"Synthetic"allopolyploids recreated by interspecific hybridization play an important role in providing novel genomic variation for crop improvement.Such synthetic allopolyploids often undergo rapid genomic structural variation(SV).However,how such SV arises,is inherited and fixed,and how it affects important traits,has rarely been comprehensively and quantitively studied in advanced generation synthetic lines.A better understanding of these processes will aid breeders in knowing how to best utilize synthetic allopolyploids in breeding programs.Here,we analyzed three genetic mapping populations(735 DH lines)derived from crosses between advanced synthetic and conventional Brassica napus(rapeseed)lines,using whole-genome sequencing to determine genome composition.We observed high tolerance of large structural variants,particularly toward the telomeres,and preferential selection for balanced homoeologous exchanges(duplication/deletion events between the A and C genomes resulting in retention of gene/chromosome dosage between homoeologous chromosome pairs),including stable events involving whole chromosomes("pseudoeuploidy").Given the experimental design(all three populations shared a common parent),we were able to observe that parental SV was regularly inherited,showed genetic hitchhiking effects on segregation,and was one of the major factors inducing adjacent novel and larger SV.Surprisingly,novel SV occurred at low frequencies with no significant impacts on observed fertility and yield-related traits in the advanced generation synthetic lines.However,incorporating genome-wide SV in linkage mapping explained significantly more genetic variance for traits.Our results provide a framework for detecting and understanding the occurrence and inheritance of genomic SV in breeding programs,and support the use of synthetic parents as an important source of novel trait variation. 展开更多

In the United States,schools offer special education services to children who are diagnosed with a learning or neurodevelopmental disorder and have difficulty meeting their learning goals.Pediatricians may play a key role in helping children access special education services.The number of children ages 6-21 in the United States receiving special education services increased 10.4%from 2006 to 2021.Children receiving special education services under the autism category increased 242%during the same period.The demand for special education services for children under the developmental delay and other health impaired categories increased by 184%and 83%respectively.Although student enrollment in American schools has remained stable since 2006,the percentage distribution of children receiving special education services nearly tripled for the autism category and quadrupled for the developmental delay category by 2021.Allowable heavy metal residues remain persistent in the American food supply due to food ingredient manufacturing processes.Numerous clinical trial data indicate heavy metal exposures and poor diet are the primary epigenetic factors responsible for the autism and attention deficit hyperactivity disorder epidemics.Dietary heavy metal exposures,especially inorganic mercury and lead may impact gene behavior across generations.In 2021,the United States Congress found heavy metal residues problematic in the American food supply but took no legislative action.Mandatory health warning labels on select foods may be the only way to reduce dietary heavy metal exposures and improve child learning across generations. 展开更多

Many populations of the two-spotted spider mite, Tetranychus urticae Koch, have developed high levels of resistance to the pesticide abamectin in China and other countries. This study developed a near-isogenic line to understand better the inheritance, cross-resistance, and fitness costs associated with abamectin resistance in the field population of T. urticae in China. We introduced the trait that confers extremely high abamectin resistance in a field-collected population of T. urticae into a susceptible laboratory strain(IPP-SS) to generate an abamectin-resistant near-isogenic line(NIL-Aba).This process was carried out through multiple backcrossing to IPP-SS and via parthenogenesis and abamectin screening. Compared with IPP-SS, the NIL-Aba strain had a 25 147-fold resistance to abamectin and a high level of cross-resistance to bifenthrin(288.17-fold), an intermediate level to emamectin benzoate(42.57-fold), and low levels to bifenazate, chlorfenapyr, cyflumetofen, cyenopyrafen, and cyetpyrafen with resistance ranging from 3.18-to 9.31-fold.But it had no cross-resistance to profenofos. The resistance to abamectin in NIL-Aba was autosomal, incompletely dominant, and polygenic. Based on two sex life table parameters, no fitness cost was found in NIL-Aba. Establishing the NIL-Aba strain provides a reliable basis for an in-depth study of abamectin resistance in T. urticae. New information on toxicological characteristics and fitness cost should facilitate the management of abamectin resistance in field populations of T. urticae. 展开更多

