AIM:To assess the expression of nestin and glial fibrillary acidic protein(GFAP)in rat retina after optic nerve transection.METHODS:Rats were randomly divided into normal control group,sham group and operation group,and used for establishing an animal model of optic nerve transection.Retinal specimen of each group was collected at 3,48h,7and 14d postoperative.Nestin and GFAP expressions on sagittal sections were analyzed by immunohistochemical staining,and protein extraction was analyzed by Western blot.RESULTS:Immunohistochemical analysis showed that nestin positive staining was rarely detected in normal control group and sham group,while sham group showed weak positive staining at 3h postoperative,the reaction gradually increased at 48h postoperative,and reached its maximum at 7d postoperative,and then decreased at 14d postoperative.Compared to the expression of GFAP,there was not statistically significant obvious difference among three groups(P〉0.05).Result of Western blot method was consistent with that of immunohistochemical method.CONCLUSION:The expression of nestin increased in a time dependent fashion in Müller cells of retina following optic nerve transection,which was statistically significant,but there was no obvious difference in GFAP expression.The results indicate that an increase in colloid synthesis in retina following optic nerve transection can improve the retinal neurons’environment. 展开更多

Dynamic changes of glial fibrillary acidic protein and nestin in the hippocampus of adult rat brain following ischemic vascular dementia

Vascular dementia produced by permanent ligation of bilateral common carotid arteries involves progressive deterioration of intellectual and cognitive function in rats, which are closely associated with the hippocampus. This study used immunohistochemical analysis to detect the expression of glial fibrillary acidic protein and nestin in the hippocampus in a vascular dementia model. The results revealed that both glial fibrillary acidic protein and nestin expression were increased 1 day after permanent ligation of the bilateral common carotid arteries, compared with a sham-operated group. The expression of glial fibrillary acidic protein peaked at 7 days post-surgery. The expression of nestin was a little weaker than that of glial fibrillary acidic protein, and peaked at 14 days (P 〈 0.01). The expression of both proteins slightly decreased at 21 and 28 days, accompanied by recovery of cerebral blood flow. In conclusion, this study demonstrated that glial fibrillary acidic protein and nestin exhibited dynamic expression in the rat hippocampus after permanent ligation of bilateral common carotid arteries. This finding suggests that dynamic alterations in protein expression play an important role in the pathogenesis of vascular dementia. 展开更多

Expression of Nestin and GFAP in different period (1,3,5d ays;1-8 weeks) and different area(injury locus and its surrounding tissue ) afte r spinal injury were observed pathologicaly using immunofluorescence and LeicaQ5 00IW imaging processing system. Results There was expression of Nestin and GFAP in injured area 1 day after injury.The expression increased not only in in injured area but its sourrounding 3-7 days later and gradually got t o peak value. As the time went on, expression of Nestin and GFAP dereased. Conclusion Spinal injury can induce the expression of Nestin. Nerve stem cell has response to spinal injury. There is positive correlation between e xpression of Nestin and hyperplasia of reactivity astrocyte. Nerve stem cell may be invovled in the repair of central nervous system (CNS). 展开更多

目的观察电针对脑缺血再灌注损伤后大鼠齿状回巢蛋白(nestin)及胶质纤维酸性蛋白(GFAP)表达的影响。方法将SD雄性大鼠随机分为假手术组、模型组和电针组,每组再分为3、7、14和21d等4个亚组,每个亚组6只大鼠,采用大脑中动脉线栓法制作脑... 目的观察电针对脑缺血再灌注损伤后大鼠齿状回巢蛋白(nestin)及胶质纤维酸性蛋白(GFAP)表达的影响。方法将SD雄性大鼠随机分为假手术组、模型组和电针组,每组再分为3、7、14和21d等4个亚组,每个亚组6只大鼠,采用大脑中动脉线栓法制作脑缺血再灌注模型。电针组选取"百会"、"大椎"穴给予2Hz电针刺激,持续30min,每天1次。采用免疫组织化学染色法显示各组大鼠齿状回nestin及GFAP的表达。结果①模型组nestin的表达在3d、7d、14d时明显高于假手术组(P<0.01),21d时和假手术组比较无显著性差异(P>0.05);电针组各时间点nestin的表达均显著高于模型组(P<0.01或P<0.05)。②模型组GFAP的表达在各时间点均明显高于假手术组(P<0.01);电针组在7d、14d时GFAP的表达强度明显低于模型组(P<0.01或P<0.05),但细胞数变化不明显(P>0.05),21d时GFAP阳性细胞数及表达强度较模型组均显著降低(P<0.01)。结论电针能明显增强急性脑缺血再灌注大鼠齿状回nestin的表达,促进神经再生;并可以下调GFAP的持续过度表达,抑制星型胶质细胞的过度活化和增殖。 展开更多

