This study aims to investigate grain quality and nutritional values of rice(Pokkali,a salt-tolerant cultivar;RD73,a new cultivar improved from KDML105 introgressed with Saltol QTL from Pokkali,and KDML105,a moderately...This study aims to investigate grain quality and nutritional values of rice(Pokkali,a salt-tolerant cultivar;RD73,a new cultivar improved from KDML105 introgressed with Saltol QTL from Pokkali,and KDML105,a moderately salt-susceptible cultivar)grown under non-saline(0.04–0.87 dS/m)and slightly saline(1.08–4.83 dS/m)field conditions.The results revealed that salinity caused significant reduction in grain size but significant increments in reducing sugar and total protein contents in the grains.Nevertheless,the amounts of starch in the grains of KDML105 and Pokkali rice genotypes were unaffected by the stress.The starch granule size distribution was also unaffected by salinity.Interestingly,only starch from Pokkali was significantly diminished in amylose content,from 19.18%to 16.99%.Accordingly,parameters relating to starch gelatinization,retrogradation,and pasting properties of KDML105 and RD73 were unaffected by salinity;only Pokkali showed a significant increase in percentage of retrogradation along with a significant reduction in gelatinization enthalpy.In the saline field,total phenolic content and antioxidant capacity in the grains of all rice cultivars tended to increase,particularly in Pokkali.On average,essential element contents in grains from the saline-treated plants showed a 33%,32%,32%,22%,20%,11%,and 10%increase in total P,N,K,Mg,Zn,Fe,and Ca content,respectively.Interestingly,total Fe content exhibited the greatest percentage of increments in KDML105(187%).Taken together,cultivation of rice in the slightly saline field did not alter its eating and cooking qualities,while enhanced some nutritional properties such as proteins,minerals,and secondary metabolites like phenolic compounds.展开更多

Here, we studied the activities of enzymes related to starch metabolism in rice stems and grains, and the microstructures related to carbohydrate accumulation and transportation to investigate the effects of different water regimes on grain filling. Two ‘super’ rice cultivars were grown under two irrigation regimes of well-watered(WW) and alternate wetting and moderate soil drying(AWMD). Compared with the WW treatment,the activities of ADP glucose pyrophosphorylase(AGPase), starch synthase(StSase) and starch branching enzyme(SBE), and the accumulation of non-structural carbohydrates(NSCs) in the stems before heading were significantly improved, and more starch granules were stored in the stems in the AWMD treatment. After heading, the activities of α-amylase, β-amylase, sucrose phosphate synthase(SPS) and sucrose synthase in the synthetic direction(SSs)were increased in the stems to promote the remobilization of NSCs for grain filling under AWMD. During grain filling, the enzymatic activities of sucrose synthase in the cleavage direction(SSc), AGPase, StSase and SBE in the inferior spikelets were increased, which promoted grain filling, especially for the inferior spikelets under AWMD.However, there were no significant differences in vascular microstructures. The grain yield and grain weight could be improved by 13.1 and 7.5%, respectively, by optimizing of the irrigation regime. We concluded that the low activities of key enzymes in carbon metabolism is the key limitation for the poor grain filling, as opposed to the vascular microstructures, and AWMD can increase the amount of NSC accumulation in the stems before heading, improve the utilization rate of NSCs after heading, and increase the grain filling, especially in the inferior spikelets, by altering the activities of key enzymes in carbon metabolism.展开更多

To understand the difference in cooking and nutrition qualities among the grains at different positions within a rice panicle, the distributions of gel consistency (GC), amylose content (AC) and crude protein content (CPC) of grains in a panicle in association with different nitrogen levels (0, 120 and 240 k c/ha) were investigated by using two rice varieties, Yangdao 6 (indica) and Wuyujing 3 (japonica). In general, the grains at the basal part of a panicle had lower GC and higher AC than those at the upper or middle part of a panicle. The 1^st grain on the secondary branch with earlier flowering exhibited the highest GC, whereas the 2nd grain on the primary branch with later flowering showed the highest AC. For Yangdao 6, CPC in the grains on the primary branches was lower at the middle part of a panicle than at the upper or basal part of a panicle. For Wuyujing 3, there were no significant differences in CPCs in the grains among the upper, middle and basal parts of a panicle. GC in the grains was increased, whereas AC was reduced from zero nitrogen application (ON) to low amount of nitrogen application (LN), and the result was reversed from LN to medium amount of nitrogen application (MN). The results showed that changes in activities of ADPGPase, SSase and Q-enzyme exhibited a single peak duringgrain filling, and the time of the activity peaks for the former two enzymes was earlier than that of the maximum grain-fillingrate (Tmax), and the time reaching the peak for Q-enzyme was synchronous with Tmax. The activities at early grain fillingstage, and the mean and maximum activities of each enzyme during grain filling period were positively and significantly orvery significantly correlated with the mean and maximum grain filling rate and starch content (mg grain-1) in the grains.Activities of ADPGPase at all grain filling stages and those of Q-enzyme at the early and mid filling stages were notsignificantly correlated the cooking quality (GC, ASV and AC). SSase activities at the early filling stage were significantlyand negatively correlated with GC and ASV, and positively correlated with AC. Activities of SSase at mid and late grainfilling stages and Q-enzyme at the late filling stage were significantly and positively correlated with GC and ASV, andnegatively correlated with AC. Mottaikaruppan) and to improve germination rate using gibberellic acid and surfactants [sodium dodecyl sulfate (SDS) (1.0 g/L) and Triton-X-100 (1.0 mL/L)], whole rice grains soaked in distilled water for 12 h at 30℃were germinated in the dark at 30℃ for five days. The highest germination rate (77.1%) was obtained on the 5th day. An increase in the content of reducing sugars from 7.3 to 58.1 mg/g DM (dry matter) was observed from the 1st day of germination. Free amino acids and soluble protein contents increased to 3.69 and 5.29 mg/g DM, respectively on the 5^th day of germination. Total protein content decreased from 100.5 to 91.0 g/kg DM during germination. Increases in amylolytic (1.1 to 190.0 U/g DM) and proteolytic (0 to 0.12 U/g DM) activities were observed during germination. Effects of different concentrations of gibberellic acid on the germination of rice grains were evaluated and 0.1 g/L was found to promote germination. When effects of gibberellic acid (0.1 g/L) and surfactants were evaluated individually and together, higher germination rate was observed in the control experiment (grains germinated in distilled water), whereas giberellic acid and surfactants decreased the germination rate. Therefore, the flour obtained from the grains germinated for four days using distilled water to obtain high content of soluble materials and enzyme activities can be used in preparation of bakery items.展开更多