Nowadays, the rapid intelligentization of media has ushered in the era of intellectual media, with characteristics of “massive media, man-machine integration and self-evolution”. In the era of intellectual media, traces of intelligent technologies can be seen everywhere, and the ecology of the inheritance of intangible cultural heritage has been reconstructed. Under this circumstance, the author firstly makes clarification of the advancement of the era, which has given a new logic of intangible cultural heritage inheritance, and various intelligent powers should be developed including intelligent acquisition, processing and judgement ability of content value. Then, the technology empowerment is analyzed, which has made reconstruction of intangible cultural heritage inheritance: while being digitalized,the inheritance of intangible cultural heritage has become systematized in way of “Points, Lines and Planes” at the same time. This editorial,comments on the article by Spartalis et al published in the recent issue of the World Journal of Cardiology.We here provide an outlook on potential ethical concerns related to the future application of gene therapy in the field of inherited arrhythmias.As monogenic diseases with no or few therapeutic options available through standard care,inherited arrhythmias are ideal candidates to gene therapy in their treatment.Patients with inherited arrhythmias typically have a poor quality of life,especially young people engaged in agonistic sports.While genome editing for treatment of inherited arrhythmias still has theoretical application,advances in CRISPR/Cas9 technology now allows the generation of knock-in animal models of the disease.However,clinical translation is somehow expected soon and this make consistent discussing about ethical concerns related to gene editing in inherited arrhythmias.Genomic off-target activity is a known technical issue,but its relationship with ethnical and individual genetical diversity raises concerns about an equitable accessibility.Meanwhile,the costeffectiveness may further limit an equal distribution of gene therapies.The economic burden of gene therapies on healthcare systems is is increasingly recognized as a pressing concern.A growing body of studies are reporting uncertainty in payback periods with intuitive short-term effects for insurance-based healthcare systems,but potential concerns for universal healthcare systems in the long term as well.Altogether,those aspects strongly indicate a need of regulatory entities to manage those issues. 展开更多

The authors evaluated 57 parental inbred lines of maize hybrids disseminated in Southwest China for drought tolerance under drought-stressed and well-watered conditions. Multiple regression analyses between drought tolerant coefficients of the grain yield per plant and 15 morphological and physiological traits measured from a subset of 12 selected lines, identified traits 1 and 5, which were important for drought tolerance, at the seedling and reproductive stages respectively. Gene effects, combining abilities, and heritabilities of these traits were estimated using generation mean and diallel cross methods. Dominance effect was more important than additive effect for the plant height, anthesis-silking interval (ASI), root weight, and the grain yield per plant, whereas, they were about equal for the leaf emergence rate. The variances of special combining ability (SCA) were about double that of the general combining ability (GCA) for plant height, ASI and grain yield per plant, although they were about equal for leaf emergence rate and root weight. Narrow sense heritabilities of the five traits for the reproductive stage were not high (12.8-29.6%), although broad sense heritabilities for plant height, ASI, and grain yield were as high as 70-85%. A segregating population consisting of 183 F2 plants from the cross N87-1 (drought tolerant) × 9526 (susceptible), was genotyped at 103 SSR loci and the F2:4 families were evaluated under two water regimes. Twelve quantitative trait loci (QTLs) (two for plant height, five for ASI, four for root biomass, and one for grain yield) were identified, most of which had overdominant gene action. Some chromosomal regions, such as those linked to markers umcl051 (bin 4.08), umc2881 (bin 4.03), and phi034 (bin 7.02), had overlapping QTLs. 展开更多