目的探讨丹龙醒脑方对大鼠脑缺血损伤后海马CA1区巢蛋白(Nestin)和胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)表达的影响。方法将48只大鼠随机分为假手术组、模型组、依达拉奉组、丹龙醒脑方组。采用大脑中动脉线栓法制... 目的探讨丹龙醒脑方对大鼠脑缺血损伤后海马CA1区巢蛋白(Nestin)和胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)表达的影响。方法将48只大鼠随机分为假手术组、模型组、依达拉奉组、丹龙醒脑方组。采用大脑中动脉线栓法制备大脑中动脉闭塞再灌注(MCAO)模型,对大鼠进行神经功能缺损评分,采用HE染色观察海马CA1区细胞的形态学改变,采用免疫组化法检测海马CA1区Nestin蛋白和GFAP蛋白的表达。结果与假手术组比较,模型组治疗3 d后和7 d后的神经功能缺损评分明显升高,差异有统计学意义(P<0.05);与模型组比较,依达拉奉组和丹龙醒脑方组治疗3 d后和7 d后的神经功能缺损评分明显降低,差异有统计学意义(P<0.05);丹龙醒脑方组在治疗3 d和7 d后Nestin蛋白的平均光密度增加(P<0.05),GFAP蛋白的平均灰度值减少(P<0.05)。结论丹龙醒脑方对海马CA1区病理形态有保护作用。丹龙醒脑方抗脑缺血损伤可能与促进海马区神经细胞增殖和分化有关。 展开更多

Nestin蛋白是一种神经干细胞标志物,参与组织修复,并且在一些肿瘤细胞中表达。最近研究报道称nestin可能参与肿瘤细胞恶性增殖及细胞侵袭。在急性淋巴细胞白血病(acute lymphoid leukemia,ALL)小鼠模型中,白血病增殖细胞(leukemia-propagating cells,LPCs)可形成一个抗药性niche,其中nestin阳性细胞参与该niche形成。为探索nestin基因在白血病患者的临床表达情况,通过RT-PCR和Western-blot实验方法 ,检测了50位髓系白血病患者的nestin基因的临床表达情况。结果发现,在50位髓系白血病患者(38位AML,12位CML)中,42位患者表达nestin基因mRNA,6位患者表达nestin蛋白,并且nestin基因表达水平和患者血红蛋白、血小板、细胞因子和T淋巴细胞数并不存在相关性,但是高白细胞白血病患者表现出nestin基因mRNA水平高表达。虽然nestin基因和患者完全缓解率(complete remission,CR)并不存在联系,但是nestin基因在髓系白血病异常表达,说明其可能作为一种诊断AML或CML的生物标记物。 展开更多

目的观察骨形态发生蛋白-7(BMP-7)对大鼠局灶性脑缺血/再灌注损伤后巢蛋白(nestin)和胶质纤维酸性蛋白(GFAP)表达的影响。方法成年♂SD大鼠40只,随机分为假手术组、对照组、BMP-7治疗组,每组10只。应用线栓法制备大脑中动脉阻塞2 h模型... 目的观察骨形态发生蛋白-7(BMP-7)对大鼠局灶性脑缺血/再灌注损伤后巢蛋白(nestin)和胶质纤维酸性蛋白(GFAP)表达的影响。方法成年♂SD大鼠40只,随机分为假手术组、对照组、BMP-7治疗组,每组10只。应用线栓法制备大脑中动脉阻塞2 h模型,治疗组尾静脉注射BMP-7(0.1 mg.kg-1)250μl,对照组和假手术组尾静脉注射等量的无菌注射用水。再灌注24h后行神经功能评分,2,3,5-三苯四氮唑(TTC)染色观察脑梗死体积,TUNEL法检测细胞凋亡,免疫组织化学方法测定nestin和GFAP表达。结果与对照组相比,BMP-7组大鼠神经功能评分(t=4.66,P<0.01)和脑梗死体积百分比降低(t=6.98,P<0.01),TUNEL阳性细胞数降低(P<0.01)。缺血侧脑组织nestin和GFAP表达增加(P<0.01)。结论 BMP-7可上调大鼠脑缺血/再灌注损伤后nestin和GFAP表达,促进神经再生,起神经保护作用。 展开更多