In this study, a double haploid(DH) population derived from Zhongjiazao 17(YK17)(an early-season indica cultivar)×D50(a tropical japonica cultivar) was used to identify quantitative trait loci(QTLs) associated with Cd concentration in brown rice(CCBR) and Cd concentration in milled rice(CCMR). Continuous and wide variation for CCBR and CCMR were observed among the DH population. Correlation analysis revealed a positive and highly significant correlation between the two traits. A total of 18 QTLs for CCBR and 14 QTLs for CCMR were identified in five different pot and field trials. Two pairs of QTLs for CCBR(qCCBR2-1 and qCCBR2-2, qCCBR9-1 and qCCBR9-2) and one pair of QTLs for CCMR(qCCMR5-1 and qCCMR5-2) were detected in multiple trials. The alleles increasing CCBR at the qC CBR2-1/qC CBR2-2 and qC CBR9-1/qC CBR9-2 QTLs were contributed by YK17 and D50, respectively, whereas the D50 allele at the qCCMR5-1/qCCMR5-2 QTLs increased CCMR. Eight pairs of QTLs for CCBR and CCMR, qCCBR2-2 and qCCMR2-2, qCCBR3 and qCCMR3, qCCBR4-2 and qCCMR4-1, qCCBR4-3 and qCCMR4-2, qCCBR4-4 and qCCMR4-3, qCCBR5 and qCCMR5-2, qCCBR7 and qCCMR7, and qCCBR11-1 and qCCMR11-2, co-localized on chromosomes 2, 3, 4, 5, 7, and 11, respectively. For all of these QTL pairs, except qCCBR5/qCCMR5-2, the additive effects came from YK17. In addition, four CCMR QTLs showing significant additive×environment interaction and two pairs of CCMR QTLs with bi-allelic epistatic interactions were identified. The results of this study could facilitate marker-assisted selection of breeding rice varieties with low Cd accumulation in grain.展开更多

Significant genotypic variation was detected in the five heavy metal concentrations in grains, indicating the possibility to reduce the concentration of these heavy metals in grains through breeding approach. The environmental effect varied with metal, with Pb and Ni having greater variation than the other three metals. There was significant genotype-environment (location) interaction of the concentrations of all five heavy metals in grains, suggesting the importance of cultivar choice in producing rice with low heavy metal concentrations in grains for a given location. Correlation analysis showed that Cd and As, Cr and Ni, and As and Pb con-centrations in rice grains were closely associated, and that Ni concentration in grains was negatively correlated with Zn concen-tration.展开更多

A kind of nematode of Aphelenchoides was isolated from the grains of rice variety Wuyujing 3 which performed 'small grains and erect panicles' symptom. There were 2014 nematodes in one hundred grains infected, and up to 74 in single grain, 92 percent of the grains tested had nematodes in the infested panicles. The diagnosis characters of nematode include lateral fields about one-fourth as wide as body, with 4 incisures. The terminus bears a mucro of diverse shape with 3-4 pointed processes. The female post-vulval uterine sac extends less than 50% of distance from vulva to anus, no sperm in it. Oocytes usually arrange in 2-4 rows. The male spicules have a moderately developed rostrum. Morphological measurements showed it to be conspecific with Aphelenchoides besseyi Christie, 1942.展开更多

The activities of AGPP, SSS and SBE gradually increased and then declined as a single-peak curve with the process of grain filling in the progenies with high and low amylose contents in grains. The progenies with high amylose content peaked earlier in the AGPP, SSS and SBE activities and had higher AGPP, SSS and SBE activities at the early grain filling stage than those with low amylose content. The GS activity peaked earlier and was higher at the late stage of grain filling in the progenies with high protein content than in those with low protein content. It is suggested that the activities of key enzymes for starch synthesis and glutamine synthetase could be changed in oriented breeding for amylose and protein contents in grains.展开更多

Four cultivars with white grains(hereafter, white rice) and five cultivars with dark colors (hereafter dark rice) were selected for this experiment. Three levels of soil Cd concentrations, background(0), 5 and 10 mg/kg, were used. After harvest, plant biomass, tissue concentrations of Cd, Ca, Fe, Cu and Zn were analyzed. The results showed that Cd concentrations are significantly different between different genotypes, but when comparing the Cd concentrations for the two groups, no significant difference was found. For other divalent cations, Ca concentrations in dark rice were higher than those in white ones(P<0.001 for shoots, P=0.037 for roots); Fe concentrations in dark rice were also higher than those in white ones(P=0.001 either in shoot or root); Zn concentrations in shoot of dark rice were higher than those in white ones, but no significant difference in roots. The total molar concentrations of divalent cations in dark rice were also significantly higher than in white rice. The potential benefit of higher Ca and Fe concentrations in dark rice and similar Cd concentrations in both groups is also discussed in this paper.展开更多

