Stem cell-based therapies have been proposed as a potential treatment for neural regeneration following closed head injury.We previously reported that induced neural stem cells exert beneficial effects on neural regen...Stem cell-based therapies have been proposed as a potential treatment for neural regeneration following closed head injury.We previously reported that induced neural stem cells exert beneficial effects on neural regeneration via cell replacement.However,the neural regeneration efficiency of induced neural stem cells remains limited.In this study,we explored differentially expressed genes and long non-coding RNAs to clarify the mechanism underlying the neurogenesis of induced neural stem cells.We found that H19 was the most downregulated neurogenesis-associated lnc RNA in induced neural stem cells compared with induced pluripotent stem cells.Additionally,we demonstrated that H19 levels in induced neural stem cells were markedly lower than those in induced pluripotent stem cells and were substantially higher than those in induced neural stem cell-derived neurons.We predicted the target genes of H19 and discovered that H19 directly interacts with mi R-325-3p,which directly interacts with Ctbp2 in induced pluripotent stem cells and induced neural stem cells.Silencing H19 or Ctbp2 impaired induced neural stem cell proliferation,and mi R-325-3p suppression restored the effect of H19 inhibition but not the effect of Ctbp2 inhibition.Furthermore,H19 silencing substantially promoted the neural differentiation of induced neural stem cells and did not induce apoptosis of induced neural stem cells.Notably,silencing H19 in induced neural stem cell grafts markedly accelerated the neurological recovery of closed head injury mice.Our results reveal that H19 regulates the neurogenesis of induced neural stem cells.H19 inhibition may promote the neural differentiation of induced neural stem cells,which is closely associated with neurological recovery following closed head injury.展开更多

Methods: Immunofluorescence was used for the detection of myelin-associated glycoprotein (MAG), a characteristic protein of rat oligodendrocytes (OLN-93 cells). To evaluate the potential protective effects of SSFs on OLN-93 cells injured by Aβ<sub>1-42</sub>, an injury model was established by subjecting OLN-93 cells to Aβ<sub>1-42</sub> exposed. Cell morphology was examined using an inverted microscope, while cell viability was assessed using the colorimetric method of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT). Additionally, lactate dehydrogenase (LDH) was measured using the pyruvic acid reduction assay. The Ginkgo biloba leaf extract (GBE) injection was used as a positive control. Results: A total of >95% of the MAG immunofluorescence-positive cells were identified as oligodendrocytes. Gradually increasing concentrations of SSFs impaired the cells, and the maximum nondetrimental dose for OLN-93 cells was 75 mg/L. This study assessed the effects of SSFs on OLN-93 cells damaged by Aβ<sub>1-42</sub>. The results indicated that SSFs significantly improved OLN-93 cell morphological abnormal changes, increased the OLN-93 cell survival rate, and reduced LDH release. Conclusion: SSFs can alleviate Aβ<sub>1-42</sub>-induced damage of OL.展开更多

It is necessary to investigate the characteristics of Mesenchymal stem cells(MSCs)derived exosomes,and especially their application in tissue regeneration.Previous studies have shown that inflammatory stimulation enha...It is necessary to investigate the characteristics of Mesenchymal stem cells(MSCs)derived exosomes,and especially their application in tissue regeneration.Previous studies have shown that inflammatory stimulation enhanced the secretion of MSC-derived exosomes with stronger anti-inflammatory protein,cytokine profiles,and functional RNA via altering COX2/PGE2 pathway.Recently,accumulating evidence has also revealed that biophysical cues(especially biomechanical cues)in cell microenvironment have significant effects not only on cells but also on their exosomes.It has been reported that applying bi-axial strain to MSCs induces formation of a stiffer cytoskeleton through mTORC2 signaling,which biases against adipogenic differentiation and toward osteoblastogenesis.At the same time,For example,dimensionality,composition and stiffness of the extracellular matrix(ECM)has been proved to affect the size and composition of exosomes secreted by cancer cells.However,the effects of biomechanical cues in the three-dimensional(3D)microenvironment on stem cell-derived exosomes remains to be unveiled.Therefore,it is important to understand the roles of 3D cell mechanical microenvironment in regulating the characteristics of stem cell-derived exosomes and develop more efficient approaches to enhance their functions.This study aimed to explore the changes in characteristics of exosomes secreted by MSCs in periodontium in response to the matrix strain in 3D.Periodontal ligament stem cells(PDLSCs)were cultured in a 3D strain microenvironment engineered with microscale magnetically stretched collagen hydrogels.The morphology,particle distribution,marker protein expression of PDLSC-derived exosomes were analyzed.Then the pro-osteogenic property of exosomes was evaluated by assessing cell viability,proliferation,migration and osteogenic differentiation of target cells,for instance human bone marrow mesenchymal stem cells(hBMSCs).Detailed characterizations revealed that PDLSC-derived exosomes in the 3D strain mi-croenvironment were with similar morphology,particle distribution and surface markers.Notably,Exosomes secreted by PDLSCs in strain microenvironment were more endocytosed by hBMSCs and were more potent in improving proliferation and migration of hBMSCs,comparing with PDLSCs in non-strain environment.Alizarin red staining and molecular biology experiments confirmed that treatment of exosomes secreted by PDLSCs under mechanical stimulation led to a significant increase in osteogenic differentiation of hBMSCs in vitro.Meanwhile,in vivo study also indicated that PDLSC-derived exosomes obtained from the 3D strain microenvironment could obviously promote new bone formation.Our findings revealed that mechanical cues profoundly affected the characteristics of PDLSC-derived exosomes,especially for their bio-activity,providing a foundation for using the 3D mechanical microenvironment to enhance the osteo-inductive functions of stem cell-derived exosomes in cell-free therapy for bone regeneration.展开更多

