One hundred and fifty Miichthys miiuy weighing approximately 200-260 g were divided into five groups and reared in 15 tanks with closed circuiting culture system. The animals were fed 5 diets: basal diet only (control) or supplemented of the basal diet with C. butyricum at doses of 10^3 (CB1), 10^5 (CB2), 10^7 (CB3) or 10^9 (CB4) CFU/g. Compared with the control, the serum phenoloxidase activity was significantly increased by the supplementation (P〈0.05), acid phosphatases activity was increased significantly (P〈0.05) at the doses of 10^9 CFU/g. Serum lysozyme activity peaked at dose of 10^7 CFU/g and in the skin mucus at dose of 10^9 CFU/g. Immunoglobulin M level in the serum and skin mucus was increased except at dose of 10^3 CFU/g (P〈0.05). The growth at the dose of 10^9 CFU/g was higher than that of the control (P〈0.05). It is concluded that supplementation of C. butyricum can mediate the humoral immune responses and improve the growth performance in Miichthys miiuy.展开更多

Nine hundred Miichthys miiuy fries were divided into 3 groups, each with triplicates. The basal diet and the basal diet supplemented with carnitine groups were considered as the negative and positive controls respectively. Results showed that the nitrogen concentration in excreted feces decreased significantly in fries fed the diet supplementation with 1000×10?6 fructooligosaccharides and 200×10?6 carnitine (P<0.05). The ammonic-nitrogen concentration decreased significantly in the carnitine group only (P<0.05), indicating the decreasing tendency caused by the supplementation with fructooligosaccha-rides. Supplementation with both did not have significant effects on the concentration of phosphorus in feces of Miichthys miiuy fries.展开更多

Histamine is a type of biogenic amine,which plays a major role in the health problems associated with seafood consumption.Legislative limits of histamine level in seafood have been set in many countries.This study foc...Histamine is a type of biogenic amine,which plays a major role in the health problems associated with seafood consumption.Legislative limits of histamine level in seafood have been set in many countries.This study focuses on investigating the feasibility and potentiality of determining histamine concentration in fish(Miichthys miiuy)by surface-enhanced Raman spectroscopy(SERS)combined with density functional theory(DFT).Both a gold colloid and a silver colloid were used to determine the enhancement effect for SERS detection of histamine standard solution,and the gold colloid exhibited more effective as compared to the silver colloid.The protocol on extraction of histamine with 12%trichloroacetic acid and adjustment of pH with NaOH solution(5 mol/L)could significantly shorten sample preparation(20 min)and provide clear SERS spectra of histamine.The peaks of histamine molecules were classified using the DFT and five spectra(953 cm^(-1),992 cm^(-1),1106 cm^(-1),1262 cm^(-1)and 1317 cm^(-1))were selected as the characteristic bands of histamine discrimination.Moreover,the intensity of the peak at 1262 cm^(-1)had a good linear relationship with histamine concentration at 5-400 mg/kg with R^(2)=0.9755.It is concluded that the SERS-DFT approach will be a potential method for rapidly and reliably detecting histamine at levels from 5 mg/kg to 400 mg/kg in fresh fish.展开更多
Inflammation is a self-protection mechanism that can be triggered when innate immune cells detect infection.Eradication of pathogen infection requires appropriate immune and inflammatory responses,but excessive inflam...Inflammation is a self-protection mechanism that can be triggered when innate immune cells detect infection.Eradication of pathogen infection requires appropriate immune and inflammatory responses,but excessive inflammatory responses can cause uncontrolled inflammation,autoimmune diseases,or pathogen dissemination.Mounting evidence has shown that microRNAs(miRNAs)in mammals act as important and versatile regulators of innate immunity and inflammation.However,miRNAmediated regulation networks are largely unknown in inflammatory responses in lower vertebrates.Here,miR-144 and miR-217 are identified as negative regulators in teleost inflammatory responses.We find that Vibrio harveyi and lipopolysaccharide(LPS)treatment significantly upregulate the expression of fish miR-144 and miR-217.Upregulated miR-144 and miR-217 suppress LPS-induced inflammatory cytokine expression by targeting nucleotide-binding oligomerization domain-containing protein 1(NOD1),thereby avoiding excessive inflammatory responses.In addition,miR-144 and miR-217 regulate inflammatory responses through NOD1-induced nuclear factor kappa(NF-κB)signaling pathways.These findings demonstrate that miR-144 and miR-217 play regulatory roles in inflammatory responses by modulating the NOD1-induced NF-κB signaling pathway.展开更多
