The genetic diversity and relationships of seven Chinese indigenous pig breeds (Meishan, Erhualian, Hezuo, Bamei, Qingping, Tongcheng, and Huainan) and three exotic pig breeds (Large White, Landrace, and Duroc) were analyzed using the DNA differential display technique by means of eight primer combinations. A total of 123 reproducible bands were used to calculate mean Nei's gene diversity, and mean Shannon's information index for each pig population. Based on these the Nei's standard genetic identity and distance were estimated, which was used to construct a dendrogram tree for the 10 pig breeds. The experimental results obtained and the method used in this study for evaluating the genetic diversity and relationships of pigs were also discussed. 展开更多

Nine Chinese yak breeds (Maiwa, Tianzhu White, Qinghai Plateau, Sibu, Zhongdian, Pali, Tibetan High Mountain, Jiulong, and Xinjiang) and Gayal were analyzed by means of 16 microsatellite markers to determine the level of genetic variation within populations, genetic relationship between populations, and population structure for each breed. A total of 206 microsatellite alleles were observed. Mean F-statistics (0.056) for 9 yak breeds indicated that 94.4% of the genetic variation was observed within yak breeds and 5.6% of the genetic variation existed amongst breeds. The Neighbor-Joining phylogenetic tree was constructed based on Nei's standard genetic distances and two clusters were obtained. The Gayal separated from the yaks far away and formed one cluster and 9 yak breeds were grouped together. The analysis of population structure for 9 yak breeds and the Gayal showed that they resulted in four clusters; one cluster includes yaks from Tibet Autonomous Region and Qinghai Province, one cluster combines Zhongdian, Maiwa, and Tianzhu White, and Jiulong and Xinjiang come into the third cluster. Pali was mainly in the first cluster (90%), Jiulong was mainly in the second cluster (87.1%), Zhongdian was primarily in the third cluster (83%), and the other yak breeds were distributed in two to three clusters. A total of seventeen caprine microsatellite markers were used on 169 goats to investigate genetic diversity of eight Cameroon native goat populations and to assess genetic differentiation with the East African small goat. All microsatellites showed a high polymorphic content (PIC) of more than 0.5 in almost all ecotypes. These microsatellite markers were useful for native goat ecotypes variability assessment. Expected heterozygosity of all ecotypes was above 0.5, varying from 0.2 to 0.7. Only goats from Eastern rainforest ecotype deviated from Hardy-Weinberg equilibrium test (P 〉 0.001). Although geographic distribution was a good indication of differentiation, there appeared a tendency of genetic exchange between various ecotypes in Cameroon native goats. 展开更多

[Method] From 27 pairs of porcine microsatellite primers recommended by Food and Agriculture Organization of the United Nations (FAO) and international Society for Animal Genetics (ISAG), six pairs (S0155, SW240, IGF1, SW951, SW857, SW24) were selected for microsatellite DNA detection of three Guizhou local pig breeds, including Nuogu Pig, Kele Pig and Guanling Pig. Subsequently, their genetic diversities were analyzed. [ Result] The three pig breeds were high polymorphic at the six microsatellite loci (PIC 〉0.5). The Nei's standard genetic distance of them was 0.206 3 -0.481 5. The genetic distance between Nuogu Pig and Kele Pig Was the closest, and that between Nuogu Pig and Guanling Pig was the furthest. China is rich in chicken genetic resources,; many indigenous breeds can be found throughout the country. China is rich in chicken genetic resources,; many indigenous breeds can be found throughout the country. Due to poor productive ability, some of them are threatened by the commercial varieties from domestic; foreign breeding companies. In a large-scale investigation into the current status of Chinese poultry genetic resources, 78 indigenous chicken breeds were surveyed; their blood samples collected. The genomes of these chickens were screened using microsatellite analysis. A total of 2740 individuals were genotyped for 27 microsatellite markers on 13 chromosomes. The number of alleles of the 27 markers ranged from 6 to 51 per locus with a mean of 18.74. Heterozygosity (H) values of the 78 chicken breeds were all more than 0.5. The average H value (0.622); polymorphism information content (PIC, 0.573) of these breeds suggested that the Chinese indigenous chickens possessed more genetic diversity than that reported in many other countries. The fixation coefficients of subpopulations within the total population (F ST) for the 27 loci varied from 0.065 (LEI0166) to 0.209 (MCW0078), with a mean of 0.106. For all detected microsatellite loci, only one (LEI0194) deviated from Hardy-Weinberg equilibrium (HWE) across all the populations. As genetic drift or non-random mating can occur in small populations, breeds kept on conservation farms such as Langshan chicken generally had lower H values, while those kept on large populations within conservation regions possessed higher polymorphisms. The high genetic diversity in Chinese indigenous breeds is in agreement with great phenotypic variation of these breeds. Using Nei’s genetic distance; the Neighbor-Joining method, the indigenous Chinese chickens were classified into six categories that were generally consistent with their geographic distributions. The molecular information of genetic diversity will play an important role in conservation, supervision,; utilization of the chicken resources. 展开更多

