AIM: To investigate how hepatitis C virus (HCV) G1b infection influences the particle number of lipoproteins.METHODS: The numbers of lipoprotein particles in fasting sera from 173 Japanese subjects, 82 with active HCV G1b infection (active HCV group) and 91 with cleared HCV infection (SVR group), were examined. Serum lipoprotein was fractionated by high-performance liquid chromatography into twenty fractions. The cholesterol and triglyceride concentrations in each fraction were measured using LipoSEARCH. The number of lipoprotein particles in each fraction was calculated using a newly developed algorithm, and the relationship between chronic HCV G1b infection and the lipoprotein particle number was determined by multiple linear regression analysis.RESULTS: The median number of low-density lipoprotein (LDL) particles was significantly lower in the active HCV group [1182 nmol/L, interquartile range (IQR): 444 nmol/L] than in the SVR group (1363 nmol/L, IQR: 472 nmol/L, P &#x0003c; 0.001), as was that of high-density lipoprotein (HDL) particles (14168 nmol/L vs 15054 nmol/L, IQR: 4114 nmol/L vs 3385 nmol/L, P = 0.042). The number of very low-density lipoprotein (VLDL) particles was similar between the two groups. Among the four LDL sub-fractions, the number of large LDL particles was similar between the two groups. However, the numbers of medium (median: 533.0 nmol/L, IQR: 214.7 nmol/L vs median: 633.5 nmol/L, IQR: 229.6 nmol/L, P &#x0003c; 0.001), small (median: 190.9 nmol/L, IQR: 152.4 nmol/L vs median: 263.2 nmol/L, IQR: 159.9 nmol/L; P &#x0003c; 0.001), and very small LDL particles (median: 103.5 nmol/L, IQR: 66.8 nmol/L vs median: 139.3 nmol/L, IQR: 67.3 nmol/L, P &#x0003c; 0.001) were significantly lower in the active HCV group than in the SVR group, respectively. Multiple linear regression analysis indicated an association between HCV G1b infection and the decreased numbers of medium, small, and very small LDL particles. However, active HCV infection did not affect the number of large LDL particles or any sub-fractions of VLDL and HDL particles.CONCLUSION: HCV G1b infection decreases the numbers of medium, small, and very small LDL particles. 展开更多

BACKGROUND Cholesterol is related to improvements in the rate of sustained virological response and a robust immune response against the hepatitis C virus(HCV).APOE gene polymorphisms regulate cholesterol levels modifying the course of the HCV infection.The relationship between cholesterol,APOE alleles,and the outcome of HCV infection has not been evaluated in the admixed population of Mexico.AIM To investigate