青花椒雄蕊调控基因ZaPI的克隆、鉴定与表达模式分析

【目的】探究青花椒雄蕊分化关键B类基因及其核酸和蛋白序列特性,并分析其在雌雄花分化过程的表达模式,为青花椒花芽性别分化的遗传调控机制提供候选基因和数据支撑。【方法】基于青花椒转录组和基因组数据库,提取雌雄花分化过程中差异... 【目的】探究青花椒雄蕊分化关键B类基因及其核酸和蛋白序列特性,并分析其在雌雄花分化过程的表达模式,为青花椒花芽性别分化的遗传调控机制提供候选基因和数据支撑。【方法】基于青花椒转录组和基因组数据库,提取雌雄花分化过程中差异表达的所有B类MADS-box家族基因,并分析获取关键成员;克隆关键成员基因编码序列,全面剖析核酸和蛋白序列特性;并解析其在11种不同组织器官以及雌雄花分化过程的表达模式。【结果】青花椒雌雄花分化过程的差异基因中共有11个B类MADS-box家族基因,其中Zardc17043(ZaPI)的表达量最高,且在雄花中极显著特异表达。该ZaPI编码序列全长624 bp,共编码207个氨基酸,含有77个氨基酸组成的MADS-box结构域和84个氨基酸组成的K-box结构域,蛋白相对分子量为24.29 kD,是位于11号染色体包含4个外显子和3个内含子且定位于细胞核的亲水性蛋白;系统进化分析显示,青花椒ZaPI与同为芸香科(Rutaceae)的克莱门柚(Citrus clementina)和甜橙(Citrus sinensis)亲缘关系最近。此外,青花椒ZaPI基因编码序列共预测到54个转录因子结合位点,主要涉及植物生殖生长和抗逆过程等多种调控途径。顺式作用元件分析发现,ZaPI启动子区富含光信号、植物激素、逆境胁迫等相关的顺式作用元件。同时,该基因在根茎叶刺等营养器官中几乎不表达,雌花分化过程的表达量也相对较低,但在雄花分化过程中呈极显著上调趋势。【结论】研究所获得的ZaPI基因与雄蕊的分化显著相关,可能与植物激素等多种信号分子互作参与了青花椒的雄蕊形成过程。研究所获结果对于进一步探究青花椒雄蕊形成的分子机制,培育高产青花椒种质资源提供了丰富的理论支撑。 展开更多

结直肠癌组织中MAD2L1的表达及与预后的关系

目的探讨有丝分裂阻滞缺陷蛋白2(mitotic arrest deficient 2-like 1,MAD2L1)在结直肠癌组织中的表达及与预后的关系。方法收集259例结直肠癌患者的临床病理资料,采用免疫组化SP法检测结直肠癌组织中MAD2L1的表达,分析MAD2L1表达与结直... 目的探讨有丝分裂阻滞缺陷蛋白2(mitotic arrest deficient 2-like 1,MAD2L1)在结直肠癌组织中的表达及与预后的关系。方法收集259例结直肠癌患者的临床病理资料,采用免疫组化SP法检测结直肠癌组织中MAD2L1的表达,分析MAD2L1表达与结直肠癌临床病理特征的关系及其对患者5年生存期的影响。结果TCGA和GTEx数据库分析结果显示,结直肠癌组织中MAD2L1 mRNA表达量显著高于癌旁组织;免疫组化结果显示,结直肠癌组织中MAD2L1表达水平均高于癌旁正常组织。259例结直肠癌组织中148例呈MAD2L1高表达,111例呈低表达。生存分析结果显示,MAD2L1低表达组患者5年生存率高于高表达组(67.57%vs 49.32%);按病变部位分析,MAD2L1高表达的结肠癌和直肠癌患者5年生存率均低于低表达患者(47.25%vs 73.21%;52.53%vs 65.38%)。结论MAD2L1在结直肠癌组织中高表达,可能与患者预后不良有关,是预测结直肠癌的潜在标志物。 展开更多