DATA SOURCES : An online search of Pubmed database was undertaken to identify English articles about the growth of neural stem cells in vitro published from January 2000 to October 2006 by using the keywords of "neural stem cells, bone marrow mesenchymal stem cells (BMSCs), umbilical cord blood stem cells, embryonic stem cells (ESC), separation methods, neural growth factor". And relevant articles published in IEEE/IEE Electronic Library (IEL) database, Springer Link database and Kluwer Online Journals were also searched, Chinese relevant articles published between January 2000 to October 2006 were searched with the same keywords in Chinese in Chinese journal full-text database. STUDY SELECTION : The articles were primarily screened, and then the full-texts were searched. Inclusive criteria: (1) Articles relevant to the biological characteristics and classification of neural stem cells; (2) Articles about the source, separation and differentiation of the ESCs, BMSCs and umbilical cord blood stem cells. The repetitive studies and reviews were excluded. DATA EXTRACTION : Thirty articles were selected from 203 relevant articles according to the inclusive criteria Articles were excluded because of repetition and reviews. DATA SYNTHESES : Neural stem cells have the ability of self-renewing and high differentiation, and they are obtained from ESCs, nerve tissue, nerve system, BMSCs and umbilical cord blood stem cells. ESCs can be separated by means of mechanical dissociation is better than that of the trypsin digestion, BMSCs by density gradient centrifuge separation, hemolysis, whole-blood culture, etc., and umbilical cord blood stem ceils by Ficoil density gradient centrifugation, hydroxyethyl starch (HES) centrifugation sedimentation, etc. Neural growth factor (NGF) and other factors play an important role in the growth of NSCs, such as transforming growth factor (TGF) is an important player in repairing organs, NGF accelerates the process of growth, insulin-like growth factor serves importantly in the differentiation of stem cells into neuron-like cells. CONCLUSION : As unipotent stem cells, NSCs have the abilities of self-renewal and potential of high differentiation. The method of mechanical dissociation is better than trypsin digestion in e separating ESCs. As a preliminary step in the nonlinear design of shape memory alloy(SMA) composite structures,the force-displacement characteristics of the SMA layer are studied.The bilinear hysteretic model is adopted to describe ...As a preliminary step in the nonlinear design of shape memory alloy(SMA) composite structures,the force-displacement characteristics of the SMA layer are studied.The bilinear hysteretic model is adopted to describe the constitutive relationship of SMA material.Under the assumption that there is no point of SMA layer finishing martensitic phase transformation during the loading and unloading process,the generalized restoring force generated by SMA layer is deduced for the case that the simply supported beam vibrates in its first mode.The generalized force is expressed as piecewise-nonlinear hysteretic function of the beam transverse displacement.Furthermore the energy dissipated by SMA layer during one period is obtained by integration,then its dependencies are discussed on the vibration amplitude and the SMA's strain(Ms-Strain) value at the beginning of martensitic phase transformation.It is shown that SMA's energy dissipating capacity is proportional to the stiffness difference of bilinear model and nonlinearly dependent on Ms-Strain.The increasing rate of the dissipating capacity gradually reduces with the amplitude increasing.The condition corresponding to the maximum dissipating capacity is deduced for given value of the vibration amplitude.The obtained results are helpful for designing beams laminated with shape memory alloys.展开更多

The perturbation formula of magnetic force is first derived based on the perturbation technique, and is then applied to the analysis of deformation characteristics with emphasis laid on the analyses of modes, symmetry of deformation and influences of incident angle of applied magnetic field on the plate deformation. The theoretical analyses offer explanations why the configuration offer- romagnetic rectangular plate with simple supports under an oblique magnetic field is in-wavy type along the x-direction, and why the largest deformation of the ferromagnetic plate occurs at the incident angle of 45°for the magnetic field. A numerical code based on the finite element method is developed to simulate quantitatively behaviors of the nonlinearly coupled multi-field problem. Some characteristic curves are plotted to illustrate the magneto--elastic-plastic deflections, and to reveal how the deflections can be influenced by the incident angle of applied magnetic field. The deformation characteristics obtained from the numerical simulations are found in good agreement with the theoretical analyses.展开更多
The pathogenesis of myelodysplastic syndrome(MDS)may be related to the abnormal expression of microRNAs(miRNAs),which could influence the differentiation capacity of mesenchymal stem cells(MSCs)towards adipogenic and ...The pathogenesis of myelodysplastic syndrome(MDS)may be related to the abnormal expression of microRNAs(miRNAs),which could influence the differentiation capacity of mesenchymal stem cells(MSCs)towards adipogenic and osteogenic lineages.In this study,exosomes from bone marrow plasma were successfully extracted and identified.Assessment of miR-103-3p expression in exosomes isolated from BM in 34 MDS patients and 10 controls revealed its 0.52-fold downregulation in patients with MDS compared with controls(NOR)and was downregulated 0.55-fold in MDS-MSCs compared with NOR-MSCs.Transfection of MDS-MSCs with the miR-103-3p mimic improved osteogenic differentiation and decreased adipogenic differentiation in vitro,while inhibition of miR-103-3p showed the opposite results in NOR-MSCs.Thus,the expression of miR-103-3p decreases in MDS BM plasma and MDS-MSCs,significantly impacting MDS-MSCs differentiation.The miR-103-3p mimics may boost MDS-MSCs osteogenic differentiation while weakening lipid differentiation,thereby providing possible target for the treatment of MDS pathogenesis.展开更多
BACKGROUND The hypoxic environment during bone healing is important in regulating the differentiation of periosteal stem cells(PSCs)into osteoblasts or chondrocytes;however,the underlying mechanisms remain unclear.AIM...BACKGROUND The hypoxic environment during bone healing is important in regulating the differentiation of periosteal stem cells(PSCs)into osteoblasts or chondrocytes;however,the underlying mechanisms remain unclear.AIM To determine the effect of hypoxia on PSCs,and the expression of microRNA-584-5p(miR-584-5p)and RUNX family transcription factor 2(RUNX2)in PSCs was modulated to explore the impact of the miR-584-5p/RUNX2 axis on hypoxiainduced osteogenic differentiation of PSCs.METHODS In this study,we isolated primary mouse PSCs and stimulated them with hypoxia,and the characteristics and functional genes related to PSC osteogenic differentiation were assessed.Constructs expressing miR-584-5p and RUNX2 were established to determine PSC osteogenic differentiation.RESULTS Hypoxic stimulation induced PSC osteogenic differentiation and significantly increased calcified nodules,intracellular calcium ion levels,and alkaline phosphatase(ALP)activity in PSCs.Osteogenic differentiation-related factors such as RUNX2,bone morphogenetic protein 2,hypoxia-inducible factor 1-alpha,and ALP were upregulated;in contrast,miR-584-5p was downregulated in these cells.Furthermore,upregulation of miR-584-5p significantly inhibited RUNX2 expression and hypoxia-induced PSC osteogenic differentiation.RUNX2 was the target gene of miR-584-5p,antagonizing miR-584-5p inhibition in hypoxia-induced PSC osteogenic differentiation.CONCLUSION Our study showed that the interaction of miR-584-5p and RUNX2 could mediate PSC osteogenic differentiation induced by hypoxia.展开更多

