Investigating the Impact of Ocean Acidification on Anti-Stress Mechanisms in Sepia esculenta Larvae Based on Transcriptome Profiling

With the rapid development of oil,energy,power and other industries,CO_(2) emissions rise sharply,which will cause a large amount of CO_(2) in the air be absorbed by the ocean and lead to ocean acidification.The growth and development of organisms can be seriously affected by acidified seawater.Sepia esculenta is a mollusk with high nutritional and economic value and is widely cultured in offshore waters of China.Larvae are the early life forms of the organism and are more vulnerable to changes in the external environment.Too low pH will lead to some adverse reactions in larvae,which will affect metabolism,immune response and other life activities.In this study,we sequenced the transcriptome of S.esculenta subjected to acidified seawater stress and identified 1072differentially expressed genes(DEGs).The detected atypical expression of DEGs substantiates cellular malformation and translocation in S.esculenta under low pH stimulation.Simultaneously,this also substantiates the notable impact of ocean acidification on mollusks.These DEGs were used for functional enrichment analysis of GO and KEGG,and the top twenty items of the biological process classification in GO terms and 11 KEGG signaling pathways were significantly enriched.Finally,the constructed proteinprotein interaction network(PPI)was used to analyze protein-protein interactions,and 12 key DEGs and 3 hub genes were identified.The reliability of 12 genes was verified by quantitative RT-PCR.A comprehensive analysis of the KEGG signaling pathway and PPI revealed that ocean acidification leads to abnormalities in lipid metabolism in S.esculenta larvae,which can lead to cancer development and metastasis,accompanied by some degree of inflammation.The results of the study will help to further investigate the physiological processes of S.esculenta when stimulated by ocean acidification,and provide a reference to cope with the captive breeding of S.esculenta affected by acidification. 展开更多

Over recent decades, Gampaha district, Sri Lanka, has experienced significant urbanisation and industrial growth, increasing groundwater demand due to limited and polluted surface water resources. In 2013, a community uprising in Rathupaswala, a village in Gampaha district, accused a latex glove manufacturing factory of causing groundwater acidity (pH < 4). This study evaluates the spatial and temporal changes in geochemical parameters across three transects in the southern part of Gampaha district to 1) assess the impact of geological formations on groundwater;2) compare temporal variations in groundwater;and 3) explain acidification via a geochemical model. Seventy-two sample locations were tested for pH, electrical conductivity (EC), and anion concentrations (sulphate, nitrate, chloride and fluoride). Depth to the water table and distance from the sea were measured to study variations across sandy, peaty, lateritic, and crystalline aquifers. Results showed pH readings around 7 for sandy and crystalline aquifers, below 7 for peaty aquifers, and below 5 for lateritic aquifers, with significant water table fluctuations near Rathupaswala area. Principal component analysis revealed three principal components (PCs) explaining 86.0% of the variance. PC1 (40.6%) correlated with pH, EC, and sulphate (saltwater intrusion), while PC2 (32.0%) correlated with nitrates and depth to the water table (anthropogenic nutrient pollution). A geochemical transport model indicated a cone of depression recharged by acidic groundwater from peat-soil aquifers, leading to acidic groundwater in Rathupaswala area. Previous attributions of acidic pH to the over-exploitation of groundwater by the latex factory have been reevaluated;the results suggest natural acidification from prolonged water-rock interactions with iron-rich lateritic aquifers. Groundwater pH is influenced by local climate, geology, topography, and drainage systems. It is recommended that similar water-rock interaction conditions may be present throughout the wet zone of Sri Lanka, warranting detailed studies to confirm this hypothesis. 展开更多

Background:Soil acidifcationn caused by anthropogenic activities may aft soil biochemical cydling,bidiversity,productivity,and multiple eosystem-related functions in drylands.However,to date,such information is lacking to support this hypothesis.Methods Based on a transect survey of 78 naturally assembled shrub communities,we caloulated acid deposition flux in Northwest China and evaluated its likely ecological ffets by testing three altemnative hypotheses,namely:.nidche complementarity,mass ratio,and vegetation quantity hypotheses Rao's quadratic entopy and community-weighted mean traits were employed to represent the complementary aspect of niche complementarity and mass ratio effects,respectively.Resulbs:We observed that in the past four decades,the concentrations of exchangeable base cations in soil in Northwest China have decreased significantly to the extent of having faced the risk of depletion,whereas changes in the calium carbonate content and pH of soil were not significant.Adid deposition primani ly increased the aboweground biomass and shrub density in shrublands but had no sigmificant effect on shrub richness and ecasystem multifunctionality(EMF),indicating that acid deposition had positive but weak ecological effects on dryland ecosystems.Community wd ghted mean of functional traits(representing the mass ratio hypothesis)correlated negatively with EMF,whereas both Rao's quadratic entropy(representing the niche complementarity hypothesis)and aboveground biomass(representing the vegetation quantity hypothesis)correlated positively but insignifcantly with EMF.These biodiversity-EMF relationships highlight the fragility and instability of drylands relative to forest ecasystems.Concuions:The findings from this study serve as important reference points to understand the ris of soil acidification in arid regions and its impacts on biodiversity-EMF relationships. 展开更多