To assess the genetic diversity between randomly and selectively bred populations,we sequenced 438 bp of the mitochondrial DNA control region from 102 pigs.These samples represented four native pig breeds,one nucleus and one conservation herd from Yunnan,China.Twenty haplotypes with sixteen polymorphic sites were identified.The number of haplotypes in the nucleus herd of Saba pig and the conservation herd of Banna miniature pig were restricted to three and one,respectively,while the randomly bred pig populations exhibited over six haplotypes.Notably,haplotype diversity in randomly bred populations was significantly greater than the selectively bred populations(h=0.732 vs.0.425 and 0,exact test,P≤0.0036).These findings demonstrate that selective breeding generated low genetic diversity compared to randomly bred pig breeds.A timely intervention and well programmed breeding approach would stop further genetic diversity reduction in the nucleus and conservation herds of native pig breeds.Otherwise,selective breeding would dramatically reduce genetic diversity in only several years,indicating that sharp contradictions exist between breeding,conservation and genetic diversity.Genetic relationships are discussed based on net genetic distances among pig populations. 展开更多

Seventeen microsatellite loci were used to assess the genetic variation and population structure of six endangered duck populations in China. PIC, heterozygosity with two estimators of genetic differentiation (FST and GST), and Nei's standard genetic distance were evaluated. The results showed that these six endangered duck populations showed high polymorphism. The propor- tion of inter-population subdivision among the six duck populations ranged between 17.0 and 14.7%. The average heterozygosity was 0.706, 0.702, 0.691, 0.676, 0.604, and 0.660 in the Gaoyou, Liancheng, Jinding, Beijing, Shaoxing, and Jianchang ducks, respectively. The average PIC ranged from 0.561 to 0.663. Finally, the results showed higher genetic diversity and difference above the levels of genetic variation among all the populations. 展开更多

长牡蛎(Crassostrea gigas)野生与选育群体的微卫星遗传多样性分析

为了评估长牡蛎(Crassostrea gigas)2个壳长性状(掌心形)快速生长选育群体(LY2-K4、LY2-K7)、1个壳高性状(速生型)快速生长选育群体(LY2-K11)和6个野生群体(QHD、LS、HD、ZH、WD、KTD)的遗传多样性和遗传结构,用21对多态性丰富的微卫星... 为了评估长牡蛎(Crassostrea gigas)2个壳长性状(掌心形)快速生长选育群体(LY2-K4、LY2-K7)、1个壳高性状(速生型)快速生长选育群体(LY2-K11)和6个野生群体(QHD、LS、HD、ZH、WD、KTD)的遗传多样性和遗传结构,用21对多态性丰富的微卫星引物对9个长牡蛎群体的269个个体进行了遗传分析。结果显示:21个微卫星位点共检测出了460个等位基因(Na),平均等位基因数为21.905;21个微卫星位点的多态信息含量(PIC)均大于0.5,具有高度遗传多态性;选育群体LY2-K11的遗传多样性最低(Na=13,I=2.128,He=0.831,PIC=0.825),野生群体KTD的遗传多样性最高(Na=29,I=3.112,He=0.941,PIC=0.938);189个群体位点组合有66%偏离哈代-温伯格平衡,表明这些群体存在一定程度的杂合子缺失;9个群体间的遗传分化指数(F_(st))为0.012~0.064,处于较低的遗传分化水平;AMOVA分析显示遗传变异主要来自于个体内;PCoA分析结果与UPGMA聚类树一致,LY2-K11群体单独聚为一类,QHD和HD群体聚为一类,其他6个群体聚为一类。综上所述,长牡蛎3个选育群体和6个野生群体遗传多样性均较高,遗传分化水平较低;选育群体LY2-K11多样性略有下降,选育过程中应保证亲本的数量及质量,防止因近交衰退造成遗传多样性降低,苗种抗逆性变差。该结果将为长牡蛎新品种的选育和野生种质资源的保护提供科学指导。 展开更多