【目的】建立Nestin-GFP转基因小鼠的胚胎干细胞(mESC)系,并观察其示踪ES细胞系的神经分化过程。【方法】将E3.5的Nestin-GFP小鼠囊胚接种于小鼠胚胎成纤维细胞上(MEF经γ射线照射失去增殖活性)。4~6d后将自囊胚长出的内细胞团(ICM)继... 【目的】建立Nestin-GFP转基因小鼠的胚胎干细胞(mESC)系,并观察其示踪ES细胞系的神经分化过程。【方法】将E3.5的Nestin-GFP小鼠囊胚接种于小鼠胚胎成纤维细胞上(MEF经γ射线照射失去增殖活性)。4~6d后将自囊胚长出的内细胞团(ICM)继续培养并传代扩增,检测所得细胞Oct-4、SSEA-1和AKP等胚胎干细胞特异性标志物的表达及在体内外向三胚层分化的能力;之后,诱导细胞向神经元样细胞及神经胶质样细胞分化,利用免疫荧光检测其标志性抗原。【结果】免疫组化可见Nestin-GFP小鼠胚胎干细胞表达未分化细胞特异性标志Oct-4、SSEA-1,AKP染色呈强阳性,并具有向体内外三胚层分化潜能;在体外诱导其向神经分化,第6d可见到明显的绿色荧光,并与免疫组化Nestin表达部位基本吻合。第12d及第16d分别可见神经元及神经胶质细胞特异性标志物表达,而此时Nestin的表达较前明显下降。【结论】成功建立了Nestin-GFP小鼠胚胎干细胞系,该细胞具有自我更新的特性及多向分化潜能,并可用于在体外更直观的监测神经分化的阶段性变化并由此进行调控。 展开更多

BACKGROUND: Studies have demonstrated that astrocytes may possess similar properties to neural stem cells/neural precursor cells and have the potential to differentiate into neurons. OBJECTIVE: To observe neuroepithelial stem cell protein (nestin) and glial fibrillary acidic protein (GFAP) expression following spinal cord injury, and to explore whether nestin+/GFAP+ cells, which are detected at peak levels in gray and white matter around the ependymal region of the central canal in injured spinal cord, possess similar properties of neural stem cells. DESIGN, TIME AND SETTING: A randomized, controlled experiment. The study was performed at the Key Laboratory of Environment and Genes Related to Diseases (Xi'an Jiaotong University), Ministry of Education between January 2004 and December 2006. MATERIALS: Rabbit anti-rat nestin, β-tubulinⅢ, mouse anti-rat GFAP, galactocerebroside (GaLC) antibodies were utilized, as well as flow cytometry. METHODS: A total of 60 male, Sprague Dawley rats, aged 8 weeks, were randomly assigned to control (n = 12) and model (n = 48) groups. The spinal cord injury model was established in the model group by aneurysm clip compression, while the control animals were not treated. The gray and white matter around the ependymal region of the central canal exhibited peak expression of nestin+/GFAP+ cells. These cells were harvested and prepared into single cell suspension, followed by primary and passage cultures. The cells were incubated with serum-containing neural stem cell complete medium. MAINOUTCOME MEASURES: Nestin and GFAP expression in injured spinal cord was determined using immunohistochemistry and double-labeled immunofluorescence at 1, 3, 5, 7, 14, 28, and 56 days post-injury. In addition, cell proliferation and differentiation were detected using immunofluorescence cytochemistry and flow cytometry. RESULTS: Compared with the control group, the model group exhibited significantly increased nestin and GFAP expression (P 〈 0.05), which reached peak levels between 3 and 7 days. The majority of cells in the ependymal region around the central canal were nestin+/GFAP- cells, while the gray and white matter around the ependymal region were full of nestin+/GFAP+ cells, with an astrocytic-like appearance. A large number of nestin+/GFAP+cells were observed in the model group cell culture, and the cells formed clonal spheres and displayed strong nestin-positive immunofluorescence staining. Following induced differentiation, a large number of GaLC-nestin, β-tubulin Ⅲ-nestin, and GFAP-nestin positive cells were observed. However, no obvious changes were seen in the control group. Cells in S stage, as well as the percentage of proliferating cells, in the model group were significantly greater than in the control group (P 〈 0.01), CONCLUSION: Spinal cord injury in the adult rat induced high expression of nestin+/GFAP+ in the gray and white matter around the ependymal region of the central canal. These nestin+/GFAP+ cells displayed the potential to self-renew and differentiate into various cells. The cells could be neural stem cells of the central nervous system. 展开更多

