METHODS: Magnetic iron-dextran nanopartides were prepared by the reaction of a mixture of ferric and ferrous ions with dextran polymers under alkaline conditions. The particles were coated with antiserum against E.coli O157: H7 by the periodate oxidation-borohydride reduction procedure. The oxidation time, amount of antibody coating the particles, amount of nanoparticles, incubation time and isolation time were varied to determine their effects on recovery of the organisms. Finally, the optimum conditions for isolating E.coli O157:H7 from food samples were established. RESULTS: E.coli O157:H7 can be isolated from samples within 15 min with the sensitivity of 101 CFU/mL or even less. In the presence of 108 CFU/mL of other organisms, the sensitivity is 101-102 CFU/mL. Nonspecific binding of other bacteria to the particles was not observed. Two and a half hours of enrichment is enough for the particles to detect the target from the food samples inoculated with 1 CFU/g. CONCLUSION: Isolation of target bacteria by immuno magnetic nanoparticles is an efficient method with high sensitivity and specificity. The technique is so simple that it can be operated in lab and field even by untrained personnel.展开更多

The HSLDS property of the isolator can be tuned off-line or on-line. This study focuses on the characterization of the isolator using a finite element based package. Firstly using the single physics solver, the stiffness behaviours of the mechanical and magnetic springs are determined, respectively. Then using the weakly coupled multi-physics method, the stiffness behaviours of the passive isolator and the semi-active isolator are investigated, respectively. With the found stiffness models, a nonlinear differential equation governing the dynamics of the isolator is solved using the time-dependent solver. The displacement transmissibility ratios of the isolator are obtained. The study confirms that the isolation region of the isolator can be widened through off-line or on-line tuning.展开更多

In this paper, the Eulerian-Lagrangian technique is adopted to simulate the delivery of drug-loaded nanoparticles to patient-specific atherosclerotic plaque with the aid of an external magnetic field. Plaques and vascular walls are introduced as porous media formulated by the Darcy-Forchheimer model in this targeted transport process. The results demonstrate that the delivery efficiency of particles to atherosclerosis depends on the external magnetic field, such as configuration and intensity, in which the configuration angle of the current wire is a key factor and the double current wires have advantages over the single current wire. Meanwhile, the delivery efficiency gradually decreases as the distance between the plaque cap and the current wire increases. Further, although augmenting the current or magnetic susceptibility can generally improve the delivery efficiency of nanoparticles, this increase is not apparent when small-sized nanoparticles are employed as drug transport particles. The results obtained can potentially serve as the guideline to optimize regimens for the targeted therapy of atherosclerosis.展开更多

Methods: Descriptive and retrospective study of the MRIs of a pediatric ADD cohort. The included images were acquired in machines with 1.5 or 3T magnets. Low-quality images were excluded. The radiological characteristics of the lesions were described using the 2010 Revised McDonald Diagnostic Criteria regarding localization, contrast enhancement and optic nerve alterations. Results: Twenty-three patients were included (55% female). The mean age of the first clinical event was 7.7 years. Most common diagnosis was Clinically Isolated Syndrome (35%), followed by Multiple Sclerosis (30%), Neuromyelitis Optica (17%) and Acute Disseminated Encephalomyelitis (17%). Mean time elapsed until diagnosis was 1.8 years. Follow-up MRIs of ADEM patients showed complete or partial resolution of lesions; MS lesions were mostly localized in the brain and four patients had contrast enhancement of lesions in their last available MRI. All NMO (neuromyelitis optica) patients had extensive spinal lesions, and two had optic neuritis. All patients with CIS (clinically isolated syndrome) had focal spinal lesions and evolved with radiologic improvement. Conclusions: ADEM and CIS patients' MRIs showed lesion reduction, while MS and NMO patients developed new lesions during follow-up.展开更多
