Methods: Washed human spermatozoa from normozoospermic donors were treated with insulin (10 μIU) and leptin (10 nmol). Insulin and leptin effects were blocked by inhibition of their intracellular effector, phosphotidylinositol 3-kinase (PI3K), by wortmannin (10 μmol) 30 min prior to insulin and leptin being given. Computer-assisted semen analysis was used to assess motility after 1, 2 and 3 h of incubation. Viability was assessed by fluorescence-activated cell sorting using propidium iodide as a fluorescent probe. Acrosome-reacted cells were observed under a fluorescent microscope using fluorescein-isothiocyanate-Pisum sativum agglutinin as a probe. NO was measured after treating the sperm with 4,5-diaminofluorescein-2/diacetate (DAF-2/DA) and analyzed by fluorescence-activated cell sorting. Results: Insulin and leptin significantly increased total motility, progressive motility and acrosome reaction, as well as NO production. Conclusion: This study showed the in vitro beneficial effects of insulin and leptin on human sperm function. These hormones could play a role in enhancing the fertilization capacity of human spermatozoa.展开更多
Objective:To observe the changes of concentrations in CO, NO and H2S plasma in cases of diffuse large B-cell lymphoma (DLBCL) in non-Hodgkin's lymphoma (NHL), and to explore the relationship between them and relat...Objective:To observe the changes of concentrations in CO, NO and H2S plasma in cases of diffuse large B-cell lymphoma (DLBCL) in non-Hodgkin's lymphoma (NHL), and to explore the relationship between them and relation with IPI.Methods: The case group were 36 patients treated in our hospital,According to the IPI score, they were classified into low risk (0-1) 9 cases, low-intermediate risk (2) 8 cases, high-intermediate risk (3) 10 cases, high risk (4-5) 5 cases. And 20 cases in the control group were healthy medical examiners in the Medical Examination Center of our hospital in the same period. The concentrations of CO, NO and H2S in the plasma of the case group and control group were measured, and the relationship between them was discussed.Results: The levels of CO, NO and H2S in the plasma of patients with NHL were significantly higher than those in the normal control group, and the difference was statistically significant, The CO, NO and H2S level of IPI 0-1 group, 2 group and 3 group was lower than that of IPI4-5 group;the CO, NO and H2S level of IPI3 group was higher than that of IPI0-1 group;After complete remission of treatment, the levels of CO, NO and H2S in plasma decreased, significantly lower than before treatment, and not statistically significant compared with the normal control group;Patients in the case group had consistent changes of levels of CO, NO and H2S in plasma before and after treatment. The three results were positively correlated.Conclusion: The levels of CO, NO and H2S in the plasma of patients with diffuse large B-cell lymphoma are significantly higher, and the higher the IPI score, the higher the CO, NO and H2S level, the level of H2S in plasma decreases obviously after complete remission of treatment, and the three results are positively correlated, and CO, NO and H2S may be involved in the pathophysiological process of non-Hodgkin's lymphoma.展开更多
