mycobacterium tuberculosis.RESULTS:The positive culture rate of the AERs was 2.0%(6/300) for gastroscope AERs and 0.8%(1/120) for colonoscope AERs.All the positive cultures,including 6 from gastroscope and 1 from colonoscope AERs,showed monofloral colonization.Of the gastroscopeAER samples,50%(3/6) were colonized by aerobic bacterial and 50%(3/6) by fungal contaminations.CONCLUSION:A full reprocessing cycle of an AER with HLD is adequate for disinfection of the machine.Swab culture is a useful method for monitoring AER decontamination after each reprocessing cycle.Fungal contamination of AERs after reprocessing should also be kept in mind.

High Density Culture of Porphyridium cruentum in a 42 L Internal Airlift Loop Photobioreactor

The key limiting factors to high-density culture of Porphyridium cruentum are the uptake of light energy and nutrient by the microalgal cells. Under the optimal conditions of carrier culture, both cell mass and cell density were increased significantly up to 5.2 g/L (DW) and 5.2107/ml. Furthermore, the effects of the liquid circulation velocity, light intensity and initial cell density on cell mass productivity of P. cruentum were investigated in a 42 L internal loop airlift photobioreactor. Although the light intensity was as low as 100 mmol/(m2s), the light damage or the photoinhibition phenomenon was observed under the culture condition of low initial cell mass (0.10 g/L, DW) and high liquid circulation velocity (0.30 m/s). However, a higher cell growth rate and a high cell mass productivity were achieved with the same conditions only at high initial cell mass (about 0.80 g/L, DW). Under the optimal conditions, the cell specific growth rate, cell mass volumetric and areal output rate reached to 0.95 d-1, 0.80 g/(Ld) and 42.5 g/(m2d) respectively. Finally, a method of nutrient feeding and gradual increase of light intensity in different cultural stages was developed, which further improved the cell mass, cell mass volumetric and areal output rate to 5.9 g/L, 1.2 g/(Ld) and 61.7 g/(m2d) respectively.

Oxidative modification of high density lipoprotein induced by cultured human arterial smooth muscle cells

Objective: To observe the oxidative modification of high density lipoprotein (HDL) induced by cultured human arterial smooth muscle cells (SMCs). Methods: HDL cocultured with SMCs at 37℃ in 48 h was subjected, and native HDL (N-HDL) served as control. Oxidative modification of HDL was identified by using agarose gel electrophoresis. Absorbances of conjugated diene (CD) and lipid hydroperoxide (LOOH) were measured with ultraviolet spectrophotometry at 234 and 560 nm respectively, and fluorescence intensity of thiobarbuturic acid reaction substance (TBARS) with fluorescence spectrophotometry at 550 nm emission wavelength with excitation at 515 nm. Results: In comparison with N-HDL, the electrophoretic mobility of SMCs-cocultured HDL was increased, and the contents of CD, LOOH and TBARS HDL were very significantly higher than those of the control HDL (P<0.01). Conclusion: Oxidative modification of HDL can be induced by human arterial SMCs.

Analysis of Culture Conflicts in The Joy Luck Club from the High-context and Low-context Culture Perspective

As a work that reflects culture differences between China and America, the Joy Luck Club has drawn a wide attention from scholars whose researches are more of holistic analyses from a macro perspective of cross-cultural communication. In accordance with the degree of dependence of semantic on dialogue context and dialogue language itself, Edward Hall divides culture into high-context culture and low-context culture, and holds that Chinese culture is a high-context culture, while American culture is a low- context one. According to Hall's theory, the Sino- US cultural conflicts reflected by the mother- daughter conflicts in the Joy Luck Club can be analyzed from high-context and low-context culture perspective. And after conflicts, there finally will be a harmonious trend of Sino-US culture development, changing from clash and conflict to convergence and integration.

An Analysis of the Mode of Information Transfer in High-context Cultures in Terms of Red Cliff

Effects of Different Ridge Culture Ways on Stem Diameter,Plant Height,the Number of Leaves and Yield of Phaseolus vulgaris L. under Dry Farming Conditions

The variety "Aifeng" was used as experimental material,and the stem diameter,plant height,the number of leaves and yield of P. vulgaris under three ways of ridge culture (bedding,high ridge and M ridge) were observed and detected to study the effects of different ridge cultures on the growth and yield. The results showed that the stem diameter,plant height,the number of leaves and yield under M ridge culture were higher than that of bedding and high ridge.