The aim of our present study was to construct genetic structure and relationships among Chinese fine-wool sheep breeds. 46 individuals from 25 breeds or strains were genotyped based on the Illumina Ovine 50K SNP array. Meanwhile, genetic variations among 482 individuals from 9 populations were genotyped with 10 microsatellites. In this study, we found high genetic polymorphisms for the microsatellites, while 7 loci in the Chinese superfine Merino strain (Xinjiang types) (CMS) and 5 loci in Gansu alpine superfine-wool sheep strain (GSS) groups were found deviated from Hardy-Weinberg equilibrium (HWE). Genetic drift FsT=0.019 (P〈0.001) and high gene flows were detected in all the 7 fine-wool sheep populations. Phylogenetic analysis showed fine-wool sheep populations were clustered in a group independent from the Chinese indigenous breeds such that the 7 fine-wool sheep clustered distinct from Liangshan semifine-wool sheep (LS) and Hu sheep (HY) reflected by different population differentiation analyses. Overall, our findings suggested that all fine-wool sheep populations have close genetic relationship, which is consistent with their breeding progress. These populations, therefore, can be regarded as open-breeding populations with high levels of gene flows. Furthermore, the two superfine-wool strains, viz., CMS and GSS, might be formed by strong artificial selection and with frequent introduction of Australian Merino. Our results can assist in breeding of superfine-wool sheep and provide guidance for the cultivation of new fine-wool sheep breeds with different breeding objectives. 展开更多

为明确不同选育群体中间球海胆的遗传多样性和遗传结构,利用SSR-seq技术和15个微卫星位点,对1个家系选育群体(FP)、1个群体选育群体(IP)和1个未经选育的普通养殖群体(CP)的遗传多样性及遗传结构进行了分析。结果显示,15个微卫星位点共... 为明确不同选育群体中间球海胆的遗传多样性和遗传结构,利用SSR-seq技术和15个微卫星位点,对1个家系选育群体(FP)、1个群体选育群体(IP)和1个未经选育的普通养殖群体(CP)的遗传多样性及遗传结构进行了分析。结果显示,15个微卫星位点共检测出112个等位基因,FP、IP、CP 3个群体的平均观测等位基因数(N_(a))分别为5.077、5.133和6.133个,平均有效等位基因(N_(e))分别为2.816、2.873和3.638个,平均观测杂合度(H_(o))分别为0.522、0.441和0.501,平均期望杂合度(H_(e))分别为0.595、0.599和0.667,平均多态性信息含量(PIC)分别为0.546、0.543和0.623。家系选育群体(FP)H_(e)与H_(o)的差值(0.073)低于IP(0.158)和CP(0.166),平均固定指数(F)(0.115)低于IP(0.248)和CP(0.246)。3个群体间遗传分化系数(F_(st))介于0.018~0.176,为中低等程度的遗传分化。分子方差分析(AMOVA)结果显示,3个群体的遗传变异主要源于个体间。主成分分析(PCoA)和聚类进化树结果均显示,3个群体之间的亲缘关系较近,其中IP群体遗传分化程度最高。研究表明,3个中间球海胆群体均具有较高的遗传多样性,多代家系选育和群体选育均未明显降低群体的遗传多样性,家系选育更有利于保持群体的杂合度和控制群体的近交水平,群体选育则会提升群体的遗传分化程度。 展开更多