Soil samples were collected from the depth 0 - 0.10 m yearly in the spring (5 samples from each locality). In the dry soil, samples were determined actively and exchanged soil reaction. The most significant negative changes (decreases of soil reaction) were determined in Haplic Stagnosols group and Cambisols group. The pH value in topsoil is primarily controlled by soil type and soil substrate, soil management and land use, and to a lesser extent by climatic region. 展开更多

However, problems of low penetration depth, local overheating and fracture closure exist. Coal demineralisation by acids has advantages in coal unblocking and permeability improvement, while it is difficult for acid to enter microcracks.Microwave-asisted acidification may offer an alternative. In this work, XRD,^(1)H-NMR, and SEM were used to evaluate the effect of microwave-assisted acidification on the microstructure of coal. Results show that kaolinite, calcite, and dolomite can be dissolved by acid. After microwave irradiation, the graphitization of microcrystalline structure of carbon improves. Microwave-assisted acidification erodes minerals in coal and enhances the graphitization degree of microcrystalline structure. Compared to individual microwave irradiation or acidification, the pore volume and pore connectivity can be greatly enhanced by microwave-assisted acidification. The NMR permeability of coal increased by 28.05%. This study demonstrates the potential of microwave-assisted acidification for coalbed methane recovery. 展开更多

Based on soil monitoring data in nine sites of Jinhu, Xuyi and Hongze counties on the south bank of Hongze Lake from 1982 to 2013, changes in soil p H, total nitrogen(TN) content, available phosphorus content, organic matter(OM)content and cation exchange capacity(CEC) were analyzed. The results show that due to excessive application of chemical fertilizer in soil on the south bank of Hongze Lake, soil p H reduced by about 2 on average, while TN content and available phosphorus content in soil increased by more than one time and 2-5 times respectively. Soil acidification caused by agricultural production was very serious. In addition, low soil p H resulted in serious loss of soil cation, so that soil CEC in2013 accounted for less than 50% of that in 1982 and affected mineral nutrient metabolism of crops. Therefore, application of calcium, potassium and trace-element fertilizer should be paid more attention to during agricultural production in future. 展开更多

Ecological experiments were conducted to examine the effects of seawater containing elevated par- tial pressure of carbon dioxide (p CO2 800 × 10 -6 , 2 000 × 10 -6 , 5 000 × 10 -6 and 10 000 × 10 -6 ) on the survival and reproduction of female Acartia pacifica, Acartia spinicauda, Calanus sinicus and Centropages tenuiremis, which are the dominant copepods in the southern coastal waters of China. The results show that the effects of elevated p CO2 on the survival rates of copepods were species-specific. C. sinicus, which was a macro-copepod, had a higher survival rate (62.01%–71.96%) than the other three species (5.00%–26.67%) during the eight day exposure. The egg production rates of C. sinicus, A. spinicauda and C. tenuiremis were significantly inhibited by the increased p CO2 and the exposure time duration. There were significantly negative impacts on the egg hatching success of A. spinicauda and C. tenuiremis in the p CO2 2 000 × 10 -6 and 10 000 × 10 -6 groups, and, in addition, the exposure time had noticeably impacts on these rates too. This study indicates that the reproductive performances of copepods were sensitive to elevated p CO2 , and that the response of different copepod species to acidified seawater was different. Furthermore, the synergistic effects of seawater acidification and climate change or other pollutant stresses on organisms should be given more attention. 展开更多

We studied the effect of pH (pH 5, 6, 7 and 8) on the hatching percentage, survival and reproduction of Artemia strains in Bohai Bay salt ponds. Strains included parthenogenetic Artemia from Bohai Bay (BHB), Artemiafranciscana from San Francisco Bay, and A. franciscana artificially produced in salt ponds in Vietnam. The latter was included as a potential inoculum for biological management of salt ponds. The hatching percentage of cysts after 24 h and the survival rate of the tested Artemia strains were significantly reduced when exposed to a culture medium at pH 5 for 18 d (P〈0.05). The tolerance of Artemia to 48 h acid exposure varied with developmental stage, increasing in the following order: juvenile, nauplii, pre-adult, with maximum tolerance in adults. All strains of Artemia tested could not reproduce at pH 5. At pH levels from pH 6-8, a higher pH generally resulted in a shorter brood interval and enhanced ovoviviparity. Hence, we suggest that brine acidification has a negative impact on Artemia populations in the Bohai Bay saltworks. Inoculation of Artemia with either local parthenogenetic Artemia or exotic A. franeiscana should be feasible at pH 7-8. 展开更多