Large gas generation potential of multiple sets of thick humic-sapropelic source rocks in high maturity stage in Bozhong depression was the basis of large gas field formation. The multi-stage tectonic evolution since Indosinian period formed large-scale buried hill traps. The Tanlu fault activity formed multi-type reservoirs, and buried hill metamorphic rock of Archean and sand-conglomerate of Kongdian Formation were high-quality reservoirs. Thick overpressure lacustrine mudstone and weak neotectonic movement provided good preservation conditions. Bozhong 19-6 gas reservoir was a condensate gas reservoir with very high condensate oil content, and the gas origin was humic-sapropelic and kerogen-cracking gas, and the gas field had large gas thickness, high gas column characteristics and the accumulation process was first oil and then gas. The buried hill reservoir was a massive reservoir and the Kongdian reservoir was a stratified reservoir. The gas field had multi-channel hydrocarbon intense charge from overpressure source rocks, atmospheric-weak overpressure reservoir favorable for accumulation, thick overpressure mudstone caprock favorable for preservation, and natural gas ultra-late rapid accumulation model.展开更多

The results showed that, firstly, plant height, gravity center height and basal internode length of the 2 rice cultivars increased with the increase of nitrogen application rate, while wall thickness and internode filling degree decreased. The breaking-resistance strength per stem and thrust-resistance strength of plants declined with the increase of nitrogen application rate. Secondly, as nitrogen input increased, the number of vascular bundles and the area of vascular bundles increased between the 2 rice cultivars. Thirdly, with the increase of nitrogen application rate, the stem section area of Guangliangyou 1128, which had strong lodging resistance, gradually increased, while that of Zhunliangyou 527 increased firstly and then decreased. The maximum application amount of nitrogen was 240 kg/hm 2. Nitrogen fertilizer mainly affected the relative gravity center height, stem wall thickness and internode filling degree of the 2 cultivars, thereby reducing the basal stem breaking resistance and plant thrust-resistance strength.展开更多

The current-voltage (I - V) characteristics are nonlinear from 300 down to 60 K. Some step-like structures in the I - V curves and oscillation peaks in the differential conductance (dI/dV - V) curves have been observed even at room temperature. It proposes that the observed behaviour can be attributed to Coulomb-blockade transport in the one-dimensional CdS nanowires with diameters of 6-10 nm.展开更多

It is found that the LFO the band the interannual oscillations of the SST monthly anomaly (SSTA) interact each other and they all relate to the occurrence and development of El Nino events closely. Before El Nino event happens, it contributes to the SST's wanning up and to the SST's quasi-biennial oscillation (called QBO for brevity) and three and half years oscillation (called SO for short) being in warm water phase in the equatorial central and eastern Pacific (ECP and EEP) that the LFO in the equatorial western Pacific (EWP) enhances and propagates eastward; When El Nino event takes place, the LFO, SSTA and SSTA's QBO and SO in the EEP interact and strengthen each other; But the warmer SST and the SSTA's QBO and SO in the warm water phase in the EEP contribute to the LFO's weakening in the equatorial Pacific. Moreover, these contribute to the SST in the EEP becoming cold and the SSTA's QBO and SO in the EWP being in cold water phase and then impel the El Nino event to end.展开更多

It has been passaged for more than 212 generations. Moderate round cells were predominant and most of them were mononuclear. Some characteristics of malignant cells and A-type viral-like particles were observed by electron microscopy. The results of cytochemical studies (DNA, RNA, SDH, 5' AMPase, ACP etc.) were comparable to the ultramicroscopic results. It multiplied approximately 27.4 fold on day 5 with mitotic index reaching 1.8% on day 3. This cell line was a hyperdiploid with karyotype of 45 or 45, -2X, tril2, tri17, +M1-5. Cell agglutination was observed when treated with ConA (≥7 fig, ml). Spontaneous agglutination might also take place without adding any ConA. After 5×106 cells of Ca 761-86 suspension were transplanted into the normal inbred 615 mice by different ways (subcutan eous, intrafoot-pad or intraperitoneal), the transplan lability rate reached 100%. Spontaneous remission was never observed and its metastatic ability reserved. PPLO were not detected. Ca 761-86 may be of value for practical purposes.展开更多

