One holds that "the local laws of cities with subordinate districts should take precedence over the regulations of provincial governments," while the other supports the exact opposite. This is a value judgment issue in legislation. To reach a solution, we need to clarify the premises based on the characteristics of the laws in question so that a basic common ground can be established for discussion. The first premise for traditional legislation is that a law should be based on experience as well as logic; the second is that the experience of authority subjects, plus the three aspects of logic should outweigh the experience of social subjects, plus the three aspects of logic. With respect to postmodern legislation, the first premise is that experience should override logic, and the second is that the experience of the authority subject should take precedence over that of social subject, with no requirements for logical consistency. Since Legislation Law fal s into the category of postmodern legislation, according to the premises, the argument that the local laws of cities with subordinate districts should take precedence enjoys wider acceptance, but the view is logically challenged in terms of conceptual consistency, system consistency and principle consistency. Regulation is currently perceived as an art and craft of governance where the regulatory processes are an outcome of interdependences between political actors,bureaucrats,business community,and the public at large.How...Regulation is currently perceived as an art and craft of governance where the regulatory processes are an outcome of interdependences between political actors,bureaucrats,business community,and the public at large.However,it is now extensively recognized that this interdependency is beyond one level of government particularly in the era of decentralization where regulatory outcomes are a result of a complex set of interacting levels.This study concedes that regulatory processes are intrinsically political endeavor.Principally,local government regulatory processes are characterized by multiple actors at different levels of governance,i.e.,the local government itself and stakeholders within its jurisdiction and the central government.In fact power dispersion has a vertical direction consisting of actors stemming from different governmental levels as a consequence of decentralization.The complexities of the relationships of multiple actors at different levels of government and across the same level of government create a number of risks that jeopardize quality local regulation that need to be managed calling for multilevel regulatory governance anchored on effective stakeholder engagement and coordination.Particularly,engagement with local government during the design of new regulations is largely poor resulting in a missed opportunity to have informed decisions that enhance quality local regulation.To augment multilevel governance,it is imperative to improve the interface between central and local government with local authorities recognized as“co-producers”of regulatory outcomes.Effective implementation of Regulatory Impact Assessment(RIA)that safeguards proper analysis of proposed and existing regulation is of paramount importance to ensure evidenced based regulations,analysis of cost implications for the local government,the capacity of LGAs in to implement as well as legitimacy of regulations.Therefore this study examines the political tradeoffs between the central and local government and among other stakeholders.展开更多

However, with the transition of economy in China and industrial upgrading in recent years, many labor problems have occurred which do harm to the sustainable development of local economy. The paper researches on the labor market issues of China from the perspective of local labor market regulation. Firstly, it reviews the theories of local labor market regulation. And then the main components of local labor market regulation of China are identified and the evaluation index system is established. The results of analysis of the calculated output show that (1) the local labor market regulation of China has an obvious character of gradient distribution geographically and decreases gradually from east to west; (2) of all the regulations, the regulation of the development of human capital has the most significant impact on local economy currently which is followed by the regulation of labor relation and the regulation of market participation. The pursuit of high energy density has promoted the development of high-performance lithium metal batteries(LMBs).However,the underestimated but non-negligible dendrites of Li anode have been observed to shorten batte...The pursuit of high energy density has promoted the development of high-performance lithium metal batteries(LMBs).However,the underestimated but non-negligible dendrites of Li anode have been observed to shorten battery lifespan.Herein,a composite separator(TiO_(2-x)@PP),in which TiO_(2)with electron-localized oxygen vacancies(TiO_(2-x))is coated on a commercial PP separator,is fabricated to homogenize lithium ion transport and stabilize the lithium anode interface.With the utilization of TiO_(2-x)@PP separators,the symmetric lithium metal battery displays enhanced cycle stability over 800 h under a high current density of 8 m A cm^(-2).Moreover,the LMBs assembled with high-loading LiFePO_(4)(9.24 mg cm^(-2))deliver a stable cycling performance over 900 cycles at a rate of 0.5 C.Comprehensive theoretical studies based on density functional theory(DFT)further unveil the mechanism.The favorable TiO_(2-x)is beneficial for facilitating fast Li+migration and impeding anions transfer.In addressing the Li dendrite issues,the use of TiO_(2-x)@PP separator potentially provides a facile and attractive strategy for designing well-performing LMBs,which are expected to meet the application requirements of rechargeable batteries.展开更多