目的 探讨干扰有丝分裂阻滞缺陷2样蛋白1(MAD2L1)在结直肠癌组织中的表达及与免疫浸润的关系。方法 基于TCGA数据库分析结直肠癌组织及正常结直肠组织中MAD2L1的表达差异,生存分析评价MAD2L1表达水平与患者预后的关系,采用临床相关性分... 目的 探讨干扰有丝分裂阻滞缺陷2样蛋白1(MAD2L1)在结直肠癌组织中的表达及与免疫浸润的关系。方法 基于TCGA数据库分析结直肠癌组织及正常结直肠组织中MAD2L1的表达差异,生存分析评价MAD2L1表达水平与患者预后的关系,采用临床相关性分析研究结直肠癌与临床资料的相关性。使用Timer数据库gene模块分析探究MAD2L1与免疫细胞群之间的关系。使用correlation模块评估与免疫检查点水平的相关性。使用correlation模块进行免疫细胞浸润的关联分析,分析结直肠癌组织中MAD2L1的表达与免疫细胞浸润的关系。结果 与正常组织相比,结直肠癌组织中MAD2L1表达上调;MAD2L1高表达组与低表达组结直肠癌患者相比,N分期患者比例不同,差异具有统计学意义(P<0.05)。预后分析显示,MAD2L1高表达组的患者生存期、无进展生存期均高于低表达组的患者,差异具有统计学意义(P<0.05)。多因素COX回归分析显示,年龄和T分期可作为影响结直肠癌患者预后的独立预后因子(P<0.05)。MAD2L1与结直肠癌中CD8^(+)T细胞(结肠癌:r=0.315,P<0.001;直肠癌r=0.352,P<0.001)、嗜中性粒细胞(结肠癌:r=0.195,P<0.001;直肠癌:r=0.278,P<0.001)浸润水平均呈正相关,与CD4^(+)T细胞(结肠癌:r=-0.174,P<0.001;直肠癌:r=-0.381,P<0.001)浸润水平呈负相关。其中还与结肠癌中B细胞水平(r=0.125,P<0.05)呈正相关。在结肠癌中,MAD2L1与PD-L1(r=0.159,P<0.05)呈正相关。在直肠癌中,MAD2L1与PD-1(r=-0.241,P<0.05)呈负相关。结论 MAD2L1在结直肠癌中高表达,与结直肠癌良好的预后及肿瘤免疫浸润相关,可能是结直肠癌患者潜在的治疗靶点。 展开更多

Pyrus pyrifolia Nakai‘Whangkeumbae'is a sand pear fruit with excellent nutritional quality and taste.However,the industrial development of pear fruit is significantly limited by its short shelf life.Salicylic acid(SA),a well-known phytohormone,can delay fruit senescence and improve shelf life.However,the mechanism by which SA regulates CONSTANS-LIKE genes(COLs)during fruit senescence and the role of COL genes in mediating fruit senescence in sand pear are poorly understood.In this study,22 COL genes were identified in sand pear,including four COLs(Pp COL8,Pp COL9a,Pp COL9b,and Pp COL14)identified via transcriptome analysis and 18 COLs through genome-wide analysis.These COL genes were divided into three subgroups according to the structural domains of the COL protein.Pp COL8,with two B-box motifs and one CCT domain,belonged to the first subgroup.In contrast,the other three Pp COLs,Pp COL9a,Pp COL9b,and Pp COL14,with similar conserved protein domains and gene structures,were assigned to the third subgroup.The four COLs showed different expression patterns in pear tissues and were preferentially expressed at the early stage of fruit development.Moreover,the expression of Pp COL8 was inhibited by exogenous SA treatment,while SA up-regulated the expression of Pp COL9a and Pp COL9b.Interestingly,Pp COL8 interacts with Pp MADS,a MADS-box protein preferentially expressed in fruit,and SA up-regulated its expression.While the production of ethylene and the content of malondialdehyde(MDA)were increased in Pp COL8-overexpression sand pear fruit,the antioxidant enzyme(POD and SOD)activity and the expression of Pp POD1 and Pp SOD1 in the sand pear fruits were down-regulated,which showed that Pp COL8 promoted sand pear fruit senescence.In contrast,the corresponding changes were the opposite in Pp MADS-overexpression sand pear fruits,suggesting that Pp MADS delayed sand pear fruit senescence.The co-transformation of Pp COL8 and Pp MADS also delayed sand pear fruit senescence.The results of this study revealed that Pp COL8 can play a key role in pear fruit senescence by interacting with Pp MADS through the SA signaling pathway. 展开更多

