Based on the samples collected from the west wall of the ABYT2004 at Yuhuicun Site in Bengbu, Anhui Province, the multi-environmental substitute indexes such as the grain-size, Rb/Sr, zircon crystal morphology and mag...Based on the samples collected from the west wall of the ABYT2004 at Yuhuicun Site in Bengbu, Anhui Province, the multi-environmental substitute indexes such as the grain-size, Rb/Sr, zircon crystal morphology and magnetic susceptibility were analyzed. The age of archaeological strata was determined by AMS^14C dating. According to the results, combined with the information of the ancient human activities and the cultural heritage data obtained from the Longshan cultural archaeological strata, it can be concluded that (1) Before 4500 aBP, the climate was humid and water level was high in the Huaihe River and its tributaries, the drainage basin and its surrounding areas were even flooded, so the site area was submerged and there were no human activities around the site. (2) About 4500 aBP, the climate began to become drier and water level of the Huaihe River and its tributaries began to decline, thus floodplain gradually appeared, Yuhuicun Site and Longshan Culture began to appear. (3) Around 4100 aBP, the climate was humid and rainfall was abundant, which resulted in frequent flood disasters in the whole Huaihe River Basin. (4) After 4100 aBP, the climate gradually transited to be cold and dry, agricultural production was constrained to reduce the food source, Yuhuicun Site was almost abandoned. (5) The environmental information in sedimentary strata and historical records based on ancient books agree with each other. In addition, Yuhuicun Site was related to Dayu Zhishui (Yu the Great in taming the floods) in ancient books.展开更多
For establishing the favourable media for growth of Phaeoacremonium parasiticum, the fungus was inoculated in five different media (Potato Dextrose Broth (PDB), Host Extract (50%) + PDB (50%) (HE + PDB), and the solid...For establishing the favourable media for growth of Phaeoacremonium parasiticum, the fungus was inoculated in five different media (Potato Dextrose Broth (PDB), Host Extract (50%) + PDB (50%) (HE + PDB), and the solid media viz., Rice Bran (RB), Maize Meal (MM) and Wheat Bran (WB)) at pH of 6.5 and incubated at a temperature of 30℃± 1℃. The solid media were added with water at the rate of 70 ml/100g of solid substrate. After 30 days of inoculation, conidial population of P. parasiticum in liquid media was lower compared to conidial population in solid media. In solid media, MM media showed the highest conidial population (9.56 log·cfu/g) followed by WB (9.50 log·cfu/g) and RB (9.38 log·cfu/g). Hence, Maize Meal media (MM: Water = 100 g:70 ml) at pH of 6.5, incubation temperature: 30℃± 1℃for 30 days would be the standard technique for mass production of P. parasiticum.展开更多

Additionally, it is being spread by a big wave of informatization, characteristic of the digital revolution. Traditional rural customs are always important contents of folklore study, but nowadays and in the future, the research focus of Chinese folklore studies needs to be adjusted. 17-β-estradiol (estrogen) is a steroid hormone important to human development;however, high levels of this molecule are associated with increased risk of breast cancer primarily due to estrogen’s ability to bind and...17-β-estradiol (estrogen) is a steroid hormone important to human development;however, high levels of this molecule are associated with increased risk of breast cancer primarily due to estrogen’s ability to bind and activate the estrogen receptor (ER) and initiate gene transcription. Currently, estrogen mechanisms of action are classified as genomic and non-genomic and occur in an ER-dependent and ER-independent manner. In this study, we examine estrogen signaling pathways, by measuring changes in protein expression as a function of time of exposure to estrogen in both ER-positive (MCF-7) and ER-negative (MDA-MB-231) cell lines. Using a robust experimental design utilizing isotopic labeling, two-dimensional LC-MS, and bioinformatics analysis, we report genomic and non-genomic ER regulated estrogen responsive proteins. We find a little over 200 proteins differentially expressed after estrogen treatment. Cell proliferation, transcription, actin filament capping and cell to cell signaling are significantly enriched in the MCF-7 cell line alone. Translational elongation and proteolysis are enriched in both cell lines. Subsets of the proteins presented in this study are for the first time directly associated with estrogen signaling in mammary carcinoma cells. We find that estrogen affected the expression of proteins involved in numerous processes that are related to tumorigenesis such as increased cellular division and invasion in an ER-dependent manner. Moreover, we identified negative regulation of apoptosis as a non-genomic process of estrogen. This study complements gene expression studies and highlights the need for both genomic and proteomic analyses in unraveling the complex mechanisms by which estrogen affects progression of breast cancer.展开更多

目的探讨基质辅助激光解吸电离飞行时间质谱(matrix-assisted laser desorption/ionization time of flight mass spectrometry,MALDI-TOF MS)结合短时培养在快速鉴定血流感染病原菌中的应用。方法对南京医科大学附属淮安第一医院微生物...目的探讨基质辅助激光解吸电离飞行时间质谱(matrix-assisted laser desorption/ionization time of flight mass spectrometry,MALDI-TOF MS)结合短时培养在快速鉴定血流感染病原菌中的应用。方法对南京医科大学附属淮安第一医院微生物室2020年11月至2021年12月血培养报告阳性的标本进行革兰染色分类,并采用4h和6h短时培养法及MALDI-TOF MS技术快速鉴定感染病原菌并实时报送病区单元。结果4h短时培养革兰阴性菌共351株,339株(96.6%)细菌鉴定到种属,与24h鉴定结果相同,11株无鉴定结果,1株鉴定错误;6h短时培养革兰阳性菌共216株,212株(98.1%)鉴定到种属,与24h鉴定结果相同,3株无鉴定结果,1株鉴定错误。跟踪321株革兰阴性菌的临床反馈,287例(89.4%)给予临床关注,89例(27.7%)进行药物调整,238例(74.1%)临床疗效良好;跟踪183株革兰阳性菌的临床反馈,其中150例(82.0%)给予临床关注,49例(26.8%)进行药物调整,131例(71.6%)临床疗效良好。结论MALDI-TOF MS技术结合短时培养方法可有效提高血流感染病原菌鉴定准确率,改善血流感染患者抗菌药物治疗时机。展开更多

The results of experiments show that the isolated cells can survive,divide and regenerate well on t...Vegetative cells of Porphyra yezoensis are isolated with sea snail enzyme and cultured on the solidified agar medium. The results of experiments show that the isolated cells can survive,divide and regenerate well on the medium solidified with agar. The first division on the solid medium starts after 7 days' culture, 4 days later than the liquid culture. The survival rate of isolated cells is 71.3% on the solid medium, lower than the 86.2% of that in seawater.Thalli, thalloids,conchocelis, spermatangia and multicellular masses are developed on the solid/medium in the first month, slowly but normally. Spermatangia sacs disappear within 4 weeks. Without adding nutrient liquid onto the surface of solid medium or injecting seawater under the agar layer in order to keep moisture, the thalli and cell groups release monospores to form new thalli instead of enlarging their areas after 5 weeks' culturing. Some monospores regenerate new thalli. Other monospores lose their pigments and minimize their volume and divide quickly to form light pink calli. After 16 weeks, numerous calli can be seen on the solid medium and after 24 weeks' culturing, almost only calli and conchocelis can be seen. If the calli are immersed in seawater, the monospores are released and may develop into young thallus.展开更多