Teaching English Literature Through Popular Culture in China: Record and Reflection

Classic English literature is high culture of a sort, too high for tertiary English majors in the 21st China, who have grown up with total exposure to popular culture in an electronic way, to reach so that they show indifference to literature class. To solve this problem, this paper intends to consider the implications and feasibilities of integrating popular culture into literature education in English departments of Chinese universities. After carefully reviewing the current English literature teaching connected with popular culture both in China and other parts of the world, this paper concludes that the choice of the popular culture texts shall be neither too random to include any form of popular culture nor too limited to only cover the film/TV adaptations of literary classics. Subsequently, the paper records the procedure and progress of the author’s classic-literature and popular-culture class and reflects upon the students’ substantial responses to the class. From the record and the reflection, three major findings are drawn: The students know little about how classic literature inspire popular culture and when they know, they become excited and want to know more;when further exploring those original texts which inspired their favourite pop, the students find themselves greatly challenged and stimulated as well;the students have strong desire to reach the “highness” of classic literature and would like to read challenging texts with their own spirits. A conclusion can be drawn from these findings: Both academic and popular literacy of English majors in Chinese universities should be enhanced, and it is workable to incorporate popular culture into literature classroom with careful preparations for classroom activities and guarantees of the smooth implements of these activities.

A Study of the Cultivation of English Cultural Awareness in High School——Based on the Core Competencies and Values of English

Cultural awareness is the basis of communication,and it includes our own cultural values,beliefs and the ability to perceive the world.When students learn English and communicate with people from English culture,cultural awareness becomes very important.Meanwhile,National English Curriculum Standards for General High School puts cultural awareness in the key position of the Core Competencies and Values of English subjects.At present,the academic research and analysis on the Core Competencies and Values of English mainly focus on the language ability,learning ability and students’learning motivation.This paper intends to explore the cultivation of high school students’English cultural awareness through qualitative research and literature collation and there are 5 parts totally in this paper.The first part is the introduction,introducing the background of cultural consciousness and the significance of research.The second part is a literature review,analyzing the previous opinions both domestically and internationally.The third part is a review of cultural consciousness,summarizing the theory,historical development and main genres of cultural consciousness.The fourth part is the exploration of the cultivation of cultural awareness,including the requirements,the problems and methods of the cultivation of cultural awareness in high school English teaching.The fifth part is the conclusion,which advocates the cultivation of cultural consciousness and the development of English subject through the summary and reflection.The significance of this paper lies in the majority of teachers to create a harmonious teaching environment,facilitate the students’cultural awareness,improve high school students’cultural understanding and cultural awareness,as well as improve high school students’English discipline Core Competencies and Values so that it could benefit the development of English in our country.

Cultural Infiltration of Foreign Literary Works in Senior High School Chinese Textbooks

Foreign literary works are an important part of Chinese textbooks in high schools.However,the exam-oriented education results in a marginalized and unsatisfactory teaching of foreign literary works.Based on the high school Chinese textbook by PEP(People's Education Press),teaching practices,and requirements of the new curriculum standards,this article analyzes the foreign literary works in the textbook,explores its value and the infiltration of cultural spirit in teaching,discusses the problems in teaching,as well as provides suggestions to them in hope to gain attention in the teaching of foreign literary works,broaden students' horizons,and cultivate their humanistic spirit.

A comparative study on the contents of vitamins in the natural and the cultured prawn

A systematically comparative study on the contents of vitamins in the natural and the cultured prawn, Penaeus chinensis, was made by using High-efficiency Liquid Phase Chromatography in 1989. The results show that the vitamin C (Vc) content in the natural prawn was 8. 19% higher than that of the cultured prawn.As the natural prawn's body length increasing, the Vc content in cephalothorax increased continuously, but after August 10, the Vc content in the cultured prawn fell down rapidly. No significant differences in vitamin B1, B2,B6, nicotinic acid (NAc), nicotinamide (NAm) and folio acid (FAc) were found between the natural and the cultured prawn. Therefore, Vc probably is one of major factors limiting the growth and development of Chinese prawn.