Oceanic uptake of anthropogenic carbon dioxide results in a decrease in seawater pH, a process known as "ocean acidification". The pearl oyster Pinctada fucata, the noble scallop Chlamys nobilis, and the green-lipped mussel Perna viridis are species of economic and ecological importance along the southern coast of China. We evaluated the effects of seawater acidification on clearance, respiration, and excretion rates in these three species. The animals were reared in seawater at pH 8.1 (control), 7.7, or 7.4. The clearance rate was highest at pH 7.7 for P. fucata and at pH 8.1 for C. nobilis and P. viridis. The pH had little effecton the respiration rate of P. fucata and P. viridis. In contrast, the respiration rate was significantly lower atpH 7.4 in C. nobilis. The excretion rate was significantly lower at pH 7.4 than pH 8.1 for all species. Theresults indicate that the reduction in seawater pH likely affected the metabolic process (food intake, oxygenconsumption, and ammonia excretion) of these bivalves. Different species respond differently to seawateracidification. Further studies are needed to demonstrate the exact mechanisms for this effect and evaluateadaptability of these bivalves to future acidified oceans. 展开更多

General pretreatment processes of ammonia stripping and phenols solvent extraction can reduce the concentration of toxic compounds of the coal gasification wastewater for the following biological treatment. However, some emulsified coal tar still exists in the influent and many substances in coal tar are refractory and toxic to microorganisms. This study is mainly on the removal of emulsified coal tar by acidification demulsion. The experimental results show that the acidification process of the wastewater by pure hydrochloric acid can reduce the chemical oxygen demand (COD), total organic carbon (TOC), total phenolics and oil about 3.1%-11.3%, 6%-- 10.8%, 5.3%--8.6% and 25.2%--57.4% respectively with pH value in the range of 4 to 7. The analysis of molecular weight distribution indicates that compounds removed from the wastewater by this process are large molecular substances. The experiment also shows that the efficiency of COD removal in the demulsion process by different acids is different and the phosphoric acid is prominent. The preserved time of the wastewater also affects the efficiency of demulsion. Small amount low-cost solid additives including kaolin and diatomite can improve the rate of coal tar sedimentation and enhance the removal efficiency of organics in the phosphoric acidification process. 展开更多

The cost-effective treatment of activated sludge that is generated by refining petroleum is a challenging industrial problem.In this study, semi-continuous stirred tank reactors(CSTRs) containing petroleum refinery excess activated sludge(PREAS)were used to comparatively investigate hydrolysis and acidification rates, after the addition of heneicosane(C_(21)H_(44))(R1)and 1-phenylnaphthalene(C16 H12)(R2) to different and individual reactors. Operation of the reactors using a sludge retention time(SRT) of 6 days and a pH of 5.0, resulted in the maintenance of stable biological activity as determined by soluble chemical oxygen demand(SCOD), volatile fatty acids(VFAs) production and oil removal efficiency. The optimum conditions for hydrogen production include a SRT of 8 days, at pH 6.5. Under these conditions, hydrogen production rates in the control containing only PREAS were 1567 mL/L(R0), compared with 1365 mL/L in Rl and 1454 mL/L-PREAS in R2.Coprothermobacter, Fervidobacterium, Caldisericum and Tepidiphilus were the dominant bacterial genera that have the potential to degrade petroleum compounds and generate VFAs. This study has shown that high concentrations of heneicosane and 1-phenylnaphthalene did not inhibit the hydrolytic acidification of PREAS. 展开更多

Increasing atmospheric CO2 can reduce seawater pH and carbonate ions, which may adversely affect the survival of the larvae of calcareous animals. Cyclina sinensis is a commercially and ecologically important species in several Asian countries. Living in coast shallow waters, this species has experienced the coastal environmental changes frequently throughout its life cycle. In this study, we simulated possible future seawater pH values including 8.2, 7.8 and 7.4 and examined the effects of ocean acidification on the early development of C. sinensis. Clam embryos were incubated for 48 h (2 d) in control and high-CO2 seawater to compare embryo- genesis, larval growth and swimming behavior. Fertilization rate was quite sensitive to pH, and moderate acidification could induce a significant decrease in fertilization rate. However, only extreme acidification could bring significant negative effect to hatching rate, body size, and average path velocity of trochophora. Moreover, with seawater acidification, C. sinensis needs much more time to reach the same developmental stage, which increases the risk of larva survival. Together with recent studies demonstrating negative impacts of high CO2 on fertilization and larva swimming behavior, the results imply a future decrease of C. sinensis populations in oceans if its acclimation to the predicted environmental alteration does not occur. 展开更多