With rising living standards,there is an increasing demand for high-quality rice.Rice quality is mainly defined by milling quality,appearance quality,cooking and eating quality,and nutrition quality.Among them,chalkin...With rising living standards,there is an increasing demand for high-quality rice.Rice quality is mainly defined by milling quality,appearance quality,cooking and eating quality,and nutrition quality.Among them,chalkiness is a key trait for appearance quality,which adversely affects cooking and eating quality,head rice yield,and commercial value.Therefore,chalkiness is undesirable,and reducing chalkiness is a major goal in rice quality improvement.However,chalkiness is a complex trait jointly influenced by genetic and environmental factors,making its genetic study and precision improvement a huge challenge.With the rapid development of molecular techniques,much knowledge has been gained about the genes and molecular networks involved in chalkiness formation.The present review describes the major environmental factors affecting chalkiness and summarizes the quantitative trait loci(QTL)associated with chalkiness.More than 150 genes related to chalkiness formation have been reported.The functions of the genes regulating chalkiness,primarily those involved in starch synthesis,storage protein synthesis,transcription regulation,organelle development,grain shape regulation,and hightemperature response,are described.Finally,we identify the challenges associated with genetic improvement of chalkiness and suggest potential strategies.Thus,the review offers insight into the molecular dynamics of chalkiness and provides a strong basis for the future breeding of high-quality rice varieties.展开更多

Increasing effective panicle number per plant(EPN)is one approach to increase yield potential in rice.However,molecular mechanisms underlying EPN remain unclear.In this study,we integrated mapbased cloning and genome-...Increasing effective panicle number per plant(EPN)is one approach to increase yield potential in rice.However,molecular mechanisms underlying EPN remain unclear.In this study,we integrated mapbased cloning and genome-wide association analysis to identify the EPN4 gene,which is allelic to NARROW LEAF1(NAL1).Overexpression lines containing the Teqing allele(TQ)of EPN4 had significantly increased EPN.NIL-EPN4^(TQ) in japonica(geng)cultivar Lemont(LT)exhibited significantly improved EPN but decreased grain number and flag leaf size relative to LT.Haplotype analysis indicated that accessions with EPN4-1 had medium EPN,medium grain number,and medium grain weight,but had the highest grain yield among seven haplotypes,indicating that EPN4-1 is an elite haplotype of EPN4 for positive coordination of the three components of grain yield.Furthermore,accessions carrying the combination of EPN4-1 and haplotype GNP1-6 of GNP1 for grain number per panicle showed higher grain yield than those with other allele combinations.Therefore,pyramiding of EPN4-1 and GNP1-6 could be a preferred approach to obtain high yield potential in breeding.展开更多
Lysine content is a criterion of the nutritional quality of rice.Understanding the process of lysine biosynthesis in early-flowering superior grain(SG)and late-flowering inferior grain(IG)of rice would advance breedin...Lysine content is a criterion of the nutritional quality of rice.Understanding the process of lysine biosynthesis in early-flowering superior grain(SG)and late-flowering inferior grain(IG)of rice would advance breeding and cultivation to improve nutritional quality.However,little information is available on differences in lysine anabolism between SG and IG and the underlying mechanism,and whether and how irrigation regimes affect lysine anabolism in these grains.A japonica rice cultivar was grown in the field and two irrigation regimes,continuous flooding(CF)and wetting alternating with partial drying(WAPD),were imposed from heading to the mature stage.Lysine content and activities of key enzymes of lysine biosynthesis,and levels of brassinosteroids(BRs)were lower in the IG than in the SG at the early grainfilling stage but higher at middle and late grain-filling stages.WAPD increased activities of these key enzymes,BR levels,and contents of lysine and total amino acids in IG,but not SG relative to CF.Application of 2,4-epibrassinolide to rice panicles in CF during early grain filling reproduced the effects of WAPD,but neither treatment altered the activities of enzymes responsible for lysine catabolism in either SG or IG.WAPD and elevated BR levels during grain filling increased lysine biosynthesis in IG.Improvement in lysine biosynthesis in rice should focus on IG.展开更多
Objective:To determine distribution of arthropods in rice grains obtained from different sources.Methods: Rice samples were randomly collected from public in urban areas,farmers in rice field areas,aborigines in undev...Objective:To determine distribution of arthropods in rice grains obtained from different sources.Methods: Rice samples were randomly collected from public in urban areas,farmers in rice field areas,aborigines in undeveloped areas and retailers in commercial premises.Random samples of rice were taken out from each sample for isolation of arthropods using a modified Berlese Tullgren Funnel Method.Mites were mounted prior to identification; weevils were directly identified.Results:Samples of rice from retailers in commercial premises had the highest infestation by arthropods followed by samples from urbanites,aborigines and rice farmers.Two species of weevils,Sitophilus oryzae(S.oryzae) and Sitophilus granarius(S.granarius),were found.Samples from commercial premises had the least percentage of weevils compared to those collected from domestic premises. Depending on the source of samples,densities of S.granarius and S.oryzae ranges from 11-103 weevils/ kg and 7- 80 weevils/kg,respectively.Important species of mites in stored rice identified were mainly members of the families Cheyletidae,Echimyopodidae,Pyroglyphidae,Saproglyphidae and Tenuipalpidae.Among the species of mites identified were Austroglycyphagus malaysiensis,Cheyletus fortis,Cheyletus malaccensis,Dermatophagoides pteronyssinus,Grammolichus malukuensis and Suidasia pontifica.Average density of most of the mites was less than 40 mites/kg of rice grains.In this study,the highest number of mites in rice samples was recovered from commercial premises,followed by samples from urbanites.Samples from farmers and aborigines contained lesser mites.Conclusion:This study demonstrated the presence of 3 allergenic mite species in rice, i.e A.malaysiensis,D.pteronyssinus and S.pontifica.Weevils,S.oryzae and S.granarius that are known to be allergenic,were also found.展开更多
Applying iodine fertilizers to cultivate iodine-rich crops for daily intake is an effective approach for iodine supplementation,especially for aromatic rice.Field experiments were conducted during the early growing se...Applying iodine fertilizers to cultivate iodine-rich crops for daily intake is an effective approach for iodine supplementation,especially for aromatic rice.Field experiments were conducted during the early growing seasons of 2021 and 2022 to evaluate the impacts of foliar application of iodine fertilizer on aromatic rice and to explore the optimal iodine fertilizer concentration.At the full heading stage,six different concentrations of sodium iodide solutions of 0%(CK),0.010%(T1),0.025%(T2),0.050%(T3),0.075%(T4),and 0.100%(T5)were applied to indica aromatic rice cultivars Meixiangzhan 2 and Xiangyaxiangzhan.The results showed that sodium iodide treatments significantly increased the iodine and sodium contents in both leaves and grains.Compared with the CK,the T1 and T2 treatments increased the 2-acetyl-1-pyrroline(2-AP)content in mature grains by 8.41%-101.66%and 13.58%-74.60%,respectively.Improvements in the contents of 1-pyrroline-5-carboxylic acid,proline,1-pyrroline,and methylglyoxal,as well as the activity of proline dehydrogenase were also detected.Additionally,sodium iodide treatments remarkably decreased the chalky grain rate,chalkiness area,and chalkiness degree of aromatic rice,with the T2 treatment exhibiting a 17.79%-47.42%decrease in chalkiness degree compared with the CK.Meanwhile,T1 and T2 treatments showed beneficial impacts on chlorophyll content,photosynthetic characteristics,and yield components,while T3,T4,and T5 treatments exhibited adverse effects on leaf and grain yields.The linear discriminant analysis revealed significant differences between treatments.The correlation analysis and piecewise structural equation modeling showed that the iodine and sodium influenced the photosynthetic characteristics and chlorophyll content of the leaves,thereby regulating the 2-AP biosynthesis and yield components,ultimately affecting the 2-AP content and yield.Overall,this study suggests that foliar application of 0.025%sodium iodide is an effective method to enrich the iodine content in rice grains,improve the grain aroma and appearance quality of aromatic rice,without detrimental effects on grain yield.展开更多
Twenty-four rice genotypes were examined to assess genetic variability,heritability,and correlations for seven-grain quality traits,eight nutritional elements,and protein.ANOVA revealed significant differences for the ...Twenty-four rice genotypes were examined to assess genetic variability,heritability,and correlations for seven-grain quality traits,eight nutritional elements,and protein.ANOVA revealed significant differences for the quality traits studied.For every trait under study,the phenotypic coefficient of variation was higher than the correspon-dence genotypic coefficient of variation.Heritability in a broad sense varied from 29.75%for grain length to 98.31%for the elongation trait.Hulling percentage recovery had a highly significant positive correlation with milling and head rice percentage.Consequently,milling percentage had a highly positive correlation with head rice percentage.In amylose percentage,all the genotypes belonged to low amylose except the Hassawi-1 variety,which had intermediate amylose content.Mineral nutrition contents of magnesium(Mg),sodium(Na),potas-sium(K),calcium(Ca),copper(Cu),manganese(Mn),zinc(Zn),iron(Fe),or protein percentage gave different variations for 24 rice genotypes under all the nutritional elements.Among the 24 genotypes,ten rice genotypes–HighNutrient-1,HighNutrient-2,HighNutrient-9,HighNutrient-8,HighNutrient-3,Hassawi-2,HighNutrient-7,HighNutrient-6,Hassawi-1,and HighNutrient-4–had the highest heist value for all nutritional and protein con-tents,and could be used as a donor to improving new varieties.There was a positive and significant correlation between magnesium Mg,K,Zn and Fe.Consequently,K had a positive correlation with zinc Zn,Fe,and protein percentage.Clustering analysis was divided into two groups:thefirst group included all genotypes rich in nutri-ents,while the remaining genotypes with low nutritional content were included in the second group.展开更多

