Rac1 relieves neuronal injury induced by oxygen-glucose deprivation and re-oxygenation via regulation of mitochondrial biogenesis and function

Certain microRNAs(miRNAs)can function as neuroprotective factors after reperfusion/ischemia brain injury.miRNA-142-3p can participate in the occurrence and development of tumors and myocardial ischemic injury by negatively regulating the activity of Rac1,but it remains unclear whether miRNA-142-3p also participates in cerebral ischemia/reperfusion injury.In this study,a model of oxygen-glucose deprivation/re-oxygenation in primary cortical neurons was established and the neurons were transfected with miR-142-3p agomirs or miR-142-3p antagomirs.miR-142-3p expression was down-regulated in neurons when exposed to oxygen-glucose deprivation/re-oxygenation.Over-expression of miR-142-3p using its agomir remarkably promoted cell death and apoptosis induced by oxygen-glucose deprivation/re-oxygenation and improved mitochondrial biogenesis and function,including the expression of peroxisome proliferator-activated receptor-γcoactivator-1α,mitochondrial transcription factor A,and nuclear respiratory factor 1.However,the opposite effects were produced if miR-142-3p was inhibited.Luciferase reporter assays verified that Rac Family Small GTPase 1(Rac1)was a target gene of miR-142-3p.Over-expressed miR-142-3p inhibited NOX2 activity and expression of Rac1 and Rac1-GTPase(its activated form).miR-142-3p antagomirs had opposite effects after oxygen-glucose deprivation/re-oxygenation.Our results indicate that miR-142-3p down-regulates the expression and activation of Rac1,regulates mitochondrial biogenesis and function,and inhibits oxygen-glucose deprivation damage,thus exerting a neuroprotective effect.The experiments were approved by the Committee of Experimental Animal Use and Care of Central South University,China(approval No.201703346)on March 7,2017. 展开更多

A1203-SiC-C iron trough castables were prepared using high alumina bauxite clinker GL-90 ( 〉 1 mm ) or homogenized bauxite GL-88 to replace the traditional brown corundum aggregates ( 〉 1 mm ) to reduce the cost of iron trough castables. Effects of the two bauxite aggregates on properties of iron trough eastables were investigated. The results show that the two kinds of bauxite affect flowability, bulk density, strength and slag corrosion resistance of trough castables differently. Compared with homogenized bauxite, high alumina bauxite clinker is more suitable to substitute brown corundum. The castables prepared from the bauxite clinker have similar performances with those prepared from brown corundum, especially used in medium or small blast furnaces with weak thermal impact and lower temperature hot metal. However, in large blast furnaces with severe thermal impact and high temperature hot metal, the performances of the two castables are significantly different. So it is suggested to use the brown corundum based castables in the front gyral zone of the slag skimmer. 展开更多

Objective: With regard to the 2010 edition of Dietary Reference Intakes for Japanese (DRIs-2010), we investigated whether the DRIs for two age groups, breast-fed infants aged 6-8 and 9-11 months, can be fulfilled for every nutrient in actual dietary practice. Design: We evaluated (1) whether the DRIs for all nutrients can be fulfilled in a formula with energy and protein exceeding their DRIs, (2) whether the DRIs for all nutrients can be fulfilled in a formula prepared in accordance with Japanese government-recommended weaning guidelines, and (3) what kinds of formulas can be prepared if the DRIs for all nutrients are fulfilled without referring to the weaning guidelines. Setting: Simulation of diet menu on the basis of published data in our university and survey of diet menu in a university hospital attached to a national medical school. Subjects: The three types of formulas were planned for ten days. Results: It was impossible to simultaneously fulfil the DRIs for 6 - 8-month-old infants concerning pantothenic acid, vitamin D, and iron and those for 9 - 11-month-old infants concerning these nutrients plus protein. Recent studies have shown that chlorogenic acid(CGA),which is present in coffee,has protective effects on the nervous system.However,its role in neonatal hypoxic-ischemic brain injury remains unclear.In this study,we established a newborn mouse model of hypoxic-ischemic brain injury using a modified Rice-Vannucci method and performed intraperitoneal injection of CGA.We found that CGA intervention effectively reduced the volume of cerebral infarct,alleviated cerebral edema,restored brain tissue structure after injury,and promoted axon growth in injured brain tissue.Moreover,CGA pretreatment alleviated oxygen-glucose deprivation damage of primary neurons and promoted neuron survival.In addition,changes in ferroptosis-related proteins caused by hypoxic-ischemic brain injury were partially reversed by CGA.Furthermore,CGA intervention upregulated the expression of the key ferroptosis factor glutathione peroxidase 4 and its upstream glutamate/cystine antiporter related factors SLC7A11 and SLC3A2.In summary,our findings reveal that CGA alleviates hypoxic-ischemic brain injury in neonatal mice by reducing ferroptosis,providing new ideas for the treatment of neonatal hypoxic-ischemic brain injury. 展开更多

