In this study the three-dimensional (3-D) model of the ligand-binding domain (V106-P322) of human interleukin-6 receptor (hlL-6 R) was constructed by computer-guided ho-mology modeling technique using the crystal structure of the ligand-binding domain (K52-L251) of human growth hormone receptor (hGHR) as templet. Furthermore, the active binding region of the 3-D model of hlL-6R with the ligand (hlL-6) was predicted. In light of the structural characteristics of the active region, a hydrophobic pocket shielded by two hydrophilic residues (E115 and E505) of the region was identified by a combination of molecular modelling and the site-directed or double-site mutation of the twelve crucial residues in the ligand-binding domain of hIL-6R (V106-P322). We observed and analyzed the effects of these mutants on the spatial conformation of the pocket-like region of hlL-6 R. The results indicated that any site-directed mutation of the five Cys residues (four conservative Cys residues: Cyst 21, Cys132, Cys165, Cys176; near membrane Cys residue: Cys193) or each double-site mutation of the five residues in WSEWS motif of hIL-6R (V106-P322) makes the corresponding spatial conformation of the pocket region block the linkage between hlL-6 R and hlL-6. However, the influence of the site-directed mutation of Cys211 and Cys277 individually on the conformation of the pocket region benefits the interaction between hlL-6R and hlL-6. Our study suggests that the predicted hydrophobic pocket in the 3-D model of hIL-6R (V106-P322) is the critical molecular basis for the binding of hlL-6R with its ligand, and the active pocket may be used as a target for designing small hlL-6R-inhibiting molecules in our further study. 展开更多

目的:探究胃癌患者病灶组织角蛋白7(CK7)、人表皮生长因子受体-2(HER2)、错配修复蛋白MutL同源物1(MLH1)、错配修复蛋白MutS同源物2(MSH2)及错配修复蛋白MutS同源物6(MSH6)的表达变化情况。方法:选取2020年2月—2023年1月苏州市相城人... 目的:探究胃癌患者病灶组织角蛋白7(CK7)、人表皮生长因子受体-2(HER2)、错配修复蛋白MutL同源物1(MLH1)、错配修复蛋白MutS同源物2(MSH2)及错配修复蛋白MutS同源物6(MSH6)的表达变化情况。方法:选取2020年2月—2023年1月苏州市相城人民医院收治的120例胃癌患者为研究对象,比较病灶组织及癌旁组织的CK7、HER2、MLH1、MSH2及MSH6的表达情况,并比较不同性别、年龄、TNM分期、分化程度、淋巴结转移情况及病灶位置者的病灶组织CK7、HER2、MLH1、MSH2及MSH6表达情况。结果:胃癌患者病灶组织CK7、HER2阳性率及MLH1、MSH2、MSH6表达缺失率显著高于癌旁组织,差异有统计学意义(P<0.05)。不同性别、年龄及病灶位置病灶组织的CK7、HER2、MLH1、MSH2及MSH6表达比较,差异无统计学意义(P>0.05);Ⅲ期、Ⅳ期病灶组织CK7、HER2阳性率高于Ⅰ期、Ⅱ期,MLH1、MSH2、MSH6表达缺失率均显著低于Ⅰ期、Ⅱ期,差异有统计学意义(P<0.05)。分化程度较低者病灶组织的CK7、HER2阳性率显著高于分化程度较高者,MLH1、MSH2及MSH6表达缺失率显著低于于分化程度较高者,差异有统计学意义(P<0.05)。有淋巴结转移者CK7、HER2阳性率显著高于无淋巴结转移者,MLH1、MSH2及MSH6表达缺失率显著低于无淋巴结转移者,差异有统计学意义(P<0.05)。结论:胃癌患者病灶组织的CK7、HER2、MLH1、MSH2及MSH6的表达相对异常,且不同TNM分期、分化程度及淋巴结转移情况者的差异较大。 展开更多

