Cytological Mechanism of Cytoplasmic Inheritance in Pinus tabulaeformis: Ⅱ. Transmission of Male and Female Organelles During Fertilization and Proembryo Development

In an earlier report the ultrastructure and nucleoid organelles of male gamete in Pinus tabulaeformis Carr. have been described. Presently, the ultrastructure of the cytoplasm of the egg cell and pollen tube—immediately before fertilization and during cytoplasmic transmission of male gametophyte—has been described for the same species. The fate of parental plastids and mitochondria in the proembryo has also been followed. The mature egg cell contains a large amount of mitochondria, but seems to lack normal plastids. Most plastids have transformed into large inclusions. Apart from the large inclusions, there are abundant small inclusions and other organelles in the egg cell. During fertilization, pollen tube penetrates into the egg cell at the micropylar end and thereafter the contents are released. Plastid and mitochondrion of male origin are lacking near the fusing sperm_egg nuclei. The second sperm nucleus—not involved in karyogamy—remains at a site near the receptive vacuole. This nucleus is surrounded by large amount of male cytoplasm containing mixed organelles from the sperm cell, tube cell, and egg cell. At the free nuclear proembryo stage, organelles of male and female origin are visible in the perinucleus_cytoplasmic zone. Most of the mitochondria have the same morphological features as those in the egg cell. Some of the mitochondria appear to have originated from the sperm and tube cells. Plastids are most likely of male gametophyte origin because they have similar appearance as those of the sperm and tube cell. Large inclusions in the egg cell become vacuole_like. Paternal plastids have been incorporated into the neocytoplasm of the proembryo. In the cellular proembryo, maternal mitochondria are more abundant. Plastids resembling those of the sperm and tube cell are still present. These cytological results clearly show that in P. tabulaeformis, plastids are inherited paternally and mitochondria bipaternally. The cytological mechanism of plastid and mitochondrion inheritance in gymnosperm is discussed.

Xianning City is a main tea producing area in Hubei Province,the capital of"border-selling tea"in China,and an important starting point for the Ten-Thousand-Mile Tea Road across the Eurasian continent.This paper studies the tea industry in Xianning from the aspects of top 100 counties of tea industry,important agricultural cultural heritage,famous Chinese historical and cultural villages,China’s time-honored brands,major brands,and designated production enterprises of border-selling tea.Besides,it introduces intellectual property resources of the tea industry from traditional knowledge,biological genetic resources,new plant varieties,patents,geographical indications,trademarks,etc.Then,it analyzes major problems in the inheritance,innovation and high-quality development of Xianning tea,especially the border-selling tea.Finally,it comes up with constructive recommendations of starting the cultural inheritance and development project and upgrade project of the tea industry to build the capital of China’s border-selling tea.

Inheritance,Innovation and Development of Wuling Mountain Area in Hubei Province Based on Intellectual Property Rights

Using the methods of field survey and literature collection,this paper firstly introduces the intellectual property resources of the Wuling Mountain Area in Hubei Province from folk literature and arts,traditional knowledge,biological genetic resources,new plant varieties,patents,geographical indications,and trademarks.Then,it analyzes the main problems in the protection and inheritance,innovation and development of intellectual property rights(IPRs).In addition,it discusses the strategy of inheritance,innovation and high-quality development of this ethnic area in the context of the strategy of strengthening the country with intellectual property rights and the strategy of rural revitalization.Finally,it comes up with recommendations including carrying forward and inheriting folk literature and art,protecting traditional knowledge of traditional craftsmanship,inheriting and innovating to develop traditional Chinese medicine and ethnic medicine,improving the protection and use of geographical indications,and strengthening the IPR protection.

The discovery of highly effective medicine"Qingfei Paidu Decoction"for COVID-19 is a new era gift of traditional Chinese medicine to the world.The traditional Chinese medicine Scutellaria baicalensis Georgi,as a medicinal plant in the family of Labiatae,is the dominant variety of genuine medicinal material in North China.It is the leading traditional Chinese medicine in the advantageous area of agricultural products with Chinese characteristics in Shangdang of Changzhi City,and it is one of the 21 herbal medicines in the new national prescription"Qingfei Paidu Decoction"for the rapid and effective treatment of COVID-19.This paper introduces S.baicalensis Georgi and its traditional Chinese medicine GAP base,ecologically suitable planting county and medicinal value in poor areas,and studies the intellectual property resources related to S.baicalensis Georgi,such as traditional knowledge,biological genetic resources,patents,new plant varieties,geographical indications,and trademarks.This paper mainly discusses the inheritance essence,innovation and high-quality development strategy of S.baicalensis Georgi under the rural revitalization strategy and traditional Chinese medicine development strategy from the aspects of intellectual property rights,important agricultural cultural heritage,advantageous areas of characteristic agricultural products and so on.

