Scanning e-lectron microscopy (SEM) results show that there are a lot of fibrous materials distributing in the interface, which connects the dispersed phase with the matrix and obtains better interfacial strength for prominent mechanical properties. The addition of compatibilizers results in the decrease of crystallinity of the blends and the disappearance of an obvious yield phenomenon, which was proved by the differential scanning calorimeter (DSC) test and X-ray diffraction (XRD) characterization Although the crystallinity of the blends decreases, the tensile strength and tensile strain of the blends significantly increases, especially for the HDPE/GTR/DCP/HY-2045 blends, which is possibly attributed to the good compatibility of the blends owing to the in-situ interface crosslinking. Scavenging of ROS microenvironment to inhibit the osteoclastogenesis is highly valued in the therapeutic process of osteoarthritis. Despite the excellent ability of polyphenolic colloidal to scavenge reactive oxygen species and its affinity for macrophages, the preparation of polyphenolic colloidal nanoparticles is limited by the complex intermolecular forces between phenol molecules and the lack of understanding of polymerization/sol-gel chemistry. Herein, our work introduces a novel poly-tannin-phenylboronic colloidal nanoparticle (PTA) exclusively linked by ROS-responsive bondings. Nanocolloidal PTA has a uniform particle size, is easy and scalable to synthesize, has excellent scavenging of ROS, and can be slowly degraded. For in vitro experiments, we demonstrated that, PTA could eliminate ROS within RAW264.7 cells and impede osteoclastogenesis and bone resorption. RNA sequencing results of PTA-treated RAW264.7 cells further reveal the promotion of antioxidant activity and inhibition of osteoclastogenesis. For in vivo experiments, PTA could eliminate the ROS environment and reduce the number of osteoclasts in the subchondral bone, thereby alleviating the damage of subchondral bone and symptoms of osteoarthritis. Our research, by delving into the formation of polyphenol colloidal nanoparticles and validating their role in ROS scavenging to inhibit osteoclastogenesis in subchondral bone, may open new avenues for OA treatment in the future.展开更多

文摘Earthquake-related hydrochemical changes in thermal springs have been widely observed;however, quantitative modeling of the reactive transport process is absent. In the present study,we apply reactive transport simulation to capture the hydrochemical responses in a thermal spring following the Wenchuan Ms 8.0 and Lushan Ms 7.0 earthquakes. We first constrain deep reservoir geothermal fluid compositions and temperature by multicomponent geothermometry, and then a reactive geochemical transport model is constructed to reproduce the hydrochemical evolution process. The results show that the recharge from the shallow aquifer increases gradually until it reaches a peak because of the permeability enhancement caused by the Lushan earthquake, which may be the mechanism to explain the earthquake-related hydrochemical responses. In contrast to the postseismic effect of the Wenchuan earthquake, the chemical evolution can be considered as hydrochemical anomalies related to the Lushan earthquake. This study proves that the efficient simulation of reactive transport processes is useful for investigating earthquake-related signals in hydrochemical time series.

文摘The excessive reactive oxygen species (ROS) accumulation and overactivated osteoclastogenesis in subchondral bone has proved to be a major cause of osteoarthritis (OA). Our research, by delving into the formation of polyphenol colloidal nanoparticles and validating their role in ROS scavenging to inhibit osteoclastogenesis in subchondral bone, may open new avenues for OA treatment in the future.

文摘Acute pancreatitis(AP)is a common acute gastrointestinal disorder affecting approximately 20%of patients with systemic inflammatory responses that may cause pancreatic and peripancreatic fat necrosis. This condition often progresses to multiple organ failure, significantly increasing morbidity and mortality.Oxidative stress, characterized by an imbalance between the body’s reactive oxygen species(ROS)and antioxidants, activates the inflammatory signaling pathways.Although the pathogenesis of AP is not fully understood,ROS are increasingly recognized as critical in the disease's progression and development.Modulating the oxidative stress pathway has shown efficacy in mitigating the progression of AP.Despite numerous basic studies examining this pathway, comprehensive reviews of recent research remain sparse. This systematic review offers an in-depth examination of the critical role of oxidative stress in the pathogenesis and progression of AP and evaluates the therapeutic potential of antioxidant interventions in its management.