Methods: Fifty rabbits were randomly divided into 5 groups (n=10) according to the treatment: sham operation group (group A), ischemla-reperfusion group (group B), low-dose remifentanil group (group C), mediate-dose remifentanil group (group D), and high-dose remlfentanil group (group E) Myocardial TLR4 mRNA levels, NF-r.B protein expression and serum levels of IL-6 were observed in 120 min after reperfusion. Results: The myocardial expressions of TLR4 mRNA, NF-rd3 protein and IL-6 level in sera of groups B, C, D and E were elevated compared with group A. However, remifentanil significantly reduced the levels of TLR4 mRNA, NF- r.B protein expression and serum IL-6 in groups C, D and E compared with group B. There were remarkable differences between the groups (P〈O.O1). Conclusion: Intravenous remifentanil has protective effect against rabbit myocardial ischemia/reperfusion injury. Objective: To evaluate the expression and its clinical significance of interleukin 6 (IL-6), soluble glycoprotein 130 (sgp130), interleukin 8 (IL-8) and type A interleukin 8 receptor (IL-8RA) in acute promyelocytic leukemia (APL) patients during all-trans retinoic acid (ATRA) induction treatment. Methods: Plasma and bone marrow mononuclear cell (MNC) culture supernatant IL-6, sgp130, IL-8 concentration of 18 cases with APL were kinetically measured in vivo and in vitro (ELISA). Bone marrow MNC IL-8RA was measured by flow cytometry after being cultured with ATRA (10?6mmol/L). Results: Plasma IL-6, sgp130, IL-8 levels were higher than normal (P<0.05), IL-6, spg130 levels correlated with white blood cell (WBC) counts (P<0.05) while IL-8 levels correlated with body temperature (P<0.05) at initial diagnosis. After 72-hour incubation with ATRA, concentration of IL-6 of bone marrow MNC culture supernatant did not change, that of sgp130 mildly decreased, and IL-8 significantly decreased while the positive rate of IL-8RA on bone marrow MNC increased. During ATRA treatment, plasma IL-6 changes were correlated with WBC counts. Peak levels of IL-6 and WBC were lower in patients who received intermittent therapy than those who received continuous therapy. Plasma IL-6 and IL-8 were increased when complicated with infection and IL-8 seemed more sensitive. Conclusion: Plasma IL-6, sgp130, IL-8 levels may reflect patients' responsiveness to ATRA treatment, and could be used to predict hyperleukocytosis and intercurrent infection. ATRA induces APL cell differentiation possibly via sgp130 signal transducer chain. Measurement of sgp130 had certain meaning to prognosis. 展开更多

In the present study it was proved first that human recombinant interleukin-6(HrIL-6) significantly augmented natural killer(NK) cell activity derived from human fetal spleens against K562 target cells in a 4 hours 51Cr release assay. The enhancement of NK activity with 24 hours preincubation in HrlL-6 was dose-dependent, and significantly higher than that of fresh NK cells and controls cultured with RPMI-1640 medium alone (P<0.001). We also found that IL-6 was able to augment NK activity from different fetal spleens at 20 to 40 weeks of gestation (up to 2.24 to 2.78 times), and no difference of NK activity of fetal splenocytes treated by HrIL-6 was observed between different fetal age (32.3% to 45.4%, P>0.05). Furthermore, IL-6-augmented NK activity of fetal splenocytes was very similar to adult levels (P>0.05). These finding strongly indicated that IL-6 plays an important role in the development of NK cell function during the gestational period, suggesting that IL-6 may be of importance in the regulation of host defense mechanisms against malignancies and viral diseases. 展开更多

In order to provide the experimental basis for the further studies on the oncogenic mechanism of the E6 protein from human papillomavirus type 16 (HPV16), the eukaryotic expression vector pcDNA3.1 (-)/E6 was used for the study on the effect of E6 protein to influence the secretory activity of LPS-induced 3MP-1-macrophages, and the reconstructed plasmid pcDNA3.1 (-)/E6 was transfected into THP-1-macrophages. The expression of E6 gene was assayed in macrophage lysates by using Western blot analysis and the level of TNF-α or IL-1β was examined by ELISA. All of data were analyzed by SPSS12.0. As demonstrated by Western blot analysis, the expression of E6 protein with a molecular weight of about 18 kDa by plasmid pcDNA3.1 (-)/E6 in THP-1-macrophages could be detected. However, as demonstrated by ELISA assay, the level of TNF-α or IL-1β in lysates of THP-1-macrophages showed an obvious difference between the pcDNA3.1 (-)/E6 group and the LPS control group or the pcDNA3.1 (-) control group (P 〈 0.01), but no significant difference existed between pcDNA3.1 (-) control group and LPS control group ( P 〉 0.05). All these results illustrate that the transient over-expression of HPV6 E6 protein reduces the production of TNF-α and IL-1β induced by LPS in THP-1-macrophages. 展开更多

