目的 建立一种快速、简便、灵敏的蛋白质含量测定的新方法。方法 含有巯基或二硫基的蛋白质与 0 5mol·L- 1 氢氧化钠、1 5× 10 - 4mol·L- 1 Pb2 + 和 0 0 2 %四丁基碘化铵的混合溶液在沸水浴中反应 5min ,采用单扫... 目的 建立一种快速、简便、灵敏的蛋白质含量测定的新方法。方法 含有巯基或二硫基的蛋白质与 0 5mol·L- 1 氢氧化钠、1 5× 10 - 4mol·L- 1 Pb2 + 和 0 0 2 %四丁基碘化铵的混合溶液在沸水浴中反应 5min ,采用单扫描极谱法 ,测定 - 0 6 6V(vsSCE)处一价导数波高。结果 牛血清白蛋白 (BSA)、人血清白蛋白 (HSA)的浓度在 7 5×10 - 1 0 ~ 3 0× 10 - 7mol·L- 1 与其波高呈线性关系 ,BSA和HSA检测限均为 3 0× 10 - 1 0 mol·L- 1 ;溶菌酶 (Lyso)的浓度在1 4× 10 - 8~ 1 3× 10 - 6 mol·L- 1 与其波高呈线性关系 ,Lyso的检测限为 7 0× 10 - 9mol·L- 1 。结论 该方法取样量少、体系简单、灵敏度高 。 展开更多

目的探讨UVB(ultraviolet B)暴露对子代大鼠血清白细胞介素1(IL-1)、白细胞介素2(IL-2)水平及溶菌酶含量的影响。方法选取健康刚出生(子代)大鼠20只,随机分为对照组和实验组,每组10只。称取体质量并记录,在310 nm波长紫外灯下,每天对实... 目的探讨UVB(ultraviolet B)暴露对子代大鼠血清白细胞介素1(IL-1)、白细胞介素2(IL-2)水平及溶菌酶含量的影响。方法选取健康刚出生(子代)大鼠20只,随机分为对照组和实验组,每组10只。称取体质量并记录,在310 nm波长紫外灯下,每天对实验组子代大鼠进行2 h的暴露,持续暴露8周(到性成熟期),每周一称取体质量并记录;8周后解剖各组大鼠,对子代大鼠血清中IL-1,IL-2水平及溶菌酶的含量进行测定。结果经UVB暴露后,试验组子代大鼠体质量增长量及血清中的溶菌酶含量与对照组比较无统计学差异(P>0.05);试验组子代大鼠血清中的IL-1,IL-2水平与对照组比较有统计学差异(P<0.05),试验组子代大鼠与对照组比较无统计学差异(P>0.05)。结论UVB暴露可降低子代大鼠血清中的IL-1,IL-2水平。 展开更多

Predicting value of serum soluble ST2 and interleukin-33 for risk stratification and prognosis in patients with acute myocardial infarction 被引量:28

Background Acute myocardial infarction (AMI) is a common cardiac emergency with high mortality.Serum soluble ST2 (sST2) is a new emerging biomarker of cardiac diseases.The present study is to investigate the predictive value of sST2 and interleukin-33 (IL-33) for risk stratification and prognosis in patients with AMI.Methods Fifty-nine patients with AMI,whose chief complaint was chest pain or dyspnea,were selected for our study.Physical examination,chest radiograph,electrocardiograph (ECG),biomarkers of myocardial infarction,NT-proBNP,echocardiography and other relevant examinations were performed to confirm the diagnosis of AMI.Thirty-six healthy people were chosen as the control group.Serum samples from these subjects (patients within 24 hours after acute attack) were collected and the levels of sST2 and IL-33 were assayed by enzyme-linked immuno-sorbent assay (ELISA) kit.The follow-up was performed on the 7th day,28th day,3rd month and 6th month after acute attack.According to the follow-up results we defined the end of observation as recurrence of AMI or any causes of death.Results Median sST2 level of the control group was 9.38ng/ml and that of AMI patients was 29.06ng/ml.Compared with the control group,sST2 expression in the AMI group was significantly different (P〈0.001).In contrast,the IL-33 level showed no significant difference between the two groups.Serum sST2 was a predictive factor independent of other variables and may provide complementary information to NT-proBNP or GRACE risk score.IL-33 had no relationship to recurrence of AMI.Both sST2 and the IL-33/sST2 ratio were correlated with the 6-month prognosis; areas under the ROC curve were 0.938 and 0.920 respectively.Conclusions Early in the course (〈24 hours) of AMI,sST2 usually increases markedly.The increase of sST2 has an independent predictive value for the prognosis in AMI patients and provides complementary information to NT-proBNP or GRACE risk score.The IL-33/sST2 ratio correlates with the 6-month prognosis of AMI patients.However,there is no significant relationship between IL-33 and the prognosis of AMI patients. 展开更多

Extracellular supplement of Ca and Si accelerates osteogenisis and angiogenisis during bone repair.CaSi-based ceramics are biodegradable and can release Ca and Si ions efficiently.However,locally high alkalinity induced by excessively rapid degradation limits their application.In this paper,NaCa_(2)HSi_(3)O_(9)(NCS)nanorods were fabricated on Ti by micro-arc oxidation and hydrothermal treatment;protein layers consisted of different rations of bovine serum albumin(BSA)and lysozyme(LYS)were prepared on NCS nanorods by spinning coating and thermal stabilization.The microstructure,ion releasing and wettability of coatings were observed;osteoblast and endotheliocyte behaviors,as well as bacteria response in vitro,were evaluated.The protein layer did not change microstructure and phase composition of nanorods but acted as a barrier isolating nanorods from contacting medium,and it results in slowing down the degradation of nanorods.The reduced increase of pH value,released Ca and Si ions and incorporation of LYS in BSA protein layer gave NCS nanorods improved MC3 T3-E1 and HUVECs response,compared with Ti.Meanwhile,the protein layers except pure LYS endowed nanorods with a highly antibacterial effect against staphylococcus aureus and escherichia coli simultaneously,indicating the key role of BSA in resisting antibacterial adhesion.NCS nanorods with BSA and LYS composited protein layers should be a potential coating applied on Ti implants for increasing osseointegration. 展开更多
