Preparation, Characterization of Superacid SO_4^(2-)/ZrO_2-SiO_2 and Its Activity on Catalytic Synthesis of Methyl p-Nitrobenzoate 被引量:2

Solid superacid SO42-/ZrO2-SiO2 was prepared by dip-impregnation method, and its catalytic activity was tested with esterification of p-nitrobenzoic acid and methanol. The optimum conditions were also found, that is, the molar ratio between silica and zirconia was 10:1, the calcination temperature was 550 ℃ and the soaked consistency of H2SO4 was 1.0 mol.L-1. Under the conditions that the ratio of methanol to aeid(mL/ g) was 12:1, the amount of catalyst was 0.5 g, the reaction time was 5 h and the stirring speed was 800 r.min-1, the yield of methyl p-nitrobenzoate could reach up to 90.5%. Then the characterizations of cataslyst, including the acidity and types of acidic centers, specific surface area and surface structure was respectively examined by Hammer indicator, in-situ pyridine IR, BET method, FT-IR(KBr), transmission electron microscope (TEM), and X-ray diffraction (XRD). The results showed that the catalyst SO42-/ZrO2-SiO2 was superacid with high specific surface area due to Ho〈-12.70. It contained acidic sites of Lewis and BrФnsted, and its surface structure changed after esterification. The zirconia crystal was mainly tetragonal and silica crystal was not found. 展开更多

Polycyclic aromatic hydrocarbons(PAHs),dimethyl beazanthracene(DMBA)and benzo-(a)-pyrene(BaP),stimulate the expression of the P1-450 gene in human lung tumor cells(ChaGo)in culture.A concentration-and time-dependent increase in the level of P1-450 specific mRNA sequences hasbeen observed in ChaGo cells treated with sublethal concentrations of DMBA.Results presentedsuggest that the parent compound causes the induction of P1-450 gene expression.The methylationpattern of the internal“-C-” residues of the “-CCGG-” sequence has been studied in a stretch ofabout 8000 base pairs(bp)of DNA sequence in and around the P1-450 gene in the control andDMBA-treated ChaGo cells.A comparative Southern blot analysis of the Msp Ⅰ/Hpa Ⅱ digestedDNA of the control and DMBA-treated ChaGo cells followed by hybridization with ^(32)P-labelledregion-specific probes,revealed that i)the internal “-C-” residues of the “-CCGG-” sequences ofdifferent regions of P1-450 gene were methylated to different degrees;ii)DMBA treatment of thecells induced hypomethylation of only the 5' end “-CCGG-” sequences of the gene;and iii)themethylation patterns of “-CCGG-” sequences of most of the coding region and of the 3' end regionof P1-450 gene were not affected by such DMBA treatment. 展开更多

Aberrant CpG methylation of specificpromoters results in inactivation of tumor suppressor genes and therefore plays an important role in carcinogenesis. Thep53-regulated gene 14-3-3σ undergoes frequent epigenetic silencing in several types of cancer, including carcinoma ofthe breast, prostate, and skin, suggesting that the loss of 14-3-3σ expression may be causally involved in tumor progression.Functional studies demonstrated that 14-3-3σ is involved in cell-cycle control and prevents the accumulation of chro-mosomal damage. The recent identification of novel 14-3-3σ-associated proteins by a targeted proteomics approachimplies that 14-3-3σ regulates diverse cellular processes, which may become deregulated after silencing of 14-3-3σexpression in cancer cells. 展开更多

以ZrOCl2·8H2O和正硅酸乙酯(TEOS)为原料,采用溶胶-凝胶法和透析法制备固体超强酸SO42-/ZrO2-SiO2催化剂,考察了制备条件对硅、锆凝胶化时间的影响,并以合成对硝基苯甲酸甲酯为探针反应,研究了催化剂制备条件对催化活性的影响,最... 以ZrOCl2·8H2O和正硅酸乙酯(TEOS)为原料,采用溶胶-凝胶法和透析法制备固体超强酸SO42-/ZrO2-SiO2催化剂,考察了制备条件对硅、锆凝胶化时间的影响,并以合成对硝基苯甲酸甲酯为探针反应,研究了催化剂制备条件对催化活性的影响,最后采用Hammett指示剂法、BET法、FT-IR、TEM和XRD分别对催化剂的酸强度、比表面、反应前后骨架结构和形貌变化、晶相进行了表征。结果表明:(1)当水浴温度为80℃,水10mL时,锆凝胶化时间最短;(2)水浴温度为80℃,水8mL,pH=3.0-4.0时硅凝胶化时间最短;(3)当硅、锆原子摩尔比为10:1,H2SO4浸渍液浓度为1.0mol·L-1,焙烧温度550℃时,催化剂的活性最高,酯产率为70%左右;(4)催化剂酸强度Ho<-12.70,反应前后催化剂比表面变化较大且结构形态存在较大差异,催化剂中的SiO2为无定形态,ZrO2为四方晶态。 展开更多

The a-amino acid derivatives constitute a class of compounds of particular medicinal and synthetic attention and considerable interest has been devoted to their synthesis in recent years.In the present work,we develop the computational study of the synthesis reaction of new pyrazolyl a-amino esters derivatives using the Gaussian 09based on the DET/B3LYP density functional theorv method,with the base 6-31G(d.p)to ensure the possibility of carrying out these reactions within the laboratory of synthesis.Indeed,this research has encouraged us to establish an economical synthesis strategy of these products in overall vields of 73.5%to 87%to have access to new active biomolecule through the O-alkvlation reaction between methyl a-azidoglycinate N-benzoylated and primary pyrazole alcohols(3,5-dimethyl-1 H-pyrazol-1-yl)methanol,(1H-pyrazol-1-yl)methanol and(3-ethoxy-5-methyl-1H-pyrazol-1yl)methanoll under different operating conditions.The structure of the prepared heterocyclic systems was characterized by conventional spectroscopic techniques,like H NMR.I3℃NMR,and MS.The results revealed that the experimental study is in good correlation with the computational one. 展开更多
