To more realistically simulate the actual de...An efficient reflux extraction of polyethylene wax(PEW) in soil is presented, followed by molecular structure characterization methods to explore its degradation mechanism. To more realistically simulate the actual degradation of PE film powders in soil, low density PE(M=5 000) powders, being used as simulated PEW residue sample, were uniformly mixed with soil and then recovered by reflux extraction with decahydronaphthalen(decalin) at 90 ?C for 60 min. The average recovery of PEW from fortified soils was 96.5% with the developed reflux extraction procedure. The recovered PEW residue samples were characterized by infrared spectroscopy(IR), element analysis(EA), X-ray fluorescence(XFR), and high-temperature gel permeation chromatography(GPC). The results from spectra analysis show that there were no significant changes in molecular structures and molecular mass distribution of PEW samples after the reflux extraction, which demonstrate the reliability of this method. These results also indicate that the reflux extraction procedure and analytical methods of characterization could serve as a novel measurement technique to evaluate the degradation of low-density PE powders in soil over time.展开更多

However, its effects on rat hippocampal neurons and the mechanism of protection remain unclear....The alcohol and n-butanol extract of Potentilla anserine L. significantly protects myocardium from acute ischemic injury. However, its effects on rat hippocampal neurons and the mechanism of protection remain unclear. In this study, primary cultured hippocampal neurons from neonatal rats were incubated in 95% N2 and 5% CO2 for 4 hours. Results indicated that hypoxic injury decreased the viability of neurons, increased the expression levels of caspase-9 and caspase-3 mRNA, as well as cytochrome c, Caspase-9, and Caspase-3 protein. Pretreatment with 0.25, 0.062 5, 0.015 6 mg/mL n-butanol extract of Potentilla anserine L. led to a significant increase in cell viability. Expression levels of caspase-9 and caspase-3 mRNA, as well as cytochrome c, Caspase-9, and Caspase-3 protein, were attenuated. The neuroprotective effect of n-butanol extract of Potentilla anserine L. was equivalent to tanshinone IIA. Our data suggest that the n-butanol extract of Potentilla anserine L. could protect primary hippocampal neurons from hypoxic injury by deactivating mitochondrial cell death.展开更多

In this study, the activity of n-butanol fractions of B. monosperma floral extract was examined on inhibiting aberrant crypt foci(ACF) formation in azoxymethane induced Wistar albino rats. The n-butanol extracts(150 mg/kg) decreased the ACF formation(per rat) by 92% and78% in short- and long-term in vivo treatments, respectively. All the compounds in the n-butanol extract were isolated and purified using column and reverse-phase high pressure liquid chromatography(HPLC).Their structures were characterized using UV–visible spectroscopy, nuclear magnetic resonance(NMR)and electrospray–ionisation mass spectrometry(ESI–MS) to determine important flavonoids, namely isocoreopsin, butrin and isobutrin. These compounds were studied for their free radical scavenging and anticancer activities. The compound isocoreopsin showed significantly greater efficacy in cell death on human colon and liver cancer cell lines(50 μg/m L in HT-29 and 100 μg/m L in Hep G2) than butrin(100 μg/m L in HT-29 and 500 μg/m L in Hep G2) and isobutrin(80 μg/m L in HT-29 and 150 μg/m L in Hep G2). These results suggest that isocoreopsin, butrin and isobutrin are the important key compounds for the chemoprevention of colon cancer and isocoreopsin can be considered as a promising novel drug.展开更多

