用核糖体ITS区序列验证自然杂交种Meconopsis×cookei G.Taylor 被引量:10

对被认为是自然杂交种的绿绒蒿植物Meconopsis×cookeiG .Taylor及其可能的亲本红花绿绒蒿(M punicea)和五脉绿绒蒿 (M .quintuplinervia)的核糖体DNAITS区进行了序列测定 ,所得序列的长度为 6 6 7~ 6 6 8bp ,其中红花绿绒蒿的序... 对被认为是自然杂交种的绿绒蒿植物Meconopsis×cookeiG .Taylor及其可能的亲本红花绿绒蒿(M punicea)和五脉绿绒蒿 (M .quintuplinervia)的核糖体DNAITS区进行了序列测定 ,所得序列的长度为 6 6 7~ 6 6 8bp ,其中红花绿绒蒿的序列长度为 6 6 7bp ,另外两个种的序列长度均为 6 6 8bp。利用软件ClustalX对所得序列进行排序和碱基比较 ,排序后的序列长度为 6 6 8bp ,其中ITS1长度为 2 5 4bp ,5 8S长度为 16 2bp ,ITS2长度为 2 5 2bp。整个ITS区序列共有 16个变异位点 ,占序列总长度的 2 4 0 % ,其中ITS1的变异位点 9个 ,占 5 6 2 5 % ,占整个序列长度的 1 35 % ;ITS2的变异位点 6个 ,占 37 5 0 % ,占整个序列长度的 0 89% ;5 8S的变异位点 1个 ,占 6 2 5 % ,占整个序列长度的 0 15 %。分析结果表明 ,M .×cookei同时具有红花绿绒蒿和五脉绿绒蒿的两种ITS序列 ,也就是说M .×cookei与红花绿绒蒿和五脉绿绒蒿之间在ITS基因上的变化规律符合孟德尔遗传学定律 ,从而从分子水平上证明M .×cookei是红花绿绒蒿和五脉绿绒蒿的杂交后代。 展开更多

A molecular structural characterization (MSC) method called reduced molecular electronegativity-distance vector (MEDVR) was used to describe the molecular structures of 55 components of meconopsis integrifolia flowers. By use of stepwise multiple regression (SMR) and partial least square (PLS) methods, a model with the correlation coefficient (R1) of 0.987 and the standard deviation (SD1) of 1.377 could be obtained. Then through multiple linear regression (MLR), another model with the correlation coefficient (R2) of 0.989 and standard deviation (SD2) of 1.395 could be constructed. Furthermore, in virtue of variable screening by the stepwise multiple regression technique (SMR), 8 vectors were selected to build up another model with its correlation coefficient (R3) and standard deviation (SD3) of 0.989 and 1.366, respectively. Then all the three models were evaluated by performing cross-validation with the leave-one-out (LOO) procedure, and the correlation coefficients (QCV) were 0.981, 0.976 and 0.979, respectively. The results show that the models constructed could provide estimation stability and favorable predictive ability. 展开更多

In the paper, using the method of Lagrange interpolation five formulas, the second derivative written is obtained from XRD (X-ray diffraction) spectrum graph of MATLAB. Methodology of XRD SD (X-ray diffraction second derivative) was adapted to extract low frequency signal from original XRD patterns of traditional Chinese medicine. The results indicated that the XRD SD fingerprints obtained had obvious peak with sharp profile, which could distinctly embody the digital characteristics of different Meconopsis. Otherwise, the results also showed that XRD SD fingerprints might be a promising method for identification and quantity control for traditional Chinese medicine and could effectively solve the problem of XRD spectrum analytical dispersion type plant medicine and had a function of magnifying glass. 展开更多
