Cationic hydrogels have received great attention to control eutrophication and recycle phosphate.In this study,a type of La(OH)_(3) loaded magnetic MAPTAC-based cationic hydrogel(La(OH)_(3)@MMCH)was developed as a potential adsorbent for enhanced phosphate removal from aqueous environment.La(OH)_(3)@MMCH exhibited high adsorption capacity of 105.72±5.99 mg P/g,and reached equilibrium within 2 hr.La(OH)_(3)@MMCH could perform effectively in a wide pH range from 3.0 to 9.0 and in the presence of coexisting ions(including SO_(4)^(2-),Cl^(-),NO_(3)^(-),HCO_(3)^(-),SiO_(4)^(4-) and HA).The adsorption-desorption experiment indicated that La(OH)_(3)@MMCH could be easily regenerated by using NaOH-NaCl as the desorption agent,and 73.3%adsorption capacity remained after five cycles.Moreover,La(OH)_(3)@MMCH was employed to treat surface water with phosphate concentration of 1.90 mg/L and showed great removal efficiency of 95.21%.Actually,MMCH showed high surface charge density of 34.38-59.38 meq/kg in the pH range from 3.0 to 11.0 and great swelling ratio of 3014.57%within 24 h,indicating that MMCH could produce the enhanced Donnan membrane effect to pre-permeate phosphate.Furthermore,the bifunctional structure of La(OH)_(3)@MMCH enabled it to capture phosphate through electrostatic attraction and ligand exchange.All the results prove that La(OH)_(3)@MMCH is a promising adsorbent for eutrophication control and phosphate recovery. 展开更多

Here we report a new method to detect DNA point mutations.The method is based on the formation and deformation of double-stranded DNA(dsDNA)membranes on a gold surface.It can encage reporter molecules between the gold surface and the double-stranded DNA or keep them away from the gold surface.In these systems,Fe(CN)63−was used as the reporter.As the temperature increases,a sharp electrochemical signal change in the melting curve of wild-type dsDNA appears.At a special temperature,the method gives 100:1 selectivity for the perfect complement and single base mutation target.Thus,the system provides a simple and sensitive method to detect DNA point mutations without labeling targets. 展开更多