Rice(Oryza sativa L.) DNA extraction methods often use seedlings or leaves rather than the grains and tend to be time-consuming, involve multiple steps, and use hazardous chemicals and expensive enzymes. Rice grains offer several benefits over seedlings and leaves as a source of DNA for genetic analysis. However, these benefits are underutilized because the bulk of a rice grain is made up of starch. It is particularly important, but difficult to get rid of the starch while extracting DNA from rice grains. This co-precipitated polysaccharide is a known inhibitor of DNA polymerase activity in polymerase chain reaction(PCR). We describe here a very simple and highly affordable Chelex~?-100 based DNA extraction method from rice grains. It does not require any hazardous chemicals or enzymes. This method reproducibly extracts DNA with good purity indices(A_(260)/A_(230) and A_(260)/A_(280) values), but requires only a few steps.展开更多

文摘This study aims to investigate grain quality and nutritional values of rice(Pokkali,a salt-tolerant cultivar;RD73,a new cultivar improved from KDML105 introgressed with Saltol QTL from Pokkali,and KDML105,a moderately salt-susceptible cultivar)grown under non-saline(0.04–0.87 dS/m)and slightly saline(1.08–4.83 dS/m)field conditions.The results revealed that salinity caused significant reduction in grain size but significant increments in reducing sugar and total protein contents in the grains.Nevertheless,the amounts of starch in the grains of KDML105 and Pokkali rice genotypes were unaffected by the stress.The starch granule size distribution was also unaffected by salinity.Interestingly,only starch from Pokkali was significantly diminished in amylose content,from 19.18%to 16.99%.Accordingly,parameters relating to starch gelatinization,retrogradation,and pasting properties of KDML105 and RD73 were unaffected by salinity;only Pokkali showed a significant increase in percentage of retrogradation along with a significant reduction in gelatinization enthalpy.In the saline field,total phenolic content and antioxidant capacity in the grains of all rice cultivars tended to increase,particularly in Pokkali.On average,essential element contents in grains from the saline-treated plants showed a 33%,32%,32%,22%,20%,11%,and 10%increase in total P,N,K,Mg,Zn,Fe,and Ca content,respectively.Interestingly,total Fe content exhibited the greatest percentage of increments in KDML105(187%).Taken together,cultivation of rice in the slightly saline field did not alter its eating and cooking qualities,while enhanced some nutritional properties such as proteins,minerals,and secondary metabolites like phenolic compounds.