In this study, we evaluated the effects of CO_2-induced seawater acidification on fertilization, embryogenesis and early larval development in the sea urchin Glyptocidaris crenularis, that inhabits subtidal coastal areas in northern China. The range in seawater p H used in experiments was based on the projections of the Intergovernmental Panel on Climate Change(IPCC), to the year 2100. A natural seawater treatment(p H_(nbs) =7.98±0.03) and three laboratory-controlled acidified treatments(OA_1, ΔpH_(nbs) =-0.3 units; OA_2, ΔpH_(nbs) =-0.4 units; OA_3, ΔpH_(nbs) =-0.5 units) were used in experiments. Results show that:(1) there was a negative effect of seawater acidification on fertilization and on the percentage of abnormal fertilized eggs;(2) the size of early cleavage stage embryos decreased in a dose-dependent manner with decreasing p H;(3) both the hatching rate of blastulae and the survival rate of four-armed pluteus larvae decreased as pH declined;(4) larval abnormalities including asymmetrical development, changes in the length of skeletal elements, and corroded spicules were observed in all seawater acidified-treatments compared with the control. These data indicate that seawater acidification has a negative impact on the early development of G. crenularis, and supports the hypothesis that the response of echinoderms to ocean acidification(OA) varies among species. Further research is required to clarify the specific cellular mechanisms involved. 展开更多

However, the impacts of acidification on the cycling of ethylene in forest soils have not been fully taken into consideration in global change studies. Forest topsoils (0-5 cm) under four temperate forest stands were sampled to study the effects of a pH change on the emissions of ethylene and carbon dioxide from the soils and concentrations of dissolved organic carbon (DOC) released into the soils. Increasing acidification or alkalinization of forest soils could increase concentrations of DOC released into the soils under anoxic and oxic conditions. The ethylene emission from these forest topsoils could significantly increase with a decreasing pH, when the soils were acidified experimentally to a pH〈4.0, and it increased with an increasing concentration of DOC released into the soils, which was different from the carbon dioxide emission from the soils. Hence, the short-term stimulating responses of ethylene emission to a decreasing pH in such forest soils resulted from the increase in the DOC concentration due to acidification rather than carbon mineralization. The results would promote one to study the effects of soil acidification on the cycling of ethylene under different forest stands, particularly under degraded forest stands with heavy acid depositions. 展开更多

To sort out reasonable agricultural practices favorable to abating soil acidification, the task of this study was directed to a long-term field trial in Chongqing, dudng which chemical fertilizer, organic fertilizer were applied to different crop rotations and the soil pH value was measured. The results indicated that all treatments decreased pH value in the 0 to 20 cm soil layer after ten years. Problems were more serious when chlorine-containing fertilizer, excessive chemical fertilizer and mixed fertilizer were applied. It is demonstrated that balance rates of N, P and K fertilizers, application of muck in field are advantageous to abating soil acidification. Oil plants affect soil acidification more than cereal in different crop rotation. 展开更多

The increasing soil acidity induced by excessive nitrogen (N) fertilization in farmlands has profoundly impacted the soil carbon dynamics. However, the way in which changes in soil pH regulating the soil carbon dynamics in a deep soil profile is still not well elucidated. In this study, through a 12-year field N fertilization experiment with three N fertilizer treatments (0, 120, and 240 kg N/(hm-2·a)) in a dryland agroecosystem of China, we explored the soil pH changes over a soil profile up to a depth of 200 cm and determined the responses of soil organic carbon (SOC) and soil inorganic carbon (SIC) to the changed soil pH. Using a generalized additive model, we identified the soil depth intervals with the most powerful statistical relationships between changes in soil pH and soil carbon dynamics. Hierarchical responses of SOC and SIC dynamics to soil acidification were found. The results indicate that the changes in soil pH explained the SOC dynamics well by using a non-linear relationship at the soil depth of 0-80 cm (P=0.006), whereas the changes in soil pH were significantly linearly correlated with SIC dynamics at the 100-180 cm soil depth (P=0.015). After a long-term N fertilization in the experimental field, the soil pH value decreased in all three N fertilizer treatments. Furthermore, the declines in soil pH in the deep soil layer (100-200 cm) were significantly greater (P=0.035) than those in the upper soil layer (0-80 cm). These results indicate that soil acidification in the upper soil layer can transfer excess protons to the deep soil layer, and subsequently, the structural heterogeneous responses of SOC and SIC to soil acidification were identified because of different buffer capacities for the SOC and SIC. To better estimate the effects of soil acidification on soil carbon dynamics, we suggest that future investigations for soil acidification should be extended to a deeper soil depth, e.g., 200 cm. 展开更多
