This paper examines temperature effects on the rheological properties of two types of high-density water-based drilling fluids (fresh water-based and brine-based) under high temperature and high pressure (HTHP) with a Fann 50SL rheometer. On the basis of the water-based drilling fluid systems formulated in laboratory, this paper mainly describes the influences of different types and concentration of clay, the content of a colloid stabilizer named GHJ-1 and fluid density on the rheological parameters such as viscosity and shear stress. In addition, the effects of aging temperature and aging time of the drilling fluid on these parameters were also examined. Clay content and proportions for different densities of brine-based fluids were recommended to effectively regulate the rheological properties. Four theological models, the Bingham, power law, Casson and H-B models, were employed to fit the rheological parameters. It turns out that the H-B model was the best one to describe the rheological properties of the high-density drilling fluid under HTHP conditions and power law model produced the worst fit. In addition, a new mathematical model that describes the apparent viscosity as a function of temperature and pressure was established and has been applied on site.展开更多

Basic helix-loop-helix(bHLH) transcription factors(TFs) have been shown to play important roles in regulating high-temperature resistance in plants. However, the bHLH TFs responsible for high-temperature tolerance in cucumbers have not been identified. We used transcriptome profiling to screen the high temperature-responsive candidate bHLH TFs in cucumber. Here, we found that the expression of 75 CsbHLH genes was altered under high-temperature stress. The expression of the CsSPT gene was induced by high temperatures in TT(Thermotolerant) cucumber plants. However, the Csspt mutant plants obtained by the CRISPR-Cas9 system showed severe thermosensitive symptoms, including wilted leaves with brown margins and reduced root density and cell activity. The Csspt mutant plants also exhibited elevated H_(2)O_(2) levels and down-regulated photosystem-related genes under normal conditions.Furthermore, there were high relative electrolytic leakage(REC), malondialdehyde(MDA), glutathione(GSH), and superoxide radical(O_(2)^(·-)) levels in the Csspt mutant plants, with decreased Proline content after the high-temperature treatment. Transcriptome analysis showed that the photosystem and chloroplast activities in Csspt mutant plants were extremely disrupted by the high-temperature stress compared with wildtype(WT) plants. Moreover, the plant hormone signal transduction, as well as MAPK and calcium signaling pathways were activated in Csspt mutant plants under high-temperature stress. The HSF and HSP family genes shared the same upregulated expression patterns in Csspt and WT plants under high-temperature conditions. However, most bHLH, NAC, and bZIP family genes were significantly down-regulated by heat in Csspt mutant plants. Thus, these results demonstrated that CsSPT regulated the high-temperature response by recruiting photosynthesis components, signaling pathway molecules, and transcription factors. Our results provide important insights into the heat response mechanism of CsSPT in cucumber and its potential as a target for breeding heat-resistant crops.展开更多

With this super-amphiphobic agent as the core agent, a super-amphiphobic, strong self-cleaning and high-performance water-based drilling fluid system has been developed by combining with other agents based on drilled formation, and compared with high-performance water-based drilling fluid and typical oil based drilling fluid commonly used in oilfields. The results show that the super-amphiphobic, strong self-cleaning and high-performance water-based drilling fluid has better rheology, and high temperature and high pressure filtration similar with that of oil-based drilling fluid, inhibiting and lubricating properties close to oil based drilling fluid. Besides, the super-amphiphobic system is non-toxic, safe and environmentally friendly. Field tests show this newly developed drilling fluid system can prevent wellbore collapse, reservoir damage and pipe-sticking, increase drilling speed and lower drilling cost, meeting the requirement of safe, high efficient, economic and environmentally friendly drilling. Compared with other drilling fluids, this new drilling fluid system can reduce downhole complexities by 82.9%, enhance the drilling speed by about 18.5%, lower drilling fluid cost by 39.3%, and increase the daily oil output by more than 1.5 times in the same block.展开更多