the role of APOE-ε2,-ε3,and-ε4 alleles and the metabolic profile in the outcome of HCV infection.METHODS A total of 299 treatment-na?ve HCV patients were included in this retrospective study.Patients were stratified in chronic hepatitis C(CHC)(n=206)and spontaneous clearance(SC)(n=93).A clinical record was registered.Biochemical tests were assessed by dry chemistry assay.APOE genotypes were determined using a Real-Time polymerase chain reaction assay.RESULTS Total cholesterol,low-density lipoprotein cholesterol(LDL-c),triglycerides,and hypercholesterolemia were higher in SC than CHC patients as well as the frequency of the APOEε4 allele(12.4%vs 7.3%).SC patients were overweight(54.8%).Theε4 allele was associated with SC(OR=0.55,95%CI:0.31-0.98,P=0.042)and mild fibrosis(F1-F2)in CHC patients(OR 0.091,95%CI 0.01-0.75,P=0.020).LDL-c≥101.5 mg/dL(OR=0.20,95%CI:0.10-0.41,P<0.001)and BMI≥26.6 kg/m2(OR=0.37,95%CI:0.18-0.76,P<0.001)were associated with SC status;while ALT≥50.5 IU/L was negatively associated(OR=5.67,95%CI:2.69-11.97,P<0.001).CONCLUSION In SC patients,the APOEε4 allele and LDL-c conferred a protective effect in the course of the HCV infection in the context of excess body weight. 展开更多

Heparin has been considered to be a potentially useful ligand for low-density lipoprotein(LDL) detection and analysis in a clinical context.In order to construct an affinity surface for preferential adsorption of LDL,heparin-modified gold surface(GS-Hep) was fabricated by a self-assembling method and hydrophobic-modified gold surfaces(GS-Hydro) was used as a control.The morphologies of the modified gold surfaces were investigated by atomic force microscopy(AFM) and the quantity of heparin bound to gold surface was assayed by the toluidine blue(TB) colorimetric method.Water contact angles were determined to investigate wettability on GS-Hep and GS-Hydro.Surface plasmon resonance(SPR) technique was used subsequently to detect the selective binding of LDL with heparin.And the investigation on the effect of pH on LDL adsorption suggests that lower pH lead to higher quantities of LDL adsorption on GS-Hep.Compared with GS-Hydro,GS-Hep is selective for LDL from both single and binary protein solutions.Moreover,adsorbed LDL on GS-Hep could be washed off by injecting elution solution,such as NaCl solution,for the purpose of the regeneration of GS-Hep for further LDL adsorption. 展开更多