Nestin is a class Ⅵ intermediate filament protein that was originally described as a neuronal stem cell marker during central nervous system (CNS) development, and is currently widely used in that capacity. Nestin is also expressed in non-neuronal immature or progenitor cells in normal tissues. Under pathological conditions, nestin is expressed in repair processes in the CNS, muscle, liver, and infarcted myocardium. Furthermore, increased nestin expression has been reported in various tumor cells, including CNS tumors, gastrointestinal stromal tumors, pancreatic cancer, prostate cancer, breast cancer, malignant melanoma, dermatofibrosarcoma protuberances, and thyroid tumors. Nestin is reported to correlate with aggressive growth, metastasis, and poor prognosis in some tumors; however, the roles of nestin in cancer cells have not been well characterized. Furthermore, nestin is more specifically expressed in proliferating small-sized tumor vessels in glioblastoma and gastric, colorectal, and prostate cancers than are other tumor vessel markers. These findings indicate that nestin may be a marker for newly synthesized tumor vessels and a therapeutic target for tumor angiogenesis. It has received a lot of attention recently as a cancer stem cell marker in various cancer cells including brain tumors, malignant rhabdoid tumors, and uterine, cervical, prostate, bladder, head and neck, ovarian, testicular, and pancreatic cancers. The purpose of this review is to clarify the roles of nestin in cancer cells and in tumor angiogenesis, and to examine the association between nestin and cancer stem cells. Nestin has the potential to serve as a molecular target for cancers with nestin-positive cancer cells and nestin-positive tumor vasculature. 展开更多

Our preliminary studies confirmed that an active principle region of Buyang Huanwu decoction, comprising alkaloid, polysaccharide, aglycon, glucoside and volatile oil, can induce bone marrow mesenchymal stem cell differentiation into neurons. Mitogen-activated protein kinase signaling was identified as one of the key pathways underlying this differentiation process. The present study shows phosphorylated extracellular signal-regulated protein kinase and phosphorylated p38 protein expression was increased after differentiation. Cellular signaling pathway blocking agents, PD98059 and SB203580, inhibited extracellular signal-regulated protein kinase and p38 in mitogen-activated protein kinase signaling pathways respectively, mRNA and protein expression of the neuronal marker, neuron specific enolase, and neural stem cell marker, nestin, were decreased in bone marrow mesenchymal stem cells after treatment with the active principle region of Buyang Huanwu decoction. Experimental findings indicate that, extracellular signal-regulated protein kinase and p38 in mitogen-activated protein kinase signaling pathways participate in bone marrow mesenchymal stem cell differentiation into neuron-like cells, induced by the active principle region of Buyang Huanwu decoction. 展开更多

目的 :观察巢蛋白 (nestin)在局灶脑缺血区的反应模式 ,以探讨其在脑梗塞灶修复过程中的作用。方法 :采用局灶脑缺血模型和免疫组织化学染色方法 ,探查 36只成年雄性SD大鼠的大脑不同区域的巢蛋白阳性细胞的形态、反应时程和分布形式。... 目的 :观察巢蛋白 (nestin)在局灶脑缺血区的反应模式 ,以探讨其在脑梗塞灶修复过程中的作用。方法 :采用局灶脑缺血模型和免疫组织化学染色方法 ,探查 36只成年雄性SD大鼠的大脑不同区域的巢蛋白阳性细胞的形态、反应时程和分布形式。结果 :假手术大鼠的巢蛋白阳性反应存在小血管、微血管和室管膜上皮 ,在第三脑室底壁内有许多细的阳性纤维 ,胞体不明显。在脑缺血 3d组 ,大量巢蛋白阳性细胞分布于缺血侧大脑半球 ,以大脑皮质缺血区、视前区、尾壳核及第三脑室底部最为显著。缺血区周围的大脑皮质浅层和视前区皮质的阳性细胞呈纤维状 ,而皮质深层的阳性细胞则呈星状。巢蛋白阳性细胞的形态和数量变化在脑缺血 1周时最显著。阳性细胞显示高度的肥大和增生性变化 ,其数量亦明显增加 ,以大脑皮质缺血区和尾壳核区最显著。皮质缺血区和尾壳核区的巢蛋白阳性反应持续到脑缺血 6周。随后 ,其反应稍有减弱。结论 :局灶脑缺血诱导反应型星形胶质细胞重表达巢蛋白 ,提示其可能参与脑缺血性损伤的修复过程。 展开更多