目的探讨吴茱萸碱对结直肠癌小鼠的抑瘤作用及对白介素-6受体(interleukin-6 receptor,IL-6R)/信号转导转录激活因子3(signal transducer and activator of transcription 3,STAT3)通路的影响。方法将59只BALA/C小鼠随机留取9只作为对照... 目的探讨吴茱萸碱对结直肠癌小鼠的抑瘤作用及对白介素-6受体(interleukin-6 receptor,IL-6R)/信号转导转录激活因子3(signal transducer and activator of transcription 3,STAT3)通路的影响。方法将59只BALA/C小鼠随机留取9只作为对照组,其余皮下接种CT-26结直肠癌细胞株建立结直肠癌小鼠模型,将建模成功的40只小鼠随机分为模型组、顺铂组、吴茱萸碱组和顺铂+吴茱萸碱组,每组10只。顺铂组用15 mg·kg^(-1)·d^(-1)顺铂灌胃,顺铂+吴茱萸碱组用15 mg·kg^(-1)·d^(-1)顺铂+15 mg·kg^(-1)·d^(-1)吴茱萸碱灌胃,对照组、模型组用等量生理盐水灌胃。各组均每日灌胃1次,连续19周。末次灌胃24 h后,处死小鼠,称肿瘤质量并计算肿瘤体积、肿瘤抑制率;用酶联免疫吸附实验(enzyme linked immunosorbent assay,ELISA)检测小鼠肿瘤组织的白细胞介素-6(interleukin-6,IL-6)、白细胞介素-1β(interleukin-1β,IL-1β)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平;用HE染色观察小鼠肿瘤组织的病理学变化;用免疫组织化学法检测肿瘤组织的微血管密度(microvesse density,MVD)和血管内皮生长因子(vascular endothelial growth factor,VEGF)水平;用Western blot检测肿瘤组织中IL-6R/STAT3通路相关蛋白的表达水平。结果从模型组到吴茱萸碱组、顺铂组、顺铂+吴茱萸碱组,小鼠肿瘤的质量依次减轻、肿瘤体积依次减小,IL-6、IL-1β、TNF-α水平依次降低(P<0.05)。模型组小鼠肿瘤组织中可见大片灶性坏死,细胞核增大、深染,大小不一,排列紊乱,有异形表现,呈典型恶性肿瘤细胞形态特征,肿瘤细胞密集;吴茱萸碱组、顺铂组和顺铂+吴茱萸碱组的肿瘤细胞核固缩、变小,肿瘤细胞形态破坏,密度依次降低,肿瘤组织内坏死区依次增大,MVD依次减小(P<0.05),VEGF阳性表达依次减弱(P<0.05)。从模型组到吴茱萸碱组、顺铂组、顺铂+吴茱萸碱组,小鼠肿瘤组织中IL-6R、p-STAT3、G 1/S-特异性周期蛋白-D1(G 1/S specific cyclin D1,Cyclin D1)、生存素(survivin)的蛋白水平依次下降(P<0.05)。结论吴茱萸碱可能通过抑制IL-6R/STAT3通路,发挥抗血管生成效应,从而抑制肿瘤的生长。 展开更多