METHODS: Reverse transcription polymerase chain reaction was performed to amplify the 14-3-3σ gene from the mRNA of colon cancer stem cells. The gene was then cloned into the pGEM-T vector. After being sequenced, the target gene 14-3-3σ was cut from the pGEM-T vector and cloned into the pGBKT7 yeast expression plasmid. Then, the bait plasmid pGBKT7-14-3-3σ was transformed into the yeast strain AH109. After the expression of the pGBKT7-14-3-3σ fusion protein in the AH109 yeast strain was accomplished, a yeast two-hybrid screening assay was performed by mating AH109 with Y187 that contained a HeLa cDNA library plasmid. The interaction between the 14-3-3σ protein and the proteins obtained from positive colonies was further confirmed by repeating the yeast two-hybridscreen. After extracting and sequencing the plasmids from the positive colonies, we performed a bioinformatics analysis. A coimmunoprecipitation assay was performed to confirm the interaction between 14-3-3σ and the proteins obtained from the positive colonies. Finally, we constructed 14-3-3σ and potassium channel modulatory factor 1 (KCMF1) siRNA expression plasmids and transfected them into colon cancer stem cells. RESULTS: The bait plasmid pGBKT7-14-3-3σ was constructed successfully, and the 14-3-3σ protein had no toxic or autonomous activation effect on the yeast. Nineteen true-positive colonies were selected and sequenced, and their full-length sequences were obtained. We searched for homologous DNA sequences for these sequences from GenBank. Among the positive colonies, four coding genes with known functions were obtained, including KCMF1 , quinone oxidore-ductase (NQO2 ), hydroxyisobutyrate dehydrogenase (HIBADH ) and 14-3-3σ . For the subsequent coimmu-noprecipitation assay, the plasmids PCDEF-Flag-14-3-3σ, PCDEF-Myc-KCMF1, PCDEF-Myc-NQO2 and PCDEF-Myc-HIBADH were successfully constructed, and the sequences were further confirmed by DNA sequencing. The Fugene 6 reagent was used to transfect the plasmids, and fluorescence-activated cell sorting analysis showed the transfection efficiency was 97.8% after 48 h. The HEK 293FT cells showed the stable expression of the PCDEF-Flag-14-3-3σ, PCDEF-Myc-KCMF1, PCDEF-Myc-NQO2 and PCDEF-Myc-HIBADH plasmids. After anti-Myc antibody immunoprecipitation with Myc-KCMF1, Myc-NQO2 and Myc-HIBADH from cell lysates, the presence of Flag-14-3-3σ protein in the immuno-precipitated complex was determined by western blot analysis. The knock-down expression of the 14-3-3σ and KCMF1 proteins significantly inhibited cell proliferation and colony formation of SW1116csc. CONCLUSION: Genes of the proteins that interactedwith 14-3-3σ were successfully screened from a HeLa cDNA library. In this research work, fiber extracted from the bark of <i>Cola</i> <i>lepidota</i> (<i>CL</i>) plant, grown in the flora of Southern part of Cameroon, was investigated for composit...In this research work, fiber extracted from the bark of <i>Cola</i> <i>lepidota</i> (<i>CL</i>) plant, grown in the flora of Southern part of Cameroon, was investigated for composites reinforcement. The investigation was carried via evaluation of <span>water absorption capacity, moisture content, real density, porosity, chemical composition, chemical structure and thermal behaviour. It was discovered that the new fiber has relatively low moisture content and water absorption capacity similar to those of other investigated natural fibers such as flax, sisal, coconut, hemp and jute. Its porosity was found appropriate for composite production and the fiber was found to be thermally stable up to 230°C, with maximum degradation temperature of 325°C. The main constituents of the fibre include cellulose, hemicellulose and lignin. In conclusion, based on the properties investigated, this fiber is considered suitable for composite manufacture.展开更多
The regulation of adult neural stem cells(NSCs) is critical for lifelong neurogenesis. MicroRNAs(miRNAs) are a type of small, endogenous RNAs that regulate gene expression post-transcriptionally and influence signalin...The regulation of adult neural stem cells(NSCs) is critical for lifelong neurogenesis. MicroRNAs(miRNAs) are a type of small, endogenous RNAs that regulate gene expression post-transcriptionally and influence signaling networks responsible for several cellular processes. In this study, mi R-103-3 p was transfected into neural stem cells derived from embryonic hippocampal neural stem cells. The results showed that mi R-103-3 p suppressed neural stem cell proliferation and differentiation, and promoted apoptosis. In addition, mi R-103-3 p negatively regulated Nud E neurodevelopment protein 1-like 1(Ndel1) expression by binding to the 3′ untranslated region of Ndel1. Transduction of neural stem cells with a lentiviral vector overexpressing Ndel1 significantly increased cell proliferation and differentiation, decreased neural stem cell apoptosis, and decreased protein expression levels of Wnt3 a, β-catenin, phosphor-GSK-3β, LEF1, c-myc, c-Jun, and cyclin D1, all members of the Wnt/β-catenin signaling pathway. These findings suggest that Ndel1 is a novel mi R-103-3 p target and that mi R-103-3 p acts by suppressing neural stem cell proliferation and promoting apoptosis and differentiation. 文摘In order to study the combustion characteristics of tar in biomass gasifier inner wall and gasification gas,“tobacco stem semi-tar inside furnace”,“tobacco stem tar inside furnace”and“tobacco stem tar out-of-furnace”were subjected to thermogravimetric experiments,and the combustion characteristics and kinetic characteristics were analyzed.The result shows that“tobacco stem semi-tar inside furnace”has the highest value and“tobacco stem tar out-of-furnace”is has the lowest value on ignition characteristics,combustion characteristics and combustible stability;“tobacco stem semi-tar inside furnace”has the lowest value and“tobacco stem tar outside furnace”has the highest value on burnout characteristics;“tobacco stem tar outside furnace”has the highest value and“tobacco stem tar inside furnace”has the lowest value on integrated combustion characteristics.

