And yet this could be the missing l...How managers’ knowledge and beliefs of human nature are formed and manifested has not been fully explored in the context of Chinese society going through rapid transition nowadays. And yet this could be the missing link in our discourse on Chinese managers. Based on a qualitative study conducted in Quanzhou, China, this study found certain assumptions of human nature that are deeply embedded in their managers’ intellectual framework, which in turn guide their managerial behaviours in diverse aspects of their work. Unless the managers are prepared to examine their own thoughts, especially those at the sub-conscious level, they would remain prisoners of their own thought, and all the efforts directed at transforming managers would be seriously compromised.展开更多

METHODS: Epstein Barr virus (EBV) was used to transform the hepatitis ...AIM: To study persistence and replication of hepatitis C virus (HCV) in patients' peripheral blood mononuclear cells (PBMC) cultured in vitro. METHODS: Epstein Barr virus (EBV) was used to transform the hepatitis C virus from a HCV positive patient to permanent lymphoblastoid cell lines (LCL). Positive and negative HCV RNA strands of the cultured cells and growth media were detected by reverse transcriptase-polymerase chain reaction (RT-PCR) each month. Core and NS5 proteins of HCV were further tested using immunohistochemical SP method and in situ RT-PCR. RESULTS: HCV RNA positive strands were consistently detected the cultured cells for one year. The negative-strand RNA in LCL cells and the positive-strand RNA in supernatants were observed intermittently. Immunohistochemical results medicated expression of HCV NS3 and C proteins in LCL cytoplasm mostly. The positive signal of PCR product was dark blue and mainly localized to the LCL cytoplasm. The RT-PCR signal was eliminated by overnight RNase digestion but not DNase digestion. CONCLUSION: HCV may exist and remain functional in a cultured cell line for a long period.展开更多

METHODS: Tolerance towards human...AIM: To determine whether normal genetically immunocompetent rodent hosts could be manipulated to accept human hepatocyte transplants with long term survival without immunosuppression. METHODS: Tolerance towards human hepatocytes was established by injection of primary human hepatocytes or Huh7 human hepatoma cells into the peritoneal cavities of fetal rats. Corresponding cells were subsequently transplanted into newborn rats via intrasplenic injection within 24h after birth. RESULTS: Mixed lymphocyte assays showed that spleen cells from non-tolerized rats were stimulated to proliferate when exposed to human hepatocytes, while cells from tolerized rats were not. Injections made between 15 d and 17 d of gestation produced optimal tolerization. Transplanted human hepatocytes in rat livers were visualized by immunohistochemical staining of human albumin. By dot blotting of genomic DNA in livers of tolerized rats 16 weeks after hepatocyte transplantation, it was found that approximately 2.5 X 10(5) human hepatocytes survived per rat liver. Human albumin mRNA was detected in rat livers by RT-PCR for 15 wk, and human albumin protein was also detectable in rat serum. CONCLUSION: Tolerization of an immuno-competent rat can permit transplantation, and survival of functional human hepatocytes.展开更多