BACKGROUND: Phycocyanin can relieve decrease of mitochondrial membrane potential through reducing production of active oxygen so as to protect neurons after hypoxia/reoxygenation. OBJECTIVE: To observe the effect of phycocyanin on activity of PC12 cells and mitochondrial membrane potential after hypoxia/reoxygenation. DESIGN: Randomized controlled study SETTING : Cerebrovascular Disease Institute of Affiliated Hospital, Medical College of Qingdao University MATERIALS: The experiment was carried out at the Key Laboratory of Prevention and Cure for cerebropathia in Shandong Province from October to December 2005. PC12 cells, rat chromaffin tumor cells, were provided by Storage Center of Wuhan University; phycocyanin was provided by Ocean Institute of Academia Sinica; Thiazoyl blue tetrazolium bromide (MTT) and rhodamine 123 were purchased from Sigma Company, USA; RPMI-1640 medium, fetal bovine serum and equine serum were purchased from Gibco Company, USA. METHODS: ① Culture of PC12 cells: PC12 cells were put into RPMI-1640 medium which contained 100 g/L heat inactivation equine serum and 0.05 volume fraction of fetal bovine serum and incubated in CO2 incubator at 37℃. Number of cells was regulated to 4 × 10^5 L 1, and cells were inoculated at 96-well culture plate. The final volume was 100μL. ② Model establishing and grouping: Cultured PC12 cells were randomly divided into three groups: phycocyanin group, model control group and non-hypoxia group. At 24 hours before hypoxia, culture solution in phycocyanin group was added with phycocyanin so as to make sure the final concentration of 3 g/L , but cells in model control group did not add with phycocyanin. Cells in non-hypoxia group were also randomly divided into adding phycocyanin group (the final concentration of 3 g/L) and non-adding phycocyanin group. Cells in model control group and phycocyanin group were cultured with hypoxia for 1 hour and reoxygenation for 1, 2 and 3 hours; meanwhile, cells in non-hypoxia group were cultured with oxygen and were measured at 1 hour after hypoxia/reoxygenation. ③ Detecting items: At 1, 2 and 3 hours after reoxygenation, absorbance (A value) of PC12 cells was measured with MTT technique so as to observe activity and quantity of cells. Fluorescence intensity of PC12 cells marked by rhodamine 123 was measured with confocal microscope in order to observe changes of mitochondrial membrane potential. MAEN OUTCOME MEASURES: Comparisons between quantity and activity of PC12 cells and mitochondria membrane potential at 1, 2 and 3 hours after reoxygenation. RESULTS: ① Effect of phycocyanin on quantity and activity of PC12 cells: A value was 0.924±0.027 in adding phycocyanin group and 0.924±0.033 in non-adding phycocyanin group. A value was lower in model control group and phycocyanin group than that in non-hypoxia group at 1, 2 and 3 hours after reoxygenation (0.817±0.053, 0.838±0.037, 0.875±0.029; 0.842±0.029, 0.872±0.025, 0.906±0.023, P 〈 0.05). A value was higher in phycocyanin group than that in model control group at 1, 2 and 3 after culture (P 〈 0.05). With culture time being longer, A value was increased gradually in phycocyanin group and model control group after reoxygenation (P 〈 0.05). ~ Effect of phycocyanin on mitochondrial membrane potential of PC12 cells: Fluorescence intensity was 2.967±0.253 in adding phycocyanin group and 2.962±0.294 in non-adding phycocyanin group. Fluorescence intensity was lower in model control group and phycocyanin group than that in non-hypoxia group at 1, 2 and 3 hours after hypoxia/reoxygenation (1.899±0.397, 2.119±0.414, 2.287±0.402; 2.191±0.377, 2.264±0.359, 2.436±0.471, P 〈 0.05); but it was higher in phycocyanin group than that in model control group at 1, 2 and 3 after reoxygenation (P 〈 0.05). With culture time being longer, fluorescence intensity was increased gradually in phycocyanin group and model control group after reoxygenation (P 〈 0.05). CONCLUSION: Phycocyanin and reoxygenation can protect PC12 cells after hypoxia injury through increasing mitochondrial membrane potential and cellular activity, and the effect is improved gradually with prolonging time of reoxygenation. 展开更多

Alveolar macrophages (AM) from BCG activated Wistar rat were irradiated with different doses of Gamma rays in vitro. The effects of radiation on their immunological functions and membrane damage were studied. The non-specific cytotoxicity and specific phagocytosis of AM irradiated with dose of 0, 100, 300 and 500 Gy decreased with the increase in dose. The relative fractions of Lactate Dehydrogenase and Beta-glucuronidase (β-glu) activity in supernatant increased with the increase in dose. There was a correlation between the suppression of immunological functions and the degree of damage of cytoplasmic and lysosomal membranes of AM after irradiation. Na2SeO3, a protective agent of cell membranes, alleviated this effect on the suppressive cytotoxicity indices of irradiated AM. 展开更多

Physiotherapeutic effects of infrared lasers have been proved in clinic.These infrared-based regulations of the bioelectrical activities can roughly be classied into enhancement and suppression of action potential(AP),which are described by sodium(Na)and potassium(K)transmembrane current equations,named as Hodgkin and Huxley(HH)-model.The enhancement effect is able to evoke or strengthen the AP when infrared light is applied.Its corresponding mechanism is commonly ascribed to the changes of the cell membrane capacitance,which is transiently increased in response to the infrared radiation.The distinctive feature of the suppression effect is to inhibit or reduce the AP by the designed protocols of infrared radiation.However,its mechanism presents more complexity than that in enhancement cases.HH-model describes how the Na current determines the initial phase of AP.So,the enhancement and suppression of AP can be also ascribed to the regulations of the corresponding Na currents.Here,a continuous infrared light at the wavelength of 980 nm(CIS-980)was employed to stimulate a freshly isolated hippocampal neuron in vitro and a suppression effect on the Na currents of the neuron cell was observed.Both Na and K currents,which are named as whole cell currents,were simultaneously recorded with the cell membrane capacitance current by using a patch clamp combined with infrared irradiation.The results demonstrated that the CIS-980 was able to reversibly increase the capacitance currents,completely suppressed Na currents,but little changed K currents,which forms the steady outward whole cell currents and plays a major role on the AP repolarization.A conrmation experiment was designed and carried out by synchronizing tens of milliseconds of infrared stimulation on the same kinds of hippocampal neuron cells.After the blocked K channel,a reduction of Na current amplitude was still recorded.This proved that infrared suppression of Na current was irrelevant to K channel.A membrane capacitance mediation process was preliminarily proposed to explain the Na channel suppression process. 展开更多