目的探讨Essen卒中风险评分联合氧化型低密度脂蛋白胆固醇(Oxidized low-density lipoprotein,OX-LDL)、血清泛素C末端水解酶L1(Ubiquitin carboxy-terminal hydrolase-L1,UCH-L1)对非房颤人群首发急性缺血性脑卒中(Acute ischemic stro... 目的探讨Essen卒中风险评分联合氧化型低密度脂蛋白胆固醇(Oxidized low-density lipoprotein,OX-LDL)、血清泛素C末端水解酶L1(Ubiquitin carboxy-terminal hydrolase-L1,UCH-L1)对非房颤人群首发急性缺血性脑卒中(Acute ischemic stroke,AIS)患者短期预后的预测价值。方法根据改良Rankin量表(Modified rankin scale,mRS)评分,将132例非房颤人群首发AIS患者分为预后良好组(mRS≤2分)93例,预后不良组(mRS>2分)39例,在入院治疗前进行Essen卒中风险评分和血清OX-LDL、UCH-L1检测。采用Spearman相关性分析Essen卒中风险评分与OX-LDL、UCH-L1水平的差异,采用Logistic回归分析预后不良的危险因素,采用受试者工作特征(Receiver operating characteristic,ROC)曲线分析Essen卒中风险评分、OX-LDL、UCH-L1水平及三者联合对非房颤人群首发AIS预后评估的效果。结果预后不良组Essen卒中风险评分高于预后良好组(Z=-5.365,P<0.001),血清中OX-LDL、UCH-L1水平高于预后良好组(Z=-6.152,P<0.001;Z=-7.020,P<0.001)。血清中OX-LDL、UCH-L1水平与Essen评分呈正相关(r=0.629,P<0.001;r=0.598,P<0.001)。Logistic回归分析显示,Essen卒中风险评分、OX-LDL、UCH-L1是脑卒中预后的独立影响因素。Essen卒中风险评分联合OX-LDL、UCH-L1对非房颤人群首发AIS预后的ROC曲线下面积为0.896,灵敏度为88.54%,特异度为92.93%。结论Essen卒中风险评分联合OX-LDL、UCH-L1对非房颤人群首发AIS预后有良好的预测作用,Essen卒中风险评分、OX-LDL、UCH-L1水平越高,提示预后不良的风险越大。 展开更多

Objectives Bai Ku Yao(White-trousers Yaos)is a special branch of Yao minority in China.They are now living in both Lihu and Baxu villages,Nandan County, Guangxi,China.The population size is about 30,000.The special customs and culture of Bai Ku Yao,including their special clothing,intra-ethnic marriages and alcohol intake are still completely conserved to the present day.In previous epidemiologic studies,we found that the serum lipid levels and the prevalence of hyperlipidaemia were lower in Bai Ku Yao than in Han Chinese from the same region.This ethnic difference in serum lipid profiles is still not well known.We hypothesized that there may be significant differences in some genetic polymorphismsssociation of low density lipoprotein receptor (LDL-R) genepolymorphism and several environmental factors with serum lipid levels in the Guangxi Bai Ku Yao and Han populations.Methods A total of 1024 subjects of Bai Ku Yao and 792 participants of Han Chinese were stud- ied by a stratified randomized cluster sampling.Epidemiological survey was carried out using internationally standardized methods.Information on demographics,socioeconomic status, and lifestyle factors was collected with standardized questionnaires. The height,weight,waist circumference,blood pressure, and serum total cholesterol(TC),triglyceride(TG), high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),apolipoprotein(Apo) A1, and ApoB were measured.Body massindex(BMI,kg/m2) was calculated.Genotyping of the LDL-RAvaⅡwas performed by polymerse chain reaction and restriction fragment length polymorphism combined with gel electrophoresis,and then confirmed by direct sequencing.Results(l)The height,weight,serum TC,HDL-C,LDL-C,ApoAl levels and the ratio of ApoAl to ApoB were lower in Bai Ku Yao than in Han Chinese(P【0.01 for all),whereas the percentage of subjects who consumed alcohol or smoked cigarettes was higher in Bai Ku Yao than in Han Chinese(P【0.01 for each).