By means of the Penn State-NCAR Mesoscale Model Version 5(MM5)with a horizontal resolution of 20 km.the maintainable mechanism of Meso-β scale Convective System(Mβ CS)has been investigated on the basis of simulation...By means of the Penn State-NCAR Mesoscale Model Version 5(MM5)with a horizontal resolution of 20 km.the maintainable mechanism of Meso-β scale Convective System(Mβ CS)has been investigated on the basis of simulation of the temporal and spatial thermodynamics structure of the MβCS which occurred in Wuhan and its surroundings on 21 July 1998.The occurrence of the significant warm-core in the center of the MβCS happened in Changjiang River Basin between the Mufu Mountain and the Dabie Mountain.To the southern side of the MβCS,there exist the southwest low-level jet(LLJ)and the vertical secondary circulation in the low and middle troposphere respectively.In respect to the northern of the MβCS,the northwest jet emerges in the upper troposphere,accompanied with cold and dry atmosphere downdrafts,resulting in another secondary circulation.The foregoing mentioned vertical wind shear provides a favorable dynamical environment for the intensification and maintenance of the MβCS.Equally important,the latent heat release associated with the MβCS produces the warm center in the middle troposphere and the pressure falls.The pressure drop then accelerates air parcels toward the low leading to strong convergence as well as the intensified convection,establishing a positive feedback between the convection and the latent heat release,which is the thermodynamic mechanism of the development and maintenance of the MβCS.展开更多

Poor bone quality is a major factor in skeletal fragility in elderly individuals.The molecular mechanisms that establish and maintain bone quality,independent of bone mass,are unknown but are thought to be primarily d...Poor bone quality is a major factor in skeletal fragility in elderly individuals.The molecular mechanisms that establish and maintain bone quality,independent of bone mass,are unknown but are thought to be primarily determined by osteocytes.We hypothesize that the age-related decline in bone quality results from the suppression of osteocyte perilacunar/canalicular remodeling(PLR),which maintains bone material properties.We examined bones from young and aged mice with osteocyte-intrinsic repression of TGFβsignaling(TβRII^(ocy−/−))that suppresses PLR.The control aged bone displayed decreased TGFβsignaling and PLR,but aging did not worsen the existing PLR suppression in male TβRII^(ocy−/−)bone.This relationship impacted the behavior of collagen material at the nanoscale and tissue scale in macromechanical tests.The effects of age on bone mass,density,and mineral material behavior were independent of osteocytic TGFβ.We determined that the decline in bone quality with age arises from the loss of osteocyte function and the loss of TGFβ-dependent maintenance of collagen integrity.展开更多
Background Previous studies havedemonstrated that excitatory repetitive transcranial magnetic stimulation(rTMS)can improve the cognitive function of patients with Alzheimer's disease(AD).Intermittent theta burst s...Background Previous studies havedemonstrated that excitatory repetitive transcranial magnetic stimulation(rTMS)can improve the cognitive function of patients with Alzheimer's disease(AD).Intermittent theta burst stimulation(iTBS)is a novel excitatory rTMS protocol for brain activity stimulation with the ability to induce long-term potentiation-like plasticity and represents a promising treatment for AD.However,the long-term effects of iTBS on cognitive decline and brain structure in patients with AD areunknown.Aims We aimed to explore whether repeating accelerated iTBS every three months could slow down the cognitive decline in patients with AD.Methods In this randomised,assessor-blinded,controlled trial,iTBS was administered to the left dorsolateral prefrontal cortex(DLPFC)of 42 patients with AD for 14days every 13weeks.Measurements included the Montreal Cognitive Assessment(MoCA),a comprehensive neuropsychological battery,and the grey matter volume(GMV)of the hippocampus.Patients were evaluated at baseline and after follow-up.The longitudinal pipeline of the Computational Anatomy Toolbox for SPM was used to detect significant treatment-related changes over time.Results The iTBS group maintained MoCA scores relative to the control group(t=3.26,p=0.013)and reduced hippocampal atrophy,which was significantly correlated with global degeneration scale changes.The baseline Mini-Mental State Examination(MMSE)score,apolipoprotein E genotype and Clinical Dementia Rating were indicative of MoCA scores at follow-up.Moreover,the GMV of the left(t=0.08,p=0.996)and right(t=0.19,p=0.977)hippocampus were maintained in the active group but significantly declined in the control group(left:t=4.13,p<0.001;right:t=5.31,p<0.001).GMV change in the left(r=0.35,p=0.023)and right(r=0.36,p=0.021)hippocampus across the intervention positively correlated with MoCA changes;left hippocampal GMV change was negatively correlated with global degeneration scale(r=-0.32,p=0.041)changes.Conclusions DLPFC-iTBS maybe a feasible and easy-to-implement non-pharmacological intervention to slow down the progressive decline of overall cognition and quality of life in patients with AD,providing a new AD treatment option.Trial registration number NCT04754152.展开更多

文摘Fibroblasts are diverse mesenchymal cells that participate in tissue homeostasis and disease by producing complex extracellular matrix and creating signaling niches through biophysical and biochemical cues.Transcriptionally and functionally heterogeneous across and within organs,fibroblasts encode regional positional information and maintain distinct cellular progeny.We summarize their development,lineages,functions,and contributions to fibrosis in four fibroblast-rich organs:skin,lung,skeletal muscle,and heart.We propose that fibroblasts are uniquely poised for tissue repair by easily reentering the cell cycle and exhibiting a reversible plasticity in phenotype and cell fate.