目的:观察红景天苷(salidroside Sal.)对氯化钴(cobaltous chloride,Co Cl2·6H2O)诱导的缺氧人肾小管上皮细胞(human kidney tubular epithelia cell,HKC)Toll样受体2(toll-like receptor 2,TLR-2)表达、白细胞介素-6(interleukin-... 目的:观察红景天苷(salidroside Sal.)对氯化钴(cobaltous chloride,Co Cl2·6H2O)诱导的缺氧人肾小管上皮细胞(human kidney tubular epithelia cell,HKC)Toll样受体2(toll-like receptor 2,TLR-2)表达、白细胞介素-6(interleukin-6,IL-6)的分泌及细胞凋亡的影响。方法:HKC细胞置于六孔板培养,随机分为正常对照组、缺氧模型组、红景天苷(salidroside Sal.)干预组(25μg/ml、50μg/ml、100μg/ml)。其中缺氧模型采用300μmol/L氯化钴处理6 h得到,红景天苷干预组在此模型基础上再用不同浓度的红景天苷处理缺氧细胞24 h。RT-PCR和Western blot检测TLR-2的表达,ELISA检测各组细胞上清中IL-6含量,流式细胞仪检测缺氧HKC凋亡情况。结果:氯化钴诱导HKC TLR-2蛋白(P<0.05)及RNA表达上调(P<0.01)、IL-6分泌增加(P<0.01)及细胞凋亡增加(P<0.01)。25μg/ml和50μg/ml的红景天苷能降低缺氧HKC细胞IL-6的水平(P<0.01),50μg/ml红景天苷抑制作用强于25μg/ml(P<0.05)。与缺氧模型组比较,尽管100μg/ml红景天苷能降低缺氧HKC细胞IL-6的绝对值水平,但差异无统计学意义(P>0.05)。25μg/ml,50μg/ml和100μg/ml红景天苷均能抑制TLR-2蛋白表达(P<0.01),且100μg/ml较25μg/ml红景天苷抑制作用显著(P<0.05),但25μg/ml与50μg/ml、50μg/ml与100μg/ml红景天苷比较,它们抑制作用的强弱差异无统计学意义(P>0.05)。三种浓度的红景天苷也能抑制缺氧HKC TLR-2的RNA表达(P<0.01),50μg/ml及100μg/ml红景天苷抑制作用强于25μg/ml红景天苷(P<0.05),但50μg/ml与100μg/ml红景天苷比较,二者抑制作用差异无统计学意义(P>0.05)。50μg/ml和100μg/ml红景天苷能抑制缺氧HKC凋亡(P<0.01),但25μg/ml红景天苷未表现出抑制凋亡的作用(P>0.05),50μg/ml与100μg/ml红景天苷比较,两者抑制凋亡的作用差异无统计学意义(P>0.05)。结论:红景天苷对缺氧HKC的TLR-2蛋白及RNA表达、IL-6的分泌及细胞凋亡均有抑制作用,但其抑制效应呈现非剂量依赖性。 展开更多

乳腺癌是全球女性最常见的恶性肿瘤,也是导致女性死亡的主要原因之一。仅2022年,中国约有34.1万的乳腺癌新发病例,呈逐年快速增长趋势。据统计,激素受体阳性/人类表皮生长因子受体-2阴性乳腺癌约占所有乳腺癌的70%。内分泌治疗是该类乳... 乳腺癌是全球女性最常见的恶性肿瘤,也是导致女性死亡的主要原因之一。仅2022年,中国约有34.1万的乳腺癌新发病例,呈逐年快速增长趋势。据统计,激素受体阳性/人类表皮生长因子受体-2阴性乳腺癌约占所有乳腺癌的70%。内分泌治疗是该类乳腺癌治疗的基石,其联合CDK4/6抑制剂的治疗方案已得到国内外专家的一致共识,成为激素受体阳性/人类表皮生长因子受体-2阴性乳腺癌的标准治疗。截止目前国内已上市的CDK4/6抑制剂包括达尔西利、哌柏西利、瑞波西利和阿贝西利。对于激素受体阳性/人类表皮生长因子受体-2阴性局部晚期和转移性乳腺癌,CDK4/6抑制剂联合芳香化酶抑制剂或氟维司群一线或二线及以上治疗均可大幅度降低疾病进展或死亡风险,奠定了CDK4/6抑制剂在晚期一线或后线治疗地位。在早期辅助治疗阶段,阿贝西利、瑞波西利已显示出显著延长患者iDFS及OS疗效,而需要接受新辅助治疗、内脏危象及CDK4/6抑制剂经治等患者研究目前仍在探索之中。该文系统性介绍CDK4/6抑制剂的药理学特征及临床研究进展,总结CDK4/6抑制剂联合内分泌治疗的破局和困局。 展开更多

