In this study, we isolated polysaccharides from four species of seaweeds, such as Grateloupia fi licina(GFP), Eucheuma spinosum(ESP), Ulva pertusa(UPP), and A scophyllum nodosum(ANP), and characterized their structures and prebiotic ef fects in vitro. The results showed that these polysaccharides were dif ferent in total sugar and sulfate contents as well as monosaccharide composition. GFP and ESP significantly promoted bifi dobacterium proliferation and 0.1% ESP and 0.4% GFP resulted in the highest proliferation rates of beneficial bacteria, whereas UPP and ANP inhibited the growth of beneficial bacteria at all tested concentrations(0.1%–0.5%). The different behaviors of the four seaweed-originated polysaccharides might be refl ected by differences in monosaccharide composition and structure. Therefore, polysaccharides isolated from GFP and ESP could be utilized as prebiotics. However, more studies must be carried out in vivo.展开更多

[Method] Different nanometallic materials were added into Agrobacterium medium and Anthurium callus medium, to investigate the effects of their effects on Agrobacterium growth, callus growth and differentiation, and Agrobacterium contamination. [Result] Among the 4 nanometallic materials, NanoAg-2 showed a significant inhibitory effect on the growth of Agrobacterium, with a minimal inhibitory concentration of 25 mg/L. Even for the Anthurium calli or transgenic material contaminated by the Agrobacterium, a good antibacterial effect could be achieved after treating with 25 mg/L NanoAg-2 for 1 d with oscillation, the antibacterial rate reached 100%, and the Anthurium calli could grow and differentiate normally. [Conclusioa] NanoAg-2 could effectively inhibit Agrobacterium contamination, and its an- tibacterial effect is significantly better than cephalosporin and carbenicillin.展开更多

After phytochemical analysis, this study evaluated extracts’ anti-inflammatory, antioxidant and antibacterial properties from the ACAZY recipe. Three extractions, an aqueous macerate (AM), an aqueous decoction (AD) and an hydroethanolic macerate (HEM) of the ACAZY recipe powder were carried out. Phytochemical screening of the extracts was carried out using high-performance thin-layer chromatography (HPTLC) and the determination of phenolic compounds. The anti-inflammatory potential was assessed in vitro using pro-inflammatory enzyme inhibition tests. 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and Ferric-reducing antioxidant power (FRAP) antioxidant properties were also determined. The antibacterial activity was evaluated on Staphylococcus aureus and Streptococcus pneumoniae strains. Phytochemical analysis revealed the presence of flavonoids, saponins, tannins, anthracenosids, sterols and triterpenes in the extracts. The extracts inhibited pro-inflammatory enzymes by more than 40% at only 100 µg/mL. The extracts also showed potent antibacterial activity with a minimum inhibitory concentration 1 mg/mL on Staphylococcus aureus and 2 mg/mL on Streptococcus pneumoniae. The extracts in the ACAZY formula have shown anti-inflammatory and antioxidant properties in vitro. The AD also showed an antibacterial activity. This justifies its use in traditional medicine to treat acute respiratory infections.展开更多

The applied 13.56 MHz radiofrequency (RF) is a carrier of the low-frequency modulation. The high-frequency part was chosen to select the malignant lesion using the specialties of the tumor: the higher conductivity and dielectric constant of the tumor than its host. The electric field selects the tumor, and the low-frequency amplitude modulation polarizes and excites the transmembrane proteins of the malignant cells. The dominant absorption of the energy by the microscopic clusters of the membrane rafts acts like nanoparticle heating. Exciting the membrane produces various apoptotic signals. The processes were modeled using silico and phantom experiments, which proved the concept. The preclinical verification was made in vitro and in vivo, and in the end, clinical proofs validated the method. Our objective is to follow all the development steps from the laboratory to the clinics in a trilogy of articles. Microtubule-associated protein 2 (MAP2) is of importance for neuronal, axonal, and dendritic generation, extension, and stabilization, as well as for the regulation of synaptic plasticity. OBJECTIVE: To investigate the antagonistic effects of natural-cerebrolysin-containing serum on beta amyloid protein 1-40 (Aβ1-40)-induced neurotoxicity from the standpoints of cell proliferation, synaptogenesis, and cytoskeleton formation (MAP2 expression). DESIGN, TIME AND SETTING: A paralleled, controlled, neural cell, and molecular biology experiment was performed at the Institute of Integrated Chinese