Elaborate regulation of gene expression is required for plants to maintain normal growth,development,and reproduction.MicroRNAs(miRNAs)and transcription factors are key players that control gene expression in plant re...Elaborate regulation of gene expression is required for plants to maintain normal growth,development,and reproduction.MicroRNAs(miRNAs)and transcription factors are key players that control gene expression in plant regulatory networks.The TEOSINTE BRANCHED1/CYCLOIDEA/PROLIFERATING CELL FACTOR(TCP)family comprises plantspecific transcription factors that contain a conserved TCP domain of 59 amino acids.Some members of this family are targeted by miR319,one of the most ancient and evolutionarily conserved miRNAs in plants.Accumulating evidence has revealed that miR319-regulated TCP(MRTCP)genes participate extensively in plant development and responses to environmental stress.In this review,the structural characteristics and classifications of TCP transcription factors and the regulatory relationships between TCP transcription factors and miRNAs are introduced.Current knowledge of the regulatory functions of MRTCP genes in multiple biological pathways including leaf development,vascular formation,flowering,hormone signaling,and response to environmental stresses such as cold,salt,and drought is summarized.This review will be beneficial for understanding the roles of the MRTCP-mediated regulatory network and its molecular mechanisms in plant development and stress response,and provides a theoretical basis for plant genetic improvement.展开更多

However, little is known about the inter- play between the developmental regulators and the cell cycle machinery--a link that ultimately affects leaf form and size. miR319 is a conserved microRNA that regulates TCP transcription factors involved in multiple developmental pathways, including leaf development and senescence, organ curvature, and hormone biosynthesis and signaling. Here, we analyze the participation of TCP4 in the control of cell proliferation. A small increase in TCP4 activity has an immediate impact on leaf cell number, by significantly reducing cell proliferation. Plants with high TCP4 levels have a strong reduction in the expression of genes known to be active in G2-M phase of the cell cycle. Part of these effects is mediated by induction of miR396, which represses Growth-Regulating Factor (GRF) transcription factors. Detailed analysis revealed TCP4 to be a direct regulator of MIR396b. However, we found that TCP4 can control cell proliferation through additional pathways, and we identified a direct connection between TCP4 and ICK1/KRP1, a gene involved in the progression of the cell cycle. Our results show that TCP4 can activate different pathways that repress cell proliferation.展开更多

However, the roles and action modes of specific miRNAs involved in viral infection and host susceptibility remain largely unclear. In this study, we show that Rice ragged stunt virus (RRSV) infection caused increased accumulation of miR319 but decreased expression of miR319-regulated TCP (TEOSINTE BRANCHED/ CYCLOIDEA/PCF) genes, especially TCP21, in rice plants. Transgenic rice plants overexpressing miP,319 or downregulating TCP21 exhibited disease-like phenotypes and showed significantly higher susceptibility to RRSV in comparison with the wild-type plants. In contrast, only mild disease symptoms were observed in RRSV-infected lines overexpressing TCP21 and especially in the transgenic plants overexpressing miR319- resistant TCP21. Both RRSV infection and overexpression of miR319 caused the decreased endogenous jasmonic acid (JA) levels along with downregulated expression of JA biosynthesis and signaling-related genes in rice. However, treatment of rice plants with methyl jasmonate alleviated disease symptoms caused by RRSV and reduced virus accumulation. Taken together, our results suggest that the induction of miR319 by RRSV infection in rice suppresses JA-mediated defense to facilitate virus infection and symp- tom development.展开更多