可移植性大鼠急性粒系白血病(Brown Norway rat acute myelocytic leukemia,BNML)是研究人急性粒系白血病的颇为完备的实验模型。由于重组人IL-6(rhIL-6)可作用于大鼠,然而大鼠BNML细胞系LT12缺乏rhIL-6受体(rhIL-6R)表达。为了在BNML... 可移植性大鼠急性粒系白血病(Brown Norway rat acute myelocytic leukemia,BNML)是研究人急性粒系白血病的颇为完备的实验模型。由于重组人IL-6(rhIL-6)可作用于大鼠,然而大鼠BNML细胞系LT12缺乏rhIL-6受体(rhIL-6R)表达。为了在BNML模型探讨rhIL-6在体外以及体内对白血病细胞增殖和分化的影响,本文应用改良的磷酸钙共沉淀法将携带rhIL-6R cDNA的逆转录病毒表达质粒XM6/IL-6RNeo导入LT12细胞,经过G418筛选获得G418抗性克隆,建立了表达rhIL-6R的LT12亚细胞系(称之为R cell lines)。我们分别应用三种不同的方法(rhIL-6R的单克隆抗体和FACScan检测、^(125)I-rhIL-6的配基结合分析测定、地高辛配基标记的rhIL-6R cDNA探针的细胞原位杂交技术)分折了R细胞系在体外不同条件下传代过程中及给BN大鼠尾静脉回输后其rhIL-6R的表达。研究结果表明,用改良的磷酸钙共沉淀法将编码rhIL-6R cDNA和Neo^R基因的逆转录病毒表达质粒XM6/IL-6RNeo导入LT12细胞系后,受体细胞在体外含适当浓度G418的基质液中传代培养,可长期稳定表达rhIL-6R,给BN大鼠尾静脉回输后20天左右,其脾脏和骨髓细胞均可检测到rhIL-6R的明显表达,而且~3H-TdR掺入分析证明细胞膜表面所表达的rhIL-6R可以介导外源性rhIL-6信号传到细胞核内。 展开更多

目的探讨STAT3信号通路在IL-6抑制人单核细胞来源树突状细胞(human monocytederived dendritic cells,mo DCs)趋化因子受体7(C-C chemokine receptor type 7,CCR7)表达中的作用。方法采用CD14+磁珠分离法从外周血中分离得到单核细胞,加... 目的探讨STAT3信号通路在IL-6抑制人单核细胞来源树突状细胞(human monocytederived dendritic cells,mo DCs)趋化因子受体7(C-C chemokine receptor type 7,CCR7)表达中的作用。方法采用CD14+磁珠分离法从外周血中分离得到单核细胞,加入rh GM-CSF和rh IL-4在体外诱导培养树突状细胞。LPS用于刺激mo DCs表达CCR7,RT-PCR检测不同浓度IL-6处理后再用LPS刺激后的mo DCs CCR7 mRNA表达。Western blot检测IL-6处理mo DCs和IL-6预处理后再加入STAT3磷酸化特各异性抑制剂JSI-124的mo DCs STAT3、p-STAT3表达。IL-6预处理并抑制STAT3磷酸化后,LPS刺激48 h,RT-PCR检测mo DCs CCR7 mRNA的表达,流式细胞仪检测mo DCs CCR7蛋白表达,细胞迁移实验检测mo DCs细胞迁移功能。结果与单独LPS刺激组比较,IL-6明显抑制LPS刺激的mo DCs CCR7mRNA的表达(P<0.05)。Western blot结果显示,与单独LPS刺激组比较,IL-6导致mo DCs细胞内STAT3高度活化;而抑制STAT3信号高度活化后,与IL-6+LPS处理组比较,mo DCs CCR7 mRNA表达、蛋白分子表达、细胞迁移能力明显升高(P<0.05),与LPS刺激组无差异(P>0.05)。结论 IL-6通过高度活化STAT3信号通路抑制人单核细胞来源树突状细胞CCR7表达,导致树突状细胞的迁移能力受损,而靶向干预STAT3通路可能成为纠正树突状细胞迁移障碍的潜在手段。 展开更多

Objective: To investigate the effect of activated protein C (APC) on inflammatory responses in human umbilical vein endothelial cells (HUVEC) stimulated with lipopolysaccharide (LPS). Methods: The second passage of collagenase digested HUVEC was divided into the following groups: serum free medium control group (SFM control), phosphate buffer solution control group (PBS control), LPS group with final concentration of 1 μg/ml (LPS group), APC group with final concentration of 7 μg/ml, Pre-APC group (APC pretreatment for 30 min prior to LPS challenge), and Post-APC group (APC administration 30 min after LPS challenge). Supernatant was harvested at 0, 4, 8, 12 and 24 h after LPS challenge. Interleukin-6 (IL-6) and Interleukin-8 (IL-8) levels were analyzed with ELISA. Cells were harvested at 24 h after LPS challenge, and total RNA was extracted. Mes-senger RNA levels for IL-6 and IL-8 were semi-quantitatively determined by RT-PCR. Results: Compared with control group, IL-6 and IL-8 levels steadily increased 4 to 24 h after LPS stimulation. APC treatment could increase LPS-induced IL-6 and IL-8 production. The mRNA levels of IL-6 and IL-8 exhibited a similar change. Conclusion: APC can further increase the level of IL-6 and IL-8 induced by LPS. The effect of these elevated cytokines is still under investigation. 展开更多