The development of high GNPS germplasm is widely emphasized in wheat-yield breeding. This paper reported two high GNPS wheat germplasm lines, Pubing 3228 and Pubing 3504, which had a stable and wide adaptability to different ecological regions. By exploring a nested cross design with reciprocals using Pubing 3228 or Pubing 3504 as a common parent and investigating the GNPS phenotypes of F1 hybrids in 2007-2008 and F2 populations in 2008-2009 of different cross combinations, the narrow-sense GNPS heritability was up to 49.58 and 52.23%, respectively. Genetic model analysis predictions suggested that GNPS in Pubing 3228 and Pubing 3504 was mainly controlled by additive genetic effects. Correlation analysis results between GNPS and 1 000- kernel weight (TKW) of F2 populations showed that TKW was not influenced with the increase of GNPS. The good coordination among three yield components of spike number per plant (SNPP), GNPS, and TKW in the F2 segregating population implied that selection of good candidate individuals in breeding programs would be relatively straightforward. Overall, our results indicated that Pubing 3228 and Pubing 3504 are two potential germplasm lines for yield improvement of GNPS in pedigree selection of wheat breeding. 展开更多

Apple bot canker [Botryosphaeria dothidea(Moug.) Ces.et de Not.] is distributed worldwide,resulting in a serious crop loss every year in apple(Malus domestica Borkh.) production.The resistance of each seedling derived from a hybrid population(Jonathan×Golden Delicious) was evaluated by disease index either from natural infection in the field or from inoculation with five isolates of B.dothidea,Ls1,Lw023,Lw048,Mx1,and Zz26.The inheritance of the resistance to bot canker was analyzed via frequency distribution analysis,and microsatellite and AFLP markers linked to the resistance loci were screened.From the binary frequency distribution patterns,it was found that the segregation ratio of resistant/susceptible genotypes infected by pathogen isolates Lw023 and Ls1 was 1:15;and that by Zz26 and Mx1 was 15:1.The variation of resistance was involved in the segregation of two to four alleles of major genes,the resistance was recessive when infected by Lw023 and Ls1,but was dominant when infected with Mx1 and Zz26.A microsatellite maker,CH02a04-450,and two AFLP markers,E-AG/M-GAC-280 and E-AGG/M-CTT-110,were identified,and their map distances to the resistance loci were 5.1,5.1 and 6.2 cM,respectively.The three markers are located in different linkage groups,while CH02a04-450 is on linkage group 2 or 7.E-AG/M-GAC-280 was successfully converted into SCAR159.Finally,CH02a04-450 and SCAR159 were re-examined in inoculated segregation population and presented a good reliability on predicting phenotypes of resistance. 展开更多

Inheritance and QTL Mapping of Salt Tolerance in Rice 被引量:6

An F2 population derived from the cross between Jiucaiqing (Japonica) and IR36 (indica) was used to analyze the inheritance of salt tolerance in rice by genetic model of major-genes plus polygenes, and to map the corresponding QTLs by SSR molecular markers. Rice plants of P1, P2, F1 and F2 at 5- to 6- leaf stage were treated under 140 mmol/LNaCI for 10 days. Three indices representing the ability of salt tolerance of rice seedlings were measured, including salt tolerance rating (STR), Na+/K+ ratio in roots and dry matter weight of shoots (DWS). STR, Na+/K+ and DWS were all controlled by two major genes with modification by polygenes. Heritability of these traits from major genes was 17.8, 53.3 and 52.3%, respectively. The linkage map constructed by 62 SSR molecular markers covered a total length of about 1 142 cM. There were three QTLs detected for STR located on chromosome 1, 5 and 9, two QTLs for DWS on chromosomes 8 and 9, and two QTLs for Na+/K+ on chromosomes 2 and 6, one on each chromosome respectively. Single QTL accounted for 6.7 to 19.3% of phenotypic variation. Identification method of salt tolerance in rice and breeding of rice varieties with salt tolerance based on molecular markers assisted selection had been discussed. 展开更多