文摘Recently developed ‘super’ rice cultivars with greater yield potentials often suffer from the problem of poor grain filling, especially in inferior spikelets. Here, we studied the activities of enzymes related to starch metabolism in rice stems and grains, and the microstructures related to carbohydrate accumulation and transportation to investigate the effects of different water regimes on grain filling. Two ‘super’ rice cultivars were grown under two irrigation regimes of well-watered(WW) and alternate wetting and moderate soil drying(AWMD). Compared with the WW treatment,the activities of ADP glucose pyrophosphorylase(AGPase), starch synthase(StSase) and starch branching enzyme(SBE), and the accumulation of non-structural carbohydrates(NSCs) in the stems before heading were significantly improved, and more starch granules were stored in the stems in the AWMD treatment. After heading, the activities of α-amylase, β-amylase, sucrose phosphate synthase(SPS) and sucrose synthase in the synthetic direction(SSs)were increased in the stems to promote the remobilization of NSCs for grain filling under AWMD. During grain filling, the enzymatic activities of sucrose synthase in the cleavage direction(SSc), AGPase, StSase and SBE in the inferior spikelets were increased, which promoted grain filling, especially for the inferior spikelets under AWMD.However, there were no significant differences in vascular microstructures. The grain yield and grain weight could be improved by 13.1 and 7.5%, respectively, by optimizing of the irrigation regime. We concluded that the low activities of key enzymes in carbon metabolism is the key limitation for the poor grain filling, as opposed to the vascular microstructures, and AWMD can increase the amount of NSC accumulation in the stems before heading, improve the utilization rate of NSCs after heading, and increase the grain filling, especially in the inferior spikelets, by altering the activities of key enzymes in carbon metabolism.
文摘Cooking and nutrition qualities of grain are two of the most important aspects of rice quality. To understand the difference in cooking and nutrition qualities among the grains at different positions within a rice panicle, the distributions of gel consistency (GC), amylose content (AC) and crude protein content (CPC) of grains in a panicle in association with different nitrogen levels (0, 120 and 240 k c/ha) were investigated by using two rice varieties, Yangdao 6 (indica) and Wuyujing 3 (japonica). In general, the grains at the basal part of a panicle had lower GC and higher AC than those at the upper or middle part of a panicle. The 1^st grain on the secondary branch with earlier flowering exhibited the highest GC, whereas the 2nd grain on the primary branch with later flowering showed the highest AC. For Yangdao 6, CPC in the grains on the primary branches was lower at the middle part of a panicle than at the upper or basal part of a panicle. For Wuyujing 3, there were no significant differences in CPCs in the grains among the upper, middle and basal parts of a panicle. GC in the grains was increased, whereas AC was reduced from zero nitrogen application (ON) to low amount of nitrogen application (LN), and the result was reversed from LN to medium amount of nitrogen application (MN). CPC was increased with the increase in the amount of nitrogen application.

文摘With 10 rice cultivars (lines) as materials, the changes in activities of adenosine diphosphoglucose pyrophosphorylase(ADPGPase), starch synthase (SSase) and starch branching enzyme (Q-enzyme) in the grains during grain filling, and theirrelationships with the filling rate, gel consistency (GC), alkali spreading value (ASV) and amylose content (AC) werestudied. The results showed that changes in activities of ADPGPase, SSase and Q-enzyme exhibited a single peak duringgrain filling, and the time of the activity peaks for the former two enzymes was earlier than that of the maximum grain-fillingrate (Tmax), and the time reaching the peak for Q-enzyme was synchronous with Tmax. The activities at early grain fillingstage, and the mean and maximum activities of each enzyme during grain filling period were positively and significantly orvery significantly correlated with the mean and maximum grain filling rate and starch content (mg grain-1) in the grains.Activities of ADPGPase at all grain filling stages and those of Q-enzyme at the early and mid filling stages were notsignificantly correlated the cooking quality (GC, ASV and AC). SSase activities at the early filling stage were significantlyand negatively correlated with GC and ASV, and positively correlated with AC. Activities of SSase at mid and late grainfilling stages and Q-enzyme at the late filling stage were significantly and positively correlated with GC and ASV, andnegatively correlated with AC. Spraying zeatin or abscisic acid at early grain filling stage could obviously regulate theactivities of ADPGPase, SSase and Q-enzyme in the grains.
文摘To determine biochemical changes during the germination of rice grains (Oryza sativa L. subsp, indica var. Mottaikaruppan) and to improve germination rate using gibberellic acid and surfactants [sodium dodecyl sulfate (SDS) (1.0 g/L) and Triton-X-100 (1.0 mL/L)], whole rice grains soaked in distilled water for 12 h at 30℃were germinated in the dark at 30℃ for five days. The highest germination rate (77.1%) was obtained on the 5th day. An increase in the content of reducing sugars from 7.3 to 58.1 mg/g DM (dry matter) was observed from the 1st day of germination. Free amino acids and soluble protein contents increased to 3.69 and 5.29 mg/g DM, respectively on the 5^th day of germination. Total protein content decreased from 100.5 to 91.0 g/kg DM during germination. Increases in amylolytic (1.1 to 190.0 U/g DM) and proteolytic (0 to 0.12 U/g DM) activities were observed during germination. Effects of different concentrations of gibberellic acid on the germination of rice grains were evaluated and 0.1 g/L was found to promote germination. When effects of gibberellic acid (0.1 g/L) and surfactants were evaluated individually and together, higher germination rate was observed in the control experiment (grains germinated in distilled water), whereas giberellic acid and surfactants decreased the germination rate. Therefore, the flour obtained from the grains germinated for four days using distilled water to obtain high content of soluble materials and enzyme activities can be used in preparation of bakery items.

