目的:探讨检测血清中溶性白介素-2受体(SIL-2R)mRNA和人骨髓内皮细胞标记物-1(HBME-1)mRNA的表达在鉴别诊断甲状腺结节性质中的应用价值。方法:选取2019年6月至2021年1月期间广东省东莞市人民医院收治的136例甲状腺结节患者及同期在该... 目的:探讨检测血清中溶性白介素-2受体(SIL-2R)mRNA和人骨髓内皮细胞标记物-1(HBME-1)mRNA的表达在鉴别诊断甲状腺结节性质中的应用价值。方法:选取2019年6月至2021年1月期间广东省东莞市人民医院收治的136例甲状腺结节患者及同期在该医院接受体检的50例健康人作为研究对象。根据病理检查的结果,将136例甲状腺结节患者分为恶性组(50例)和良性组(86例)。将50例健康人作为对照组。使用qPT-PCR法检测三组研究对象血清中sIL-2R mRNA、HBME-1 mRNA的表达情况,然后使用荧光侧流免疫分析法检测136例甲状腺结节患者白细胞介素-35(IL-35)和白细胞介素-17(IL-17)的水平。比较三组研究对象血清中sIL-2R mRNA和HBME-1 mRNA的表达情况,采用Pearson法检验恶性甲状腺结节患者血清中sIL-2R mRNA、HBME-1 mRNA的表达与IL-17、IL-35等指标的相关性,采用logistics回归模型检测血清sIL-2R mRNA、HBME-1 mRNA等指标与恶性甲状腺结节患者病情严重程度的相关性。结果:恶性组患者血清中sIL-2R mRNA和HBME-1 mRNA的表达显著高于良性组患者和对照组健康人,P<0.05。良性组患者血清中sIL-2R mRNA、HBME-1 mRNA的表达显著高于对照组健康人,P<0.05。恶性甲状腺结节患者血清中sIL-2R mRNA、HBME-1 mRNA的表达与其血清中IL-17的水平、IL-35的水平、甲状腺结节病灶的个数、癌细胞发生淋巴结转移的情况和病情的分期呈正相关性。血清sIL-2R mRNA、HBME-1 mRNA呈高表达状态是导致恶性甲状腺结节患者病情加重的独立危险因素。结论:恶性甲状腺结节患者血清中HBME-1 mRNA和SIL-2R mRNA的表达可明显高于良性甲状腺结节患者和正常人,而且血清sIL-2R mRNA和HBME-1 mRNA呈高表达状态是导致恶性甲状腺结节患者病情加重的独立危险因素。因此,血清sIL-2R mRNA和HBME-1 mRNA可作为临床上鉴别诊断甲状腺结节性质的重要指标。 展开更多

目的:利用TaqMan实时荧光定量逆转录聚合酶链反应(FQ-RT-PCR)检测强直性脊柱炎患者外周血单个核细胞白细胞介素-2受体α(IL-2Rα)mRNA的表达水平,并进行疾病的活动性相关分析。方法:用 Primer Express v2.0软件,根据GenBank提供的序列,... 目的:利用TaqMan实时荧光定量逆转录聚合酶链反应(FQ-RT-PCR)检测强直性脊柱炎患者外周血单个核细胞白细胞介素-2受体α(IL-2Rα)mRNA的表达水平,并进行疾病的活动性相关分析。方法:用 Primer Express v2.0软件,根据GenBank提供的序列,在人IL-2Rα mRNA的第2和第3外显子之间设计一对引物和一条TaqMan探针。提取总RNA,加入已优化的一步法FQ-RT-PCR反应体系,扩增产物经凝胶电泳后切胶回收,并与pUCm-T载体连接。制备好的重组质粒转化感受态细胞进行克隆。经测序分析确定为特异性片段后,用T7RNA聚合酶转录为cRNA,作为FQ-RT-PCR系列浓度标准品。评价FQ-RT-PCR检测 IL-2Rα mRNA的特异性、线性、精密度和探针稳定性等。定量测定HLA-B27阳性活动组与阴性非活动组强直性脊柱炎患者外周血单个核细胞IL-2Rα mRNA基因的表达水平,结合血浆可溶性白细胞介素-2受体 (sIL-2R)浓度,探讨与炎症活动的相互关系。结果:成功构建了cRNA标准品。该方法的线性范围为7~107 cells/ml,其批内CV8.4%,批间CV9.6%;扩增产物克隆的双向测序结果经BLAST比较、分析,证实为IL- 2Rα mRNA特异性片段。对各30例HLA-B27阳性与阴性的强直性脊柱炎的测定表明:与正常人对照组结果比较,HLA-B27阳性组与HLA-B27阴性组IL-2Rα mRNA和sIL-2R都有明显增加(P<0.01)。IL-2Rα mRNA对炎症活动评价的灵敏度为96.7%。结论:FQ-RT-PCR具有灵敏、特异、结果重复性好等优点;IL- 2Rα mRNA与sIL-2R比较更能反映强直性脊柱炎患者的免疫状态,可能为免疫药物的疗效评价和药物筛选提供基因表达水平上的信息。 展开更多

为了探讨中草药复方对断奶仔兔肠道发育及肠道免疫水平的影响,试验选取30日龄、体重相近且健康状况良好的断奶伊拉兔仔兔60只,公母各半,随机分为2组,每组3个重复,每个重复10只,对照组饲喂基础日粮,试验组饲喂基础日粮+1%中草药复方,预试... 为了探讨中草药复方对断奶仔兔肠道发育及肠道免疫水平的影响,试验选取30日龄、体重相近且健康状况良好的断奶伊拉兔仔兔60只,公母各半,随机分为2组,每组3个重复,每个重复10只,对照组饲喂基础日粮,试验组饲喂基础日粮+1%中草药复方,预试期5 d,试验期30 d。试验结束后,取小肠制作石蜡切片,观察小肠组织结构;测定空肠消化酶活性及小肠白细胞介素2(interleukin-2,IL-2)mRNA表达量。结果表明:(使用中草药复方后,试验组十二指肠、空肠和回肠的绒毛高度均较对照组显著增加(P<0.05),空肠的隐窝深度显著降低(P<0.05),三段肠段的绒毛高度与隐窝深度比值均极显著升高(P<0.01)。)试验组淀粉酶、胰蛋白酶活性较对照组显著或极显著提高(P<0.05或P<0.01);糜蛋白酶活性也有所提高,但差异不显著(P>0.05)。试验组空肠和回肠IL-2 mRNA表达量较对照组显著升高(P<0.05);十二指肠IL-2 mRNA表达量有升高趋势,但差异不显著(P>0.05)。说明中草药复方能够有效促进仔兔小肠发育,并增强其肠道免疫力。 展开更多