[ Method] Pomegranate peels were used as materials for single factor experiment to determine the optimal technique for effective ingredient extraction from pomegranate peels and explore the inhibition of the extracts on tyrosinase and whitening effect of the effective ingredient of pomegranate peels. [ Result] The optimal conditions for ultrasound-assisted extraction of effective ingredient in pomegranate peels were: liquid-to-solid ratio of 1:20 with 60% ethanol, extraction temperature of 60℃, ultrasonic intensity of 7 and pro- cessing time of 40 min, which led to relatively strong inhibition effect of pomegranate peel extracts on tyrosinase. The inhibition rate of extracts obtained with com- mon solvent reflux extraction method was only around 60%, while the inhibition rate of extracts obtained with ultrasound-assisted extraction method on tyrosinase ac- tivity had achieved around 90%, showing a very significant the difference, which indicated that during the process of extraction of effective ingredient from pome- granate peels, appropriate ultrasound-assistance was advantageous to the extraction of active ingredients; in addition, the inhibition effect of extracts on tyrosinase had increased with the increasing concentration of extracts, and the inhibition rate could achieve 100%. This activity was attributed to active ingredients extracted with N-butyl alcohol, named Schisandra N-butanol extract. In this study, ovariectomized mice were pretreated with Schisandra N-butanol extract given by intragastric administration. This treatment led to the enhancement of learning, and an increase in hippocampal CA1 synaptic, surface and postsynaptic density. A decrease in the average size of the synaptic active zone was also observed. These experimental findings showing that Schisandra N-butanol extract improved synaptic morphology indicate an underlying mechanism by which the ability of learning is enhanced in ovariectomized mice.展开更多
[Objectives]This study was conducted to investigate the extraction method of Impatiens balsamina L.(Tougucao),and to optimize the extraction process conditions and parameters.[Methods]Different ethanol concentrations,...[Objectives]This study was conducted to investigate the extraction method of Impatiens balsamina L.(Tougucao),and to optimize the extraction process conditions and parameters.[Methods]Different ethanol concentrations,ethanol amounts and extraction time were investigated by an orthogonal test L_(9)(3^(4))and high performance liquid chromatography(HPLC),and the extraction process parameters were evaluated by the yield of extract and the content of quercetin.[Results]Under the three optimal extraction conditions of impregnation,heating reflux and ultrasound,the extract yield and quercetin contents of I.balsamina were 8.27%,9.50%,7.09%and 1.60,3.05,3.03μg/g,respectively.Heating reflux extraction and ultrasonic extraction showed higher extract yields and quercetin contents of I.balsamina,while the values were lower under the impregnation extraction conditions.The primary and secondary factors were:ethanol amount(C)>ethanol concentration(A)>extraction time(B).The optimal extraction process of I.balsamina was as follows:adding 10 times of 60%ethanol and ultrasonically extracting for 30 min.[Conclusions]The method is simple,convenient,accurate and reliable,and provides a scientific basis for the research on the extraction process of I.balsamina and its preparations.展开更多
Objective:To determine the extraction suitable conditions of total phenolic content(TPC) by heat-reflux system,antioxidant activities and HPLC characterization of the aqueous-ethanolic extracts of Jatropha dioica(J.di...Objective:To determine the extraction suitable conditions of total phenolic content(TPC) by heat-reflux system,antioxidant activities and HPLC characterization of the aqueous-ethanolic extracts of Jatropha dioica(J.dioica)(Dragon's blood).Flourensia cernua(F.cernua)(Tar bush).Eucalyptus camaldulensis(E.camaldulensis)(Eucalyptus) and Turnera diffusa(T.diffusa)(Damiana).Methods:TPC was evaluated by the well-known colorimetric assay using FolinCiocalteu reagent.The antioxidant activities were assayed by three methods based on scavenging of DPPH,ABTS and by lipid oxidation inhibition.The chemical composition of the extracts obtained was subject to HPLC analysis.Results:TPC in the plant extracts ranged from 2.3 to14.12 mg gallic acid equivalents/g for J.dioica and E.camaldulensis,respectively.The plant extracts of F.cernua,E.camaldulensis and T.diffusa showed similar strong antioxidant activities on scavenging of DPPH and lipid oxidation inhibition.In contrast,J.dioica extracts had lowest potential antioxidant in three assays used.HPLC assay showed the presence of several phenolic compounds in the extracts used.Conclusions:The results obtained suggest that F.cernua,E.camaldulensis and T.diffusa are potential sources to obtain bioactive phenolic compounds with high antioxidant properties which can be used in the factories as antioxidant agents or for treatments in diseases.展开更多