文摘Cadmium(Cd) contamination in rice has been a hot topic of research because of its potential risk to human health. In this study, a double haploid(DH) population derived from Zhongjiazao 17(YK17)(an early-season indica cultivar)×D50(a tropical japonica cultivar) was used to identify quantitative trait loci(QTLs) associated with Cd concentration in brown rice(CCBR) and Cd concentration in milled rice(CCMR). Continuous and wide variation for CCBR and CCMR were observed among the DH population. Correlation analysis revealed a positive and highly significant correlation between the two traits. A total of 18 QTLs for CCBR and 14 QTLs for CCMR were identified in five different pot and field trials. Two pairs of QTLs for CCBR(qCCBR2-1 and qCCBR2-2, qCCBR9-1 and qCCBR9-2) and one pair of QTLs for CCMR(qCCMR5-1 and qCCMR5-2) were detected in multiple trials. The alleles increasing CCBR at the qC CBR2-1/qC CBR2-2 and qC CBR9-1/qC CBR9-2 QTLs were contributed by YK17 and D50, respectively, whereas the D50 allele at the qCCMR5-1/qCCMR5-2 QTLs increased CCMR. Eight pairs of QTLs for CCBR and CCMR, qCCBR2-2 and qCCMR2-2, qCCBR3 and qCCMR3, qCCBR4-2 and qCCMR4-1, qCCBR4-3 and qCCMR4-2, qCCBR4-4 and qCCMR4-3, qCCBR5 and qCCMR5-2, qCCBR7 and qCCMR7, and qCCBR11-1 and qCCMR11-2, co-localized on chromosomes 2, 3, 4, 5, 7, and 11, respectively. For all of these QTL pairs, except qCCBR5/qCCMR5-2, the additive effects came from YK17. In addition, four CCMR QTLs showing significant additive×environment interaction and two pairs of CCMR QTLs with bi-allelic epistatic interactions were identified. The results of this study could facilitate marker-assisted selection of breeding rice varieties with low Cd accumulation in grain.

文摘Genotypic and environmental variation in Cd, Cr, As, Ni and Pb concentrations of grains, and the relationships between these heavy metals and Fe, Zn were investigated using 9 rice genotypes grown in 6 locations for two successive years. Significant genotypic variation was detected in the five heavy metal concentrations in grains, indicating the possibility to reduce the concentration of these heavy metals in grains through breeding approach. The environmental effect varied with metal, with Pb and Ni having greater variation than the other three metals. There was significant genotype-environment (location) interaction of the concentrations of all five heavy metals in grains, suggesting the importance of cultivar choice in producing rice with low heavy metal concentrations in grains for a given location. Correlation analysis showed that Cd and As, Cr and Ni, and As and Pb con-centrations in rice grains were closely associated, and that Ni concentration in grains was negatively correlated with Zn concen-tration.
文摘The abnormal rice with small grains and erect panicles were found on a large scale in China, which showed shortend rice panicle and decreased number of grains in comparison with normal rice, and the grain was small and black-brown, and some of them were distorted, while the flag leaf was normal. A kind of nematode of Aphelenchoides was isolated from the grains of rice variety Wuyujing 3 which performed 'small grains and erect panicles' symptom. There were 2014 nematodes in one hundred grains infected, and up to 74 in single grain, 92 percent of the grains tested had nematodes in the infested panicles. The diagnosis characters of nematode include lateral fields about one-fourth as wide as body, with 4 incisures. The terminus bears a mucro of diverse shape with 3-4 pointed processes. The female post-vulval uterine sac extends less than 50% of distance from vulva to anus, no sperm in it. Oocytes usually arrange in 2-4 rows. The male spicules have a moderately developed rostrum. Morphological measurements showed it to be conspecific with Aphelenchoides besseyi Christie, 1942.

文摘The progenies differed in amylose and protein contents in grains, which derived from a rice cross, Dongnong 423×Toukei 180, were used to study changes in the activities of ADP-glucose pyrophosphorylase (AGPP), soluble starch synthetase (SSS), starch branching enzyme (SBE) and glutamine synthetase (GS) in rice grains during grain filling. The activities of AGPP, SSS and SBE gradually increased and then declined as a single-peak curve with the process of grain filling in the progenies with high and low amylose contents in grains. The progenies with high amylose content peaked earlier in the AGPP, SSS and SBE activities and had higher AGPP, SSS and SBE activities at the early grain filling stage than those with low amylose content. The GS activity peaked earlier and was higher at the late stage of grain filling in the progenies with high protein content than in those with low protein content. It is suggested that the activities of key enzymes for starch synthesis and glutamine synthetase could be changed in oriented breeding for amylose and protein contents in grains.
文摘A pot experiment was conducted to investigate cadmium(Cd) uptake by different rice cultivars that produce white or dark grains. Four cultivars with white grains(hereafter, white rice) and five cultivars with dark colors (hereafter dark rice) were selected for this experiment. Three levels of soil Cd concentrations, background(0), 5 and 10 mg/kg, were used. After harvest, plant biomass, tissue concentrations of Cd, Ca, Fe, Cu and Zn were analyzed. The results showed that Cd concentrations are significantly different between different genotypes, but when comparing the Cd concentrations for the two groups, no significant difference was found. For other divalent cations, Ca concentrations in dark rice were higher than those in white ones(P<0.001 for shoots, P=0.037 for roots); Fe concentrations in dark rice were also higher than those in white ones(P=0.001 either in shoot or root); Zn concentrations in shoot of dark rice were higher than those in white ones, but no significant difference in roots. The total molar concentrations of divalent cations in dark rice were also significantly higher than in white rice. The potential benefit of higher Ca and Fe concentrations in dark rice and similar Cd concentrations in both groups is also discussed in this paper.