A single-crystal X-ray study shows that the iron atoms lie in a trigonally distorted octahedral environment coordinated to the hydroxy and ketone oxygen atoms of three ligands in the mer configuration Mr=773.57(C42H46N3O8Fe). The crystal is hexagonal with space group P31c; a=15.943(2), c=17.612(4)? V=3877.0(12)?, Z=4, Dc=1.325g/cm3, m=0.445mm-1, F(000)=1634, R=0.0446, wR= 0.1154 for 3085 reflections with I >2s(I). The bond lengths from iron to oxygens are 1.980(1)?for the ketone oxygens and 2.071(1)?for the hydroxy oxygens. The molecule exhibits the expected propeller shape, and the angle of the trigonal twist is 48.37. The results show that : ( 1 ) the corro- sion amolult of magnesia -chrome brick by slag without FeO is higher than that by the slag with 5% FeO, and the vorrosion amount increases when FeO content increa- ses from 5% to 15% ; (2) the test temperature is one of the important factors affecting the slag corrosion resist- ance of magnesia - chrome brick, and the corrosion of smelting reduction slag to brick increases with the temperature rising. 展开更多

Reaction of FeCIz'6H20, pyridine-2,4,6-tricarboxylic acid(H3pyta) and NaOH in the hydrothermal system at 185 ℃ resulted in the formation of 1D coordination polymer[Fe(Hpyta)(H2O)2](1) based on Fe-Hpyta chains. When the abpt[abpt=-4-amino-3,5-bis(4-pyridyl)-l,2,4-triazole] ligand was employed in the reaction system, a 3D supramolecular porous network (H2bpt)[Fe(pyta)(H2O)2]·4H2O(2) was obtained. The framework was constructed with negative (Fe-pyta) chains and the positive H3bpt^+ in situ, formed from the abpt reagent, to form 1D channels filled with guest water molecules via hydrogen-bonds. X-Ray diffraction crystal structure analysis shows that complex 1 crystallized in the monoclinic system, space group C2/c, a=1.1490(5) nm, b=0.9008(4) nm, c=1.0058(5) nm, B=107.254(9)°, V=0.9942(8) nm3, Z=4; and complex 2 crystallizes in the triclinic system, space group P1 with a=0.90392(11) nm, b=0.96027(11 ) nm, c=1.55540(18) nm, a=73.558(2)°,B= 86.126(2)°,r= 68.745(2)°, V=1.2059(2) nm^3 and Z=2. 展开更多

CDGSH iron sulfur domain 2 can inhibit ferroptosis,which has been associated with cerebral ischemia/reperfusion,in individuals with head and neck cancer.Therefore,CDGSH iron sulfur domain 2 may be implicated in cerebral ischemia/reperfusion injury.To validate this hypothesis in the present study,we established mouse models of occlusion of the middle cerebral artery and HT22 cell models of oxygen-glucose deprivation and reoxygenation to mimic cerebral ischemia/reperfusion injury in vivo and in vitro,respectively.We found remarkably decreased CDGSH iron sulfur domain 2 expression in the mouse brain tissue and HT22 cells.When we used adeno-associated virus and plasmid to up-regulate CDGSH iron sulfur domain 2 expression in the brain tissue and HT22 cell models separately,mouse neurological dysfunction was greatly improved;the cerebral infarct volume was reduced;the survival rate of HT22 cells was increased;HT22 cell injury was alleviated;the expression of ferroptosis-related glutathione peroxidase 4,cystine-glutamate antiporter,and glutathione was increased;the levels of malondialdehyde,iron ions,and the expression of transferrin receptor 1 were decreased;and the expression of nuclear-factor E2-related factor 2/heme oxygenase 1 was increased.Inhibition of CDGSH iron sulfur domain 2 upregulation via the nuclear-factor E2-related factor 2 inhibitor ML385 in oxygen-glucose deprived and reoxygenated HT22 cells blocked the neuroprotective effects of CDGSH iron sulfur domain 2 up-regulation and the activation of the nuclear-factor E2-related factor 2/heme oxygenase 1 pathway.Our data indicate that the up-regulation of CDGSH iron sulfur domain 2 can attenuate cerebral ischemia/reperfusion injury,thus providing theoretical support from the perspectives of cytology and experimental zoology for the use of this protein as a therapeutic target in patients with cerebral ischemia/reperfusion injury. 展开更多