文摘Stem cell-based therapies have been proposed as a potential treatment for neural regeneration following closed head injury.We previously reported that induced neural stem cells exert beneficial effects on neural regeneration via cell replacement.However,the neural regeneration efficiency of induced neural stem cells remains limited.In this study,we explored differentially expressed genes and long non-coding RNAs to clarify the mechanism underlying the neurogenesis of induced neural stem cells.We found that H19 was the most downregulated neurogenesis-associated lnc RNA in induced neural stem cells compared with induced pluripotent stem cells.Additionally,we demonstrated that H19 levels in induced neural stem cells were markedly lower than those in induced pluripotent stem cells and were substantially higher than those in induced neural stem cell-derived neurons.We predicted the target genes of H19 and discovered that H19 directly interacts with mi R-325-3p,which directly interacts with Ctbp2 in induced pluripotent stem cells and induced neural stem cells.Silencing H19 or Ctbp2 impaired induced neural stem cell proliferation,and mi R-325-3p suppression restored the effect of H19 inhibition but not the effect of Ctbp2 inhibition.Furthermore,H19 silencing substantially promoted the neural differentiation of induced neural stem cells and did not induce apoptosis of induced neural stem cells.Notably,silencing H19 in induced neural stem cell grafts markedly accelerated the neurological recovery of closed head injury mice.Our results reveal that H19 regulates the neurogenesis of induced neural stem cells.H19 inhibition may promote the neural differentiation of induced neural stem cells,which is closely associated with neurological recovery following closed head injury.
文摘Aim: This study aimed to investigate the protective effects of flavonoids from the stem and leaves of Scutellaria baicalensis Georgi (SSFs) against Aβ<sub>1-42</sub>-induced oligodendrocytes (OL) damage. Methods: Immunofluorescence was used for the detection of myelin-associated glycoprotein (MAG), a characteristic protein of rat oligodendrocytes (OLN-93 cells). To evaluate the potential protective effects of SSFs on OLN-93 cells injured by Aβ<sub>1-42</sub>, an injury model was established by subjecting OLN-93 cells to Aβ<sub>1-42</sub> exposed. Cell morphology was examined using an inverted microscope, while cell viability was assessed using the colorimetric method of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT). Additionally, lactate dehydrogenase (LDH) was measured using the pyruvic acid reduction assay. The Ginkgo biloba leaf extract (GBE) injection was used as a positive control. Results: A total of >95% of the MAG immunofluorescence-positive cells were identified as oligodendrocytes. Gradually increasing concentrations of SSFs impaired the cells, and the maximum nondetrimental dose for OLN-93 cells was 75 mg/L. This study assessed the effects of SSFs on OLN-93 cells damaged by Aβ<sub>1-42</sub>. The results indicated that SSFs significantly improved OLN-93 cell morphological abnormal changes, increased the OLN-93 cell survival rate, and reduced LDH release. Conclusion: SSFs can alleviate Aβ<sub>1-42</sub>-induced damage of OL.

文摘[Objectives]To establish a HPLC characteristic spectrum of honeysuckle stem formula granules,and to evaluate the quality consistency based on the standard decoction.[Methods]Using Agilent ZORBAX SB-C_(18)(5μm,250 mm×4.6 mm)chromatographic column;gradient elution with acetonitrile-0.4%phosphoric acid solution(0.2:99.8,V/V),column temperature at 25℃;detection wavelength at 236 nm;flow rate 1.0 mL/min;using"Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine",to evaluate the similarity of the characteristic chromatogram of crude drug,standard decoction and formula granules of honeysuckle stem.[Results]The similarity of characteristic chromatogram was higher than 0.94,and 6 components were identified from 10 characteristic peaks.[Conclusions]The established method can more comprehensively reflect the overall appearance of the chemical substances in the honeysuckle stem and the transmission law,and can provide an experimental basis for the consistency of the quality of the honeysuckle stem formula granules.