and Western Medicine, Shenzhen Hospital, Southern Medical University between February 2006 and April 2008. MATERIALS: PC12 cells, derived from the rat central nervous system, were purchased from Shanghai Institute of Cell Biology, Chinese Academy of Sciences, China. A β1-40 was provided by Sigma, USA. Natural-cerebrolysin was provided by Shenzhen Institute of Integrated Chinese and Western Medicine, China. The natural-cerebrolysin was predominantly composed of Renshen (Radix Ginseng), Tianma (Rhizoma Gastrodiae), and Yixingye (Ginkgo Leaf) in a proportion of 1:2:2. Following conventional water extraction technology, an extract (1:20) was prepared. Each gram of extract equaled 20 grams of crude drug. In a total of 12 adult male New Zealand rabbits, six underwent intragastric administration of natural-cerebrolysin extract for 1 month to prepare natural-cerebrolysin-containing serum, and the remaining six rabbits received intragastric administration of physiological saline to prepare normal blank serum. METHODS: An AIzheimer's disease in vitro model was induced in PC12 cells using Aβ1-40. The cells were incubated with varying doses of natural-cerebrolysin-containing serum (2.5%, 5%, and 10%). Normal blank serum-treated PC12 cells served as a blank control group. MAIN OUTCOME MEASURES: Through the use of inverted phase contrast microscope, cell morphology and neurite growth were observed, neurite length was measured, and the percentage of neurite-positive cells was calculated. Cell proliferation rate was determined by MTT assay, and MAP 2 expression was detected by fluorescent immunocytochemistry. RESULTS: Following Aβ1-40 treatments, some PC12 cells were apoptotic/dying, and only a few short neurites were observed. Following interventions with natural-cerebrolysin-containing serum, the PC12 cells proliferated, there was an increased number of neurites, and neurite length was enhanced. After middle- and high-dose natural-cerebrolysin treatments, the percentage of neurite-positive cells, as well as the average length of neurites, was significantly greater than the normal blank serum-treated PC12 cells (P 〈 0.05 or P 〈 0.01). Compared with the blank control group, MAP2 expression in the Aβ1-40-treated PC12 cells was significantly inhibited, and the cell proliferation rate was significantly decreased (P 〈 0.01). Following incubations with natural-cerebrolysin-containing serum, MAP2 expression and cell proliferation rate in the PC12 cells were significantly increased in a dose-dependent manner, compared with treatments with blank control serum (P 〈 0.05 or P 〈 0.01 ). CONCLUSION: Natural-cerebrolysin exhibited antagonistic effects on neurotoxicity in Aβ1-40 induced Alzheimer's disease in vitro models. These effects were likely related to cell proliferation and the upregulation of intracellular MAP2 expression.展开更多

The pre...Oral health problems such as periodontal diseases, dental caries, and endodontic infections have a significant negative impact on oral health and impose a substantial financial burden on the global population. The prevalence of these issues is increasing due to the buildup of bacterial plaque and the growing resistance of bacteria to antimicrobial treatments. The aims of this study to evaluate the anti-bacterial activity of four types of antibiotics (Amoxicillin, Augmentin, Azithromycin and Metronidazole) and four types of toothpastes (Sensodyne, ipana, denta and cariax Gingival Kin) on two oral pathogenic bacteria (Streptococcus mutans and Staphylococcus epidermidis). Bacterial samples of previously isolated Streptococcus mutans and Staphylococcusepidermidis were used as test organisms and the Kirby-Bauer disc diffusion method was employed to assess the antibacterial efficacy of various antibiotics and evaluate the impact of different toothpastes using a filter paper disc agar measurement technique. Each filter disc was saturated with toothpaste solution in a test tube for approximately 30 to 40 seconds, after which they were placed on Mueller-Hinton broth bacterial cultures in petri dishes. These Petri dishes were then incubated at 37°C for 24 hours, and the clear zone’s diameter (inhibition zone in mm) was subsequently measured and the results were recorded. The results demonstrated that Sensodyne toothpaste and Metronidazole antibiotic were ineffective against both types of bacteria, while Augmentin and Amoxicillin were effective by high diameter inhibition zones of growth against S. mutans and Azithromycine against S. epidermidis. Also Ipana, Denta, and Cariax Gingival Kin toothpastes exhibited a moderate effect against the two bacteria. This study suggests that certain antibiotics and toothpastes can effectively inhibit the growth of harmful oral bacteria, but not all of them are effective.展开更多