Piled embankments have many advantages that have been applied in high-speed railway construction engineering.However,the load transfer mechanism of piled embankments,such as soil arching and tension membranes,is still unclear,especially under dynamic loads.To investigate the soil arching and tension membrane under dynamic train loads on high-speed railways,a large-scale piled embankment model test with X-shaped piles as vertical reinforcement was performed,in which twenty-eight earth pressure cells were installed in the piled embankment and an M-shaped wave was adopted to simulate the high-speed railway train load.The results show that dynamic soil arching only occurs when two bogies of a carriage pass by and disappears at other times.The dynamic soil arching and membrane effect are the most significant under the concrete base.The arching height,stress concentration ratio and pile-soil load sharing ratio have a minimal value at 25 Hz.The dynamic soil arching degrades severely at 25 Hz,whose height at 25 Hz is only 0.35 times that at 5 Hz.The arching height fluctuates over a narrow range with increasing loading amplitude.The stress concentration ratio and the pile-soil load sharing ratio increase monotonically as the loading amplitude increases. 展开更多

Methods: The RBClife span of 30 normal human subjects and 30patients with CAA were measured by sir labelled technique before and after TCM--WMtherapy. The morphology and distribution ofRBC membrane protein granules were observed by freeze fracture etching and transmission electron microscope. Results: The halflife of erythrocytes (RBC TI/2)was shortenedin CAA cases and there was a significant difference compared to healthy control (P <0. 01). After therapy, the RBC life span prolonged and approached the normal level. Before treatment, there existed abnormal in morphology, decrease in amount and uneven indistribution of protein granules in protoplasmicface (PF) and extracellular face (EF) of RBCmembrane. After treatment, the protein granules of RBC membrane was improved and approached to control. Conclusions: The morphology, amount, quality and distribution ofRBC membrane protein granule were closelyrelated to its life span. The therapeutic effectof TCM--WM was better than that of WMalone and it had a function both in stabilizingmembrane protein and extending the RBC lifespan. 展开更多

The removal of iodide(I^(−))from source waters is an effective strategy to minimize the formation of iodinated disinfection by-products(DBPs),which are more toxic than their brominated and chlorinated analogues.In this work,a nanocomposite Ag-D201 was synthesized by multiple in situ reduction of Ag-complex in D201 polymer matrix,to achieve highly efficient removal of iodide from water.Scanning electron microscope/energy dispersive spectrometer characterization showed that uniform cubic silver nanoparticles(AgNPs)evenly dispersed in the D201 pores.The equilibrium isotherms data for iodide adsorption onto Ag-D201 was well fitted with Langmuir isotherm with the adsorption capacity of 533 mg/g at neutral pH.The adsorption capacity of Ag-D201 increased with the decrease of pH in acidic aqueous solution,and reached themaximum value of 802 mg/g at pH 2.This was attributed to the oxidization of I^(−),by dissolved oxygen under the catalysis of AgNPs,to I2 which was finally adsorbed as AgI3.However,the aqueous solutions at pH 7–11 could hardly affect the iodide adsorption.The adsorption of I^(−)was barely affected by real water matrixes such as competitive anions(SO_(4)^(2−),NO_(3)^(−),HCO3−,Cl^(−))and natural organic matter,of which interference of NOM was offset by the presence of Ca^(2+).The proposed synergistic mechanism for the excellent performance of iodide adsorption by the absorbent was ascribed to the Donnan membrane effect caused by the D201 resin,the chemisorption of I^(−)by AgNPs,and the catalytic effect of AgNPs. 展开更多