Regulated cell death is a genetically determined form of programmed cell death that commonly occurs during the development of living organisms.This process plays a crucial role in modulating homeostasis and is evolutionarily conserved across a diverse range of living organisms.Ferroptosis is a classic regulatory mode of cell death.Extensive studies of regulatory cell death in Alzheimer’s disease have yielded increasing evidence that fe rroptosis is closely related to the occurrence,development,and prognosis of Alzheimer’s disease.This review summarizes the molecular mechanisms of ferroptosis and recent research advances in the role of ferro ptosis in Alzheimer’s disease.Our findings are expected to serve as a theoretical and experimental foundation for clinical research and targeted therapy for Alzheimer’s disease. 展开更多

目的 :探讨铁超载对铁剥夺对Ara C诱导HL 60细胞凋亡的影响。方法 :细胞培养法、台盼蓝活细胞拒染实验、细胞活力测定、光镜形态学观察、DNA琼脂糖电脉及流式细胞仪 (FCM )等方法检测HL 60细胞凋亡。观察不同浓度的三氯化铁 (FeCl3 )、... 目的 :探讨铁超载对铁剥夺对Ara C诱导HL 60细胞凋亡的影响。方法 :细胞培养法、台盼蓝活细胞拒染实验、细胞活力测定、光镜形态学观察、DNA琼脂糖电脉及流式细胞仪 (FCM )等方法检测HL 60细胞凋亡。观察不同浓度的三氯化铁 (FeCl3 )、铁剥夺剂 去铁胺 (DFO)等对HL 60细胞的作用 ,选择 10 0 μmol/LFeCl3 和 10 μmol/LDFO与Ara C共同作用于HL 60细胞 ,并以单用Ara C及生理盐水作对照。结果 :①FeCl3 (10 0 μmol/L) +Ara C(10 0 μg/ml) 6h ,12h ,2 4h ,APO %和Sub -G1%低于单用Ara C(10 0 μg/ml)组 ,两组相比有显著差异性 (P <0 0 5 )。②DFO(10 μmol/L) +Ara C(10 0 μg/ml)组APO %、DNA电泳ladder数目 ,FCM检测Sub -G1%均比单用Ara C高 (P <0 0 1)。③等摩尔的DFO与等摩尔FeCl3 共同作用于HL 60细胞 ,结果与生理盐水组相同 ;等摩尔DFO加等摩尔FeCl3 和Ara C(10 0 μg/ml)与单用Ara C(10 0 μg/ml)结果相同。 结论 :铁超载抑制化疗药物Ara C诱导HL 60细胞凋亡作用 ,铁剥夺 (DFO)可促进化疗药物Ara C诱导HL 60细胞凋亡作用。临床上可采用铁剥夺的方法协同治疗白血病 。 展开更多

Previous studies have suggested that miR-324-3p is related to the pathophysiology of cerebral ischemia,but the mechanism underlying this relationship is unclea r.In this study,we found that miR-324-3p expression was decreased in patients with acute ischemic stroke and in in vitro and in vivo models of ischemic stro ke.miR-324-3p agomir potentiated ischemic brain damage in rats subjected to middle cerebral artery occlusion,as indicated by increased infarct volumes and cell apoptosis rates and greater neurological deficits.In a PC12 cell oxygen-glucose deprivation/reoxygenation model,a miR-324-3 p mimic decreased cell viability and expression of the anti-apoptotic protein BCL2 and increased expression of the pro-apoptotic protein BAX and rates of cell apoptosis,whereas treatment with a miR-324-3p inhibitor had the opposite effects.Silencing miR-324-3p increased adenosine A1 receptor(A1R)expression thro ugh regulation of GATA binding protein 2(GATA2).These findings suggest that silencing miR-324-3p reduces ischemic brain damage via the GATA2/A1R axis. 展开更多

Two one-dimensional chain-like cyanide-bridged compounds, [Fe(Phen)2(CN)2Ni(Cyclam)](ClO4)2·DMF-2H2O 1 (Cyclam = 1,4,8,11-tetraaza-cyclotetradecane, Phen = 1,10-phenanthroline, and DMF = N,N-dimethylformamide) and [Fe(Phen)2(CN)2Zn(Cyclam)](PF6)2·CH3CN 2, were prepared by the reaction of [Fe(Phen)2(CN)2]·2H2O with M(Cyclam)^2+ (M = Ni, Zn). In complexes 1 and 2, the metal atoms are connected alternatively with CN groups. X-ray structure and IR analyses for 1 and 2 were performed. Structure analysis revealed that both complexes are centrosymmetric and the geometry around each metal atom is an octahedron. The two complexes crystallize in triclinic with space group of P^1-. For 1, a = 10.439(4), b = 14.976(7), c = 15.914(8)A,α = 83.168(15), β = 74.338(15), γ = 78.023(15)°, V = 2338.3(18)A^3, Z = 2, Mr = 1035.37, Dc = 1.471 g/cm^3, F(000) = 1076,μ = 0.895 mm^-1, the final R = 0.0616 and wR = 0.1414 for 5849 observed reflections (1 〉 2σ(I)). Application Research of Dense Sintered Alumina in Iron Runner Castables