(2) The frequency of A+ allele in Bai Ku Yao was 34.5%,and the frequencies of A-A-,A-A+ and A+A + genotypes were 42.6%,45.9%and 11.5%;respectively. The frequency of A+ allele in Han Chinese was 19.3%(P【0.001),and the frequencies of A-A-,A-A + and A+A+ genotypes were 64.9%,31.6%and 3.5%(P【0.001);respectively. The frequencies of A-A-,A-A+ and A+A+ genotypes in Bai Ku Yao were significant difference between males and females,between normal TC and high TC subgroup, and between normal LDL-C and high LDL-C subgroup (P【0.05 for all),whereas the frequencies of A- and A+ ? alleles in Han Chinese were significant difference between males and females(P【0.05).(3) Serum LDL-C levels in Bai Ku Yao were significant difference among the A-A-, A-A+ and A+A+ genotypes(P【0.05),the A+ carriers had higher serum LDL-C levels.Serum HDL-C levels in Han Chiese were significant difference among the A-A-,A-A + and A+A+ genotypes(P【0.01),the A+ carriers had higher serum HDL-C levels.(4) After adjusting other factors,the prevalence of LDL-C abnormality was still higher in Han Chiese than in Bai Ku Yao.The prevalence of TC abnormality in Han Chinese was almost twice high as in Bai Ku Yao. The age and diet were common risk factor for TC abnormality. No effect of AvaⅡgenotype or alcohol consumption on the TC abnormality was found,but the combination of geno-type and alcohol consumption can increase the prevalence of TC abnormality[Exp(B) =(1.154)].Age was negatively cor- related with TG level.Conclusions Serum TC and LDL-C levels were lower in Bai Ku Yao than in Han Chinese.There were significant differences in the AvaⅡallele and genotype frequencies between the he A+ carriers in Bai Ku Yao had higher serum LDL-C levels,whereas the A+ carriers in Han had higher serum HDL-C levels.Interactions between alcohol consumption or cigarette smoking and the LDL-R AvaⅡgenotype were also observed.The differences in the serum lipid profiles between the two ethnic groups might partly result from different genotypic frequency of LDL-R AvaⅡpolymorphism or differentgene-enviromental interactions.Bai Ku Yao and Han population,the frequency of A + allele was higher in Bai Ku Yao than in Han.T between the two ethnic groups.Therefore,the aim of the present study was to detect the 展开更多

