Microwave-assisted extraction(MAE)was used for extraction of effective components of sarcandra glabra(Thunb.),and then chromatographic fingerprint of sarcandra glabra(Thunb.)was studied by high performance liquid chro...Microwave-assisted extraction(MAE)was used for extraction of effective components of sarcandra glabra(Thunb.),and then chromatographic fingerprint of sarcandra glabra(Thunb.)was studied by high performance liquid chromatography/diode array detector(HPLC/DAD).The conditions of MAE were optimized by an orthogonal experiment,and then the authentication and validation of the chromatographic fingerprint were conducted.Nine peaks were identified as common peaks in the fingerprint chromatograms,and isofraxidin was considered as a reference compound and quantified.Relative standard deviations of retention time and peak area of each component were less than 3% and 8%,respectively.Similarity and difference analysis were conducted by use of PCA and relation coefficient.Twenty batches of sarcandra glabra(Thunb.)samples from two different producing areas could be classified into two different groups in the PCA model.The results showed that MAE-HPLC/DAD method was simple,efficient and stable for the study of complex chromatographic fingerprint of sarcandra glabra(Thunb.),which could provide more reliable and precise information for quality evaluation.展开更多

The results showed that broken efficiency us...The broken efficiency of cell wall and the release amount of Pichia pastoris intracellular protein under different cell breaking conditions were investigated in this paper. The results showed that broken efficiency using hot alkali combined with high-pressure homogenizing method was higher than that of enzyme hydrolysis, hot alkali treatment and high-pressure homogenation, respectively. Suspended medium had little effect on the broken efficiency of yeast cell, but had significant effect on the protein release yield. The results indicated that optimal condition for intracellular proteins extraction was 30% (wet weight, w/v) of yeast cells suspend in 50 mM phosphate buffer (pH 10.0), water bathed at 60?C for 2 hours, homogenized twice at 100 MPa pressure. The broken efficiency of Pichia pastoris cell could reach 87.6% and the protein yield was 35.48 g per 100 g cells.展开更多

The method used online microwave-assisted steam extraction coupled with solid phase extraction(MASE-SPE), which was followed by the analysis using high performance liquid chromatography-mass spectro- metry(HPLC-MS/MS). The SAs residues were extracted successively with water steam under microwave irradiation, and thus directly introduced into an SPE colunm containing cation-exchange resin. The SAs were then eluted with methanol-ammonia(90:10, volume ratio) from the SPE column and followed by HPLC-MS/IVIS. The limits of detec- tion(LODs) for the analytes ranged from 0.24 ng/g to 0.49 ng/g. The limits of quantification(LOQs) ranged from 0.82 ng/g to 1.63 ng/g. Average recoveries of SAs were 76.3%--92.1%. The developed method was a reliable and envi-ronmentally friendly alternative to previous methods with respect to time, solvent and labor consumption for the analysis of SAs in animal foodstuffs.展开更多
To improve the gas extraction efficiency of single seam with high gas and low air permeability,we developed the"fracturing-sealing"integration technology,and carried out the engineering experiment in the3305...To improve the gas extraction efficiency of single seam with high gas and low air permeability,we developed the"fracturing-sealing"integration technology,and carried out the engineering experiment in the3305 Tunliu mine.In the experiment,coal seams can achieve the aim of antireflection effect through the following process:First,project main cracks with the high energy pulse jet.Second,break the coal body by delaying the propellant blasting.Next,destroy the dense structure of the hard coal body,and form loose slit rings around the holes.Finally,seal the boreholes with the"strong-weak-strong"pressurized sealing technology.The results are as follows:The average concentration of gas extraction increases from8.3%to 39.5%.The average discharge of gas extraction increases from 0.02 to 0.10 m^3/min.The tunneling speeds up from 49.5 to 130 m/month.And the permeability of coal seams improves nearly tenfold.Under the same conditions,the technology is much more efficient in depressurization and antireflection than common methods.In other words,it will provide a more effective way for the gas extraction of single seam with high gas and low air permeability.展开更多
为挖掘混合类型热电联产机组的节能潜力、降低发电成本,通过EBSILON软件搭建60MW双抽(double-extraction condensing unit,CC)-抽背(extraction condensing unit with a high back-pressure,CB)热电联产机组的仿真模型,研究该联合机组...为挖掘混合类型热电联产机组的节能潜力、降低发电成本,通过EBSILON软件搭建60MW双抽(double-extraction condensing unit,CC)-抽背(extraction condensing unit with a high back-pressure,CB)热电联产机组的仿真模型,研究该联合机组的运行特性并建立基于可解释增强机和鸟群算法的双抽-抽背热电联产机组负荷优化模型,最后以典型日热电负荷优化任务为例,给出双机热电负荷优化结果。结果表明:当保持双抽机组的中压流量不变,存在中压流量极限值10.39t/h,使低压流量与电功率的运行区域只受到最大主蒸汽流量、最小凝汽量以及最小主蒸汽流量的限制;存在中压流量极限值59.26t/h,使运行区域只受最大主蒸汽流量和最小凝汽量限制;当双机总中压流量一定时,双抽-抽背机组的联合运行区域可以用极限工况即抽背机组承担最大中压流量,双抽机组承担剩余中压流量来近似表示。该优化方法与热电负荷平均分配方案对比,典型日可以降低1148.58GJ热耗,发电标准煤耗率由212.10g/(kW·h)降低为209.05g/(kW·h),可以节省标煤3.05g/(kW·h)。展开更多