In a world where caffeinated beverages are evidently the adult drug of choice...The benefits or detriments of caffeine on the human cardiovascular system have not been thoroughly studied and are still poorly understood. In a world where caffeinated beverages are evidently the adult drug of choice (e.g. coffee, energy drinks, soda, tea),?investigating its effects on our bodies is of great importance. In this study,?we examined the effects of caffeine, taken as a tablet, on pressure-flow autoregulation. Young adults between 18 and 21 years of age were the experimental subjects. They were instrumented to monitor systemic arterial blood pressure, peripheral blood flow, calculated peripheral vascular resistance, and the electrocardiogram during an autoregulatory maneuver in the absence and presence of caffeine. Caffeine-mediated vasoconstriction was observed as early as 15 minutes after its consumption. Sixty minutes post-caffeine, vasoconstriction was so prominent that autoregulation was abolished. This was reflected, in part, as a significant reduction in blood flow that accompanied a 3-fold increase in calculated peripheral resistance and a significant increase in systemic arterial pressure. Heart rate was unaffected by caffeine under our experimental conditions. We conclude that caffeine has the ability to inhibit significant cardiovascular properties including pressure-flow autoregulation. Even though more work is needed, the significant caffeine-mediated changes in flow, pressure and resistance during autoregulation could have serious consequences for the cardiovascular system specifically, and for one’s overall health in general.展开更多

As a preliminary study, we investigated the aggregation of an HSV-1 immediate-early protein, infected-cell protein 22 (ICP22), in the nucleus by observing the localization of ICP22-EGFP fusion protein Results showed that, in high-level expression conditions, ICP22-EGFP gradually concentrates in the nucleus, persists throughout the cell cycle without disaggregation even in the cell division phase, and is finally distributed to daughter cells. We subsequently constructed a mammalian cell expression system, which had tetracycline- dependent transcriptional regulators. Consequently, the location of ICP22-EGFP in the nucleus changed with distinct induction conditions. This suggests that the cellular location of ICP22 is also influenced by promoter regulation, in addition to its own structure. Our findings provide new clues for the investigation of transcriptional regulation of viral genes. In addition, the non-protease reporter system we constructed could be utilized to evaluate the role of intemal ribosome entry sites (IRES) on transcriptional regulation.展开更多

ClassⅡhistone deacetylases(cⅡ-HDACs) are localized primarily in the nucleus in unstimulated cardiac myocytes and negatively regulate cardiac hypertrophy by interacting with pro-hypertrophic transcription factors,including myocyte enhancer factor 2(MEF2),calmodulin-binding transcription activator (CAMTA),and nuclear factor activated T cells (NFATs).Nuclear localization of cII-HDACs is regulated not only by phosphorylation but also through oxidation of conserved cysteine residues(Ago et al 2007),suggesting that posttranslational modulation plays an important role in mediating pathological cardiac hypertrophy and heart failure. In this presentation,I will discuss how reactive oxygen species(ROS) are produced in the heart under stress and how ROS regulate the subcellular localization of cII-HDACs,thereby causing pathological hypertrophy.The NADPH oxidase family is a group of transmembrane proteins producing superoxide and hydrogen peroxide.Nox4 is localized primarily in mitochondria,endoplasmic reticulum,and nucleus, whose expression is upregulated by pressure overload and heart failure.Nox4 plays an essential role in mediating increases in ROS in the failing heart.Increased oxidative stress induces oxidation of cysteine resides in cⅡ-HDACs,such as C667 and C669 in HDAC4,which in turn causes nuclear exit of cⅡ-HDACs.The cⅡ-HDACs are further phospho-rylated by the HDAC kinases,including Ca<sup>2+</sup> calmodulin kinase and PKD,leading to prolonged cytoplasmic localization of cⅡ-HDACs and consequent cardiac hypertrophy.Thioredoxin 1(Trx1),an anti-oxidant, reduces the critical cysteine residues in cⅡ-HDACs, thereby restoring the nuclear localization of cⅡ-HDACs and inhibiting pathological hypertrophy. Downregulation of Nox4 enhances nuclear localization of HDAC4,thereby inhibiting cardiac hypertrophy, suggesting that endogenous Nox4 mediates oxidation of HDAC4. In summary,oxidative stress plays an important role in regulating the subcellular localization of cⅡ-HDACs, critical regulators of pathological cardiac hypertrophy.Nox4 is upregulated in response to cardiac hypertrophy and heart failure,thereby playing a major role in mediating oxidative stress in the failing heart.Both Trxl and Nox4 play an essential role in regulating the oxidation status of cⅡ-HDACs, thereby critically regulating pathological hypertrophy and heart failure.I will also discuss pharmacological interventions targeting phosphorylation and/or oxidation of cⅡ-HDACs,which may be effective in treating pathological hypertrophy and heart failure in patients.展开更多