目的比较低密度脂蛋白胆固醇(LDL-C)和非高密度脂蛋白胆固醇(non-HDL-C)在家族性高三酰甘油血症(FHTG)和家族性混合型高脂血症(FCHL)鉴别诊断中的应用价值。方法选取9个FHTG家系共94人、24个FCHL家系共94人,根据血脂异常情况将2个家系... 目的比较低密度脂蛋白胆固醇(LDL-C)和非高密度脂蛋白胆固醇(non-HDL-C)在家族性高三酰甘油血症(FHTG)和家族性混合型高脂血症(FCHL)鉴别诊断中的应用价值。方法选取9个FHTG家系共94人、24个FCHL家系共94人,根据血脂异常情况将2个家系再分为受累组和未受累组;另设10个对照家系(共57人)。比较各家系血清三酰甘油(TAG)、总胆固醇(TC)、HDL-C、LDL-C、non-HDL-C等血脂指标水平;FHTG家系和FCHL家系按TAG水平分层后,分析non-HDL-C和LDL-C的相关性;最后用受试者工作特征曲线(ROC曲线)分析比较nonHDL-C和LDL-C用于FHTG家系和FCHL家系鉴别诊断的cut-off值。结果 FHTG家系受累组TAG、TC、nonHDL-C水平均高于未受累组和对照组(P<0.01或P<0.05);FCHL家系受累组TAG、TC、HDL-C、LDL-C、nonHDL-C水平均高于未受累组和对照组(P<0.01或P<0.05);FHTG家系受累组TAG水平显著高于FCHL家系受累组(P<0.01),而TC、HDL-C、LDL-C、non-HDL-C水平低于FCHL家系(P<0.01或P<0.05)。FHTG家系和FCHL家系内non-HDL-C和LDL-C均呈正相关,但随TAG水平升高其相关性均呈下降趋势;LDL-C和non-HDL-C用于FHTG家系和FCHL家系鉴别诊断的最佳cut-off值分别为3.575、4.525mmol/L。结论除常规血脂指标外,non-HDL-C可能是鉴别诊断FHTG家系和FCHL家系的新指标;其鉴别诊断价值可能优于LDL-C。 展开更多

目的探讨同型半胱氨酸(Hcy)、C反应蛋白(CRP)、小而密低密度脂蛋白胆固醇(sdLDL-C)及血脂相关指标在不同脑卒中患者血清中的表达及诊断价值。方法选取脑卒中患者652例为研究对象,其中脑梗死患者393例(脑梗死组),脑出血患者259例(脑出血... 目的探讨同型半胱氨酸(Hcy)、C反应蛋白(CRP)、小而密低密度脂蛋白胆固醇(sdLDL-C)及血脂相关指标在不同脑卒中患者血清中的表达及诊断价值。方法选取脑卒中患者652例为研究对象,其中脑梗死患者393例(脑梗死组),脑出血患者259例(脑出血组),另选取健康体检者120例作为对照组。分别比较3组一般资料及血清sdLDL-C、Hcy、CRP、总胆固醇、三酰甘油、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、载脂蛋白A-Ⅰ(ApoA-Ⅰ)、载脂蛋白B水平。采用受试者工作特征曲线分析sdLDL-C、Hcy、CRP联合检测及各指标单独检测诊断脑出血与脑梗死的效能。结果脑梗死组、脑出血组LDL-C、CRP、sdLDL-C、Hcy水平均高于对照组(P<0.05),HDL-C、ApoA-Ⅰ水平均低于对照组(P<0.05)。Hcy、CRP和sdLDL-C 3项指标联合检测诊断脑梗死的曲线下面积(AUC),为0.965,灵敏度为83.5%,特异度为99.0%,高于其他各指标单独检测。Hcy、CRP和sdLDL-C 3项指标联合检测诊断脑出血的AUC为0.962,灵敏度为85.2%,特异度为99.0%,高于其他各指标单独检测。结论与单一指标相比,Hcy、CRP、sdLDL-C 3项指标联合检测诊断脑出血、脑梗死的效能更高,可在临床进一步推广应用。 展开更多

目的:分析动脉粥样硬化性脑梗死(atherosclerotic cerebral infarction,ASCI)患者血清超敏C反应蛋白(hypersensitive c-reactive protein,hs-CRP)、载脂蛋白-B(apolipoprotein-B,Apo-B)、氧化低密度脂蛋白(oxidized low-density lipopro... 目的:分析动脉粥样硬化性脑梗死(atherosclerotic cerebral infarction,ASCI)患者血清超敏C反应蛋白(hypersensitive c-reactive protein,hs-CRP)、载脂蛋白-B(apolipoprotein-B,Apo-B)、氧化低密度脂蛋白(oxidized low-density lipoprotein,ox-LDL)及基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)水平与颈动脉粥样硬化性斑块(carotid atherosclerotic plaques,CAP)形成的相关性。方法:选取本院神经内科确诊的125例ASCI患者为观察组,另同期选取125例健康者为对照组。比较两组血清hs-CRP、Apo-B、ox-LDL及MMP-9水平,并对病例组行颈动脉超声检查,分析血清hs-CRP、Apo-B、ox-LDL及MMP-9水平与ASCI患者CAP形成的相关性。结果:与对照组相比较,观察组血清hs-CRP、Apo-B、ox-LDL及MMP-9水平显著升高(P<0.01)。观察组中伴CAP形成的患者血清hs-CRP、Apo-B、ox-LDL及MMP-9水平比无CAP形成的患者显著升高(P<0.01)。经多因素Logistic回归分析结果显示,血清hs-CRP(OR=4.76,95%CI:2.35~9.18)、Apo-B(OR=3.16,95%CI:1.59~7.32)、ox-LDL(OR=1.48,95%CI:1.15~2.01)及MMP-9(OR=3.86,95%CI:1.63~9.14)水平是ASCI患者CAP形成的独立危险因素(P<0.05)。结论:血清hs-CRP、Apo-B、ox-LDL及MMP-9的水平可反映ASCI患者CAP的炎症程度以及斑块的不稳定程度;ASCI患者血清hs-CRP、Apo-B、ox-LDL及MMP-9水平显著升高,其与ASCI患者CAP的形成密切相关,可作为临床诊断ASCI及CAP形成的重要血清学生物标志物。 展开更多

