The thermal decomposition characteristics of methyl linoleate (ML) under nitrogen and oxygen atmo- spheres were investigated, using a thermogravimetric analyzer at a heating rate of 10 ~C/min from room tem- perature to 600℃. Furthermore, the pyrolytic and kinetic characteristics of ML at different heating rates were stud- ied. The results showed that the thermal decomposition characteristics of ML under nitrogen and oxygen atmo- spheres were macroscopically similar, although ML exhibited relatively lower thermal stability under an oxy- gen atmosphere than under a nitrogen atmosphere. The initial decomposition temperature, the maximum weight loss temperature, the peak decomposition temperature, and the rate of maximum weight loss of ML under an oxygen atmosphere were much lower than those under a nitrogen atmosphere and increased with increasing heating rates under either oxygen or nitrogen atmosphere. In addition, the kinetic characteristics of thermal decomposition of ML were elucidated based on the experimental results and by the multiple linear regression method. The activation energy, pre-exponential factor, reaction order, and the kinetic equation for thermal decomposition of ML were obtained. The comparison of experimental and calculated data and the analysis of statistical errors of pyrolysis ratios demonstrated that the kinetic model was reliable for pyrolysis of ML with relative errors of about 1%. Finally, the kinetic compensation effect between the pre-exponential factors and the activation energy in the pyrolysis of ML was also confirmed. 展开更多

This paper focuses on the method of synthesis of conjugated linoleic acid of high purity and the process line and conditions for its purification that can be used in large scale production. CLA of more than 95% purity was prepared by means of urea adduct purification and conjugation using safflower oil as material. The total recovery of the product adds up to more than 48%. The reactive kinetics about linoleic acid from sunflower oil converted into CLA was investigated, and its apparent kinetic model was also established, which can be used as a base for industrial designs. 展开更多

Conjugated linoleic acid (CLA) is a kind of fatty acid with physiological activities and potential application prospect. A synthesis method of conjugated linoleic acid and a purification technology were studied. CLA was prepared and purified by urea-complexation and conjugation using safflower oil as raw material. The purity of CLA and total recovery of the product was more than 95% and 48%, respectively. The main isomers produced in alkali-catalyzed conjugation were identified by gas chromatography (GC) linked to mass spectrometry (MS) and Fourier transform infrared spectroscopy (FTIR). The total amount of the two main isomers (9cis, 11trons-and 10trans, 12cis-CLA) determined by GC was more than 90% of the product. 展开更多

AIM: To determine the effect of cis -9, trans -11-conjugated linoleic acid (c9, t11-CLA) on the cell cycle of gastric cancer cells (SGC-7901) and its possible mechanism in inhibition cancer growth. METHODS: Using cell culture and immunocytochemical techniques, we examined the cell growth, DNA synthesis, expression of PCNA, cyclin A, B(1), D(1), p16(ink4a) and p21(cip/waf1) of SGC-7901 cells which were treated with various c9, t11-CLA concentrations (25, 50, 100 and 200 micromol.L(-1))of c 9, t 11-CLA for 24 and 48h, with a negative control (0.1% ethane). RESULTS: The cell growth and DNA synthesis of SGC-7901 cells were inhibited by c9, t11-CLA.SGC-7901 cells. Eight day after treatment with various concentrations of c9, t11-CLA mentioned above, the inhibition rates were 5.92%, 20.15%, 75.61% and 82.44%, respectively and inhibitory effect of c9, t11-CLA on DNA synthesis (except for 25 micromol.L, 24h) showed significantly less (3)H-TdR incorporation than that in the negative controls (P【0.05 and P【0.01). Immunocytochemical staining demonstrated that SGC-7901 cells preincubated in media supplemented with different c9, t11-CLA concentrations at various times significantly decreased the expressions of PCNA (the expression rates were 7.2-3.0%, 24h and 9.1-0.9% at 48h, respectively), Cyclin A (11.0-2.3%, 24h and 8.5-0.5%,48h), B(1) (4.8-1.8% at 24h and 5.5-0.6% at 48h)and D(1) (3.6-1.4% at 24h and 3.7%-0 at 48h) as compared with those in the negative controls(the expressions of PCNA, Cyclin A, B(1) and D(1) were 6.5% at 24h and 9.0% at 48h, 4.2% at 24h and 5.1% at 48h, 9.5% at 24h and 6.0% at 48h,respectively)(P【0.01), whereas the expressions of P16(ink4a) and P21(cip/waf1), cyclin-dependent kinases inhibitors(CDKI), were increased. CONCLUSION: The cell growth and proliferation of SGC-7901 cell is inhibited by c9, t11-CLA via blocking the cell cycle, with reduced expressions of cyclin A,B(1) and D(1) and enhanced expressions of CDKI(P16(ink4a) and p21(cip/waf1)). 展开更多