As a result, the human being has created a non-scientifically based cultural environment that is affecting his ability to survive. In effect, cultural evolution is occurring more rapidly than biological evolution that can adapt to the changes brought about in the physical and psycho-social environments. In a pluralistic cultural world, where each society has generated a different view of human nature and different ethical values, the use, misuse, or non-use of scientific and technological advances are derived from these unscientific views of human nature. Since all life depends on limiting interacting environmental and ecological factors, it is imperative that scientific information be used to govern how to minimize irreversible effects on life-sustaining ecological factors, but also scientific information bearing on understanding human nature ought to be integrated into a "global bioethics". METHODS:...AIM: To search for the biomarker of cellular immortalization, the telomere length, telomerase activity and its subunits in cultured epithelial cells of human fetal esophagus in the process of immortalization. METHODS: The transgenic cell line of human fetal esophageal epithelium (SHEE) was established with E(6)E(7) genes of human papillomavirus (HPV) type 18 in our laboratory. Morphological phenotype of cultured SHEE cells from the 6th to 30th passages, was examined by phase contrast microscopy, the telomere length was assayed by Southern blot method, and the activity of telomerase was analyzed by telomeric repeat amplification protocol (TRAP). Expressions of subunits of telomerase, hTR and hTERT, were assessed by RT-PCR. DNA content in cell cycle was detected by flow cytometry. The cell apoptosis was examined by electron microscopy (EM) and TUNEL label. RESULTS: SHEE cells from the 6th to 10th passages showed cellular proliferation with a good differentiation. From the 12th to the 16th passages, many senescent and apoptotic cells appeared, and the telomere length sharply shortened from 23kb to 17kb without expression of hTERT and telomerase activity. At the 20th passage, SHEE cells overcame the senescence and apoptosis and restored their proliferative activity with expression of telomerase and hTERT at low levels, but the telomere length shortened continuously to the lowest of 3kb. After the 30th passage cells proliferation was restored by increment of cells at S and G2M phase in the cell cycle and telomerase activity expressed at high levels and with maintenance of telomere length. CONCLUSION: At the early stage of SHEE cells, telomeres are shortened without expression of telomerase and hTERT causing cellular senescence and cell death. From the 20th to the 30th passages, the activation of telomerase and maintenance of telomere length show a progressive process for immortalization of esophageal epithelial cells. The expression of telomerase may constitute a biomarker for detection of immortalization of cells.展开更多
AIM:To research the effect of Y-27632,a selective Rhoassociated coiled-coil kinase(ROCK) inhibitor,on TGF-β1/Smad2,3 signal transduction in ocular Tenon's capsule fi broblasts(OTFs).METHODS:Primary ocular Teno...AIM:To research the effect of Y-27632,a selective Rhoassociated coiled-coil kinase(ROCK) inhibitor,on TGF-β1/Smad2,3 signal transduction in ocular Tenon's capsule fi broblasts(OTFs).METHODS:Primary ocular Tenon's capsule fibroblasts had been cultured in vitro.The effect of Y27632 on proliferation of OTF stimulated by lysophosphatidic acid(LPA) was evaluated by MTT colorimetric assay so as to sift out the proper concentrations range of Y-27632 for the next experiment.Real time-polymerase chain reactor(RT-PCR) was to analyze the changes of Smad2 and Smad3 genes of cells affected by Y-27632,though unaffected by transforming growth factorbeta1(TGF-β1).Proteins of Smad2,Smad3,phosphorylated Smad2(Ser245/250/255),and phosphorylated Smad3(Ser423/425/203) were respectively quantifi ed by Western blot after OTFs were successively incubated by TGF-β1 and Y-27632.Meanwhile,α-smooth muscular actin(α-SMA) protein was also quantified after the small intervening gene fragments of human Smad2 and Smad3 were designed,synthesized,and then transfected to OTFs.RESULTS:Y-27632 signifi cantly inhibited OTFs proliferation stimulated by LPA.Also Y-27632 signifi cantly suppressed the expressions of Smad2 m RNA,Smad2,3 proteins expressions,Smad3 phosphorylation at the carboxylic terminals of Ser423/425/203 which had been radically promoted by TGF-β1.Si RNA-Smad2,3 suppressed α-SMA expressions,but less effectively than Y-27632.CONCLUSION:The inhibition of ROCK signaling may be a potential therapeutic candidate for the treatment of the fi ltration channel fi brosis.展开更多

Because there are multiple frequency components overlapping seriously in the radar echoes from walking humans, it is a very difficult work to recognize walking humans based on radar echoes. In this paper, a recognition method of walking humans based on radar m-D signatures is proposed. In this method, the m-D spectrum is generated by generalized S transform first,and then the entropy segmentation is used to segment the interesting region from the original spectrum. Next,the m-D features are extracted from the m-D region. Lastly, the support vector machine is used to recognize different walking human targets. The simulation experiments considering two factors of height and velocity are also conducted to test the performance of this proposed method.展开更多

However, NDD...Delay differential equations (DDEs), as well as neutral delay differential equations (NDDEs), are often used as a fundamental tool to model problems arising from various areas of sciences and engineering. However, NDDEs particularly the systems of these equations are special transcendental in nature;it has therefore, become a challenging task or times almost impossible to obtain a convergent approximate analytical solution of such equation. Therefore, this study introduced an analytical method to obtain solution of linear and nonlinear systems of NDDEs. The proposed technique is a combination of Homotopy analysis method (HAM) and natural transform method, and the He’s polynomial is modified to compute the series of nonlinear terms. The presented technique gives solution in a series form which converges to the exact solution or approximate solution. The convergence analysis and the maximum estimated error of the approach are also given. Some illustrative examples are given, and comparison for the accuracy of the results obtained is made with the existing ones as well as the exact solutions. The results reveal the reliability and efficiency of the method in solving systems of NDDEs and can also be used in various types of linear and nonlinear problems.展开更多

