SCs and their derived products offer great promise for biomedical applica...Stem cells (SCs), the undifferentiated biological cells, have the infinite capacity to self-renew and the pluripotent ability to differentiate. SCs and their derived products offer great promise for biomedical applications such as cell therapy, tissue engineering, regenerative medicine and drug screening. However, the clinical applications of SCs require a large amount of SCs with high quality and the number of SCs from their tissue resources is very limited. Large-scale expansion is needed to generate homogeneous SCs with good biological characteristics for clinical application. This necessitates a bioreactor system to provide controllable and stable conditions for stem cell (SC) culture. Traditional methods of bioreactor for maintenance and expansion of cells rely on two-dimensional (2-D) culture techniques, leading to loss self-renewal ability and differentiation capacity upon long-term culture. New approaches for SC expansion with bioreactor employ three-dimensional (3-D) cell growth to mimic their environment in vivo. This paper addresses how a digital heritage project can impact the research and interpretation of a large-scale rural cultural landscape in the United States.Due to the size and scope of rural landscapes,large-scale d...This paper addresses how a digital heritage project can impact the research and interpretation of a large-scale rural cultural landscape in the United States.Due to the size and scope of rural landscapes,large-scale documentation methods are critical to advancing landscape conservation and preservation initiatives.Using an in-progress online project to document a 1935 US federally sponsored program,the Prairie States Forestry Project(PSFP),the authors show how diverse visual and textual data can be spatialised to construct a map reading of landscape change over time.To date,the PSFP is one of the largest afforestation projects in the history of the United States;the United States Forest Service and thousands of landowners undertook a series of cooperative planting agreements to plant over 200 million trees over seven years in approximately 33,000 shelterbelts from the panhandle of Texas to the North Dakota border.Due to a lack of coordinated monitoring,shelterbelt location and status was unknown,and the original archival material remained unpreserved.In the case of the Prairie States Forestry Project,the process for digitising and disseminating previously inaccessible primary source documents is an act of preservation that creates opportunities for future large-scale landscape conservation projects.The application of the archival mapping method and resulting PSFP datasets can be incorporated by individuals working on heritage documentation such as Historic American Landscapes Survey(HALS)reports,National Register nominations,or Cultural Landscape Reports for the National Parks Service.The dataset could also be used by private groups such as cooperative conservation land managers.展开更多
As a group,the halophilic archaea(class Halobacteria)are the most salt-requiring and salt-resistant microorganisms within the domain Archaea.Halophilic archaea flourish in thalassohaline and athalassohaline environmen...As a group,the halophilic archaea(class Halobacteria)are the most salt-requiring and salt-resistant microorganisms within the domain Archaea.Halophilic archaea flourish in thalassohaline and athalassohaline environments and require over 100–150 g/L NaCl for growth and structural stability.Natural hypersaline environments vary in salt concentration,chemical composition and pH,and occur in climates ranging from tropical to polar and even under-sea.Accordingly,their resident haloarchaeal species vary enormously,as do their individual population compositions and community structures.These diverse halophilic archaeal strains are precious resources for theoretical and applied research but assessing their taxonomic and metabolic novelty and diversity in natural environments has been technically difficult up until recently.Environmental DNA-based high-throughput sequencing technology has now matured sufficiently to allow inexpensive recovery of massive amounts of sequence data,revealing the distribution and community composition of halophilic archaea in different hypersaline environments.While cultivation of haloarchaea is slow and tedious,and only recovers a fraction of the natural diversity,it is the conventional means of describing new species,and provides strains for detailed study.As of the end of May 2020,the class Halobacteria contains 71 genera and 275 species,49.8%of which were first isolated from the marine salt environment and 50.2%from the inland salt environment,indicating that both thalassohaline and athalassohaline environments contain diverse halophilic archaea.However,there remain taxa that have not yet been isolated in pure culture,such as the nanohaloarchaea,which are widespread in the salt environment and may be one of the hot spots in the field of halophilic archaea research in the future.In this review,we focus on the cultivation strategies that have been used to isolate extremely halophilic archaea and point out some of the pitfalls and challenges.展开更多