文摘It is necessary to investigate the characteristics of Mesenchymal stem cells(MSCs)derived exosomes,and especially their application in tissue regeneration.Previous studies have shown that inflammatory stimulation enhanced the secretion of MSC-derived exosomes with stronger anti-inflammatory protein,cytokine profiles,and functional RNA via altering COX2/PGE2 pathway.Recently,accumulating evidence has also revealed that biophysical cues(especially biomechanical cues)in cell microenvironment have significant effects not only on cells but also on their exosomes.It has been reported that applying bi-axial strain to MSCs induces formation of a stiffer cytoskeleton through mTORC2 signaling,which biases against adipogenic differentiation and toward osteoblastogenesis.At the same time,For example,dimensionality,composition and stiffness of the extracellular matrix(ECM)has been proved to affect the size and composition of exosomes secreted by cancer cells.However,the effects of biomechanical cues in the three-dimensional(3D)microenvironment on stem cell-derived exosomes remains to be unveiled.Therefore,it is important to understand the roles of 3D cell mechanical microenvironment in regulating the characteristics of stem cell-derived exosomes and develop more efficient approaches to enhance their functions.This study aimed to explore the changes in characteristics of exosomes secreted by MSCs in periodontium in response to the matrix strain in 3D.Periodontal ligament stem cells(PDLSCs)were cultured in a 3D strain microenvironment engineered with microscale magnetically stretched collagen hydrogels.The morphology,particle distribution,marker protein expression of PDLSC-derived exosomes were analyzed.Then the pro-osteogenic property of exosomes was evaluated by assessing cell viability,proliferation,migration and osteogenic differentiation of target cells,for instance human bone marrow mesenchymal stem cells(hBMSCs).Detailed characterizations revealed that PDLSC-derived exosomes in the 3D strain mi-croenvironment were with similar morphology,particle distribution and surface markers.Notably,Exosomes secreted by PDLSCs in strain microenvironment were more endocytosed by hBMSCs and were more potent in improving proliferation and migration of hBMSCs,comparing with PDLSCs in non-strain environment.Alizarin red staining and molecular biology experiments confirmed that treatment of exosomes secreted by PDLSCs under mechanical stimulation led to a significant increase in osteogenic differentiation of hBMSCs in vitro.Meanwhile,in vivo study also indicated that PDLSC-derived exosomes obtained from the 3D strain microenvironment could obviously promote new bone formation.Our findings revealed that mechanical cues profoundly affected the characteristics of PDLSC-derived exosomes,especially for their bio-activity,providing a foundation for using the 3D mechanical microenvironment to enhance the osteo-inductive functions of stem cell-derived exosomes in cell-free therapy for bone regeneration.
文摘OBJECTIVE: To summarize the biological characteristics of neural stem cells, and the separation, purification. differentiation and source of neural stem cells. DATA SOURCES : An online search of Pubmed database was undertaken to identify English articles about the growth of neural stem cells in vitro published from January 2000 to October 2006 by using the keywords of "neural stem cells, bone marrow mesenchymal stem cells (BMSCs), umbilical cord blood stem cells, embryonic stem cells (ESC), separation methods, neural growth factor". And relevant articles published in IEEE/IEE Electronic Library (IEL) database, Springer Link database and Kluwer Online Journals were also searched, Chinese relevant articles published between January 2000 to October 2006 were searched with the same keywords in Chinese in Chinese journal full-text database. STUDY SELECTION : The articles were primarily screened, and then the full-texts were searched. Inclusive criteria: (1) Articles relevant to the biological characteristics and classification of neural stem cells; (2) Articles about the source, separation and differentiation of the ESCs, BMSCs and umbilical cord blood stem cells. The repetitive studies and reviews were excluded. DATA EXTRACTION : Thirty articles were selected from 203 relevant articles according to the inclusive criteria Articles were excluded because of repetition and reviews. DATA SYNTHESES : Neural stem cells have the ability of self-renewing and high differentiation, and they are obtained from ESCs, nerve tissue, nerve system, BMSCs and umbilical cord blood stem cells. ESCs can be separated by means of mechanical dissociation is better than that of the trypsin digestion, BMSCs by density gradient centrifuge separation, hemolysis, whole-blood culture, etc., and umbilical cord blood stem ceils by Ficoil density gradient centrifugation, hydroxyethyl starch (HES) centrifugation sedimentation, etc. Neural growth factor (NGF) and other factors play an important role in the growth of NSCs, such as transforming growth factor (TGF) is an important player in repairing organs, NGF accelerates the process of growth, insulin-like growth factor serves importantly in the differentiation of stem cells into neuron-like cells. CONCLUSION : As unipotent stem cells, NSCs have the abilities of self-renewal and potential of high differentiation. The method of mechanical dissociation is better than trypsin digestion in e separating ESCs. However, density gradient centrifuge separation is better than other methods in the separation of the BMSCs. NGF and other factors play an important role in the growth of NSCs.

文摘As a preliminary step in the nonlinear design of shape memory alloy(SMA) composite structures,the force-displacement characteristics of the SMA layer are studied.The bilinear hysteretic model is adopted to describe the constitutive relationship of SMA material.Under the assumption that there is no point of SMA layer finishing martensitic phase transformation during the loading and unloading process,the generalized restoring force generated by SMA layer is deduced for the case that the simply supported beam vibrates in its first mode.The generalized force is expressed as piecewise-nonlinear hysteretic function of the beam transverse displacement.Furthermore the energy dissipated by SMA layer during one period is obtained by integration,then its dependencies are discussed on the vibration amplitude and the SMA's strain(Ms-Strain) value at the beginning of martensitic phase transformation.It is shown that SMA's energy dissipating capacity is proportional to the stiffness difference of bilinear model and nonlinearly dependent on Ms-Strain.The increasing rate of the dissipating capacity gradually reduces with the amplitude increasing.The condition corresponding to the maximum dissipating capacity is deduced for given value of the vibration amplitude.The obtained results are helpful for designing beams laminated with shape memory alloys.

文摘In this paper, the magnetic-elastic-plastic deformation behavior is studied for a ferromagnetic plate with simple supports. The perturbation formula of magnetic force is first derived based on the perturbation technique, and is then applied to the analysis of deformation characteristics with emphasis laid on the analyses of modes, symmetry of deformation and influences of incident angle of applied magnetic field on the plate deformation. The theoretical analyses offer explanations why the configuration offer- romagnetic rectangular plate with simple supports under an oblique magnetic field is in-wavy type along the x-direction, and why the largest deformation of the ferromagnetic plate occurs at the incident angle of 45°for the magnetic field. A numerical code based on the finite element method is developed to simulate quantitatively behaviors of the nonlinearly coupled multi-field problem. Some characteristic curves are plotted to illustrate the magneto--elastic-plastic deflections, and to reveal how the deflections can be influenced by the incident angle of applied magnetic field. The deformation characteristics obtained from the numerical simulations are found in good agreement with the theoretical analyses.

文摘The pathogenesis of myelodysplastic syndrome(MDS)may be related to the abnormal expression of microRNAs(miRNAs),which could influence the differentiation capacity of mesenchymal stem cells(MSCs)towards adipogenic and osteogenic lineages.In this study,exosomes from bone marrow plasma were successfully extracted and identified.Assessment of miR-103-3p expression in exosomes isolated from BM in 34 MDS patients and 10 controls revealed its 0.52-fold downregulation in patients with MDS compared with controls(NOR)and was downregulated 0.55-fold in MDS-MSCs compared with NOR-MSCs.Transfection of MDS-MSCs with the miR-103-3p mimic improved osteogenic differentiation and decreased adipogenic differentiation in vitro,while inhibition of miR-103-3p showed the opposite results in NOR-MSCs.Thus,the expression of miR-103-3p decreases in MDS BM plasma and MDS-MSCs,significantly impacting MDS-MSCs differentiation.The miR-103-3p mimics may boost MDS-MSCs osteogenic differentiation while weakening lipid differentiation,thereby providing possible target for the treatment of MDS pathogenesis.