文摘Plasma-activated water(PAW),as an extended form of cold atmospheric-pressure plasma, greatly expands the application of plasma-based technology. The biological effects of PAW are closely related to the aqueous reactive species, which can be regulated by the activation process. In this study, surface plasma-activated air(SAA)and a He+O_(2)plasma jet(Jet)were parallelly combined(the SAA+Jet combination)or sequentially combined(the SAA→Jet combination and the Jet→SAA combination)to prepare plasma-activated saline(PAS).The PAS activated by the combinations exhibited stronger bactericidal effects than that activated by the SAA or the Jet alone. The concentrations of H_(2)O_(2)and NO_(2)^(-)were higher in the PAS activated by the Jet→SAA combination, while ONOO^(-)concentrations were close in the three kinds of PAS and^(1)O_(2)concentrations were higher in the PAS activated by the SAA+Jet combination. The analysis of scavengers also demonstrated that H_(2)O_(2),^(1)O_(2),and ONOO^(-)in the PAS activated by the SAA+Jet combination,and^(1)O_(2)in the PAS activated by the Jet→SAA combination played critical roles in bactericidal effects.Further, the effective placement time of the three PAS varied, and the PAS activated by the Jet→SAA combination could also inactivate 2.6-log_(10)of MRSA cells after placement for more than 60 min.The regulation of reactive species in plasma-activated water via different combinations of plasma devices could improve the directional application of plasma-activated water in the biomedical field.

文摘A novel and reactive oxygen species(ROS)responsive astaxanthin phenylboronic acid derivative(AstaDPBA)was constructed by grafting phenylboronic acid(PBA)onto astaxanthin succinate diester(AstaD),and its chemical structure and physicochemical property were identified.AstaD-PBA could effectively improve the ROS quenching ability in the lipopolysaccharide(LPS)-induced RAW264.7 cell inflammation model. Then, the bioactivity of AstaD-PBA was studied by 4 zebrafish ROS-responsive infl ammatory models induced by LPS,copper(Cu^(2+)),high-fat diet, and dextran sodium sulfate(DSS).The results suggest that AstaD-PBA might have high biosafety and the best effect on ulcerative colitis(UC)induced by DSS.Furtherly,AstaDPBA significantly alleviated and treated weight loss and colonic shrinkage, inhibited infl ammatory cytokines, and maintained microbiota homeostasis to improve UC in C57BL/6J mice.Alistipes and Oscillibacter were expected to be considered UC marker fl ora according to the Metastats analysis and Pearson correlation Mantel test(P<0.01)of 16S rRNA gene sequencing data.In conclusion,AstaD-PBA has been promised to be a functional compound to improve UC and maintain intestinal microbiota homeostasis.
文摘Reactive transport modeling(RTM)is an emerging method used to address geological issues in diagenesis research. However, the extrapolation of RTM results to practical reservoir prediction is not sufficiently understood. This paper presents a case study of the Eocene Qaidam Basin that combines RTM results with petrological and mineralogical evidence. The results show that the Eocene Xiaganchaigou Formation is characterized by mixed siliciclastic-carbonate-evaporite sedimentation in a semiclosed saline lacustrine environment.Periodic evaporation and salinization processes during the syngeneticpenecontemporaneous stage gave rise to the replacive genesis of dolomites and the cyclic enrichment of dolomite in the middle-upper parts of the meter-scale depositional sequences. The successive change in mineral paragenesis from terrigenous clastics to carbonates to evaporites was reconstructed using RTM simulations.Parametric uncertainty analyses further suggest that the evaporation intensity(brine salinity)and particle size of sediments(reactive surface area)were important rate-determining factors in the dolomitization,as shown by the relatively higher reaction rates under conditions of higher brine salinity and fine-grained sediments.Combining the simulation results with measured mineralogical and reservoir physical property data indicates that the preservation of original intergranular pores and the generation of porosity via replacive dolomitization were the major formation mechanisms of the distinctive lacustrine dolomite reservoirs(widespread submicron intercrystalline micropores)in the Eocene Qaidam Basin. The results confirm that RTM can be effectively used in geological studies, can provide a better general understanding of the dolomitizing fluid-rock interactions, and can shed light on the spatiotemporal evolution of mineralogy and porosity during dolomitization and the formation of lacustrine dolomite reservoirs.

