METHODS: Cell culture, MTT, Electromicroscopy, agarose gel electrophoresis, lucigenin, Western blot and electrophoretic mobility shift assay (EMSA) analysis were employed to investigate the effect of JTE-522 on cell proliferation and apoptosis in AGS cells and related molecular mechanisms. RESULTS: JTE-522 inhibited the growth of AGS cells and induced the apoptosis. Lucigenin assay showed the generation of ROS in cells under incubation with JTE-522. The increased ROS generation might contribute to the induction of AGS cells to apoptosis. EMSA and Western blot revealed that NF-kB activity was almost completely inhibited by preventing the degradation of IkBalpha. Additionally, by using Western blot we confirmed that the level of bcl-2 was decreased, whereas p53 showed a great increase following JTE-522 treatment. Their changes were in a dose-dependent manner. CONCLUSION: These findings suggest that reactive oxygen species, NF-kB, p53, bcl-2 and caspase-3 may play an important role in the induction of apoptosis in AGS cells after treatment with JTE-522.展开更多
How managers’ knowledge and beliefs of human nature are formed and manifested has not been fully explored in the context of Chinese society going through rapid transition nowadays. And yet this could be the missing l...How managers’ knowledge and beliefs of human nature are formed and manifested has not been fully explored in the context of Chinese society going through rapid transition nowadays. And yet this could be the missing link in our discourse on Chinese managers. Based on a qualitative study conducted in Quanzhou, China, this study found certain assumptions of human nature that are deeply embedded in their managers’ intellectual framework, which in turn guide their managerial behaviours in diverse aspects of their work. Unless the managers are prepared to examine their own thoughts, especially those at the sub-conscious level, they would remain prisoners of their own thought, and all the efforts directed at transforming managers would be seriously compromised.展开更多
Natural systems have been“restoring”disturbed sites(landslides,volcanic eruptions,shoreline erosion,etc.)for millions of years.By understanding how these natural systems operate they can be applied to sites humans d...Natural systems have been“restoring”disturbed sites(landslides,volcanic eruptions,shoreline erosion,etc.)for millions of years.By understanding how these natural systems operate they can be applied to sites humans disturb(mines,industrial developments,etc.).Natural systems initiate recovery using pioneering species such as willows(Salix spp.),balsam poplar(Populus balsamifera L.)or alder(Alnus spp.).The seeds of these species are designed to travel long distances and use commonly occurring conditions to get established.Balsam poplar and willows have light fluffy seeds that at some times of the year look like snow.They land on puddles or other waterbodies and are blown to the wet mud at the edges of the puddle or on the shore of the waterbody where they germinate and grow.By creating these conditions(making puddles)on a mine site these species can be encouraged to establish on sites that are being reclaimed.The cost of these treatments is a fraction of traditional reclamation costs and because the resulting vegetation is appropriate to the area and the site where it establishes,natural processes can provide effective strategies for the reclamation of mining disturbances.Examples are drawn from the author’s experience.展开更多

As a result, the human being has created a non-scientifically based cultural environment that is affecting his ability to survive. In effect, cultural evolution is occurring more rapidly than biological evolution that can adapt to the changes brought about in the physical and psycho-social environments. In a pluralistic cultural world, where each society has generated a different view of human nature and different ethical values, the use, misuse, or non-use of scientific and technological advances are derived from these unscientific views of human nature. Since all life depends on limiting interacting environmental and ecological factors, it is imperative that scientific information be used to govern how to minimize irreversible effects on life-sustaining ecological factors, but also scientific information bearing on understanding human nature ought to be integrated into a ＂global bioethics＂. While ethical values cannot be directly derived from scientific factors, it is also true that human values or our ＂ought＂ cannot be maintained in ignorance or defiance of the facts or the ＂is＂.展开更多

Experimental research has recently shown that acupuncture induces the formation of opioid-like peptides (OLPs) in animals. In order to provide further evidence, we tested the beta-endorphin levels and other parameters...Experimental research has recently shown that acupuncture induces the formation of opioid-like peptides (OLPs) in animals. In order to provide further evidence, we tested the beta-endorphin levels and other parameters (VIP, lymphocyte subsets, NK cells and monocyte phagocytosis) in a group of 90 patients suffering from various painful disorders treated with acupuncture. Zusanli (St 36) and Hegu (LI 4) acupoints were selected. A homogeneous group of 30 subjects was used as control. Evaluation of the above parameters was made with 3 series of blood tests before treatment, 30 minutes and 24 hours after acupuncture treatment. In the acupuncture group, the following results were achieved: 1) A considerable increase in beta-endorphin levels remained high even 24 hours after acupuncture treatment. In addition, we demonstrated an inverted correlation between beta-endorphins and VIP; 2) 30 minutes after acupuncture session, 80% of the treated patients showed a significant increase of CD3 and CD4 values and an increase of CD8 24 hours after stimulation; 3) Monocyte phagocytosis was increased in 45% of the treated subjects 30 minutes from starting treatment, and in 100% of them after 24 hours. The percentage of NK cells was also increased in 40% of cases after 30 minutes, and in 50% after 24 hours. However, in the control group, no such significant changes in immune parameters were found.展开更多

METHODS: Low concentration...OBJECTIVE: To explore the association of inherent cellular reactive oxygen species (ROS) levels with susceptibility of the tumor cells to apoptosis induction by arsenic trioxide (As(2)O(3)). METHODS: Low concentration (2 micromol/L) of As(2)O(3) was administered to two cultured leukemic cell lines, NB4 and U937, and two esophageal carcinoma cell lines, EC1.71 (also named EC/CUHK1) and EC1867, to confirm the difference in apoptosis susceptibility of NB4 versus U937 and of EC1.71 versus EC1867. Dihydrogenrhodamine 123 (DHR123), used as a ROS capture agent, was incubated with cells in the absence of As(2)O(3). Fluorescence intensity of rhodamine 123, the product of cellular oxidation of DHR123, was detected by flow cytometry and ROS was measured. RESULTS: Low concentration of As(2)O(3) induced apoptosis was more likely to occur in NB4 and EC1.71 cells than in U937 and EC1867 cells, or NB4 was more sensitive than U937, and EC1.71 more sensitive than EC1867 to As(2)O(3). The inherent cellular ROS level is higher in NB4 than in U937, and also higher in EC1.71 than in EC1867. CONCLUSIONS: The difference in cellular ROS level is positively associated with cellular susceptibility to apoptosis induction by As(2)O(3).