It was then conjugated w...Murine monoclonal antibody (MoAb) BB4.3, raised against the human gastric cancer cell line BGC823, was puriffied with Protein A-Sepharose CL-4B affinity chromatography and identified as IgG2a. It was then conjugated with a hematoporphyrin derivative (HPD) by using carbodiimide. The qualitative analysis of this conjugate showed that the amount of free HPD was negligible and there were no IgG aggregates among the conjugates. The conjugate retained both the antibody and photochemical activity of HPD.In vitro, the phototoxic effect of this HPD-BB4.3 conjugate on target cells was about 15 times higher than that of free HPD. The quality of selective photocytotoxicity was proven by the greater cytotoxi-city this conjugate showed than that of corresponding normal mouse IgG (NIgG) conjugated with HPD. It showed less cytotoxicity to colon cancer cell line B-80 (negative reaction to MoAb BB4.3) than to BGC825. Moreover, its cytotoxicity to BGC823 cells could be blocked specifically by excess BB4.3 antibody, but not by another MoAb 3G9, which combines with BGC823 at different binding sites from MoAb BB4.3.Nude mice inoculated with 2 × 10- BGC823 cells were given HPD-BB4.3, HPD, HPD-NIgG, HPD plus BB4.3 and PBS, respectively then exposed to light. Four out of six animals treated with the HPD-BB4.3 conjugate remained tumor-free for a long period. Although two developed tumors, there was a significant difference between the HPD-BB4.3-treated group and all the control groups in tumor induction time, tumor growth rate, and survival time (p<0.001). The HPD-BB4.3 conjugate inhibited the growth of established tumors by more than 40% in comparison with control groups (p<0.05).展开更多

After OVI was given to the mice via gastric lavage, the effects of OVI on the infected mice were observed. The minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) for dysentery bacteria were determined in vitro. The results showed that origanum volatile oil showed obvious protective effect on mice infected with Sh. sonnei and Sh. flexneri F 2a. and it had germistatic and germicidal effects on dysentry bacteria. We are led to conclude that origanum volatile oil is an effective medicine against the infection of dysentery bacteria.展开更多

However, this feed resource does not meet animal nutritional requirement due to deficiency in nitrogen, energy and minerals. In addition, at maturity lignification is the major concern since it reduces digestibility and contributes to methane emission. Thus, the objective of this study was to evaluate the effect of supplementing low-quality Eragrostis grass hay with five (9281, 11,252, 11,255, 11,595 and 11,604) selected Stylosanthes scabra accessions on in vitro ruminal fermentation and neutral detergent fiber degradation. Therefore, in vitro study was conducted on grass hay, accessions and the mixture of grass hay with each accession included at two (15%, 30%) levels. The substrates (grass hay, accessions and the mixtures) were incubated in separate serum bottles for 72 h. Neutral detergent fiber (NDF) of the accessions ranged from 300 to 350 g/kg DM with crude protein (CP) value ranging from 177.5 to 184.1 g/kg DM. Eragrostis grass hay had NDF value of 813 g/kg DM, with CP value of 34.3 g/kg DM. Grass hay fermented slowly, it took 30 h for grass hay to produce gas volume above 50 mL, while Stylosanthes scabra accessions took 12 h. Supplementing grass hay with accessions significantly improved fermentation. However, it was observed that 15% inclusion took 30 h to produce gas volume above 50 mL, whereas at 30% inclusions it took 24 h for accession 9281, 11,595 and 11,604. Accession 11,604 improve grass fermentation by almost three times the value of grass hay in 2 h. Grass hay supplemented with accession 11,604 at 30% had a positive associative effect and significantly improved NDF degradability. In conclusion, accession 11,604 may be fed strategically as forage supplement to low-quality forage for ruminants.展开更多

文摘Surface functionalization of magnesium(Mg)alloys is desired to obtain the surfaces with both improved corrosion resistance and antibacterial property.A corrosion-resistant and antimicrobial coating was prepared on Mg alloy surface by layer-by-layer(LbL)assembly of chitosan(CHI)and poly-L-glutamic acid(PGA)by electrostatic attraction.The functionalized surfaces of the Mg alloys were characterized by field-emission scanning electron microscopy(FE-SEM),Fourier transform infrared(FT-IR)spectroscopy and electrochemical tests.The bactericidal activity of the samples against Staphylococcus aureus was assessed by the zone of plate-counting method.The obtained coating on the Mg alloy substrates exhibits good corrosion resistance and antibacterial performance.