目的分析睡眠集束化(SLEEP⁃MAD)干预模式在提高重症监护病房(ICU)机械通气患者睡眠质量中的作用。方法选择2020年1月至2022年12月南通市第六人民医院ICU收治的100例机械通气患者作为研究对象,随机将其均分为对照组、观察组,每组各50例... 目的分析睡眠集束化(SLEEP⁃MAD)干预模式在提高重症监护病房(ICU)机械通气患者睡眠质量中的作用。方法选择2020年1月至2022年12月南通市第六人民医院ICU收治的100例机械通气患者作为研究对象,随机将其均分为对照组、观察组,每组各50例。对照组给予常规ICU护理,观察组给予SLEEP⁃MAD干预护理。对比分析2组患者ICU环境压力源量表(ICUESS)评分、免疫功能相关指标水平及理查兹⁃坎贝尔睡眠量表(RCSQ)评分。结果护理前,ICUESS评分、免疫功能相关指标水平、RCSQ评分组间比较差异无统计学意义(P>0.05);护理后,2组患者ICUESS评分均降低且观察组低于对照组,免疫功能相关指标水平、RCSQ评分均升高且观察组高于对照组(P<0.05)。结论在ICU机械通气患者中应用SLEEP⁃MAD干预模式可有效缓解患者环境恐惧心理,改善睡眠质量与免疫功能,有利于提高患者预后水平。 展开更多

目的:从组织学观察三七总皂苷(PNS)对大鼠跟腱异位骨化(HO)的影响,并基于转化生长因子-1(TGF-1)/Sma和Mad相关蛋白(Smad)同源物2/3信号通路探讨其潜在作用机制。方法:将126只SD大鼠随机分为正常对照组、假手术组、模型组、西药组、高剂... 目的:从组织学观察三七总皂苷(PNS)对大鼠跟腱异位骨化(HO)的影响,并基于转化生长因子-1(TGF-1)/Sma和Mad相关蛋白(Smad)同源物2/3信号通路探讨其潜在作用机制。方法:将126只SD大鼠随机分为正常对照组、假手术组、模型组、西药组、高剂量组、中剂量组、低剂量组,每组18只。HO造模术后分别给予不同浓度PNS或对照西药或生理盐水灌胃2周。于术后2周、5周及10周分别行小动物X线及CT平扫评估HO发生情况。采用一般组织学染色,包括苏木精-伊红染色、Masson三色染色及番红O固绿软骨染色,观测HO病灶内组织学改变。经免疫组织化学法比较各组病灶局部TGF-β1、磷酸化Smad同源物2(p-Smad2)及p-Smad3表达的差异。结果:术后10周,X线检测可见各手术大鼠成骨率差异无统计学意义(P>0.05)。术后2周,手术大鼠跟腱细胞显著增加(P<0.05),跟腱纤维排列紊乱;5周可见大量增殖肥大软骨细胞;10周可见大量成熟异位骨组织形成。中剂量组术后10周相对骨量显著低于模型组(P<0.05)。免疫组织化学染色显示,假手术组TGF-β1、p-Smad2及p-Smad3表达术后与正常对照组比较均显著增加,差异均有统计学意义(均P<0.01)。与模型组比较,中剂量组TGF-β1的表达在术后2周显著降低(P<0.05),表达峰值延后。结论:TGF-β1的表达在大鼠跟腱HO造模术后具有显著增加。一定浓度的PNS可降低异位骨组织的相对骨量,这可能与PNS延后TGF-β1的表达峰值并下调其表达量有关。 展开更多