However, with the transition of economy in China and industrial upgrading in recent years, many labor problems have occurred which do harm to the sustainable development of local economy. The paper researches on the labor market issues of China from the perspective of local labor market regulation. Firstly, it reviews the theories of local labor market regulation. And then the main components of local labor market regulation of China are identified and the evaluation index system is established. The results of analysis of the calculated output show that (1) the local labor market regulation of China has an obvious character of gradient distribution geographically and decreases gradually from east to west; (2) of all the regulations, the regulation of the development of human capital has the most significant impact on local economy currently which is followed by the regulation of labor relation and the regulation of market participation. As to the regulation of social security, it has no Significant impact on the development of local economy.
文摘Mitogen-activated protein kinases(MAPKs)are a family of proteins that constitute signaling pathways involved in processes that control gene expression,cell division, cell survival,apoptosis,metabolism,differentiation and motility.The MAPK pathways can be divided into conventional and atypical MAPK pathways.The first group converts a signal into a cellular response through a relay of three consecutive phosphorylation events exerted by MAPK kinase kinases,MAPK kinase,and MAPK.Atypical MAPK pathways are not organized into this three-tiered cascade.MAPK that belongs to both conventional and atypical MAPK pathways can phosphorylate both non-protein kinase substrates and other protein kinases.The latter are referred to as MAPK-activated protein kinases.This review focuses on one such MAPK-activated protein kinase,MAPK-activated protein kinase 5(MK5)or p38-regulated/activated protein kinase(PRAK).This protein is highly conserved throughout the animal kingdom and seems to be the target of both conventional and atypical MAPK pathways.Recent findings on the regulation of the activity and subcellular localization,bona fide interaction partners and physiological roles of MK5/PRAK are discussed.

文摘Yin Yang 1(YY1) is a multi-functional transcription factor that regulates gene expression in a range of cell types,including neurons.It controls neuronal differentiation,as well as neuronal specification and migration during the development of the mammalian nervous system.Besides,YY1 also mediates the transcription of genes that are required for neuronal survival.An impairment of the transcriptional function of YY1 causes neuronal death.This review summarizes recent research findings that unveil the dysfunction of YY1 in multiple neurodegenerative disorders.The expression of disease proteins perturbs the function of YY1 via distinct molecular mechanisms,including recruitment to protein aggregates,protein degradation and aberrant nuclear/cytoplasmic shuttling.Understanding the pathogenic roles of YY1 will further broaden our knowledge of the disease mechanisms in distinct neurodegenerative disorders.

文摘The pursuit of high energy density has promoted the development of high-performance lithium metal batteries(LMBs).However,the underestimated but non-negligible dendrites of Li anode have been observed to shorten battery lifespan.Herein,a composite separator(TiO_(2-x)@PP),in which TiO_(2)with electron-localized oxygen vacancies(TiO_(2-x))is coated on a commercial PP separator,is fabricated to homogenize lithium ion transport and stabilize the lithium anode interface.With the utilization of TiO_(2-x)@PP separators,the symmetric lithium metal battery displays enhanced cycle stability over 800 h under a high current density of 8 m A cm^(-2).Moreover,the LMBs assembled with high-loading LiFePO_(4)(9.24 mg cm^(-2))deliver a stable cycling performance over 900 cycles at a rate of 0.5 C.Comprehensive theoretical studies based on density functional theory(DFT)further unveil the mechanism.The favorable TiO_(2-x)is beneficial for facilitating fast Li+migration and impeding anions transfer.In addressing the Li dendrite issues,the use of TiO_(2-x)@PP separator potentially provides a facile and attractive strategy for designing well-performing LMBs,which are expected to meet the application requirements of rechargeable batteries.
文摘The benefits or detriments of caffeine on the human cardiovascular system have not been thoroughly studied and are still poorly understood. In a world where caffeinated beverages are evidently the adult drug of choice (e.g. coffee, energy drinks, soda, tea),?investigating its effects on our bodies is of great importance. In this study,?we examined the effects of caffeine, taken as a tablet, on pressure-flow autoregulation. Young adults between 18 and 21 years of age were the experimental subjects. They were instrumented to monitor systemic arterial blood pressure, peripheral blood flow, calculated peripheral vascular resistance, and the electrocardiogram during an autoregulatory maneuver in the absence and presence of caffeine. Caffeine-mediated vasoconstriction was observed as early as 15 minutes after its consumption. Sixty minutes post-caffeine, vasoconstriction was so prominent that autoregulation was abolished. This was reflected, in part, as a significant reduction in blood flow that accompanied a 3-fold increase in calculated peripheral resistance and a significant increase in systemic arterial pressure. Heart rate was unaffected by caffeine under our experimental conditions. We conclude that caffeine has the ability to inhibit significant cardiovascular properties including pressure-flow autoregulation. Even though more work is needed, the significant caffeine-mediated changes in flow, pressure and resistance during autoregulation could have serious consequences for the cardiovascular system specifically, and for one’s overall health in general.