AIM To study the role of free fatty acids (arachidonic acid and linoleic acid) in proliferation of rat hepatic stellate cells (HSC). METHODS HSC were isolated and cultured from liver of Wistar rats by in situ perfusion with pronase and collagenase, and density gradient centrifugation with Nycodenz. MTT colorimetric assay was detected for HSC proliferation. RESULTS Arachidonic acid and linoleic acid had an effect on proliferation of HSC. Arachidonic acid of 25mg/L promoted HSC proliferation ( P <0 01), but 50 and 100mg/L had an inhibitory effect on HSC ( P <0 01), and showed cytotoxity on HSC under inverted microscope; 6 25, 12 5 and 25mg/L of linoleic acid had no effect on HSC proliferation ( P >0 05), but with concentration increasing, 50 and 100mg/L of linoleic acid might promote HSC proliferation ( P <0 05 or 0 01). CONCLUSION Arachidonic acid and linoleic acid may promote HSC proliferation, but increased concentration can have cytotoxity on HSC. Arachidonic acid and linoleic acid might be associated with fatty liver and hepatic fibrogenesis by lipid peroxidation. 展开更多

Soybean bioactive peptides(SBPs) were prepared from the isolated soybean protein by proteolysis with an alkaline protease, alcalase, at 50℃ and pH = 8.0. The dependence of hydrolysis time on hydrolysis degree and molecular weight distribution were examined. The hydrolysate was fractionated on a Sephadex G-25 column and the anti-oxidative activities of the fractions were detected by the method of pyrogallol auto-oxidation. The average chain length of soybean peptides that have anti-oxidative activity was estimated to be about 7. The anti-oxidative properties of the soy- bean peptide were also studied by using linoleic acid peroxidation systems. The optimal condition of the peroxidation system was set up, Vc/Cu^2 + as the inducer at pH = 7.4 and 25 ℃. In addition, soybean peptides show higher antioxidative activity compared with GSH. 展开更多

Background: Intramuscular fat(IMF) content is a relevant trait for high-quality meat products such as dry-cured ham,but increasing IMF has the undesirable correlated effect of decreasing lean growth.Thus,there is a need to find selection criteria for IMF independent from lean growth.In pigs,the proportion of linoleic(C18:2) and arachidonic(C20:4) acids decline with fat deposition and therefore they can be considered as indicators of fatness.The aim of this research was to estimate the genetic variation for C18:2 and C20:4 in IMF and their genetic correlations with IMF and lean growth traits,with the objective to assess their potential as specific biomarkers of IMF.The analysis was conducted using a full-pedigreed Duroc resource line with 91,448 records of body weight and backfat thickness(BT) at 180 days of age and 1371 records of fatty acid composition in the muscle gluteus medius.Results: The heritability estimates for C18:2 and C20:4 in IMF,whether expressed in absolute(mg/g of muscle) or in relative(mg/g of fatty acid) terms,as well as for their ratio(C20:4/C18:2),were high(> 0.40),revealing that the C18:2 to C20:4 pathway is subjected to substantial genetic influence.Litter effects were not negligible,with values ranging from8% to 15% of the phenotypic variance.The genetic correlations of C18:2 and C20:4 with IMF and BT were negative(-0.75 to-0.66,for IMF,and-0.64 to-0.36,for BT),if expressed in relative values,but almost null(-0.04 to 0.07),if expressed in absolute values,except for C18:2 with IMF,which was highly positive(0.88).The ratio of C20:4 to C18:2 also displayed a stronger genetic correlation with IMF(-0.59) than with BT(-0.10).Conclusions: The amount of C18:2 in muscle can be used as an IMF-specific biomarker.Selection for the absolute amount of C18:2 is expected to deliver a similar response outcome as selection for IMF at restrained BT.Further genetic analysis of the C18:2 metabolic pathway may provide new insights into differential fat deposition among adipose tissues and on candidate genes for molecular markers targeting specifically for one of them. 展开更多