文摘BACKGROUND The hypoxic environment during bone healing is important in regulating the differentiation of periosteal stem cells(PSCs)into osteoblasts or chondrocytes;however,the underlying mechanisms remain unclear.AIM To determine the effect of hypoxia on PSCs,and the expression of microRNA-584-5p(miR-584-5p)and RUNX family transcription factor 2(RUNX2)in PSCs was modulated to explore the impact of the miR-584-5p/RUNX2 axis on hypoxiainduced osteogenic differentiation of PSCs.METHODS In this study,we isolated primary mouse PSCs and stimulated them with hypoxia,and the characteristics and functional genes related to PSC osteogenic differentiation were assessed.Constructs expressing miR-584-5p and RUNX2 were established to determine PSC osteogenic differentiation.RESULTS Hypoxic stimulation induced PSC osteogenic differentiation and significantly increased calcified nodules,intracellular calcium ion levels,and alkaline phosphatase(ALP)activity in PSCs.Osteogenic differentiation-related factors such as RUNX2,bone morphogenetic protein 2,hypoxia-inducible factor 1-alpha,and ALP were upregulated;in contrast,miR-584-5p was downregulated in these cells.Furthermore,upregulation of miR-584-5p significantly inhibited RUNX2 expression and hypoxia-induced PSC osteogenic differentiation.RUNX2 was the target gene of miR-584-5p,antagonizing miR-584-5p inhibition in hypoxia-induced PSC osteogenic differentiation.CONCLUSION Our study showed that the interaction of miR-584-5p and RUNX2 could mediate PSC osteogenic differentiation induced by hypoxia.

文摘Based on the study of natural gas resource, low buried hill trap formation mechanism, high quality reservoir control factors and natural gas preservation conditions, the formation conditions and reservoir accumulation characteristics of Bozhong 19-6 large condensate gas field were summarized. Large gas generation potential of multiple sets of thick humic-sapropelic source rocks in high maturity stage in Bozhong depression was the basis of large gas field formation. The multi-stage tectonic evolution since Indosinian period formed large-scale buried hill traps. The Tanlu fault activity formed multi-type reservoirs, and buried hill metamorphic rock of Archean and sand-conglomerate of Kongdian Formation were high-quality reservoirs. Thick overpressure lacustrine mudstone and weak neotectonic movement provided good preservation conditions. Bozhong 19-6 gas reservoir was a condensate gas reservoir with very high condensate oil content, and the gas origin was humic-sapropelic and kerogen-cracking gas, and the gas field had large gas thickness, high gas column characteristics and the accumulation process was first oil and then gas. The buried hill reservoir was a massive reservoir and the Kongdian reservoir was a stratified reservoir. The gas field had multi-channel hydrocarbon intense charge from overpressure source rocks, atmospheric-weak overpressure reservoir favorable for accumulation, thick overpressure mudstone caprock favorable for preservation, and natural gas ultra-late rapid accumulation model.

文摘The effects of different nitrogen application rate on the physical characteristics and anatomic structure of rice stems were investigated with rice cultivars Guangliangyou 1128 (with high resistance) and Zhunliangyou 527 (with low resistance) as materials. The results showed that, firstly, plant height, gravity center height and basal internode length of the 2 rice cultivars increased with the increase of nitrogen application rate, while wall thickness and internode filling degree decreased. The breaking-resistance strength per stem and thrust-resistance strength of plants declined with the increase of nitrogen application rate. Secondly, as nitrogen input increased, the number of vascular bundles and the area of vascular bundles increased between the 2 rice cultivars. Thirdly, with the increase of nitrogen application rate, the stem section area of Guangliangyou 1128, which had strong lodging resistance, gradually increased, while that of Zhunliangyou 527 increased firstly and then decreased. The maximum application amount of nitrogen was 240 kg/hm 2. Nitrogen fertilizer mainly affected the relative gravity center height, stem wall thickness and internode filling degree of the 2 cultivars, thereby reducing the basal stem breaking resistance and plant thrust-resistance strength.

文摘This paper studies the electronic transport in an individual helically twisted CdS nanowire rope, on which platinum microleads are attached by focused-ion beam deposition. The current-voltage (I - V) characteristics are nonlinear from 300 down to 60 K. Some step-like structures in the I - V curves and oscillation peaks in the differential conductance (dI/dV - V) curves have been observed even at room temperature. It proposes that the observed behaviour can be attributed to Coulomb-blockade transport in the one-dimensional CdS nanowires with diameters of 6-10 nm.

文摘The characteristics of 30-60 day oscillation (hereafter called LFO ) of the outgoing longwave radiation data (OLR) and its relations to the interannual oscillations of the sea surface temperature (SST) are investigated by using the daily OLR data for the period from January, 1979 to December, 1987 and the corresponding monthly SST data. It is found that the LFO the band the interannual oscillations of the SST monthly anomaly (SSTA) interact each other and they all relate to the occurrence and development of El Nino events closely. Before El Nino event happens, it contributes to the SST's wanning up and to the SST's quasi-biennial oscillation (called QBO for brevity) and three and half years oscillation (called SO for short) being in warm water phase in the equatorial central and eastern Pacific (ECP and EEP) that the LFO in the equatorial western Pacific (EWP) enhances and propagates eastward; When El Nino event takes place, the LFO, SSTA and SSTA's QBO and SO in the EEP interact and strengthen each other; But the warmer SST and the SSTA's QBO and SO in the warm water phase in the EEP contribute to the LFO's weakening in the equatorial Pacific. Moreover, these contribute to the SST in the EEP becoming cold and the SSTA's QBO and SO in the EWP being in cold water phase and then impel the El Nino event to end.
文摘A cell line designated as Ca 761-86 has been established from the solid mouse mammary cancer (Ca 761) by suspension culture. It has been passaged for more than 212 generations. Moderate round cells were predominant and most of them were mononuclear. Some characteristics of malignant cells and A-type viral-like particles were observed by electron microscopy. The results of cytochemical studies (DNA, RNA, SDH, 5' AMPase, ACP etc.) were comparable to the ultramicroscopic results. It multiplied approximately 27.4 fold on day 5 with mitotic index reaching 1.8% on day 3. This cell line was a hyperdiploid with karyotype of 45 or 45, -2X, tril2, tri17, +M1-5. Cell agglutination was observed when treated with ConA (≥7 fig, ml). Spontaneous agglutination might also take place without adding any ConA. After 5×106 cells of Ca 761-86 suspension were transplanted into the normal inbred 615 mice by different ways (subcutan eous, intrafoot-pad or intraperitoneal), the transplan lability rate reached 100%. Spontaneous remission was never observed and its metastatic ability reserved. PPLO were not detected. Ca 761-86 may be of value for practical purposes.

