METHODS: A miRNA microarray conta...AIM: To investigate the molecular mechanisms of miRNA in advanced gastric cancers(AGCs) before and after cytoreductive surgery(CRS) + hyperthermic intraperitoneal chemotherapy(HIPEC). METHODS: A miRNA microarray containing human mature and precursor miRNA sequences was used to compare expression profiles in serum samples of 5 patientswith AGC before and after CRS + HIPEC. The upregulation of miR-218 was confirmed by real-time reverse transcription polymerase chain reaction and its expression was analyzed in SGC7901 gastric cancer cells. RESULTS: miRNA microarray chip analysis found that the level of miR-218 expression was upregulated more than 8 fold after CRS + HIPEC. Furthermore, miR-218 increased gastric cancer cell chemosensitivity to cisplatin in vitro and inhibited gastric cell tumor growth in nude mice in vivo(0.5 vs 0.78, P < 0.05).CONCLUSION: Our results indicated that targeting miR-218 may provide a strategy for blocking the development of gastric cancer and reverse the multi-drug resistance of gastric cell lines.展开更多
Hepatocellular carcinoma(HCC)often develops in patients with underlying liver disease,yet HCC with syncytial giant cells(SGCs)is extremely rare.Herein,we report a 55-year-old man with a 6-year history of alcoholic cir...Hepatocellular carcinoma(HCC)often develops in patients with underlying liver disease,yet HCC with syncytial giant cells(SGCs)is extremely rare.Herein,we report a 55-year-old man with a 6-year history of alcoholic cirrhosis who during his regular checkup presented with marked elevation of alpha-fetoprotein.Clinical examination and imaging analyses revealed a tumor-like lesion in segment 4 of the liver,which was removed by limited wedge resection.Histological analysis by hematoxylin and eosin staining indicated pleomorphic and atypical nodules,with some SGCs,embedded within the boundaries of the neoplastic lesion.The adjacent liver parenchyma showed microvesicular steatosis,pericellular fibrosis,and moderate hemosiderin accumulation(grade 2,as determined by Prussian blue iron stain)in hepatocytes and Kupffer cells but no copper accumulation(as determined by orcein stain).Immunohistochemical analysis showed hepatocyte antigen-positive staining for the neoplastic cells and SGCs.The diagnosis was made for cirrhosis-related HCC with SGCs.The previous reports of pleomorphic HCC have featured osteoclast-like(i.e.,mesenchymal type)giant cells,making this case of epithelial type giant cells very rare.The patient’s 6-month history of hypericum perforatum/St John’s wort self-medication may have prompted the cirrhosis or HCC progression or the unusual SGC manifestation.展开更多

The introduction of a new technology or innovation is often accompanied by“ups and downs”in its fortunes.Gartner Inc.defined a so-called hype cycle to describe a general pattern that many innovations experience:tech...The introduction of a new technology or innovation is often accompanied by“ups and downs”in its fortunes.Gartner Inc.defined a so-called hype cycle to describe a general pattern that many innovations experience:technology trigger,peak of inflated expectations,trough of disillusionment,slope of enlightenment,and plateau of productivity.This article will compare the ongoing introduction of Open Science(OS)with the hype cycle model and speculate on the relevance of that model to OS.Lest the title of this article mislead the reader,be assured that the author believes that OS should happen and that it will happen.However,I also believe that the path to OS will be longer than many of us had hoped.I will give a brief history of the today’s“semi-open”science,define what I mean by OS,define the hype cycle and where OS is now on that cycle,and finally speculate what it will take to traverse the cycle and rise to its plateau of productivity(as described by Gartner).展开更多
OBJECTIVE: To observe the impact of total flavonoids from Ganshanbian(Herba Hyperici Attenuati)on the expression of vascular smooth muscle membrane L-type calcium channel alpha1 C subunit(CaL-α1C) and ATP-sensitive K...OBJECTIVE: To observe the impact of total flavonoids from Ganshanbian(Herba Hyperici Attenuati)on the expression of vascular smooth muscle membrane L-type calcium channel alpha1 C subunit(CaL-α1C) and ATP-sensitive K+channel(KATP)-Kir6.1mRNA, and explore the mechanisms of the antiarrhythmic effect of Ganshanbian(Herba Hyperici Attenuati)total flavonoids.METHODS: The treatment group was fed total flavonoids from Ganshanbian(Herba Hyperici Attenuati)for 7 days by gavage with 100 mg·kg﹣1·d﹣1.The blank control group and model control group were given the same amount of normal saline for 7 d.Arrhythmias were induced by performing a myocardial ischemia-reperfusion and electrocardiogram was observed. Reverse transcription-polymerase chain reaction was used to detect the expression of CaL-α1CandKATP-Kir6.1 mRNA in the myocardial cell membrane of all groups of rats. RESULTS: Total flavonoids from Ganshanbian(Herba Hyperici Attenuati) can delay the appearance of myocardial ischemia reperfusion arrhythmias,shorten the duration of myocardial ischemia reperfusion arrhythmias, reduce heart rate, reduce cell membrane expression of CaL-α1C mRNA and enhance the expression of KATP-Kir6.1 mRNAin myocardial ischemia-reperfusion arrhythmic rats.CONCLUSION: Total flavonoids from Ganshanbian(Herba Hyperici Attenuati) can alleviate arrhythmias by affecting the expression of L-type calcium channels and ATP-sensitiveK+ channels.展开更多