【目的】基于全基因组重测序研究分析广西地方猪种与西方商业猪种的遗传结构,挖掘出广西地方猪种的选择基因,为我国地方猪种质资源的保存和利用打下基础。【方法】对广西地方猪种(巴马香猪、东山猪和陆川猪)和西方商业猪种(杜洛克猪、... 【目的】基于全基因组重测序研究分析广西地方猪种与西方商业猪种的遗传结构,挖掘出广西地方猪种的选择基因,为我国地方猪种质资源的保存和利用打下基础。【方法】对广西地方猪种(巴马香猪、东山猪和陆川猪)和西方商业猪种(杜洛克猪、长白猪和大白猪)共60头猪个体进行基因组重测序分析,经质控后分别进行猪种亲缘关系、主成分层次、连锁不平衡衰减、群体遗传结构、系统发育进化树及群体遗传分化指数(Fst)等分析。【结果】西方商业猪种与广西地方猪种间的群体遗传关系较远,西方商业猪种群体聚集在一起,而广西地方猪种群体较分散;构建的系统发育进化树显示,6个猪种聚类成3个分支,其中,东山猪单独聚为一支,巴马香猪和陆川猪聚为一支,而杜洛克猪、大白猪和长白猪聚类在另一分支上。广西地方猪种组均被选择的位点共有641个受选择区域,最终定位注释到108个基因,其中Cenpp基因的受选择区域高达25个。这些基因显著注释到自然杀伤细胞分化正向调控、翻译抑制因子活性、mRNA调控元件结合、肽激素加工、正调控细胞迁移参与新生血管生成、电压门控离子通道活性及离子跨膜转运调控等GO功能条目;KEGG信号通路富集分析发现显著富集于内吞作用、cGMP-PKG信号、胆汁分泌、肌醇磷酸盐代谢和磷脂酰肌醇信号系统等通路上。【结论】广西地方猪种与西方商业猪种间存在明显差异,但广西地方猪种内部存在较高的近交现象,部分猪种可能受到西方商业猪种的影响,而导致其血统不纯。广西地方猪种共有641个受选择信号区域,定位注释到的108个基因主要参与离子跨膜转运、磷脂酰肌醇信号系统、正调控细胞迁移及新生血管生成等通路,其中Mrps27基因可能与广西地方猪种独特的生长发育有关。 展开更多

微卫星是近几年来应用较多的一种分子标记 ,可有效地进行基因鉴定与系谱分析 ,并可估算群体间的遗传距离。通过选用 5个微卫星标记 ,检测了山东省 5个地方鸡种 :日照麻鸡、寿光鸡、莱芜黑鸡、济宁百日鸡、鲁西斗鸡以及一个外来鸡种——... 微卫星是近几年来应用较多的一种分子标记 ,可有效地进行基因鉴定与系谱分析 ,并可估算群体间的遗传距离。通过选用 5个微卫星标记 ,检测了山东省 5个地方鸡种 :日照麻鸡、寿光鸡、莱芜黑鸡、济宁百日鸡、鲁西斗鸡以及一个外来鸡种———安卡黄鸡和一个外省地方鸡种———广西黄鸡共 7个鸡种的遗传多样性。根据测试结果计算了每个等位基因的频率 ,以基因频率为基础分析了品种内的遗传变异和品种间的DA 遗传距离 ,并讨论了微卫星多态性在应用于群体遗传变异及亲缘关系等方面的意义。结果表明 :共检测到 4 0个等位基因 ,其中等位基因数最多的位点为ADL0 136 (10个 ) ;等位基因数最低的位点为ADL0 14 6 (5个 ) ;而且每个位点的等位基因分布并不均匀 ,都有一种或几种优势基因存在。在 7个品种中 ,杂合度最低的为寿光鸡 ,杂合度值为 0 332 7,因为此鸡种多年来一直由寿光市慈伦种鸡场进行纯繁保种 ,未与其他鸡种杂交 ,因此杂合度最小 ;其他鸡种杂合度也都低于 0 4 ,据分析可能是由于日照麻鸡、济宁百日鸡群体较小 ;莱芜黑鸡是正在选育的一个品种 ,个体间遗传关系也不远 ;安卡黄鸡和广西黄鸡自从引入嘉明公司后 ,群体近交现象普遍 ,因此各鸡种杂合度都偏低。由此可见 ,通过对杂合度的计算 ,微卫星可以较好地? 展开更多