文摘Although prebiotic activities of alginate and agar oligosaccharides isolated from seaweeds have been reported, it remains unknown whether seaweed polysaccharides have prebiotic activity. In this study, we isolated polysaccharides from four species of seaweeds, such as Grateloupia fi licina(GFP), Eucheuma spinosum(ESP), Ulva pertusa(UPP), and A scophyllum nodosum(ANP), and characterized their structures and prebiotic ef fects in vitro. The results showed that these polysaccharides were dif ferent in total sugar and sulfate contents as well as monosaccharide composition. GFP and ESP significantly promoted bifi dobacterium proliferation and 0.1% ESP and 0.4% GFP resulted in the highest proliferation rates of beneficial bacteria, whereas UPP and ANP inhibited the growth of beneficial bacteria at all tested concentrations(0.1%–0.5%). The different behaviors of the four seaweed-originated polysaccharides might be refl ected by differences in monosaccharide composition and structure. Therefore, polysaccharides isolated from GFP and ESP could be utilized as prebiotics. However, more studies must be carried out in vivo.
文摘[Objective] This study was conducted to investigate the application of nanometallic materials in inhibiting Agrobacteriurn contamination in genetic transformation of Anthurium. [Method] Different nanometallic materials were added into Agrobacterium medium and Anthurium callus medium, to investigate the effects of their effects on Agrobacterium growth, callus growth and differentiation, and Agrobacterium contamination. [Result] Among the 4 nanometallic materials, NanoAg-2 showed a significant inhibitory effect on the growth of Agrobacterium, with a minimal inhibitory concentration of 25 mg/L. Even for the Anthurium calli or transgenic material contaminated by the Agrobacterium, a good antibacterial effect could be achieved after treating with 25 mg/L NanoAg-2 for 1 d with oscillation, the antibacterial rate reached 100%, and the Anthurium calli could grow and differentiate normally. [Conclusioa] NanoAg-2 could effectively inhibit Agrobacterium contamination, and its an- tibacterial effect is significantly better than cephalosporin and carbenicillin.

文摘In order to develop natural antistaling agent for blueberries preservation,the antibacterial activities of 10 kinds of plant essential oils were analyzed by oxford-cup test and tube dilution method,and their inhibitory abilities to two kinds of representative Gram bacteria which are Escherichia coli and Staphylococcus aureus,and three kinds of harmful fungi in blueberries,involving Alternaria alternata,Penicillium digitatum and Botrytis cinerea,were tested.The results showed that 7 kinds of essential oils can inhibit the three fungi,among which cinnamon oil,clove oil,anise oil and thyme oil have strong fungi static efficacy.Cinnamon oil and thyme oil can effectively inhibit the Gram-positive bacterium(Staphylococcus aureus);cinnamon oil and masson pine oil are strongly inhibitory to Gram-negative bacterium(E.coli).Cinnamon oil has broad-spectrum ability to inhibit fungi,Gram-negative and positive bacteria,with a minimum inhibitory concentration(MIC)ranging from 0.0391 to 0.1560 L/mL.With strong inhibition effects on fungi,cinnamon oil can be used as a compound base oil and mixed with other essential oils to retain the freshness of blueberries,thus achieving better antibacterial and anti-corrosion effects.
文摘ACAZY is a plant formula used in traditional medicine in Burkina Faso to treat respiratory infections. After phytochemical analysis, this study evaluated extracts’ anti-inflammatory, antioxidant and antibacterial properties from the ACAZY recipe. Three extractions, an aqueous macerate (AM), an aqueous decoction (AD) and an hydroethanolic macerate (HEM) of the ACAZY recipe powder were carried out. Phytochemical screening of the extracts was carried out using high-performance thin-layer chromatography (HPTLC) and the determination of phenolic compounds. The anti-inflammatory potential was assessed in vitro using pro-inflammatory enzyme inhibition tests. 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and Ferric-reducing antioxidant power (FRAP) antioxidant properties were also determined. The antibacterial activity was evaluated on Staphylococcus aureus and Streptococcus pneumoniae strains. Phytochemical analysis revealed the presence of flavonoids, saponins, tannins, anthracenosids, sterols and triterpenes in the extracts. The extracts inhibited pro-inflammatory enzymes by more than 40% at only 100 µg/mL. The extracts also showed potent antibacterial activity with a minimum inhibitory concentration 1 mg/mL on Staphylococcus aureus and 2 mg/mL on Streptococcus pneumoniae. The extracts in the ACAZY formula have shown anti-inflammatory and antioxidant properties in vitro. The AD also showed an antibacterial activity. This justifies its use in traditional medicine to treat acute respiratory infections.