Historically, the political development of the world has followed two routes: one involves inter-personal relationships, which have evolved gradually through violent and non-violent forms of revolution from one marked by the conflict between the oppressed and the oppressors and between the exploited and the exploiters, to one marked by equality and mutual respect; and the other involves the progress of the forms of production organization and society made possible by the productive forces driving the production relations forward and the change to the economic foundation that forced the superstructure to experience fundamental transformation. The latter is fundamental and recessive, while the former is dominant, which has a direct impact on states and the broader world. In such a progressive evolution, the political transformation of the world has gradually changed from a violent one to a more peaceful one, and the grander trend has changed from having the features of national independence and liberation as well as a people's revolution, to having the features of seeking development in an all-around way and forging a harmonious world free from threat, fear and anxiety.Hereinto, the dominant role of rational human self-restraint is becoming more and more obvious, with which the actors, even those who onceopposed each other, are seeking mutual tolerance, acceptance and cooperation, including human respect for nature and the environment. Experimental research has recently shown that acupuncture induces the formation of opioid-like peptides (OLPs) in animals. In order to provide further evidence, we tested the beta-endorphin levels and other parameters...Experimental research has recently shown that acupuncture induces the formation of opioid-like peptides (OLPs) in animals. In order to provide further evidence, we tested the beta-endorphin levels and other parameters (VIP, lymphocyte subsets, NK cells and monocyte phagocytosis) in a group of 90 patients suffering from various painful disorders treated with acupuncture. Zusanli (St 36) and Hegu (LI 4) acupoints were selected. A homogeneous group of 30 subjects was used as control. Evaluation of the above parameters was made with 3 series of blood tests before treatment, 30 minutes and 24 hours after acupuncture treatment. In the acupuncture group, the following results were achieved: 1) A considerable increase in beta-endorphin levels remained high even 24 hours after acupuncture treatment. In addition, we demonstrated an inverted correlation between beta-endorphins and VIP; 2) 30 minutes after acupuncture session, 80% of the treated patients showed a significant increase of CD3 and CD4 values and an increase of CD8 24 hours after stimulation; 3) Monocyte phagocytosis was increased in 45% of the treated subjects 30 minutes from starting treatment, and in 100% of them after 24 hours. The percentage of NK cells was also increased in 40% of cases after 30 minutes, and in 50% after 24 hours. However, in the control group, no such significant changes in immune parameters were found.展开更多

文摘AIM:To research the effect of Y-27632,a selective Rhoassociated coiled-coil kinase(ROCK) inhibitor,on TGF-β1/Smad2,3 signal transduction in ocular Tenon's capsule fi broblasts(OTFs).METHODS:Primary ocular Tenon's capsule fibroblasts had been cultured in vitro.The effect of Y27632 on proliferation of OTF stimulated by lysophosphatidic acid(LPA) was evaluated by MTT colorimetric assay so as to sift out the proper concentrations range of Y-27632 for the next experiment.Real time-polymerase chain reactor(RT-PCR) was to analyze the changes of Smad2 and Smad3 genes of cells affected by Y-27632,though unaffected by transforming growth factorbeta1(TGF-β1).Proteins of Smad2,Smad3,phosphorylated Smad2(Ser245/250/255),and phosphorylated Smad3(Ser423/425/203) were respectively quantifi ed by Western blot after OTFs were successively incubated by TGF-β1 and Y-27632.Meanwhile,α-smooth muscular actin(α-SMA) protein was also quantified after the small intervening gene fragments of human Smad2 and Smad3 were designed,synthesized,and then transfected to OTFs.RESULTS:Y-27632 signifi cantly inhibited OTFs proliferation stimulated by LPA.Also Y-27632 signifi cantly suppressed the expressions of Smad2 m RNA,Smad2,3 proteins expressions,Smad3 phosphorylation at the carboxylic terminals of Ser423/425/203 which had been radically promoted by TGF-β1.Si RNA-Smad2,3 suppressed α-SMA expressions,but less effectively than Y-27632.CONCLUSION:The inhibition of ROCK signaling may be a potential therapeutic candidate for the treatment of the fi ltration channel fi brosis.