Three different kinds of corundum aggregates-tabular sintered alumina, dense sintered alumina, and fused dense corundum-were introduced into the silica fume .free or silica fume containing Al2O3 -SiC - C iron runner castables to investigate their influences on the flow ability, linear change on heating, bulk density, apparent porosity, cold strength, hot modulus of rupture, therm, al shock resistance, slag resistance, oxidation resistance as well as wear resistance of Al2O3 - SiC - C iron runner castables. The results show that ( 1 ) compared with the specimens with fused dense corundum, the specimens with dense sintered alumina have equivalent installation property, slag resistance and oxidation resistance, equivalent or even higher cold modulus of rupture, cold crushing strength and hot modulus of rupture, exhibiting better thermal shock resistance and cold wear resistance ; (2) adopting bimodal alumina micropowder LISAL22RABL as well as water reducers ZX2 and ZD2 can well reduce the water requirement of silica fume free castables, solving the problem of deteriorated flow ability resulted from the lack of silica fume; since the lack of silica fume avoids the formation of low melting point liquid, the hot modulus of rupture and the thermal shock resistance of the silica fume free castables are both better than those of the silica fume containing castables ; (3) the density of the castable specimens with dense sintered alumina is 4% -6% lower than that of the castable specimens with Jhsed dense corundum so the refractories consumption of one iron runner reduces by 5% by using the tastable with dense sintered alumina, which obviously reduces the cost of refractories. 展开更多

目的研究大鼠心肌缺血/再灌注(myocardial ischemia/reperfusion,MI/R)损伤后心肌细胞的铁代谢及从肝治心组方干预对铁死亡的调节作用。方法选择体外培养的大鼠H9c2心肌细胞,并采用氧糖剥夺/复灌(oxygen glucose deprivation/reperfusio... 目的研究大鼠心肌缺血/再灌注(myocardial ischemia/reperfusion,MI/R)损伤后心肌细胞的铁代谢及从肝治心组方干预对铁死亡的调节作用。方法选择体外培养的大鼠H9c2心肌细胞,并采用氧糖剥夺/复灌(oxygen glucose deprivation/reperfusion,OGD/R)法建立体外MI/R模型。将细胞分为正常组、模型组、中药组、抑制剂组、激动剂组、联合组、空白血清组。除正常组外,其余各组均予以OGD/R诱导损伤,在复灌时分别给予完全培养基、含15%含药血清的高糖DMEM培养基、含铁死亡抑制剂Ferrostatin-1(Fer-1)(0.5μmol/L)的完全培养基、含爱拉斯汀(Erastin)(10μmol/L)的完全培养基、含Erastin(10μmol/L)的15%含药血清培养基、含15%空白血清的高糖DMEM培养基。采用CCK-8检测细胞存活率,TMRE荧光探针检测线粒体膜电位,FerroOrange检测细胞内Fe^(2+)水平,DCFH-DA荧光探针检测活性氧(reactive oxygen species,ROS)水平。结果与正常组比较,模型组与空白血清组细胞存活率及线粒体膜电位显著下降(P<0.01),Fe^(2+)及ROS水平显著升高(P<0.01)。模型组与空白血清组线粒体膜电位、Fe^(2+)水平差异无统计学意义(P>0.05)。与模型组比较,中药组及抑制剂组细胞存活率及线粒体膜电位显著升高(P<0.01),Fe^(2+)及ROS水平显著降低(P<0.01)。中药组与抑制剂组线粒体膜电位、Fe^(2+)及ROS水平差异无统计学意义(P>0.05)。与模型组相比,激动剂组细胞存活率及线粒体膜电位显著下降(P<0.01),Fe^(2+)及ROS水平显著升高(P<0.01)。与激动剂组相比,联合组细胞存活率及线粒体膜电位显著升高(P<0.01),Fe^(2+)及ROS水平显著降低(P<0.01)。结论从肝治心组方可以减少MI/R损伤,其机制可能与调节心肌细胞铁代谢、抑制铁死亡有关。 展开更多