BACKGROUND: Previous studies have indicated preventive acupuncture and moxibustion at the Guanyuan point have positive effects on the immune system of menopausal rats. OBJECTIVE: To observe the effects of preventive acupuncture or moxibustion at the Guanyuan point on interleukin-2 (IL-2) and its receptor (IL-2R mRNA) levels in hypothalamus, serum, and uterus of menopausal rats. DESIGN: Randomized control animal experiment. SETTING: School of Acupuncture and Moxibustion, Beijing University of Chinese Medicine. MATERIALS: This study was performed at the Laboratory of Beijing University of Chinese Medicine from November 2004 to July 2005. The vaginal smear method was used to determine the estrus cycle of 9.5-month-old Sprague Dawley (SD) female rats. Three continuous estrus cycles were measured, and 160 rats with estrus cycle disorder, weighing (300 _+ 20) g, were included in this study. The rats were randomly divided into four groups: preventive acupuncture group (n =48), preventive moxibustion group (n =48), model groups (n =48), and 10-month-old group (n =16). Sixteen 3.5-month-old healthy SD female rats, weighing (240 ± 20) g, were also assigned to the young group. All rats were clean grade and were purchased from the Laboratory Animal Center, Beijing University of Chinese Medicine. The experiment was in accordance with animal ethics standards. METHODS: Acupuncture was applied to the preventive acupuncture group at 10 months of age. The needle was inserted upward and perpendicularly two fen to a point that was two fen below the Guanyuan point. The rats freely moved around during needle retention (20 minutes). Acupuncture was administered twice a week and continued for eight weeks. A moxa cone (small) moxibustion was applied to the preventive moxibustion group at 10 months of age. The rats were caught gently and kept at the same position as the preventive acupuncture group. The moxa cone was ignited and held to the Guanyuan point directly. Only one moxa cone was used each time. After moxibustion, the local skin turned slightly red. Moxibustion was used twice a week and continued for eight weeks. Nothing except gently catching and lying was administered to the model group and the young group. MAIN OUTCOME MEASURES: Radioimmunoassay, in situ hybridization, and similar methods were employed to detect IL-2 in serum and the uterus, as well as IL-2R mRNA expression in the hypothalamus and uterus arcuate nuclei, of the preventive acupuncture group, preventive moxibustion group, model groups (at 12, 14, and 16 months), 10 month old group (at 10 months), and the young group (at 4 months). RESULTS: All 176 rats were included in the final analysis. (1) IL-2R mRNA: IL-2R mRNA expression decreased significantly in the uterus of 12-, 14-, and 16-month-old model groups and in the hypothalamus of 14- and 16-month-old model groups (P 〈 0.05-0.01). Compared to the same month-aged model groups, the expression of IL-2R mRNA increased significantly in the hypothalamus and uterus of 16-month-old preventive acupuncture or moxibustion groups (P 〈 0.05-0.01).