文摘Grain size is a key factor influencing grain weight in rice.In this study,a chromosome segment substitution line(CSSL9-17)was identified,that exhibits a significant reduction in both grain size and weight compared to its donor parent 93-11.Further investigation identified two quantitative trait loci(QTL)on chromosome 11,designated qGW11a and qGW11b,which contribute to 1000-grain weight with an additive effect.LOC_Os11g05690,encoding the amino acid permease OsCAT8,is the target gene of qGW11a.Overexpression of OsCAT8 resulted in decreased grain weight,while OsCAT8 knockout mutants exhibited increased grain weight.The 287-bp located within the OsCAT8 promoter region of 93-11 negatively regulates its activity,which is subsequently correlated with an increase in grain size and weight.These results suggest that OsCAT8 functions as a negative regulator of grain size and grain weight in rice.

文摘The size and shape of rice grains influence their yield and commercial value.We investigated the role of OsDA1,a rice homolog of the Arabidopsis DA1 gene,in regulating grain size and shape.OsDA1 was highly expressed in young spikelets and glumes.Its overexpression led to enlarged seeds with increased width and decreased length/width ratio(LWR)and knocking out OsDA1 reduced grain width and increased grain length and LWR.A R310K point mutation in the DA1-like domain is a potential target for breeding for increased grain width and length.OsDA1 interacted with TCP gene-family proteins to regulate grain size and shape.Our findings deepen our understanding of the molecular mechanisms underlying grain size regulation and provide useful information for improving grain yield.

文摘With rising living standards,there is an increasing demand for high-quality rice.Rice quality is mainly defined by milling quality,appearance quality,cooking and eating quality,and nutrition quality.Among them,chalkiness is a key trait for appearance quality,which adversely affects cooking and eating quality,head rice yield,and commercial value.Therefore,chalkiness is undesirable,and reducing chalkiness is a major goal in rice quality improvement.However,chalkiness is a complex trait jointly influenced by genetic and environmental factors,making its genetic study and precision improvement a huge challenge.With the rapid development of molecular techniques,much knowledge has been gained about the genes and molecular networks involved in chalkiness formation.The present review describes the major environmental factors affecting chalkiness and summarizes the quantitative trait loci(QTL)associated with chalkiness.More than 150 genes related to chalkiness formation have been reported.The functions of the genes regulating chalkiness,primarily those involved in starch synthesis,storage protein synthesis,transcription regulation,organelle development,grain shape regulation,and hightemperature response,are described.Finally,we identify the challenges associated with genetic improvement of chalkiness and suggest potential strategies.Thus,the review offers insight into the molecular dynamics of chalkiness and provides a strong basis for the future breeding of high-quality rice varieties.

文摘The development and application of the small-grain rice sterile line Zhuo201S(Z201S)has demonstrated its potential for mechanized hybrid rice seed production,leading to significant cost reductions.However,the molecular mechanism responsible for the small-grain size characteristic of Z201S remains unclear.In this study,we conducted a genetic analysis using near-isogenic lines constructed from Z210S,a small-grain rice sterile line,and R2115,a normal-grain variety.The results revealed that the small-grain trait in Z201S is governed by a single partially dominant gene which also enhances grain number.Through mapping,we localized the causal gene to the short arm of chromosome 2,within a 113 kb physical region delimited by the molecular markers S2-4-1 and LB63.Transgenic analysis and gene expression assays indicated LOC_Os02g14760 as the most likely candidate gene,suggesting that the small-grain size trait of Z201S is controlled by a novel locus that has not been previously identified.

文摘Increasing effective panicle number per plant(EPN)is one approach to increase yield potential in rice.However,molecular mechanisms underlying EPN remain unclear.In this study,we integrated mapbased cloning and genome-wide association analysis to identify the EPN4 gene,which is allelic to NARROW LEAF1(NAL1).Overexpression lines containing the Teqing allele(TQ)of EPN4 had significantly increased EPN.NIL-EPN4^(TQ) in japonica(geng)cultivar Lemont(LT)exhibited significantly improved EPN but decreased grain number and flag leaf size relative to LT.Haplotype analysis indicated that accessions with EPN4-1 had medium EPN,medium grain number,and medium grain weight,but had the highest grain yield among seven haplotypes,indicating that EPN4-1 is an elite haplotype of EPN4 for positive coordination of the three components of grain yield.Furthermore,accessions carrying the combination of EPN4-1 and haplotype GNP1-6 of GNP1 for grain number per panicle showed higher grain yield than those with other allele combinations.Therefore,pyramiding of EPN4-1 and GNP1-6 could be a preferred approach to obtain high yield potential in breeding.

文摘Lysine content is a criterion of the nutritional quality of rice.Understanding the process of lysine biosynthesis in early-flowering superior grain(SG)and late-flowering inferior grain(IG)of rice would advance breeding and cultivation to improve nutritional quality.However,little information is available on differences in lysine anabolism between SG and IG and the underlying mechanism,and whether and how irrigation regimes affect lysine anabolism in these grains.A japonica rice cultivar was grown in the field and two irrigation regimes,continuous flooding(CF)and wetting alternating with partial drying(WAPD),were imposed from heading to the mature stage.Lysine content and activities of key enzymes of lysine biosynthesis,and levels of brassinosteroids(BRs)were lower in the IG than in the SG at the early grainfilling stage but higher at middle and late grain-filling stages.WAPD increased activities of these key enzymes,BR levels,and contents of lysine and total amino acids in IG,but not SG relative to CF.Application of 2,4-epibrassinolide to rice panicles in CF during early grain filling reproduced the effects of WAPD,but neither treatment altered the activities of enzymes responsible for lysine catabolism in either SG or IG.WAPD and elevated BR levels during grain filling increased lysine biosynthesis in IG.Improvement in lysine biosynthesis in rice should focus on IG.