文摘Modulated electro-hyperthermia (mEHT) targets tissue’s natural electric and thermal heterogeneities to heat the cancer cells selectively. The applied 13.56 MHz radiofrequency (RF) is a carrier of the low-frequency modulation. The high-frequency part was chosen to select the malignant lesion using the specialties of the tumor: the higher conductivity and dielectric constant of the tumor than its host. The electric field selects the tumor, and the low-frequency amplitude modulation polarizes and excites the transmembrane proteins of the malignant cells. The dominant absorption of the energy by the microscopic clusters of the membrane rafts acts like nanoparticle heating. Exciting the membrane produces various apoptotic signals. The processes were modeled using silico and phantom experiments, which proved the concept. The preclinical verification was made in vitro and in vivo, and in the end, clinical proofs validated the method. Our objective is to follow all the development steps from the laboratory to the clinics in a trilogy of articles. This present is the first part, which deals with in silico, phantom, and in vitro research.
文摘Laboratory cultures of freshwater cyanobacterium (blue-green alga) Microcystis aeruginosa PCC 7806 was cvaluated for its hepatotoxic effects in rats. The lyophilized cell extract injected intraperitoneally at 1 and 2 LD50 (15.8 and 31.6 mg/kg, respectively) produced significant increase in liver-specific enzymes viz. plasma alkaline phosphatase,γ-glutamyl transferase, lactate dehydrogenase with a concomitant decrease in hepatic glutamic pyruvic transaminase. A corresponding increase in liver body weight index and histopathological changes in liver (degeneration of hepatocytes, congestion and hemorrhage etc.) are indicative of a dose and time dependent hepatotoxic nature of the algal extract

文摘BACKGROUND: Neuronal loss, synapse mutilation, and increasing malnourished axons are pathologically related to Alzheimer's disease. Microtubule-associated protein 2 (MAP2) is of importance for neuronal, axonal, and dendritic generation, extension, and stabilization, as well as for the regulation of synaptic plasticity. OBJECTIVE: To investigate the antagonistic effects of natural-cerebrolysin-containing serum on beta amyloid protein 1-40 (Aβ1-40)-induced neurotoxicity from the standpoints of cell proliferation, synaptogenesis, and cytoskeleton formation (MAP2 expression). DESIGN, TIME AND SETTING: A paralleled, controlled, neural cell, and molecular biology experiment was performed at the Institute of Integrated Chinese and Western Medicine, Shenzhen Hospital, Southern Medical University between February 2006 and April 2008. MATERIALS: PC12 cells, derived from the rat central nervous system, were purchased from Shanghai Institute of Cell Biology, Chinese Academy of Sciences, China. A β1-40 was provided by Sigma, USA. Natural-cerebrolysin was provided by Shenzhen Institute of Integrated Chinese and Western Medicine, China. The natural-cerebrolysin was predominantly composed of Renshen (Radix Ginseng), Tianma (Rhizoma Gastrodiae), and Yixingye (Ginkgo Leaf) in a proportion of 1:2:2. Following conventional water extraction technology, an extract (1:20) was prepared. Each gram of extract equaled 20 grams of crude drug. In a total of 12 adult male New Zealand rabbits, six underwent intragastric administration of natural-cerebrolysin extract for 1 month to prepare natural-cerebrolysin-containing serum, and the remaining six rabbits received intragastric administration of physiological saline to prepare normal blank serum. METHODS: An AIzheimer's disease in vitro model was induced in PC12 cells using Aβ1-40. The cells were incubated with varying doses of natural-cerebrolysin-containing serum (2.5%, 5%, and 10%). Normal blank serum-treated PC12 cells served as a blank control group. MAIN OUTCOME MEASURES: Through the use of inverted phase contrast microscope, cell morphology and neurite growth were observed, neurite length was measured, and the percentage of neurite-positive cells was calculated. Cell proliferation rate was determined by MTT assay, and MAP 2 expression was detected by fluorescent immunocytochemistry. RESULTS: Following Aβ1-40 treatments, some PC12 cells were apoptotic/dying, and only a few short neurites were observed. Following interventions with natural-cerebrolysin-containing serum, the PC12 cells proliferated, there was an increased number of neurites, and neurite length was enhanced. After middle- and high-dose natural-cerebrolysin treatments, the percentage of neurite-positive cells, as well as the average length of neurites, was significantly greater than the normal blank serum-treated PC12 cells (P 〈 0.05 or P 〈 0.01). Compared with the blank control group, MAP2 expression in the Aβ1-40-treated PC12 cells was significantly inhibited, and the cell proliferation rate was significantly decreased (P 〈 0.01). Following incubations with natural-cerebrolysin-containing serum, MAP2 expression and cell proliferation rate in the PC12 cells were significantly increased in a dose-dependent manner, compared with treatments with blank control serum (P 〈 0.05 or P 〈 0.01 ). CONCLUSION: Natural-cerebrolysin exhibited antagonistic effects on neurotoxicity in Aβ1-40 induced Alzheimer's disease in vitro models. These effects were likely related to cell proliferation and the upregulation of intracellular MAP2 expression.