文摘AIM: To investigate the biological function of 14-3-3σ protein and to look for proteins that interact with 14-3-3σ protein in colon cancer stem cells. METHODS: Reverse transcription polymerase chain reaction was performed to amplify the 14-3-3σ gene from the mRNA of colon cancer stem cells. The gene was then cloned into the pGEM-T vector. After being sequenced, the target gene 14-3-3σ was cut from the pGEM-T vector and cloned into the pGBKT7 yeast expression plasmid. Then, the bait plasmid pGBKT7-14-3-3σ was transformed into the yeast strain AH109. After the expression of the pGBKT7-14-3-3σ fusion protein in the AH109 yeast strain was accomplished, a yeast two-hybrid screening assay was performed by mating AH109 with Y187 that contained a HeLa cDNA library plasmid. The interaction between the 14-3-3σ protein and the proteins obtained from positive colonies was further confirmed by repeating the yeast two-hybridscreen. After extracting and sequencing the plasmids from the positive colonies, we performed a bioinformatics analysis. A coimmunoprecipitation assay was performed to confirm the interaction between 14-3-3σ and the proteins obtained from the positive colonies. Finally, we constructed 14-3-3σ and potassium channel modulatory factor 1 (KCMF1) siRNA expression plasmids and transfected them into colon cancer stem cells. RESULTS: The bait plasmid pGBKT7-14-3-3σ was constructed successfully, and the 14-3-3σ protein had no toxic or autonomous activation effect on the yeast. Nineteen true-positive colonies were selected and sequenced, and their full-length sequences were obtained. We searched for homologous DNA sequences for these sequences from GenBank. Among the positive colonies, four coding genes with known functions were obtained, including KCMF1 , quinone oxidore-ductase (NQO2 ), hydroxyisobutyrate dehydrogenase (HIBADH ) and 14-3-3σ . For the subsequent coimmu-noprecipitation assay, the plasmids PCDEF-Flag-14-3-3σ, PCDEF-Myc-KCMF1, PCDEF-Myc-NQO2 and PCDEF-Myc-HIBADH were successfully constructed, and the sequences were further confirmed by DNA sequencing. The Fugene 6 reagent was used to transfect the plasmids, and fluorescence-activated cell sorting analysis showed the transfection efficiency was 97.8% after 48 h. The HEK 293FT cells showed the stable expression of the PCDEF-Flag-14-3-3σ, PCDEF-Myc-KCMF1, PCDEF-Myc-NQO2 and PCDEF-Myc-HIBADH plasmids. After anti-Myc antibody immunoprecipitation with Myc-KCMF1, Myc-NQO2 and Myc-HIBADH from cell lysates, the presence of Flag-14-3-3σ protein in the immuno-precipitated complex was determined by western blot analysis. The knock-down expression of the 14-3-3σ and KCMF1 proteins significantly inhibited cell proliferation and colony formation of SW1116csc. CONCLUSION: Genes of the proteins that interactedwith 14-3-3σ were successfully screened from a HeLa cDNA library. KCMF1 and 14-3-3σ protein may affect the proliferation and colony formation of human colon cancer stem cells.
文摘In this research work, fiber extracted from the bark of <i>Cola</i> <i>lepidota</i> (<i>CL</i>) plant, grown in the flora of Southern part of Cameroon, was investigated for composites reinforcement. The investigation was carried via evaluation of <span>water absorption capacity, moisture content, real density, porosity, chemical composition, chemical structure and thermal behaviour. It was discovered that the new fiber has relatively low moisture content and water absorption capacity similar to those of other investigated natural fibers such as flax, sisal, coconut, hemp and jute. Its porosity was found appropriate for composite production and the fiber was found to be thermally stable up to 230°C, with maximum degradation temperature of 325°C. The main constituents of the fibre include cellulose, hemicellulose and lignin. In conclusion, based on the properties investigated, this fiber is considered suitable for composite manufacture.

文摘The regulation of adult neural stem cells(NSCs) is critical for lifelong neurogenesis. MicroRNAs(miRNAs) are a type of small, endogenous RNAs that regulate gene expression post-transcriptionally and influence signaling networks responsible for several cellular processes. In this study, mi R-103-3 p was transfected into neural stem cells derived from embryonic hippocampal neural stem cells. The results showed that mi R-103-3 p suppressed neural stem cell proliferation and differentiation, and promoted apoptosis. In addition, mi R-103-3 p negatively regulated Nud E neurodevelopment protein 1-like 1(Ndel1) expression by binding to the 3′ untranslated region of Ndel1. Transduction of neural stem cells with a lentiviral vector overexpressing Ndel1 significantly increased cell proliferation and differentiation, decreased neural stem cell apoptosis, and decreased protein expression levels of Wnt3 a, β-catenin, phosphor-GSK-3β, LEF1, c-myc, c-Jun, and cyclin D1, all members of the Wnt/β-catenin signaling pathway. These findings suggest that Ndel1 is a novel mi R-103-3 p target and that mi R-103-3 p acts by suppressing neural stem cell proliferation and promoting apoptosis and differentiation.