文摘The recognition of human movements based on radar m-D(micro-Doppler) signatures attracts great interest in the field of radar research on automatic target recognition. Because there are multiple frequency components overlapping seriously in the radar echoes from walking humans, it is a very difficult work to recognize walking humans based on radar echoes. In this paper, a recognition method of walking humans based on radar m-D signatures is proposed. In this method, the m-D spectrum is generated by generalized S transform first,and then the entropy segmentation is used to segment the interesting region from the original spectrum. Next,the m-D features are extracted from the m-D region. Lastly, the support vector machine is used to recognize different walking human targets. The simulation experiments considering two factors of height and velocity are also conducted to test the performance of this proposed method.
文摘Delay differential equations (DDEs), as well as neutral delay differential equations (NDDEs), are often used as a fundamental tool to model problems arising from various areas of sciences and engineering. However, NDDEs particularly the systems of these equations are special transcendental in nature;it has therefore, become a challenging task or times almost impossible to obtain a convergent approximate analytical solution of such equation. Therefore, this study introduced an analytical method to obtain solution of linear and nonlinear systems of NDDEs. The proposed technique is a combination of Homotopy analysis method (HAM) and natural transform method, and the He’s polynomial is modified to compute the series of nonlinear terms. The presented technique gives solution in a series form which converges to the exact solution or approximate solution. The convergence analysis and the maximum estimated error of the approach are also given. Some illustrative examples are given, and comparison for the accuracy of the results obtained is made with the existing ones as well as the exact solutions. The results reveal the reliability and efficiency of the method in solving systems of NDDEs and can also be used in various types of linear and nonlinear problems.
文摘In the contemporary world, various political actors, both within and outside of states, have continued to break through subjective and objective constraints to participate more and more widely in the political activities of the world society, which has promoted inclusiveness in the relationship between all actors and fostered reconciliation between man and nature. Historically, the political development of the world has followed two routes: one involves inter-personal relationships, which have evolved gradually through violent and non-violent forms of revolution from one marked by the conflict between the oppressed and the oppressors and between the exploited and the exploiters, to one marked by equality and mutual respect; and the other involves the progress of the forms of production organization and society made possible by the productive forces driving the production relations forward and the change to the economic foundation that forced the superstructure to experience fundamental transformation. The latter is fundamental and recessive, while the former is dominant, which has a direct impact on states and the broader world. In such a progressive evolution, the political transformation of the world has gradually changed from a violent one to a more peaceful one, and the grander trend has changed from having the features of national independence and liberation as well as a people's revolution, to having the features of seeking development in an all-around way and forging a harmonious world free from threat, fear and anxiety.Hereinto, the dominant role of rational human self-restraint is becoming more and more obvious, with which the actors, even those who onceopposed each other, are seeking mutual tolerance, acceptance and cooperation, including human respect for nature and the environment. In essence, this is indicative of the emancipation of human sociality and the reconciliation between man and nature, which is still currently under way.

文摘Abstract:The development of China’s human rights path is driv-en by both external and internal factors,influenced by general prin-ciples of human rights worldwide while also following China’s own endogenous logic.The concept of the“Two Integrations”reflects the continuous theoretical innovation of the Chinese Communist Party based on China’s national conditions,significantly illustrating the en-dogenous logic of China’s human rights path.Among these,the“basic principles of Marxism”occupy a“core”position within the endoge-nous logic of China’s human rights path,and to some extent,embody an understanding of the inherent laws of human rights.Meanwhile,“China’s specific realities”and“fine traditional Chinese culture”serve as the“living water sources”for the ongoing advancement of China’s human rights.The“Two Integrations”represent an intrinsic requirement of historical materialism and practical materialism.The logical structure of“One Core,Two Sources”explains the endogenous nature of China’s human rights path,offering both an interpretation of the driving forces behind China’s human rights path and a defense of its rationality.

文摘Experimental research has recently shown that acupuncture induces the formation of opioid-like peptides (OLPs) in animals. In order to provide further evidence, we tested the beta-endorphin levels and other parameters (VIP, lymphocyte subsets, NK cells and monocyte phagocytosis) in a group of 90 patients suffering from various painful disorders treated with acupuncture. Zusanli (St 36) and Hegu (LI 4) acupoints were selected. A homogeneous group of 30 subjects was used as control. Evaluation of the above parameters was made with 3 series of blood tests before treatment, 30 minutes and 24 hours after acupuncture treatment. In the acupuncture group, the following results were achieved: 1) A considerable increase in beta-endorphin levels remained high even 24 hours after acupuncture treatment. In addition, we demonstrated an inverted correlation between beta-endorphins and VIP; 2) 30 minutes after acupuncture session, 80% of the treated patients showed a significant increase of CD3 and CD4 values and an increase of CD8 24 hours after stimulation; 3) Monocyte phagocytosis was increased in 45% of the treated subjects 30 minutes from starting treatment, and in 100% of them after 24 hours. The percentage of NK cells was also increased in 40% of cases after 30 minutes, and in 50% after 24 hours. However, in the control group, no such significant changes in immune parameters were found.