A 90-day experiment was conducted to investigate the effects of different dietary linoleic acid (LA; 18:2n-6) and linolenic acid ratios (LNA; 18:3n-3) on growth induces, feed utilization and tissue fatty acid profile of freshwater prawn, Macrobrachium rosenbergii post-larvae (PL). The experiment was conducted in cubic indoor fiberglass tanks, each holding 700 L in triplicate. Post-larvae with an average weight of 20.8 ± 0.20 mg were stocked at 80 PL m2. Five experimental isocaloric (15.06 MJ kgl digestible energy), and isonitrogenous (30.45% digestible protein) diets were formulated by blending of soybean oil and linseed oil to containing five dietary LA/LNA ratios (7.80, 2.75, 1.28, 0.65 and 0.30). The highest survival values were recorded for prawn PL fed diet containing 0.65 LA/LAN ratios. Growth indices of PL significantly increased (P 〈 0.05) with decreased dietary LA/LAN ratios to 0.65. The same trend was observed for the highest (P ≤ 0.05) protein efficiency ratio, protein productive value, fat retention, energy retention and best feed conversion ratio. The total whole tissue polyunsaturated fatty acid composition of M. rosenbergii PL was dominated by LA followed by LAN. Post larvae fed the diets containing higher LA/LNA ratios showed a higher tissue LA/LNA ratio. The obtained findings revealed that fatty acid patterns ofM. rosenbergii PL were influenced by fatty acid profiles of diets. The diet containing 0.65 LA/LNA ratio is recommended to obtaining optimum growth performance and feed utilization for M. rosenbergii PL. 展开更多

The aims of this investigation were to isolate active ingredients from the roots/rhizomes of Cyperus articulatus used as herbal medicine in Cameroon for the treatment of human onchocerciasis and to assess the efficacy of the metabolites on the Onchocerca worm.The antifilarial activity was evaluated in vitro on microfilariae(Mfs)and adult worms of the bovine derived Onchocerca ochengi,a close relative of Onchocerca volvulus.Cytotoxicity was assessed in vitro on monkey kidney epithelial cells.The structures of the active compounds were determined using spectroscopic methods and their drug-likeness evaluated using Lipinski parameters.Two secondary metabolites,AMJ1[containing mustakone(1)as the major component]and linoleic acid or(9Z,12Z)-octadeca-9,12-dienoic acid(2)were isolated.Both compounds were found to kill both the microfilariae and adult worms of O.ochengi in a dose dependent manner.The IC50s for AMJ1 were 15.7 lg/mL for Mfs,17.4 lg/mL for adult males and 21.9 lg/mL for adult female worms while for linoleic acid the values were,15.7 lg/mL for Mfs,31.0 lg/mL for adult males and 44.2 lg/mL for adult females.The present report provides the first ever evidence of the anti-Onchocerca efficacy of AMJ1 and linoleic acid.Thus,these secondary metabolites may provide a lead for design and development of new antifilarial agents. 展开更多

Pine wilt disease (PWD), which is caused by pine wood nematodes (PWN), is one of the most serious forest diseases worldwide. To clarify the mechanism of resistance to PWD, we compared metabolites from resistant and susceptible Japanese black pine (Pinus thunbergii) families after inoculation with PWN. After 2 weeks to 1 month post inoculation, the number of PWN dramatically increased in susceptible plants, but not in resistant plants. At this PWN-proliferation phase, ethyl acetate soluble fractions extracted from PWN-inoculated plants were analyzed by gas chromatogramphy-mass spectrometry (GC-MS). Although most compounds were qualitatively and quantitatively similar between resistant and susceptible plants, resistant plants accumulated 2.0-fold more linoleic acid (LA) than susceptible plants. On the other hand, benzoic acid (BA) was barely detected in resistant plants, but it accumulated in susceptible plants as the number of PWN increased. Susceptible plants contained greater levels of the nematicidal compounds pinosylvin and pinosylvin monomethyl ether, compared with resistant plants. These results suggested that LA is involved in the resistance reaction against PWN-proliferation, and that BA could be a good biomarker for PWD. 展开更多