(2) IL-2:IL-2 in serum of 12- and 14-month-old model groups and in the uterus of 12-, 14-, and 16-month-old model groups decreased significantly compared to the young group (P 〈 0.05-0.01). IL-2 serum levels increased significantly in the 16-month-old group with preventive moxibustion; in addition, IL-2 expression levels increased significantly in the uterus of the 12-month-old group with preventive moxibustion, as well as the 12- and 16-month-old groups with preventive acupuncture (P 〈 0.05-0.01). CONCLUSION: As a rat ages and becomes menopausal, IL-2R mRNA expression in the hypothalamus and uterus, as well as IL-2 levels in the serum and uterus, decrease. However, preventive acupuncture and preventive moxibustion can repress the decrease to some extent. This effect is even greater in the late stages of menopause (16-month-old rats). 展开更多

目的:探讨促甲状腺激素(thyroid stimulating hormone,TSH)抑制疗法对分化型甲状腺癌(differentiated thyroid carcinoma,DTC)患者术后血清甲状腺球蛋白(thyroglobulin,Tg)、血管内皮生长因子(vascular endothelial growth factor,VEGF... 目的:探讨促甲状腺激素(thyroid stimulating hormone,TSH)抑制疗法对分化型甲状腺癌(differentiated thyroid carcinoma,DTC)患者术后血清甲状腺球蛋白(thyroglobulin,Tg)、血管内皮生长因子(vascular endothelial growth factor,VEGF)、肿瘤特异性生长因子(tumors pecific growth factor,TSGF)、白细胞分化抗原44变异型6(CD44V6)、可溶性白细胞介素-2受体(soluble interleukin-2receptor,sIL-2R)及T淋巴细胞亚群水平的影响。方法:选择2014年1月~2017年1月我院收治的接受甲状腺全切手术治疗的100例DTC患者,随机分为对照组和实验组,各50例。对照组患者常规给予甲状腺素替代治疗,实验组患者给予TSH抑制疗法(口服左甲状腺素钠片,控制血清TSH水平低于0.1mU/L),两组患者均给予治疗1个月。比较两组患者治疗前后血清Tg、VEGF、TSGF、CD44V6、sIL-2R水平及外周血CD3^+、CD4^+、CD8^+水平。结果:两组治疗前的血清Tg、VEGF、TSGF、CD44V6、sIL-2R水平比较,均无显著性差异(P>0.05);两组治疗后的血清Tg、VEGF、TSGF、CD44V6、sIL-2R水平相比治疗前均较低,且实验组治疗后血清Tg、VEGF、TSGF、CD44V6、sIL-2R水平变化均显著优于对照组(P<0.05)。两组治疗前的外周血CD3^+、CD4^+、CD8^+水平比较,均无显著性差异(P>0.05);两组治疗后的外周血CD3^+、CD4^+水平相比治疗前均较高、CD8^+水平相比治疗前均较低,且实验组治疗后血外周血CD3^+、CD4^+、CD8^+水平变化均优于对照组,具有显著性差异(P<0.05)。结论:DTC行甲状腺全切手术治疗后接受TSH抑制疗法能够有效降低血清Tg、VEGF、TSGF、CD44V6、sIL-2R水平,改善细胞免疫功能,值得在临床上推广应用。 展开更多

目的 研究血管紧张素Ⅱ 2型受体 (AT2R)基因可调控表达对体外培养血管平滑肌细胞(VSMC)基质金属蛋白酶 2形成的影响 ,旨在探讨AT2R基因在VSMC上条件表达在再狭窄防治中的潜在意义。方法 本研究通过常规分子生物学方法 ,建立四环素类似... 目的 研究血管紧张素Ⅱ 2型受体 (AT2R)基因可调控表达对体外培养血管平滑肌细胞(VSMC)基质金属蛋白酶 2形成的影响 ,旨在探讨AT2R基因在VSMC上条件表达在再狭窄防治中的潜在意义。方法 本研究通过常规分子生物学方法 ,建立四环素类似物Doxycycline可调控表达AT2 R基因的双重稳定VSMC细胞系。应用RT PCR和免疫细胞化学染色技术 ,观察该双重稳定表达AT2R的VSMC细胞系中基质金属蛋白酶 2的mRNA及蛋白表达情况 ;采用血管紧张素Ⅱ (AngⅡ )及其Ⅰ型、Ⅱ型其受体拮抗剂干预上述细胞 ,观测上述指标的变化。结果 Doxycycline干预前 ,AT2R的表达处于静止状态 ,通过Doxycycline(1μg/ml)的干预 ,可以使该VSMC系稳定表达AT2R。加入Doxycycline前转染组低水平基质金属蛋白酶 2 ;AngⅡ 10 - 7mol/L干预VSMC后基质金属蛋白酶 2的mRNA及蛋白表达均显著增加 (P <0 .0 1) ;在AngⅡ干预的同时加入Doxycycline后 ,基质金属蛋白酶 2的mRNA及蛋白表达受到显著抑制 (P<0 .0 1) ;ATIR拮抗剂CV 11974可进一步下调上述mRNA及蛋白的表达 ;AT2R拮抗剂PD12 3319干预组基质金属蛋白酶 2mRNA及蛋白仍旧强表达 (与AngⅡ组比较 ,P >0 .0 5 )。 结论 AngⅡ干预VSMC后通过ATIR介导引起基质金属蛋白酶 2表达增强 ; 展开更多

