We obt...According to the large-scale sequencing of cDNA library from silkworm pupae, the cDNA of a novel gene with blank research background was identified and temporarily named Bm-X. The length of this cDNA is 778 bp. We obtained its ORF for further study by bioinformatics analysis. It is 444 bp and encodes 147 amino acid residues, with a predicted molecular weight (MW) of 16.4 kD and an isoelectric point (pI) of 3.69. In this study, we successfully constructed a recombinant plasmid pET-28a(+)-Bm-X and expressed it in Escherichia coli. We used the fusion protein rBm-X which purified by Niaffinity chromatography to produce polyclonal antibodies against Bm-X for Western blot analysis. The analysis revealed that Bm-X was expressed in the larval midgut, the epidermis and the silk gland. In addition, the subcellular localization analysis of silkworm ovary epithelial cells (BmN cells) showed that Bm-X protein was located both in cytoplasm and nucleus, and the signal was stronger in cytoplasm than in nucleus. Our findings indicate that Bm-X gene is a novel species-specificity gene and its expression product can be detected in tissues of the fifth silkworm instar larvae and BmN cells.展开更多

Previous experiments have shown that in...Macroscopic productions of “non-locality” or “excess correlations” of dynamic changes within media between two spaces could be utilized as alternative communication systems. Previous experiments have shown that injections of a weak acid within one of two volumes of spring water sharing the same patterned circular magnetic fields with changing angular accelerations separated by non-traditional (5 m) distances were associated with opposite (basic) shifts in pH within the non-injected, non-local volume. In the present experiments, employing a different technology, pairs of beakers separated by 1 m containing either 25 cc, 50 cc, or 100 cc of spring water were placed within toroids generating weak (30, 300 nT) changing acceleration magnetic fields with 1 ms, 2 ms, or 3 ms point durations or a field whose point durations changed. When a proton source (weak acid) was injected into one beaker (local) pH shifts in the other (non-local) beaker exhibit increased acidity for the 3 ms point duration but increased alkalinity for the 1 ms duration. Neither intermittent point durations nor variable point durations for the same volumes of water placed between the two magnetic field-coupled beakers exhibited significant changes from baseline. Contingent upon the point duration of the applied field, the pH shift was consistent with a fixed quantity of decreased free protons (increased pH) or increased protons (decreased pH) in the non-local beakers. The opposite directions of the pH shifts at 1 ms and 3 ms that correspond to quantitative cosmological solutions for electrons and protons suggest these results may reflect a fundamental physical process.展开更多
The coronavirus disease 2019(COVID-19)coronavirus is a new strain of coronavirus that had not been previously detected in humans.As its severe pathogenicity is concerned,it is important to study it thoroughly to aid i...The coronavirus disease 2019(COVID-19)coronavirus is a new strain of coronavirus that had not been previously detected in humans.As its severe pathogenicity is concerned,it is important to study it thoroughly to aid in the discovery of a cure.In this study,the microRNAs(miRNAs)of COVID-19 were annotated to provide a powerful tool for the study of this novel coronavirus.We obtained 16 novel coronavirus genome sequences and the mature sequences of all viruses in the microRNA database(miRbase),and then used the miRNA matures sequences of the virus to perform the Basic Local Alignment Search Tool(BLAST)analysis in the coronavirus genome,extending the matched regions of approximately 20 bp to two segments by 200 bp.Six sequences were obtained after deleting redundant sequences.Then,the hairpin structures of the mature miRNAs were determined using RNAfold.The mature sequence on one hairpin arm was selected into a total of 4 sequences,and finally the relevant miRNA precursor prediction tools were used to verify whether the selected sequences are miRNA precursor sequences of the novel coronavirus.The miRNAs of the novel coronavirus were annotated by our newly developed method,which will lay the foundation for further study of this virus.展开更多

Many local breeds are now threatened by extinction and the loss of their native genetic...In recent decades, a considerable number of local breeds have been replaced by high-yielding breeds for reasons of profitability. Many local breeds are now threatened by extinction and the loss of their native genetic diversity. The need to conserve breeds and their genetic diversity has a major importance due to the necessity for genetic change within and between populations. Novel approaches have to be explored and extended to maintain this genetic diversity. The aim of this study was the identification and implementation of breed-specific traits for a small, local sheep breed in northern Germany. The data comprised pedigree information, estimated breeding values (EBVs) of several conventional traits, and phenotypic information from a field experiment for two novel traits: 1) average daily gain under extensive circumstances (ADGE) and 2) ultrasonic measurements of muscle-fat ratio (UMFR). The experimental design included a dataset of 47 progeny from 14 pure-bred rams of German White-Headed Mutton (GWM). The methodical approach was divided into four parts: 1) the analysis of the breeding programme, 2) the identification of breed-specific traits, 3) the estimation and correlation of novel breeding values, and 4) the consequences of implementing these novel traits. Genetic parameters and correlations were conducted by applying linear mixed models. The estimates for the heritability (repeatability) were between 0.70 and 0.83 (0.42 and 0.46). The genetic correlation was positive (0.61) and in accordance with the phenotypic correlation (0.62). Average daily gain under intensive circumstances (ADGI) was moderately positive correlated with muscularity (0.60), as opposed to ADGE, which was moderately negative correlated with muscularity (-0.68). The EBV of ADGE was also moderately positive correlated with UMFR (0.64). Genetic response for ADGE enhanced to values of 481.09 g/day, 639.97 g/day, >700 g/day and >850 g/day for different selection intensity scenarios. Corresponding rates of inbreeding were 1.4%, 2.7%, 5.1%, and 7.9% after 10 years of selection. Genetic response for UMFR increased to 0.92, 1.34, 2.41, and >2.75, whereas remaining rates of inbreeding increased to 1.1%, 2.2%, 5.1%, and 7.9%. ADGI and ADGE were tendentially negatively correlated (-0.11), which strengthen the assumption of a biased ADGI. ADGE has a positive influence on meat-quality aspects (UMFR). Optimal use of reference sires with predefined selection intensity achieves genetic response for ADGE and UMFR with simultaneously acceptable rates of inbreeding.展开更多