目的观察金果胃康胶囊对胃癌前病变大鼠胃黏膜HSP27、MAD2蛋白表达的影响。方法将85只大鼠随机分为空白组10只和造模组75只。空白组给予正常饮食,造模组采用N-甲基-N’-硝基-N-亚硝基胍(MNNG)联合乙醇、氨水及饥饱失常组成复合造模法。... 目的观察金果胃康胶囊对胃癌前病变大鼠胃黏膜HSP27、MAD2蛋白表达的影响。方法将85只大鼠随机分为空白组10只和造模组75只。空白组给予正常饮食,造模组采用N-甲基-N’-硝基-N-亚硝基胍(MNNG)联合乙醇、氨水及饥饱失常组成复合造模法。将造模后大鼠随机分成模型组、金果胃康胶囊高、中、低剂量组和维酶素组。各给药组分别给予相应药物灌胃,空白组和模型组给予等量生理盐水灌胃,1次·d-1,连续12 w。12 w后处死所有大鼠,取胃体组织,光镜下观察各组大鼠胃黏膜组织病理变化;免疫组化法检测胃黏膜组织中HSP27、MAD2蛋白表达情况。结果模型组大鼠HSP27、MAD2蛋白表达水平明显高于空白组(均P<0.05)。各给药组HSP27、MAD2蛋白表达水平明显低于模型组(P均<0.05),且金果胃康胶囊中高剂量组明显低于维霉素组(均P<0.05)。结论金果胃康胶囊可逆转胃癌前病变的发生,作用机制可能与其能抑制HSP27蛋白的表达及控制MAD2蛋白的表达有关。 展开更多

Background:Colon adenocarcinoma(COAD)is the second leading cause of cancer death worldwide thus,identification of COAD biomarkers is critical.Mitotic Arrest Deficient 2 Like 2(MAD2L2)is a key factor in mammalian DNA damage repair and is highly expressed in many malignant tumors.This is a comprehensive study of MAD2L2 expression,its diagnostic value,prognostic analysis,potential biological function,and impact on the immune system of patients with COAD.Methods:Gene expression,clinical relevance,prognostic analysis,diagnostic value,GO/KEGG cluster analysis,data obtained from TCGA,and bioinformatics statistical analysis were performed using the R package.Immune responses to MAD2L2 expression in COAD were analyzed using TIMER.The expression of MAD2L2 in HCT116 cells induced by the inflammatory factor TNF-αwas detected using Western blot.Results:Our results underscore the clinical diagnostic value and potential biological significance of MAD2L2 in patients with COAD.A high level of MAD2L2 expression has been found in COAD and correlated with tumor status and colon polyps.ROC curve analysis showed that MAD2L2 expression has high diagnostic value in COAD.Analysis of immune infiltration results showed that MAD2L2 expression was positively correlated with neutrophil levels.The western blot results demonstrated that MAD2L2 was dose-dependently present with TNF-α.GO/KEGG revealed that MAD2L2 overexpressed and coexpressed genes were mostly involved in biological functions,including hypoxia response,response to reduced oxygen levels,mitochondrial translation elongation,and other processes.Conclusion:MAD2L2 as a new COAD biomarker contributes to our understanding of how alterations in gene expression and the immunological environment contribute to the development of colon cancer.Following further investigation,MAD2L2 may prove to be a viable target factor for clinical diagnosis and therapy of COAD. 展开更多

The current study proposes a novel technique for feature selection by inculcating robustness in the conventional Signal to noise Ratio(SNR).The proposed method utilizes the robust measures of location i.e.,the“Median”as well as the measures of variation i.e.,“Median absolute deviation(MAD)and Interquartile range(IQR)”in the SNR.By this way,two independent robust signal-to-noise ratios have been proposed.The proposed method selects the most informative genes/features by combining the minimum subset of genes or features obtained via the greedy search approach with top-ranked genes selected through the robust signal-to-noise ratio(RSNR).The results obtained via the proposed method are compared with wellknown gene/feature selection methods on the basis of performance metric i.e.,classification error rate.A total of 5 gene expression datasets have been used in this study.Different subsets of informative genes are selected by the proposed and all the other methods included in the study,and their efficacy in terms of classification is investigated by using the classifier models such as support vector machine(SVM),Random forest(RF)and k-nearest neighbors(k-NN).The results of the analysis reveal that the proposed method(RSNR)produces minimum error rates than all the other competing feature selection methods in majority of the cases.For further assessment of the method,a detailed simulation study is also conducted. 展开更多