文摘Oral health problems such as periodontal diseases, dental caries, and endodontic infections have a significant negative impact on oral health and impose a substantial financial burden on the global population. The prevalence of these issues is increasing due to the buildup of bacterial plaque and the growing resistance of bacteria to antimicrobial treatments. The aims of this study to evaluate the anti-bacterial activity of four types of antibiotics (Amoxicillin, Augmentin, Azithromycin and Metronidazole) and four types of toothpastes (Sensodyne, ipana, denta and cariax Gingival Kin) on two oral pathogenic bacteria (Streptococcus mutans and Staphylococcus epidermidis). Bacterial samples of previously isolated Streptococcus mutans and Staphylococcusepidermidis were used as test organisms and the Kirby-Bauer disc diffusion method was employed to assess the antibacterial efficacy of various antibiotics and evaluate the impact of different toothpastes using a filter paper disc agar measurement technique. Each filter disc was saturated with toothpaste solution in a test tube for approximately 30 to 40 seconds, after which they were placed on Mueller-Hinton broth bacterial cultures in petri dishes. These Petri dishes were then incubated at 37°C for 24 hours, and the clear zone’s diameter (inhibition zone in mm) was subsequently measured and the results were recorded. The results demonstrated that Sensodyne toothpaste and Metronidazole antibiotic were ineffective against both types of bacteria, while Augmentin and Amoxicillin were effective by high diameter inhibition zones of growth against S. mutans and Azithromycine against S. epidermidis. Also Ipana, Denta, and Cariax Gingival Kin toothpastes exhibited a moderate effect against the two bacteria. This study suggests that certain antibiotics and toothpastes can effectively inhibit the growth of harmful oral bacteria, but not all of them are effective.
文摘Objective:To access the in vitro antibacterial activity of Hibiscus rosa-sinensis(H.rosa-sinensis) flower extract against human pathogens.Methods:Antibacterial activity was evaluated by using disc and agar diffusion methods.The protein was run through poly acrylmide gel electrophoresis to view their protein profile.Results:The results showed that the cold extraction illustrates a maximum zone of inhibition against Bacillus suhtillis(B.suhtillis),Escherichia coli(E.coli) viz.,(17.00±2.91),(14.50±1.71)mm.followed by hot extraction against.E.coli.Salmonella sp.as(11.66 ±3.14),(10.60±3.09)mm.In methanol extraction showed a highest zone of inhibition recorded against B.suhtillis,E.coli as(18.86±0.18),(18.00±1.63) mm pursued by ethanol extraction shower) utmost zone of inhibition recorded against Salmonella sp.at(20.40±1.54) mm.The crude protein from flower showed a maximum inhibitory zone observed against Salmonella sp.,E.coli viz.,(16.55±1.16),(14.30±2.86)mm.The flower material can be taken as an alternative source of antibacterial agent against the human pathogens.Conclusions:The extracts of the H.rosasinensis art;proved to have potential antibacterial activity,further studies arc highly need for the drug development.
文摘Preliminary exploration of the soothing,oil control,acne removing effects and its mechanisms of Artemisia annua extract.The soothing effect of Artemisia annua extract was tested by hyaluronidase biochemical reaction.The soothing and oil-controlling effects were investigated by cell model.The inhibitory effect on propionibacterium acnes was studied by suspension quantification.The results showed that Artemisia annua extract could effectively inhibit the degradation of hyaluronic acid(P<0.01).Artemisia annua extract significantly inhibited the secretion of inflammatory cytokines TNF-αand IL-6 in RAW264.7 cells(P<0.05).Artemisia annua extract at 0.5%,0.25%,0.125%could significantly inhibit the secretion of oil by SZ95 cells(P<0.05).The minimum inhibitory concentration of Artemisia annua extract against propionibacterium acnes was 0.625%,and the inhibitory rate against propionibacterium acnes increased with the increase of the concentration of Artemisia annua extract.In summary,Artemisia annua extract can achieve acne efficacy through soothing and oil control,and this function may be achieved by reducing hyaluronic acid degradation,inhibiting inflammatory pathways produced by inflammatory factors TNF-αand IL-6,inhibiting oil secretion,and inhibiting the growth of propionibacterium acnes.