Omega-3(linolenic acid(ALA),docosapentaenoic acid,eicosapentaenoic acid)and omega-6(linoleic acid(LA),arachidonic acid)polyunsaturated fatty acids are essential for health and normal physiological functioning in humans.Here we report a genome-wide association study(GWAS)on LA content in chicken meat.The 19 significant single nucleotide polymorphisms(SNPs)identified by the GWAS approach were annotated in VILL,PLCD1 and OXSR1 genes with highly polymorphic linkage blocks,and explained 4.5%of the phenotypic variation in the LA content.Specifically,the PLCD1 mRNA expression level was negatively correlated with the LA content,and significantly higher in chickens with low LA content than in those with high LA content.In addition,PLCD1 was found to be involved in metabolic pathways,etc.Furthermore,the LA content was correlated with volatile organic compounds(e.g.,octanal,etc.),but no relationship was found with intramuscular fat and triglycerides in chicken meat.The results indicated that there are key SNPs in PLCD1 that regulate the content of LA,and it has no significant effect on fat deposition,but may affect the content of volatile organic compounds(VOCs). 展开更多

Methods GPR40/FFA1 expression was detected by immunofluorescence imaging. We employed linoleic acid (LA), a free fatty acid that has a high affinity to the rat GPR40, and examined its effect on cytosolic free calcium concentration ([Ca2+]i) in primary rat β-cells by Fluo-3 intensity under confocal microscopy recording. Downregulation of GPR40/FFA1 expression by antisense oligonucleotides was performed in pancreatic β-cells, and insulin secretion was assessed by enzyme-linked immunosorbent assay. Results LA acutely stimulated insulin secretion from primary cultured rat pancreatic islets. LA induced significant increase of [Ca2+]i in the presence of 5.6 mmol/L and 11.1 mmol/L glucose, which was reflected by increased Fluo-3 intensity under confocal microscopy recording. LA-stimulated increase in [Ca2+]i and insulin secretion were blocked by inhibition of GPR40/FFA1 expression in β-cells after GPR40/FFA1-specific antisense treatment. In addition, the inhibition of phospholipase C (PLC) activity by U73122, PLC inhibitor, also markedly inhibited the LA-induced [Ca2+]i increase. Conclusion LA activates GPR40/FFA1 and PLC to stimulate Ca2+ release, resulting in an increase in [Ca2+]i and insulin secretion in rat islet β-cells. 展开更多