文摘ZrB_(2)-based ceramics typically necessitate high temperature and pressure for sintering, whereas ZrB_(2)-SiC ceramics can be fabricated at 1500℃using the process of reactive melt infiltration with Si.In comparison to the conventional preparation method, reactive synthesis allows for the more facile production of ultra-high temperature ceramics with fine particle size and homogeneous composition. In this work,ZrSi_(2),B4C,and C were used as raw materials to prepare ZrB_(2)-SiC via combination of tape casting and reactive melt infiltration herein referred to as ZBC ceramics.Control sample of ZrB_(2)-SiC was also prepared using ZrB_(2) and SiC as raw materials through an identical process designated as ZS ceramics.Microscopic analysis of both ceramic groups revealed smaller and more uniformly distributed particles of the ZrB_(2) phase in ZBC ceramics compared to the larger particles in ZS ceramics. Both sets of ceramics underwent cyclic oxidation testing in the air at 1600℃for a cumulative duration of 5 cycles, each cycle lasting 2 h.Analysis of the oxidation behavior showed that both ZBC ceramics and ZS ceramics developed a glassy SiO_(2)-ZrO_(2) oxide layer on their surfaces during the oxidation. This layer severed as a barrier against oxygen. In ZBC ceramics,ZrO_(2) is finely distributed in SiO_(2),whereas in ZS ceramics, larger ZrO_(2) particles coexist with glassy SiO_(2).The surface oxide layer of ZBC ceramics maintains a dense structure because the well-dispersed ZrO_(2) increases the viscosity of glassy SiO_(2),preventing its crystallization during the cooling.Conversely, some SiO_(2) in the oxide layer of ZS ceramics may crystallize and form a eutectic with ZrO_(2),leading to the formation of ZrSiO_(4).This leads to cracking of the oxide layer due to differences in thermal expansion coefficients, weakening its barrier effect. An analysis of the oxidation resistance shows that ZBC ceramics exhibit less increase in oxide layer thickness and mass compared to ZS ceramics, suggesting superior oxidation resistance of ZBC ceramics.

文摘Conventional blood sampling for glucose detection is prone to cause pain and fails to continuously record glucose fluctuations in vivo.Continuous glucose monitoring based on implantable electrodes could induce pain and potential tissue inflammation, and the presence of reactive oxygen species(ROS)due to inflammationmay affect glucose detection.Microneedle technology is less invasive, yet microneedle adhesion with skin tissue is limited. In this work,we developed a microarrow sensor array(MASA),which provided enhanced skin surface adhesion and enabled simultaneous detection of glucose and H_(2)O_(2)(representative of ROS)in interstitial fluid in vivo. The microarrows fabricated via laser micromachining were modified with functional coating and integrated into a patch of a three-dimensional(3D)microneedle array.Due to the arrow tip mechanically interlocking with the tissue, the microarrow array could better adhere to the skin surface after penetration into skin. The MASA was demonstrated to provide continuous in vivo monitoring of glucose and H_(2)O_(2) concentrations, with the detection of H_(2)O_(2) providing a valuable reference for assessing the inflammation state. Finally, the MASA was integrated into a monitoring system using custom circuitry. This work provides a promising tool for the stable and reliable monitoring of blood glucose in diabetic patients.

文摘Drought stress is a devastating natural disaster driven by the continuing intensification of global warming, which seriously threatens the productivity and quality of several horticultural crops, including pear.Gibberellins(GAs)play crucial roles in plant growth, development, and responses to drought stress.Previous studies have shown significant reductions of GA levels in plants under drought stress;however, our understanding of the intrinsic regulation mechanisms of GA-mediated drought stress in pear remains very limited. Here,we show that drought stress can impair the accumulation of bioactive GAs(BGAs),and subsequently identified PbrGA2ox1 as a chloroplast-localized GA deactivation gene. This gene was significantly induced by drought stress and abscisic acid(ABA)treatment, but was suppressed by GA_(3)treatment.PbrGA2ox1-overexpressing transgenic tobacco plants(Nicotiana benthamiana)exhibited enhanced tolerance to dehydration and drought stresses, whereas knock-down of PbrGA2ox1 in pear(Pyrus betulaefolia)by virus-induced gene silencing led to elevated drought sensitivity.Transgenic plants were hypersensitive to ABA,and had a lower BGAs content, enhanced reactive oxygen species(ROS)scavenging ability, and augmented ABA accumulation and signaling under drought stress compared to wild-type plants. However, the opposite effects were observed with PbrGA2ox1 silencing in pear. Moreover, exogenous GA_(3)treatment aggravated the ROS toxic effect and restrained ABA synthesis and signaling, resulting in the compromised drought tolerance of pear.In summary, our results shed light on the mechanism by which BGAs are eliminated in pear leaves under drought stress, providing further insights into the mechanism regulating the effects of GA on the drought tolerance of plants.