However, studies have indicated that high-carbohydrate(HC) diet disrupt the homeostasis of the gut–liver axis in largemouth bass, resulting in decreased intestinal acetate and butyrate level.Method Herein, we had concepted a set of feeding experiment to assess the effects of dietary sodium acetate(SA) and sodium butyrate(SB) on liver health and the intestinal microbiota in largemouth bass fed an HC diet. The experimental design comprised 5 isonitrogenous and isolipidic diets, including LC(9% starch), HC(18% starch), HCSA(18% starch;2 g/kg SA), HCSB(18% starch;2 g/kg SB), and HCSASB(18% starch;1 g/kg SA + 1 g/kg SB). Juvenile largemouth bass with an initial body weight of 7.00 ± 0.20 g were fed on these diets for 56 d.Results We found that dietary SA and SB reduced hepatic triglyceride accumulation by activating autophagy(ATG101, LC3B and TFEB), promoting lipolysis(CPT1α, HSL and AMPKα), and inhibiting adipogenesis(FAS, ACCA, SCD1 and PPARγ). In addition, SA and SB decreased oxidative stress in the liver(CAT, GPX1α and SOD1) by activating the Keap1-Nrf2 pathway. Meanwhile, SA and SB alleviated HC-induced inflammation by downregulating the expression of pro-inflammatory factors(IL-1β, COX2 and Hepcidin1) through the NF-κB pathway. Importantly, SA and SB increased the abundance of bacteria that produced acetic acid and butyrate(Clostridium_sensu_stricto_1). Combined with the KEGG analysis, the results showed that SA and SB enriched carbohydrate metabolism and amino acid metabolism pathways, thereby improving the utilization of carbohydrates. Pearson correlation analysis indicated that growth performance was closely related to hepatic lipid deposition, autophagy, antioxidant capacity, inflammation, and intestinal microbial composition. Conclusions In conclusion, dietary SA and SB can reduce hepatic lipid deposition;and alleviate oxidative stress and inflammation in largemouth bass fed on HC diet. These beneficial effects may be due to the altered composition of the gut microbiota caused by SA and SB. The improvement effects of SB were stronger than those associated with SA.展开更多

文摘The combination of nano sizes, large pore sizes and green synthesis is recognized as one of the most crucial and challenging problems in constructing metal-organic frameworks(MOFs).Herein,a water-based strategy is proposed for the synthesis of nanoscale hierarchical MOFs(NH-MOFs)with high crystallinity and excellent stability. This approach allows the morphology and porosity of MOFs to be fine tuned, thereby enabling the nanoscale crystal generation and a well-defined hierarchical system. The aqueous solution facilitates rapid nucleation kinetics, and the introduced modulator acts as a deprotonation agent to accelerate the deprotonation of the organic ligand as well as a structure-directing agent(SDA)to guide the formation of hierarchical networks. The assynthesized NH-MOFs(NH-ZIF-67)were assessed as efficient adsorbents and heterogeneous catalysts to facilitate the diffusion of guest molecules, outperforming the parent microZIF-67.This study focuses on understanding the NH-MOF growth rules, which could allow tailor-designing NH-MOFs for various functions.
文摘Difenoconazole(DIF)is a representative variety of broad-spectrum triazole fungicides and liposoluble pesticides. However, the water solubility of DIF is so poor that its application is limited in plant protection. In addition, the conventional formulations of DIF always contain abundant organic solvents, which may cause pollution of the environment. In this study, two DIF/cyclodextrins(CDs)inclusion complexes(ICs)were successfully prepared, which were DIF/β-CD IC and DIF/hydroxypropyl-β-CD IC(DIF/HP-β-CD IC).The effect of cyclodextrins on the water solubility and the antifungal effect of liposoluble DIF pesticide were investigated.According to the phase solubility test, the molar ratio and apparent stability constant of ICs were obtained.Fourier transform infrared spectroscopy, thermal gravity analysis,X-ray diffraction and scanning electron microscopy were used systematically to characterize the formation and characteristics of ICs.The results noted that DIF successfully entered the cavities of two CDs.In addition, the antifungal effect test proved the better performance of DIF/HP-β-CD IC,which exceeded that of DIF emulsifiable concentrate. Therefore, our study provides informative direction for the intelligent use of liposoluble pesticides with cyclodextrins to develop water-based environmentally friendly formulations.

文摘The stamping-out strategy has been used to control highly pathogenic avian influenza viruses in many countries, driven by the belief that vaccination would not be successful against such viruses and fears that avian influenza virus in vaccinated birds would evolve more rapidly and pose a greater risk to humans. In this review,we summarize the successes in controlling highly pathogenic avian influenza in China and make suggestions regarding the requirements for vaccine selection and effectiveness. In addition,we present evidence that vaccination of poultry not only eliminates human infection with avian influenza virus, but also significantly reduces and abolishes some harmful characteristics of avian influenza virus.

文摘High-temperature stress threatens the growth and yield of crops. Our results provide important insights into the heat response mechanism of CsSPT in cucumber and its potential as a target for breeding heat-resistant crops.