OBJECTIVE: To assess whether dietary fat intake influences Parkinson’s disease risk. DATA SOURCES: We systematically surveyed the Embase and PubMed databases, reviewing manuscripts published prior to October 2018. The following terms were used:(“Paralysis agitans” OR “Parkinson disease” OR “Parkinson” OR “Parkinson’s” OR “Parkinson’s disease”) AND (“fat” OR “dietary fat” OR “dietary fat intake”). DATA SELECTION: Included studies were those with both dietary fat intake and Parkinson’s disease risk as exposure factors. The Newcastle-Ottawa Scale was adapted to investigate the quality of included studies. Stata V12.0 software was used for statistical analysis. OUTCOME MEASURES: The primary outcomes included the relationship between high total energy intake, high total fat intake, and Parkinson’s disease risk. The secondary outcomes included the relationship between different kinds of fatty acids and Parkinson’s disease risk. RESULTS: Nine articles met the inclusion criteria and were incorporated into this meta-analysis. Four studies scored 7 and the other five studies scored 9 on the Newcastle-Ottawa Scale, meaning that all studies were of high quality. Meta-analysis results showed that high total energy intake was associated with an increased risk of Parkinson’s disease (P = 0.000, odds ratio (OR)= 1.49, 95% confidence interval (CI): 1.26–1.75);in contrast, high total fat intake was not associated with Parkinson’s disease risk (P = 0.123, OR = 1.07, 95% CI: 0.91–1.25). Subgroup analysis revealed that polyunsaturated fatty acid intake (P = 0.010, OR = 1.03, 95% CI: 0.88–1.20) reduced the risk of Parkinson’s disease, while arachidonic acid (P = 0.026, OR = 1.15, 95% CI: 0.97–1.37) and cholesterol (P = 0.002, OR = 1.09, 95% CI: 0.92–1.29) both increased the risk of Parkinson’s disease. Subgroup analysis also demonstrated that, although the results were not significant, consumption of n-3 polyunsaturated fatty acids (P = 0.071, OR = 0.88, 95% CI: 0.73–1.05),α-linolenic acid (P = 0.06, OR = 0.86, 95% CI: 0.72–1.02), and the n-3 to n-6 ratio (P = 0.458, OR = 0.89, 95% CI: 0.75–1.06) were all linked with a trend toward reduced Parkinson’s disease risk. Monounsaturated fatty acid (P = 0.450, OR = 1.06, 95% CI: 0.91–1.23), n-6 polyunsaturated fatty acids (P = 0.100, OR = 1.15, 95% CI: 0.96–1.36) and linoleic acid (P = 0.053, OR = 1.11, 95% CI: 0.94–1.32) intakes were associated with a non-significant trend toward higher PD risk. Saturated fatty acid (P = 0.619, OR = 1.01, 95% CI: 0.87–1.18) intake was not associated with Parkinson’s disease. CONCLUSION: Dietary fat intake affects Parkinson’s disease risk, although this depends on the fatty acid subtype. Higher intake of polyunsaturated fatty acids may reduce the risk of Parkinson’s disease, while higher cholesterol and arachidonic acid intakes may elevate Parkinson’s disease risk. However, further studies and evidence are needed to validate any link between dietary fat intake and Parkinson’s disease. 展开更多

To determine the effect of dietary supplementation with vegetable oilseeds on the composition of bovine milk fatty acids (FAs), 40 Holstein dairy cows were used with a complete randomized design. At the beginning of the experiment, the cows were 150±25 day in milk (DIM). Total duration of the experiment was six weeks. Measurements were made during the last three weeks. Cows in four treatments were fed with a basal diet (CT) or basal diet supplemented with either whole full fat soybean (WFS), full fat expanded soybean (FPS) or whole full fat soybean with whole cottonseed and full fat expanded soybean (MIX). The composition of the milk fat was analyzed by gas chromatography. Relative to control, the conjugated linoleic acid (CLA) concentration in milk fat from cows on FPS was significantly increased by 83.88% (P〈 0.05). The proportions of C12:0 were decreased by 35.7, 35.51, and 38.65% in milk fat from cows on WFS, MIX, and FPS compared with cows on CT. Similar decreases in C 14:0 were 23.83, 24.85, and 31.48% in WFS, MIX, and FPS treatments, respectively. Feeding vegetable oilseeds increased the proportion of healthy FAs (mainly CLA), whereas decreased the concentratiofi of C12:0 and C14:0. Therefore, milk and dairy products would have higher nutritive and therapeutic value. 展开更多

According to the classic "diet-heart" hypothesis, high dietary intake of saturated fats and cholesterol combined with low intake of polyunsaturated fats can increase levels of serum cholesterol and lead to the development of atherogenic plaques and ultimately cardio- vascular diseases. Recently, the beneficial health effects of omega-6 polyunsaturated fatty acids, particularly linoleic acid (LA), on cardiovascular health have been called into question with some scientists suggesting that consumption of LA should be reduced in Western countries. The focus of this critical review is on the controversy surrounding the effects of dietary intake of LA on cardiovascular health. Specifically, we critically examined the effects of both unadulterated and adulterated forms of LA on cardiovascular health outcomes based on findings from epidemiological studies and randomized controlled trials. Additionally, we address common concems surrounding dietary intake of LA regarding its relationship with arachidonic acid, the ratio of omega-6 to omega-3 fatty acids, and its relationship with inflammation. Our critical review indicates that unadulterated forms of LA are cardioprotective and should be consumed as part of a healthy diet. In contrast, abundant evidence now indicates that adulterated forms of LA, predominantly hydrogenated vegetable oils, are atherogenic and should not be considered part of a healthy diet. The ability to adulterate the natural omega-6 fatty acid, LA, has contributed to mixed findings regarding the effects of this fatty acid on cardiovascular health. Thus, it is critical that the source of LA be taken into account when drawing conclusions about the physiological effects of this fatty acid. The findings of the present review are in line with current dietary recommendations of the American Heart Association. 展开更多