Objective: To investigate the difference of peripheral blood sIL-2R before and after chemotherapy in breast cancer patients, and evaluate the clinical value of the sIL-2R in breast cancer's diagnosis and therapy. Methods: The peripheral blood sIL-2R levels of the breast cancer patients with or without chemotherapy were detected by ELISA. The healthy persons were made as the control group. Results: The sIL-2R levels of the breast cancer patients were higher than that of the control group(P(0.05); the sIL-2R's levels in I^II stage breast cancer were lower than that in Ill^IV stage breast cancer (P(0.05); the sIL-2R levels of the patients before chemotherapy were higher than that of the patients undergone chemotherapy(P(0.05); The level of the patient with chemotherapy was still higher than that of the control group (P(0.05); the sIL-2R levels of the patients whose chemotherapies were noneffective were higher than that of the patients received effective chemotherapies (P(0.05). There was no significant difference between the group with ER(+) or PR(+) and the group with ER(-) or PR(-)(P〉0.05). Conclusion: The breast cancer patients have the high sIL-2R levels. There is a close relationship between the cancer incidence and the patients, immune situation. The level of sIL-2R could be a clinical index which can be used for evaluating the cancer degree, because the higher levels of sIL-2R can indicate that the immune ability of patient is worse. There is a significant difference between the sIL-2R levels of the patients before chemotherapy and that of the patients undergone chemotherapy. 展开更多

The aim of this stu... Background: Cytokines are mediators of diseases. Expression levels in the blood could be of clinical relevance. Objective: sIL-2Rα is used as a marker for different malignancies in human medicine. The aim of this study was to show if sIL-2Rα is detectable and if there is any correlation to different diseases in dogs. Methods: For this purposes sIL-2Rα concentrations in the blood were measured in healthy dogs, in dogs with different non-neoplastic diseases and benign tumors and in dogs with malignant tumors. Serum levels of sIL-2Rα were measured by using a human specific enzyme linked immunosorbent assay (ELISA). Results: Measurement of sIL-2Rα was successful in most of the samples. Dogs with diseases have significantly increased serum levels of sIL-2Rα compared to healthy controls. sIL-2Rα serum levels are higher in patients with non-neoplastic diseases and benign tumors than in those with malignant neoplasia. There is a strong correlation between sIL-2Rα and leukocyte count. Conclusion: Measurements of sIL-2Rα in serum may be helpful in detecting stages and grades of inflammation in the