文摘Microwave-assisted mechanical excavation has great application prospects in mines and tunnels, but there are few field experiments on microwave-assisted rock breaking. This paper takes the Sishanling iron mine as the research object and adopts the self-developed high-power microwave-induced fracturing test system for hard rock to conduct field experiments of microwave-induced fracturing of iron ore.The heating and reflection evolution characteristics of ore under different microwave parameters(antenna type, power, and working distance)were studied, and the optimal microwave parameters were obtained.Subsequently, the ore was irradiated with the optimal microwave parameters, and the cracking effect of the ore under the action of the high-power open microwave was analyzed. The results show that the reflection coefficient(standing wave ratio)can be rapidly(<5 s)and automatically adjusted below the preset threshold value(1.6)as microwave irradiation is performed. When using a right-angle horn antenna with a working distance of 5 cm,the effect of automatic reflection adjustment reaches the best among other antenna types and working distances. When the working distance is the same, the average temperature of the irradiation surface and the area of the high-temperature area under the action of the two antennas(right-angled and equal-angled horn antenna)are basically the same and decrease with the increase of working distance. The optimal microwave parameters are:a right-angle horn antenna with a working distance of 5 cm.Subsequently,in further experiments, the optimal parameters were used to irradiate for 20 s and 40 s at a microwave power of 60 kW,respectively. The surface damage extended 38 cm×30 cm and 53 cm×30 cm,respectively, and the damage extended to a depth of about 50 cm.The drilling speed was increased by 56.2%and 66.5%,respectively,compared to the case when microwaves were not used.

文摘Based on the amphiphobic theory on underground rock surface, a super-amphiphobic agent is developed and evaluated which can form nano-micro papilla structure on rock, filter cake and metal surface, reduce surface free energy, prevent collapse, protect reservoir, lubricate and increase drilling speed. Compared with other drilling fluids, this new drilling fluid system can reduce downhole complexities by 82.9%, enhance the drilling speed by about 18.5%, lower drilling fluid cost by 39.3%, and increase the daily oil output by more than 1.5 times in the same block.

文摘Background Adequate level of carbohydrates in aquafeeds help to conserve protein and reduce cost. The improvement effects of SB were stronger than those associated with SA.

文摘High-performance water-based drilling fluids(HPWBFs)are essential to wellbore stability in shale gas exploration and development.Laponite is a synthetic hectorite clay composed of disk-shaped nanoparticles. This paper analyzed the application potential of laponite in HPWBFs by evaluating its shale inhibition, plugging and lubrication performances.Shale inhibition performance was studied by linear swelling test and shale recovery test.Plugging performance was analyzed by nitrogen adsorption experiment and scanning electron microscope(SEM)observation.Extreme pressure lubricity test was used to evaluate the lubrication property.Experimental results show that laponite has good shale inhibition property, which is better than commonly used shale inhibitors, such as polyamine and KCl.Laponite can effectively plug shale pores. It considerably decreases the surface area and pore volume of shale, and SEM results show that it can reduce the porosity of shale and form a seamless nanofilm.Laponite is beneficial to increase lubricating property of drilling fluid by enhancing the drill pipes/wellbore interface smoothness and isolating the direct contact between wellbore and drill string.Besides, laponite can reduce the fluid loss volume.According to mechanism analysis, the good performance of laponite nanoparticles is mainly attributed to the disk-like nanostructure and the charged surfaces.

文摘High spatiotemporal resolution brain electrical signals are critical for basic neuroscience research and high-precision focus diagnostic localization,as the spatial scale of some pathologic signals is at the submillimeter or micrometer level. This entails connecting hundreds or thousands of electrode wires on a limited surface. This study reported a class of flexible, ultrathin, highdensity electrocorticogram(ECoG)electrode arrays. The challenge of a large number of wiring arrangements was overcome by a laminated structure design and processing technology improvement. The flexible,ultrathin,high-density ECoG electrode array was conformably attached to the cortex for reliable, high spatial resolution electrophysiologic recordings. The minimum spacing between electrodes was 15μm,comparable to the diameter of a single neuron.Eight hundred electrodes were prepared with an electrode density of 4444 mm^(-2).In focal epilepsy surgery, the flexible,high-density,laminated ECoG electrode array with 36 electrodes was applied to collect epileptic spike waves inrabbits, improving the positioning accuracy of epilepsy lesions from the centimeter to the submillimeter level. The flexible,high-density,laminated ECoG electrode array has potential clinical applications in intractable epilepsy and other neurologic diseases requiring high-precision electroencephalogram acquisition.

文摘As the key infrastructure of space-ground integrated information networks, satellite communication networks provide high-speed and reliable information transmission. In order to meet the burgeoning service demands of the IoT and the Internet, the low-latency LEO satellite network has developed rapidly. However,LEO satellites face inherent problems such as small coverage, fast moving speed and short overhead time, which will be more severe when serving high-dynamic users,e.g.high-speed rails and airplanes. The heterogeneous network composed of GEO,MEO and LEO satellites can provide various services, whose network management and resource allocation are also more challenging.