The objective of the experiment was to improve the healthy value of milk fatty acids (FA) by feeding sunflower oil (SO) or sunflower seed (SS) combined or not with fish oil (FO) to grazing dairy cows. Four Holstein cows (515 ± 80 kg liveweight) in late lactation (230 days postpartum) fitted with ruminal cannulae were allotted to a 4 × 4 Latin square design with factorial arrangement of treatments: SS = 1.9 kg dry matter (DM)/cow/d of SS;SO = 0.8 kg/cow/d of SO;SS-FO = SS + 0.24 kg/cow/d FO and SO-FO = SO + 0.24 kg/FO. Cows grazed a high quality pasture offered at 11 kg DM/cow/day. After the afternoon milking cows also received 5.6 kg DM/cow of corn silage and during each milking time cracked corn grain (1.3 kg DM/cow) mixed with a mineral-vitamin premix was fed. Oils and SS were introduced via ruminal cannulae and SS was fed roughly grounded. Milk yield tended (p 12:0, 51% for C14:0 and 29% for C16:0. Atherogenicity index (AI) of milk was reduced particularly in SS-FO. Basal concentration of cis-9, trans-11 C18:2 (CLA) in milk was 1.39 g/100g FA and increased (p < 0.05) by 144% across treatments without differences between SS or SO. Feeding FO increased (p < 0.05) milk contents of CLA (2.86 to 3.92 g/100g FA) and linolenic acid. Comparing SO or SS with or without FO showed no changes for neutral detergent fiber (NDF) and crude protein (CP) degradation of pasture. Polyunsaturated fatty acid (PUFA) fed to grazing dairy cows had a marked effect on milk FA profile putatively enhancing its healthy value. 展开更多

This paper also discusses the impact of fatty acids in human metabolism, and the available evidence regarding its risks. Objective: To determine the effect of cis-9, trans-1 1-conjugated linoleic acid on the cell cycle of mammary cancer cells (MCF-7) and the possible mechanism of the inhibitory effect of c9,t11-CLA. Methods: Using cell culture and immunocytochemical techniques, we examined the cell growth, DNA synthesis, expression of PCNA, cyclin A, B1, D1, p16ink4a and p21cip/waf1 of MCF-7 cells at various c9,t11-CLA concentrations (25μM, 50μM, 100μM and 200μM), at 24h and 48h. 96% ethand was used as negative control. Results: The cell growth and DNA synthesis of MCF-7 cells were inhibited by c9,t11-CLA. After treatment with various doses of c9,t11-CLA mentioned above for 8 days, the inhibition frequency was 27.18%, 35.43%, 91.05%, and 92.86%, respectively. Inhibitory effect of c9,t11-CLA on DNA synthesis (except for 25μM, 24h) was demonstrated by significantly less incorporation of 3H-TdR than the negative control (P<0.05 and P<0.01). To further investigate the influence of the cell cycle progression, we found that c9,t11-CLA may arrest the cell cycle of MCF-7 cells. Immunocytochemical staining demonstrated that incubation with different concentration of c9,t11-CLA at various times significantly decreased the expression of PCNA, Cyclin A, B1, D1 in MCF-7 cells compared to the negative control (P<0.01), whereas the expression of p16ink4a and p21cip/waf1, cyclin-dependent kinases inhibitors (CDKI), were increased. Conclusions: The cell growth and proliferation of MCF-7 cells is inhibited by c9,t11-CLA via blocking cell cycle, accompanying reduced expression of cyclin A, B1, D1 and enhanced expression of CDKI (p16ink4a and p21cip/waf1). 展开更多