目的探讨急性脑梗死(ACI)患者低密度脂蛋白胆固醇(LDL-C)、同型半胱氨酸(Hcy)、新蝶呤(Npt)、C反应蛋白(CRP)水平的改变及其与颈动脉粥样硬化的关系。方法 2014年6月—2015年6月诊治ACI死患者68例为ACI组,选取同期行健康检查的体检者60... 目的探讨急性脑梗死(ACI)患者低密度脂蛋白胆固醇(LDL-C)、同型半胱氨酸(Hcy)、新蝶呤(Npt)、C反应蛋白(CRP)水平的改变及其与颈动脉粥样硬化的关系。方法 2014年6月—2015年6月诊治ACI死患者68例为ACI组,选取同期行健康检查的体检者60例为健康对照组,应用多普勒超声仪测定2组颈动脉内膜中层厚度(IMT),ELISA法测定2组血浆Hcy、Npt,酶直接法测定血浆LDL-C,乳胶免疫比浊法测定CRP。结果 ACI组患者颈动脉斑块发生率为91.17%,显著高于健康对照组10.00%(P<0.05);ACI组血浆LDL-C(4.98±0.89)mmol/L、Hcy(38.96±4.89)μmol/L、Npt(2.45±0.48)ng/ml、CRP(6.89±0.48)mg/L水平高于健康对照组(2.86±0.93)mmol/L、(18.02±3.88)μmol/L、(1.38±0.59)ng/ml、(1.58±0.56)mg/L(P<0.05),而颈动脉IMT(1.85±0.32)mm大于健康对照组(0.42±0.16),差异均有统计学意义(P<0.05)。ACI患者中软斑亚组、混合斑亚组血浆LDLC、Hcy、Npt、CRP水平显著高于硬斑亚组(P<0.05)。经Pearson单因素分析,血浆LDL-C、Hcy、Npt、CRP水平与IMT呈正相关(r=0.412、0.326、0.345、0.369,P<0.05)。结论 ACI合并颈动脉不稳定斑块患者血浆LDL-C、Hcy、Npt、CRP水平显著高于斑块不稳定患者,提示血浆LDL-C、Hcy、Npt、CRP水平可反映ACI患者颈动脉粥样硬化稳定性。 展开更多

目的探讨中国南部地区表观健康人群脂蛋白相关磷脂酶A2(Lp-PLA2)水平分布及其与低密度脂蛋白胆固醇(LDL-C)、高敏C反应蛋白(hs-CRP)的相关性。方法选取中国南部地区8家三级甲等医院健康体检者2318名,检测血清Lp-PLA2及血脂[总胆固醇(TC... 目的探讨中国南部地区表观健康人群脂蛋白相关磷脂酶A2(Lp-PLA2)水平分布及其与低密度脂蛋白胆固醇(LDL-C)、高敏C反应蛋白(hs-CRP)的相关性。方法选取中国南部地区8家三级甲等医院健康体检者2318名,检测血清Lp-PLA2及血脂[总胆固醇(TC)、三酰甘油(TG)、LDL-C、高密度脂蛋白胆固醇(HDL-C)]、hs-CRP水平,将所有对象按性别和年龄(<50岁、50~59岁、60~69岁、70~79岁、≥80岁)分别分组。采用Pearson相关分析或Spearman相关分析评估各项目之间的相关性。结果中国南部地区2318名表观健康者血清Lp-PLA2总体水平为(145.84±43.96)ng/mL,参考区间上限为217.93 ng/mL。女性血清Lp-PLA2水平低于男性(P<0.05)。<50岁组血清Lp-PLA2水平低于50~59岁组、60~69岁组及70~79岁组(P<0.05),与≥80岁组差异无统计学意义(P>0.05);50~59岁组、60~69岁组及70~79岁组之间血清Lp-PLA2水平差异均无统计学意义(P>0.05)。8家医院中,福建医科大学附属协和医院的血清Lp-PLA2检测结果最低,海南省人民医院的检测结果最高,与其他医院比较,差异均有统计学意义(P<0.05)。相关分析结果显示,Lp-PLA2与LDL-C呈正相关(r=0.163,P<0.05),与TC、TG、HDL-C和hs-CRP均无相关性(r值分别为-0.028、0.000、0.001、0.088,P>0.05)。结论初步建立了中国南部地区表观健康人群血清Lp-PLA2的参考区间。 展开更多

目的研究木豆叶、鲜地黄提取物(DMT)对氧化低密度脂蛋白(ox-LDL)诱导损伤的人脐静脉内皮细胞(HUVEC)的影响,初步探讨DMT防治动脉粥样硬化的机制。方法应用血清药理学方法制备正常对照、辛伐他汀、DMT血清,以供细胞实验使用。采用ox-LDL... 目的研究木豆叶、鲜地黄提取物(DMT)对氧化低密度脂蛋白(ox-LDL)诱导损伤的人脐静脉内皮细胞(HUVEC)的影响,初步探讨DMT防治动脉粥样硬化的机制。方法应用血清药理学方法制备正常对照、辛伐他汀、DMT血清,以供细胞实验使用。采用ox-LDL损伤HUVEC,与各组药物共培养,即空白组、模型组、辛伐他汀组(15%辛伐他汀血清)及DMT低(5%DMT血清)、中(10%DMT血清)、高剂量组(15%DMT血清),24 h后MTT法检测细胞活力;收集细胞,流式细胞术检测细胞内活性氧(ROS)含量;硝酸还原酶法检测上清液NO含量;ELISA法检测上清液内皮素(ET-1)、细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)含量;Western Blot法检测细胞凝集素样氧化低密度脂蛋白受体(Lox-1)、Caspase-3、Cytc蛋白的表达。结果与模型组比较,辛伐他汀组及DMT低、中、高剂量组细胞活力明显提高(P<0.01);DMT低剂量组ROS水平明显降低(P<0.05);辛伐他汀组细胞上清液ET-1、VCAM-1含量及Lox-1、Caspase-3、Cytc蛋白含量明显降低(P<0.05,P<0.01);DMT低剂量组细胞Caspase-3、Cytc蛋白含量明显降低(P<0.01);DMT中剂量组细胞上清液ET-1、VCAM-1含量及Lox-1、Caspase-3、Cytc蛋白含量明显降低(P<0.05,P<0.01),细胞上清液中NO含量明显升高(P<0.01);DMT高剂量组细胞上清液ET-1、VCAM-1含量及Lox-1、Cytc蛋白含量明显降低(P<0.01),NO含量明显升高(P<0.01)。结论DMT能够抑制ox-LDL诱导的HUVEC损伤,恢复其收缩舒张分泌功能,其机制可能与影响Lox-1介导的内皮细胞氧化应激有关。 展开更多