Inheritance and Sustainable Development of Concession Gardens of Tianjin City,China

Through reviewing development history of concession gardens in Tianjin City,features,advantages and problems of these gardens were analyzed.It was stressed that cultural context should be inherited,and features of the city highlighted.The gardens should be protected from the perspective of development,combined with urban green space system,integrated with cultural life of local citizens,and coordinated with contemporary architectural style of the city.In addition,plant landscapes should be enriched,and infrastructure of the gardens supplemented to properly handle the relationship between urban construction and protection of historical image,and provide reference for inheriting cultural context of the city in the urban development.

Inheritance and Sustainable Development: Countermeasures for the Development of Tianjin Concession Parks

Through the overview of the development history of Tianjin Concession Parks, this paper analyzed the historical significance of Tianjin Concession Parks and put forward its existing problems, then summed up a series of strategies of sustainable development as follows: adhering to the Florence Charter; retaining the original form and texture of the Concession Parks; adopting a dynamic philosophy of protection and preservation; integrating with citizens' social life; supplementing and improving infrastructure facilities; and achieving environmental regeneration by planting. All these efforts are expected to contribute to making a balance between the development and construction of urban cities and the preservation of historical features, as well as provide lessons for inheriting urban history and culture in the future.

Development of organelle single nucleotide polymorphism (SNP) markers and their application for the identification of cytoplasmic inheritance patterns in Pyropia yezoensis (Bangiales,Rhodophyta)

The genus Pyropia contains several important cultivated species.Genetic research in nori species has mainly focused on the cell nucleus,with few studies on organelles(chloroplast and mitochondria).Due to the high copy numbers of organelles in cells,which influence the development and traits of algae,it is necessary to study their genetic mechanism.In this study,the marine red alga Pyropia yezoensis,an important economic macroalga,was selected as the study object.To investigate organelle(chloroplast and mitochondria)inheritance in P.yezoensis,the wild type RZ(maternal strain)was crossed with the red mutant HT(paternal strain)and 30 color-sectors from 11 F1 gametophytic blades were examined.The complete chloroplast and mitochondrial genomes of the red mutant(HT)were assembled for the first time.One reliable and stable single nucleotide polymorphism(SNP)loci filtrated by bioinformatics analysis was used as a molecular marker for chloroplast and mitochondrial DNA,respectively,in subsequent experiments.PCR amplification and sequence analysis showed that the haplotypes of color-sectors detected were consistent with those of the maternal parent,confirming that both chloroplast and mitochondrial genomes were inherited maternally in P.yezoensis.The inheritance pattern of organelles in P.yezoensis can be used to guide the hybridization and breeding of nori.Additionally,the organelle SNP markers developed in this study can be applied in subsequent genetic research.

Inheritance of Powdery Mildew Resistance in Cucumber (Cucumis sativus L.) and Development of an AFLP Marker for Resistance Detection

Cucumber powdery mildew is one of the most destructive diseases of cucumber throughout the world. In the present study, inheritance of powdery mildew resistance in three crosses, and linkage of resistance with amplified fragment length polymorphism （AFLP） markers are studied to formulate efficient strategies for breeding cultivars resistant to powdery mildew. The joint analysis of multiple generations and AFLP technique has been applied in this study. The best model is the one with two major genes, additive, dominant, and epistatic effects, plus polygenes with additive, dominant, and epistatic effects （E-l-0 model）. The heritabilities of the major genes varied from 64.26% to 97.82%, and susceptibility was incompletely dominant for the two major genes in the three crosses studied. The additive effects of the two major genes and the dominant effect of the second major gene were high, and the epistatic effect of the additive-dominant between the two major genes was the highest in cross I . In cross II, the absolute value of the additive effect, dominant effect, and potential ratio of the first major gene were far higher than those of the second major gene, and the epistatic effect of the additive-additive was the highest. The genetic parameters of the two major genes in cross III were similar to those in cross II. Correlation and regression analyses showed that marker E25/M63-103 was linked to a susceptible gene controlling powdery mildew resistance. The marker could account for 19.98% of the phenotypic variation. When the marker was tested on a diverse set of 29 cucumber lines, the correlation between phenotype and genotype was not significant, which suggested cultivar specialty of gene expression or different methods of resistance to powdery mildew. The target DNA fragment was 103 bp in length, and only a small part was found to be homologous to DNA in the other species evaluated, which indicated that it was unique to the cucumber genome.