文摘Objective:To determine distribution of arthropods in rice grains obtained from different sources.Methods: Rice samples were randomly collected from public in urban areas,farmers in rice field areas,aborigines in undeveloped areas and retailers in commercial premises.Random samples of rice were taken out from each sample for isolation of arthropods using a modified Berlese Tullgren Funnel Method.Mites were mounted prior to identification; weevils were directly identified.Results:Samples of rice from retailers in commercial premises had the highest infestation by arthropods followed by samples from urbanites,aborigines and rice farmers.Two species of weevils,Sitophilus oryzae(S.oryzae) and Sitophilus granarius(S.granarius),were found.Samples from commercial premises had the least percentage of weevils compared to those collected from domestic premises. Depending on the source of samples,densities of S.granarius and S.oryzae ranges from 11-103 weevils/ kg and 7- 80 weevils/kg,respectively.Important species of mites in stored rice identified were mainly members of the families Cheyletidae,Echimyopodidae,Pyroglyphidae,Saproglyphidae and Tenuipalpidae.Among the species of mites identified were Austroglycyphagus malaysiensis,Cheyletus fortis,Cheyletus malaccensis,Dermatophagoides pteronyssinus,Grammolichus malukuensis and Suidasia pontifica.Average density of most of the mites was less than 40 mites/kg of rice grains.In this study,the highest number of mites in rice samples was recovered from commercial premises,followed by samples from urbanites.Samples from farmers and aborigines contained lesser mites.Conclusion:This study demonstrated the presence of 3 allergenic mite species in rice, i.e A.malaysiensis,D.pteronyssinus and S.pontifica.Weevils,S.oryzae and S.granarius that are known to be allergenic,were also found.

文摘Applying iodine fertilizers to cultivate iodine-rich crops for daily intake is an effective approach for iodine supplementation,especially for aromatic rice.Field experiments were conducted during the early growing seasons of 2021 and 2022 to evaluate the impacts of foliar application of iodine fertilizer on aromatic rice and to explore the optimal iodine fertilizer concentration.At the full heading stage,six different concentrations of sodium iodide solutions of 0%(CK),0.010%(T1),0.025%(T2),0.050%(T3),0.075%(T4),and 0.100%(T5)were applied to indica aromatic rice cultivars Meixiangzhan 2 and Xiangyaxiangzhan.The results showed that sodium iodide treatments significantly increased the iodine and sodium contents in both leaves and grains.Compared with the CK,the T1 and T2 treatments increased the 2-acetyl-1-pyrroline(2-AP)content in mature grains by 8.41%-101.66%and 13.58%-74.60%,respectively.Improvements in the contents of 1-pyrroline-5-carboxylic acid,proline,1-pyrroline,and methylglyoxal,as well as the activity of proline dehydrogenase were also detected.Additionally,sodium iodide treatments remarkably decreased the chalky grain rate,chalkiness area,and chalkiness degree of aromatic rice,with the T2 treatment exhibiting a 17.79%-47.42%decrease in chalkiness degree compared with the CK.Meanwhile,T1 and T2 treatments showed beneficial impacts on chlorophyll content,photosynthetic characteristics,and yield components,while T3,T4,and T5 treatments exhibited adverse effects on leaf and grain yields.The linear discriminant analysis revealed significant differences between treatments.The correlation analysis and piecewise structural equation modeling showed that the iodine and sodium influenced the photosynthetic characteristics and chlorophyll content of the leaves,thereby regulating the 2-AP biosynthesis and yield components,ultimately affecting the 2-AP content and yield.Overall,this study suggests that foliar application of 0.025%sodium iodide is an effective method to enrich the iodine content in rice grains,improve the grain aroma and appearance quality of aromatic rice,without detrimental effects on grain yield.

文摘Twenty-four rice genotypes were examined to assess genetic variability,heritability,and correlations for seven-grain quality traits,eight nutritional elements,and protein.ANOVA revealed significant differences for the quality traits studied.For every trait under study,the phenotypic coefficient of variation was higher than the correspon-dence genotypic coefficient of variation.Heritability in a broad sense varied from 29.75%for grain length to 98.31%for the elongation trait.Hulling percentage recovery had a highly significant positive correlation with milling and head rice percentage.Consequently,milling percentage had a highly positive correlation with head rice percentage.In amylose percentage,all the genotypes belonged to low amylose except the Hassawi-1 variety,which had intermediate amylose content.Mineral nutrition contents of magnesium(Mg),sodium(Na),potas-sium(K),calcium(Ca),copper(Cu),manganese(Mn),zinc(Zn),iron(Fe),or protein percentage gave different variations for 24 rice genotypes under all the nutritional elements.Among the 24 genotypes,ten rice genotypes–HighNutrient-1,HighNutrient-2,HighNutrient-9,HighNutrient-8,HighNutrient-3,Hassawi-2,HighNutrient-7,HighNutrient-6,Hassawi-1,and HighNutrient-4–had the highest heist value for all nutritional and protein con-tents,and could be used as a donor to improving new varieties.There was a positive and significant correlation between magnesium Mg,K,Zn and Fe.Consequently,K had a positive correlation with zinc Zn,Fe,and protein percentage.Clustering analysis was divided into two groups:thefirst group included all genotypes rich in nutri-ents,while the remaining genotypes with low nutritional content were included in the second group.

文摘An efficient and good DNA extraction protocol should be simple, affordable and yield enough DNA with high quality. Rice(Oryza sativa L.) DNA extraction methods often use seedlings or leaves rather than the grains and tend to be time-consuming, involve multiple steps, and use hazardous chemicals and expensive enzymes. Rice grains offer several benefits over seedlings and leaves as a source of DNA for genetic analysis. However, these benefits are underutilized because the bulk of a rice grain is made up of starch. It is particularly important, but difficult to get rid of the starch while extracting DNA from rice grains. This co-precipitated polysaccharide is a known inhibitor of DNA polymerase activity in polymerase chain reaction(PCR). We describe here a very simple and highly affordable Chelex~?-100 based DNA extraction method from rice grains. It does not require any hazardous chemicals or enzymes. This method reproducibly extracts DNA with good purity indices(A_(260)/A_(230) and A_(260)/A_(280) values), but requires only a few steps.
