Cells of Candida albicans(C.albicans) can invade humans and may lead to mucosal and skin infections or to deep-seated my coses of almost all inner organs,especially in immunocompromised patients.In this context,both the host immune status and the ability of C.albicans to modulate the expression of its virulence factors are relevant aspects that drive the candidal susceptibility or resistance;in this last case,culminating in the establishment of successful infection knownas candidiasis.C.albicans possesses a potent arma-mentarium consisting of several virulence moleculesthat help the fungal cells to escape of the host immuneresponses.There is no doubt that the secretion of aspartyl-type proteases,designated as Saps,are one of the major virulence attributes produced by C.albicans cells,since these hydrolytic enzymes participate in a wide range of fungal physiological processes as well as in different facets of the fungal-host interactions.For these reasons,Saps clearly hold promise as new potential drug targets.Corroborating this hypothesis,the introduction of new anti-human immunodeficiency virus drugs of the as party l protease inhibitor-type(HIV PIs) have emerged as new agents for the inhibition of Saps.The introduction of HIV PIs has revolutionized the treatment of HIV disease,reducing opportunistic infections,especially candidiasis.The attenuation of candidal infections in HIV-infected individuals might not solely have resulted from improved immunological status,but also as a result of direct inhibition of C.albicans Saps.In this article,we review updates on the beneficial effects of HIV PIs against the human fungal pathogen C.albicans,focusing on the effects of these compounds on Sap activity,growth behavior,morphological architecture,cellular differentiation,fungal adhesion to animal cells and abiotic materials,modulation of virulence factors,experimental candidiasis infection,and their synergistic actions with classical antifungal agents. 展开更多

目的:从分泌抗人类天冬氨酰基β-羟化酶(HAAH)单克隆抗体(mAb)的杂交瘤细胞G3/F11中,克隆出鼠源an-ti-HAAH mAb重、轻链可变区基因,构建Anti-HAAH的单链抗体(scFv),并进行scFv基因的蛋白表达。方法:提取杂交瘤细胞G3/F11的总RNA,通过RT-... 目的:从分泌抗人类天冬氨酰基β-羟化酶(HAAH)单克隆抗体(mAb)的杂交瘤细胞G3/F11中,克隆出鼠源an-ti-HAAH mAb重、轻链可变区基因,构建Anti-HAAH的单链抗体(scFv),并进行scFv基因的蛋白表达。方法:提取杂交瘤细胞G3/F11的总RNA,通过RT-PCR扩增出VH和VL基因;再利用重叠延伸PCR(SOE-PCR),通过设计在引物上的linker序列,将VH和VL拼接为完整anti-HAAHscFv基因。将测序正确的scFv基因克隆入pHEN1载体,并利用E.coliHB2151进行蛋白表达;通过SDS-PAGE,Western blot分析其表达状况,ELISA鉴定其抗原结合活性。结果:测序结果显示,本实验成功地构建出鼠源anti-HAAHVH-linker-VLscFv基因,且VH、VL均具有完整正确的小鼠抗体骨架区和互补决定区结构,所得的scFv基因片段全长744bp,编码248个氨基酸。SDS-PAGE和Westernblot分析表明,pHEN1-anti-HAAH在E.coliHB2151可表达为Mr约27000的可溶性scFv蛋白,表达量为7.8%。间接ELISA检测显示,可溶性鼠源anti-HAAHscFv蛋白具有较高的抗原结合活性。结论:成功扩增出的鼠源anti-HAAHmAbVH区和VL区基因,并构建为anti-HAAHscFv基因。然后,利用pHEN1载体对anti-HAAHscFv基因进行成功表达,为进一步研究其生物活性及应用奠定了基础。 展开更多

ASPH和CD44V6在非小细胞肺癌中的表达及临床意义 被引量:2

目的探讨人天冬胺酰基β-羟化酶(ASPH)和CD44V6在非小细胞肺癌(NSCLC)中的表达,评估其临床意义。方法收集中国人民解放军联勤保障部队第908医院2015年1月至2019年6月经病理确诊的NSCLC组织及配对的癌旁组织各90例、肺良性疾病组织20例,... 目的探讨人天冬胺酰基β-羟化酶(ASPH)和CD44V6在非小细胞肺癌(NSCLC)中的表达,评估其临床意义。方法收集中国人民解放军联勤保障部队第908医院2015年1月至2019年6月经病理确诊的NSCLC组织及配对的癌旁组织各90例、肺良性疾病组织20例,采用Leica-Bond全自动免疫组化仪检测ASPH和CD44V6的表达,并分析其与临床病理特征的关系。结果ASPH和CD44V6在NSCLC组织的阳性表达率(68.9%、72.2%)均显著高于癌旁(20.0%、11.1%)及肺良性疾病(10%、0%)组织(均P<0.01)。肿瘤大小(>5 cm)、TNM分期(Ⅲ—Ⅳ期)及淋巴结转移(有)的NSCLC组织中ASPH和CD44V6的表达更高(均P<0.05),但在年龄、性别、吸烟史、组织学分级、分化程度及组织学类型上差异无统计学意义(均P>0.05);NSCLC组织中ASPH和CD44V6的表达呈显著正相关(r=0.76,P<0.01)。结论ASPH和CD44V6可能是NSCLC及肺结节良恶性诊断与评估的重要参考指标。 展开更多

QUANTIFICATION OF P4HA2 mRNA OF FIBROBLASTS WITH SYBR GREEN BASED RT-PCR FOR CORRECTING CMV INACTIVATION EFFICIENCY IN DONOR BLOOD

Objective To quantify proline 4-hydroxylase, alpha polypeptide ii ( P4HA2 ) mRNA of human embryo lung fibroblast (HELF) with SYBR green based reversed transcript PCR (RT-PCR) for correcting cytomegalovirus (CMV) inactivation or clearance efficiency in donor blood. Methods A pair of specific primers of exon 12a of P4HA2 was designed, and the related PCR-reaction system and condition were optimized. Then the recombinant plasmid containing the target fragment was constructed for making standard curve with SYBR green based real-time RT-PCR. Finally, the sensitivity, reproducibility, and specificity of this method were fully estimated. Results The sensitivity of the method was 1.5E + 04 copies/mL of P4HA2 mRNA, corresponding to 10^3 fibroblasts. In addition, existence of 8. 67E + 06 leukocytes could not interfere with the accurate quantification of HELF in the large dynamic range. The intra-assay variability and inter-assay variability both varied in different concentrations, being higher in low concentrations and lower in high concentrations. But all of them were below 13. 76% in variation, which showed acceptable stability of this method. Conclusion SYBR green and specific primer based real-time RT-PCR show up a good quality for quantifying HELF P4HA2 mRNA with good specificity, stability, and high sensitivity. Approximate 10 copies of P4HA2 mRNA per cell in average can be detected by the method. Therefore, this method can be used to deduct fibroblast-associated CMV for correcting CMV inactivation efficiency in leukocytes. 展开更多