文摘Nuclear proteins often form punctiform structures, but the precise mechanism for this process is unknown. As a preliminary study, we investigated the aggregation of an HSV-1 immediate-early protein, infected-cell protein 22 (ICP22), in the nucleus by observing the localization of ICP22-EGFP fusion protein Results showed that, in high-level expression conditions, ICP22-EGFP gradually concentrates in the nucleus, persists throughout the cell cycle without disaggregation even in the cell division phase, and is finally distributed to daughter cells. We subsequently constructed a mammalian cell expression system, which had tetracycline- dependent transcriptional regulators. Consequently, the location of ICP22-EGFP in the nucleus changed with distinct induction conditions. This suggests that the cellular location of ICP22 is also influenced by promoter regulation, in addition to its own structure. Our findings provide new clues for the investigation of transcriptional regulation of viral genes. In addition, the non-protease reporter system we constructed could be utilized to evaluate the role of intemal ribosome entry sites (IRES) on transcriptional regulation.
文摘Oxidative stress plays an essential role in regulating growth and death of cardiac myocytes. ClassⅡhistone deacetylases(cⅡ-HDACs) are localized primarily in the nucleus in unstimulated cardiac myocytes and negatively regulate cardiac hypertrophy by interacting with pro-hypertrophic transcription factors,including myocyte enhancer factor 2(MEF2),calmodulin-binding transcription activator (CAMTA),and nuclear factor activated T cells (NFATs).Nuclear localization of cII-HDACs is regulated not only by phosphorylation but also through oxidation of conserved cysteine residues(Ago et al 2007),suggesting that posttranslational modulation plays an important role in mediating pathological cardiac hypertrophy and heart failure. In this presentation,I will discuss how reactive oxygen species(ROS) are produced in the heart under stress and how ROS regulate the subcellular localization of cII-HDACs,thereby causing pathological hypertrophy.The NADPH oxidase family is a group of transmembrane proteins producing superoxide and hydrogen peroxide.Nox4 is localized primarily in mitochondria,endoplasmic reticulum,and nucleus, whose expression is upregulated by pressure overload and heart failure.Nox4 plays an essential role in mediating increases in ROS in the failing heart.Increased oxidative stress induces oxidation of cysteine resides in cⅡ-HDACs,such as C667 and C669 in HDAC4,which in turn causes nuclear exit of cⅡ-HDACs.The cⅡ-HDACs are further phospho-rylated by the HDAC kinases,including Ca<sup>2+</sup> calmodulin kinase and PKD,leading to prolonged cytoplasmic localization of cⅡ-HDACs and consequent cardiac hypertrophy.Thioredoxin 1(Trx1),an anti-oxidant, reduces the critical cysteine residues in cⅡ-HDACs, thereby restoring the nuclear localization of cⅡ-HDACs and inhibiting pathological hypertrophy. Downregulation of Nox4 enhances nuclear localization of HDAC4,thereby inhibiting cardiac hypertrophy, suggesting that endogenous Nox4 mediates oxidation of HDAC4. In summary,oxidative stress plays an important role in regulating the subcellular localization of cⅡ-HDACs, critical regulators of pathological cardiac hypertrophy.Nox4 is upregulated in response to cardiac hypertrophy and heart failure,thereby playing a major role in mediating oxidative stress in the failing heart.Both Trxl and Nox4 play an essential role in regulating the oxidation status of cⅡ-HDACs, thereby critically regulating pathological hypertrophy and heart failure.I will also discuss pharmacological interventions targeting phosphorylation and/or oxidation of cⅡ-HDACs,which may be effective in treating pathological hypertrophy and heart failure in patients.
文摘By collecting the environmental target constraints in the annual government work report of prefecture-level cities,this paper studies its impact on local enterprises’OFDI.The main conclusions are as follows:(1)Both direct and indirect constraints have a significant positive impact on enterprises’OFDI,and the degree of direct constraints is stronger than that of indirect constraints.(2)Environmental target constraints of local governments will affect corporate OFDI behavior by affecting production costs,local economic development level and local openness,and(3)There are regional heterogeneity and investment type heterogeneity in the impact of environmental goal constraints on corporate OFDI.The suggestions are as follows:government departments can appropriately strengthen the constraint intensity of environmental targets and formulate specific restraint rules for industries with different levels of pollution,so as to effectively make use of the technology spillover effects brought about by OFDI to promote the upgrading of domestic industrial structure.When making OFDI,enterprises should clarify the investment motivation,strengthen corporate social responsibility,and make use of technology spillover effect to promote the upgrading and development of home country industry while being beneficial to their own development.