progression of disease. sIL-2Rα could actually not be used as an indicator for malignant diseases in dogs like in humans. The strong correlation between sIL-2Rα and the leukocyte count indicates the inflammatory response to the disease. This could be helpful in giving a prognosis in some cases, because the inflammatory reaction is of prognostic relevance in different diseases including malignant and non-malignant neoplasia. Although the results of our research studies were very promising, further studies should be performed with a canine ELISA. 展开更多

Plasma levels of soluble interleukin-2 receptor (sIL-2R) in patients with chronic active hepatitis B (CAHB) or severe hepatitis B (SHB) were measured quantitatively by 'sandwich' ELISA with monoclonal antibodies in order to explore the change of sIL-2R levels, its clinical significance,and its relation to liver damage. The results showed that the plasma sIL-2R levels in patients with CAHB and SHB were much higher than those in normal controls (P < 0. 01 ), and the level ofplasma sIL-2R in patients with SHB was greatly higher than that in patients with CAHB. These results suggest that there is close relation between plasma level of sIL-2R, the clinical types of hepatitis B,and the severity of liver damage. In addition, there is no significant difference in plasma levels of sIL-2R between acute severe hepatitis B (ASHB), subacute severe hepatitis B (SASHB), and chronic severe hepatitis B (CSHB). Esophageal squamous cell carcinoma(ESCC),a malignancy of the digestive system,is highly prevalent and the primary cause of cancer-related deaths worldwide due to the lack of early diagnostic biomarkers and effective therapeutic targets.Dysregulated ribonucleotide reductase(RNR)expression has been confirmed to be causally linked to tumorigenesis.This study demonstrated that ribonucleotide reductase small subunit M2(RRM2)is significantly upregulated in ESCC tissue and that its expression is negatively correlated with clinical outcomes.Mechanistically,HuR promotes RRM2 mRNA stabilization by binding to the adenine/uridine(AU)-rich elements(AREs)within the 3′UTR,resulting in persistent overexpression of RRM2.Furthermore,bifonazole is identified as an inhibitor of HuR via computational screening and molecular docking analysis.Bifonazole disrupts HuR-ARE interactions by competitively binding to HuR at F65,R97,I103,and R153 residues,resulting in reduced RRM2 expression.Furthermore,bifonazole exhibited antitumor effects on ESCC patient-derived xenograft(PDX)models by decreasing RRM2 expression and the dNTP pool.In summary,this study reveals the interaction network among HuR,RRM2,and bifonazole and demonstrated that bifonazole is a potential therapeutic compound for ESCC through inhibition of the HuR/RRM2 axis. 展开更多
