In this study, pyrosequencing was applied to compare bacteria phylogenic profile in incubated sediments with normal and exchanged organic detritus in macrophyte-dominated East Lake Taihu and algal-dominated Meiliang Bay. We observed significant bacteria species variations in sediments from two bays, regardless of treatments. RDA (Redundancy Analysis) analysis showed that sediment characteristics, especially concentrations of total nitrogen might account for this differentiation. Besides, algal-dominated Meiliang Bay sediment with addition of Vallisneria detritus exhibited higher bacterial species variations than the sediment amended with Microcystis detritus. To the contrary, sediments from macrophyte-dominated East Lake Taihu shared similar bacteria profile at all taxonomic levels and grouped together in MDS (multidimensional scaling) plots over the treatments with Vallisneria or Microcystis detritus addition into the sediment. We speculated that the different degradability of macrophyte detritus and algal detritus led to varied bacterial responses to exchanged organic resources and ultimately, the amounts, nutrient availability and degradability of organic resources may be main reasons for benthic bacteria community structure differentiation between the two states in shallow lakes.展开更多

The b...A great deal of oil contaminated the shoreline by the Qingdao oil pipeline explosion in 2013. The four oildegrading consortia were enriched from sediment samples with crude oil as sole carbon and energy sources. The biodiversity and community analysis showed that the Luteibacter, Parvibaculum and a genus belonging to Alcanivoracaceae were found predominant bacteria in the four consortia, which belonged to Proteobacteria. Nine strains exhibiting distinct 16S rRNA gene sequences were isolated from the consortia. These strains were identified to eight genera based on 16S rRNA gene sequences. Five of the nine strains degraded more than 30% of the crude oil in two weeks by gravimetric method. From the analysis of GC-MS, most of the isolated strains tended to degrade n-alkanes rather than PAHs. Five strains showed high degrading ability of the total n-alkanes. Only Strain D2 showed great PAHs degrading ability and the degrading rates ranged from 34.9% to 77.5%. The sequencing analysis of the oil-degrading consortia confirmed that the genus of Alcanivorax was one of the dominant bacteria in Consortia A and E and Strain E4 might be one of the dominant bacteria. The strains obtained in this study demonstrated the potential for oil bioremediation in oil-contaminated beach ecosystems.展开更多

The results show that the algal inhibiting agent could eliminate the blue-green algal blooms. 54 days later,chlorophyll a content in the water bodies reduced to below 0. 06 mg/L,and both COD_(Cr)and BOD_5 decreased significantly. NH_4^+-N and TP content were lower than 2 and 0. 05 mg/L respectively. The removal rate of COD_(Cr)was slightly low( 33%-48%),and the removal rates of other indicators were higher than 60%. It is clearly seen that the algal inhibiting agent could quickly eliminate the algal blooms and had a notable effect on the recovery of the eutrophic water bodies.展开更多
Magnetotactic bacteria(MTB)are morphologically and phylogenetically diverse prokaryotes commonly able to produce magnetic nanocrystals within intracellular membrane-bound organelles(i.e.,magnetosomes)and to swim along...Magnetotactic bacteria(MTB)are morphologically and phylogenetically diverse prokaryotes commonly able to produce magnetic nanocrystals within intracellular membrane-bound organelles(i.e.,magnetosomes)and to swim along geomagnetic field lines.We studied the diversity of MTB in the samples collected from a mangrove area in the Sanya River,Hainan,South China,using microscopic and microbial phylogenetic methods.Results of microanalysis and observation in microscopy and energy dispersive X-ray spectroscopy(EDXS)reveal a highly morphological diversity of MTB including unicellular cocci,vibrios,rod-shaped bacteria,and three morphotypes of multicellular magnetotactic prokaryotes(MMPs).In addition,analysis of the 16S rRNA gene showed that these MTB were clustered into 16 operational taxonomic units affi liated to the Alpha-,Delta-,and Gamma-proteobacteria classes within the Proteobacteria phylum.Meanwhile,by using the coupled fluorescence and transmission electron microscopy analysis,rodshaped bacteria,vibrio,and cocci were phylogenetically and structurally identified at the single-cell level.This study demonstrated highly diverse MTB communities in the mangrove ecosystem and provide a new insight into the overall diversity of MTB.展开更多