文摘Murine monoclonal antibody (MoAb) BB4.3, raised against the human gastric cancer cell line BGC823, was puriffied with Protein A-Sepharose CL-4B affinity chromatography and identified as IgG2a. It was then conjugated with a hematoporphyrin derivative (HPD) by using carbodiimide. The qualitative analysis of this conjugate showed that the amount of free HPD was negligible and there were no IgG aggregates among the conjugates. The conjugate retained both the antibody and photochemical activity of HPD.In vitro, the phototoxic effect of this HPD-BB4.3 conjugate on target cells was about 15 times higher than that of free HPD. The quality of selective photocytotoxicity was proven by the greater cytotoxi-city this conjugate showed than that of corresponding normal mouse IgG (NIgG) conjugated with HPD. It showed less cytotoxicity to colon cancer cell line B-80 (negative reaction to MoAb BB4.3) than to BGC825. Moreover, its cytotoxicity to BGC823 cells could be blocked specifically by excess BB4.3 antibody, but not by another MoAb 3G9, which combines with BGC823 at different binding sites from MoAb BB4.3.Nude mice inoculated with 2 × 10- BGC823 cells were given HPD-BB4.3, HPD, HPD-NIgG, HPD plus BB4.3 and PBS, respectively then exposed to light. Four out of six animals treated with the HPD-BB4.3 conjugate remained tumor-free for a long period. Although two developed tumors, there was a significant difference between the HPD-BB4.3-treated group and all the control groups in tumor induction time, tumor growth rate, and survival time (p<0.001). The HPD-BB4.3 conjugate inhibited the growth of established tumors by more than 40% in comparison with control groups (p<0.05).
文摘Objective:To assess the in-vitro antihacterial activity and anti-inflammatory activity of orally administered different extracts(Hydro-alcoholic,methanolic,ethyl acetate and hexane)of Rauvolfia tetraphylla(R.tetraphylla)root bark in Carrageetiaii induced acute inflammation in rats.Methods:In-vitro antibacterial activity was evaluated for extracts against four Gram positive and four Gram negative bacteria by using cylinder plate assay.Hydro-alcoholic extract(70%v/v ethanol)at 200,400 and 800 mg/kg doses and methanolic,ethyl acetate and hexane extracts at doses 100,200 and 400 mg/kg were tested for anti-inflammatory activity in Carrageenan induced rat paw oedema model and paw thickness was measured every one hour up to 6 hrs.Results:All extracts of R.tetraphylla root bark showed good zone of inhibition against tested bacterial strains.In Carrageenan induced inflammation model,hydro-alcoholic and methanolic extract of R.tetraphylla root bark at three different doses produced significant(P<0.00l)reduction when compared to vehicle treated control group and hexane,ethyl acetate extracts.Conclusions:In the present study extracts of R.tetraphylla root bark shows good in-vitro antibacterial activity and in-vivo anti-inflammatory activity in rats.
文摘To observe the germistatic and germicidal effects of origanum volatile oil (OVI) on the dysentery bacteria, the abdominal cavity of mice was infected with Shigella sonne (Sh. sonnei) and Shigella flexneri (Sh. flexneri) F 2a. After OVI was given to the mice via gastric lavage, the effects of OVI on the infected mice were observed. The minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) for dysentery bacteria were determined in vitro. The results showed that origanum volatile oil showed obvious protective effect on mice infected with Sh. sonnei and Sh. flexneri F 2a. and it had germistatic and germicidal effects on dysentry bacteria. We are led to conclude that origanum volatile oil is an effective medicine against the infection of dysentery bacteria.

文摘In order to understand in vitro antibacterial effect of antibiotics and EM on Aeromonas hydrophila,14 isolates of A. hydrophila were isolated from diseased fish and aquaculture in the study. PCR amplification results of aerolysin gene( Aero gene) of isolates indicated that 7 out of 14 isolates were pathogenic A.hydrophila. In vitro antibacterial test indicated that 14 isolates were resistance to penicillin G,ampicillin,cephradine,cefazolin,tetracycline,terimethoprim,lincomycin and cephalexin,but showed high sensitivity to cefotaxime and furazolidone. In vitro antibacterial effect of EM on 14 isolates of A. hydrophila was positively correlated with concentration,and EM with the concentration greater than 60% had antibacterial effect on 14 isolates.
文摘This paper reports the effects of the Yh^(3+) and its four complexes on cultured cells.The results indicate that the 5 mmol YbCl_3 has evident toxicity on cultured cells,and after it is coordinated with ligands,its toxicity is decreased.These.four complexes neither destroy the growth nor inhibit the adliesion of diploid cell to the surface of the flasket,but they inhibit the adhesion of cancer cells,and result in cell death.The complex of Yb~?(3+) with ligand(1) can destroy the cancer cells in division phase.forming aberrant mitotic cells,but the other three complexes of Yb^(3+) with ligands can destroy the cancer cells in interphase.These effects depend on the dose of the complex.
