Protective effect of long-chain polyunsaturated fatty acids on hepatorenal syndrome in rats

BACKGROUND Hepatorenal syndrome(HRS)is the most prevalent form of acute kidney injury in cirrhotic patients.It is characterized by reduced renal blood flow and represents the most severe complication in cirrhotic patients with advanced disease.Previous research has indicated that antioxidants can delay the onset of a hyperdynamic circulatory state in cirrhosis and improve renal function in HRS patients.Regular omega-3 supplementation has significantly reduced the risk of liver disease.This supplementation could represent an additional therapy for individuals with HRS.AIM To evaluated the antioxidant effect of omega-3 polyunsaturated fatty acid supplementation on the kidneys of cirrhotic rats.METHODS Secondary biliary cirrhosis was induced in rats by biliary duct ligation(BDL)for 28 d.We used 24 male Wistar rats divided into the following groups:I(control);II(treated with omega-3,1 g/kg of body weight);III(BDL treated with omega-3,1 g/kg of body weight);and IV(BDL without treatment).The animals were killed by overdose of anesthetic;the kidneys were dissected,removed,frozen in liquid nitrogen,and stored in a freezer at-80℃for later analysis.We evaluated oxidative stress,nitric oxide(NO)metabolites,DNA damage by the comet assay,cell viability test,and apoptosis in the kidneys.Data were analyzed by one-way analysis of variance,and means were compared using the Tukey test,with P≤0.05.RESULTS Omega-3 significantly decreased the production of reactive oxygen species(P<0.001)and lipoperoxidation in the kidneys of cirrhotic rats treated with omega-3(P<0.001).The activity of the antioxidant enzymes superoxide dismutase and catalase increased in the BDL+omega-3 group compared to the BDL group(P<0.01).NO production,DNA damage,and caspase-9 cleavage decreased significantly in the omega-3-treated BDL group.There was an increase in mitochondrial electrochemical potential(P<0.001)in BDL treated with omega-3 compared to BDL.No changes in the cell survival index in HRS with omega-3 compared to the control group(P>0.05)were observed.CONCLUSION The study demonstrates that omega-3 can protect cellular integrity and function by increasing antioxidant enzymes,inhibiting the formation of free radicals,and reducing apoptosis. 展开更多

为探索热加工过程中鱼肉脂肪氧化形成风味物质的过程,以鱼肉典型脂肪酸--ω-3 LCPUFAs(EPA、DPA、DHA)为代表,分别采用脂肪氧化酶处理和加热处理,采用固相微萃取-气相色谱-质谱联用技术分析。结果显示:经两种方式处理,3种脂肪酸共产生8... 为探索热加工过程中鱼肉脂肪氧化形成风味物质的过程,以鱼肉典型脂肪酸--ω-3 LCPUFAs(EPA、DPA、DHA)为代表,分别采用脂肪氧化酶处理和加热处理,采用固相微萃取-气相色谱-质谱联用技术分析。结果显示:经两种方式处理,3种脂肪酸共产生8大类107种风味特性各不相同的成分,其中具有土腥味和鱼腥味等鱼肉特征风味成分包括2-辛酮、2-壬酮、3-辛烯-2-酮、反-2-戊醇、3-辛醇、1-辛烯-3-醇、十二醇。产物中含量最大的成分是芳香族化合物,其它产物含量大致顺序为醛、酮、醇或烷烃、呋喃或酯,脂肪氧化型产物(醛、酮、醇、呋喃)占总含量的6.0%~63.7%。3种脂肪酸产物总含量顺序为EPA>DHA>DPA,推测3种脂肪酸稳定性顺序为DPA>DHA>EPA。两种处理方式相比,加热处理形成比例更高的脂肪氧化型产物,而低温脂肪氧化酶处理主要形成芳香族化合物。提示受热作用更利于鱼肉形成风味良好的产物。 展开更多

Objective To assess the net health effect caused by the consumption of specific marine species based on Benefit-Risk Analysis for Foods (BRAFO)-tiered approach. Methods Twenty species were collected from the Zhoushan Archipelago, China. Concentrations of n-3 long-chain polyunsaturated fatty acids, methyl mercury (MeHg), and dioxin-like compounds (DLCs) in the samples were analyzed for benefit risk assessment based on BRAFO-tiered approach. Results Based on the BRAFO-tiered approach, reference scenario (no intake) and alternative scenario (intake of specific species of 200 g/week) were determined. The exposure to MeHg/DLCs via alternative scenario of all studied species did not exceed provisional tolerable weekly/monthly intake. However, the adult population with high DLCs exposure in China would significantly exceed the upper limit of DLCs via an additional alternative scenario of some species such as Auxis thazard. The results of deterministic computation showed that alternative scenario of all studied species generated clear net beneficial effects on death prevention and child IQ gain. Conclusion The alternative scenario of all studied species could be recommended to population with average DLCs exposure, and the reference scenario of species with relatively high DLCs concentration could be recommended to population exposed to high DLCs. 展开更多

Elovl4 is a fatty acyl elongase which participates in the biosynthesis of long-chain polyunsaturated fatty acids(LCPUFA).Herein we isolated and functionally characterized the elovl4a gene in the large yellow croaker Larimichthys crocea,and investigated the regulatory effects of transcription factors Hnf4α,Lxrα,Pparαon the activity of elovl4a promoter.Tissue expression pattern revealed that elovl4a was widely expressed in several tissues and predominantly in brain.Heterologous expression in yeast showed that L.crocea Elovl4a could effectively elongate both C18 and C20 PUFA substrates to C22 fatty acid.In addition,the affinity of large yellow croaker Elovl4a to n-6 series fatty acids is weak and cannot prolong 18:2n-6 and 18:3n-6.Moreover,L.crocea elovl4a reporter activities were elevated by 1.31-,1.39-and 1.48-fold via over-expression of Lxrα,Pparαand Hnf4α,respectively.The above findings can enrich the knowledge of biosynthesis pathway of LC-PUFA in the large yellow croaker and contribute to elucidate the LC-PUFA anabolism mechanism in fish. 展开更多