文摘Natural pastures constitute a major component of ruminant livestock feed, and are the most cost-effective feed resource available for smallholder subsistence farmers. However, this feed resource does not meet animal nutritional requirement due to deficiency in nitrogen, energy and minerals. In addition, at maturity lignification is the major concern since it reduces digestibility and contributes to methane emission. Thus, the objective of this study was to evaluate the effect of supplementing low-quality Eragrostis grass hay with five (9281, 11,252, 11,255, 11,595 and 11,604) selected Stylosanthes scabra accessions on in vitro ruminal fermentation and neutral detergent fiber degradation. Therefore, in vitro study was conducted on grass hay, accessions and the mixture of grass hay with each accession included at two (15%, 30%) levels. The substrates (grass hay, accessions and the mixtures) were incubated in separate serum bottles for 72 h. Neutral detergent fiber (NDF) of the accessions ranged from 300 to 350 g/kg DM with crude protein (CP) value ranging from 177.5 to 184.1 g/kg DM. Eragrostis grass hay had NDF value of 813 g/kg DM, with CP value of 34.3 g/kg DM. Grass hay fermented slowly, it took 30 h for grass hay to produce gas volume above 50 mL, while Stylosanthes scabra accessions took 12 h. Supplementing grass hay with accessions significantly improved fermentation. However, it was observed that 15% inclusion took 30 h to produce gas volume above 50 mL, whereas at 30% inclusions it took 24 h for accession 9281, 11,595 and 11,604. Accession 11,604 improve grass fermentation by almost three times the value of grass hay in 2 h. Grass hay supplemented with accession 11,604 at 30% had a positive associative effect and significantly improved NDF degradability. In conclusion, accession 11,604 may be fed strategically as forage supplement to low-quality forage for ruminants.

文摘Onion(Allium cepa L.)is a common biennial herb,and contains allicin compounds which can kill harmful microorganism.The antibacterial effects of raw and mature onion juice of purple,yellow and white-skinned onions at different concentrations on Escherichia coli(E.coli)and Staphylococcus aureus(S.aureus)were studied.There were differences among the antibacterial effects of raw and mature onion juice of purple,yellow and white-skinned onions at different concentrations.What was more,the antibacterial effects of raw onion juice were better than those of the mature one.The antibacterial effects of high concentration onion juice were better than those of the low one.The content of the total flavonoids in raw purple onion was higher than that in mature onion,and reached extremely significant levels in 50%,75%and 100%,respectively.Through comparison,it was found that onion juice of purple-skinned had the best suppression effects on S.aureus(gram-positive bacterium)and E.coli(gram-negative bacterium),and gram-positive bacterium had better suppression effects than gram-negative bacterium.It wound provide the basis about bioactive function and antibacterial drugs.

文摘[Objectives]To explore the antibacterial effects of essential oils from pomegranate on two kinds of postharvest pathogenic fungi,and to provide reference for the exploration of new plant-derived preservatives for pomegranate.[Methods]The inhibitory effects of 15 kinds of plant essential oils on 2 kinds of pomegranate postharvest pathogenic fungi were studied.[Results]Tea tree oil and eucalyptus oil have good inhibitory effects on the mycelium growth of Coniella granati and Botrytis cinerea,and the fumigation treatment of pomegranate can inhibit the rot of fruit during storage to a certain extent.[Conclusions]Tea tree oil and eucalyptus oil can effectively inhibit the mycelium growth of two kinds of pathogenic fungi,and can better control the rot of pomegranate during storage.It is necessary to further study the fresh-keeping effect and antibacterial mechanism of plant essential oils on pomegranate.
文摘[Objectives]This study was conducted to investigate the antibacterial effect of the extract from Hedyotis hedyotidea(DC)Merr.[Methods]Through in-vitro antibacterial tests,the antibacterial effects of H.hedyotidea ethanol extract on Escherichia coli,Staphylococcus aureus and Bacillus subtilis were analyzed.[Results]The extract had a low-susceptibility inhibitory effect to the three kinds of bacteria,and the minimal inhibitory concentrations(MICs)corresponding to S.aureus,E.coli and B.subtilis were 200,175 and 150 mg/ml,respectively.The minimal bactericidal concentrations(MBCs)corresponding to E.coli and S.aureus were both 500 mg/ml.[Conclusions]This study provides theoretical support for the wide application of H.hedyotidea.
