A long-term field experiment (started at 2003) was conducted to determine the effects of different dce cultivation methods on growth characteristics and grain yield of late-season rice under double-rice cropping sys...A long-term field experiment (started at 2003) was conducted to determine the effects of different dce cultivation methods on growth characteristics and grain yield of late-season rice under double-rice cropping system in seasonal drought region of southeast China (Yujiang County, Jiangxi Province). The rice cultivation methods included no-tillage and flooded rice cultivation (N-F), no-tillage and non-flooded rice cultivation with straw mulching (N-SM), and no-tillage and non-flooded rice cultivation without straw mulching (N-ZM). There was no significant difference in rice grain yield between the N-SM and N-F treatments. However, the rice grain yields in the N-SM and N-F treatments were significantly higher than that in the N-ZM treatment. The late-season rice plants in the N-SM treatment had significantly higher numbers of effective panicles and total grains per hill compared with those in the N-ZM treatment. The above-ground dry matter of late-season rice was similar between the N-SM and N-F treatments. Compared with the N-F treatment, the N-ZM and N-SM treatments significantly decreased the leaf area at the heading stage. Moreover, the N-SM treatment could significantly increase total root length and root tip number at the grain-filling stage compared with the N-ZM treatment.展开更多

Long non-coding RNAs(lncRNAs) play important roles in response to various biotic and abiotic stresses. So far, systematic identification and characterization of lncRNAs have been reported in a few model plant species ...Long non-coding RNAs(lncRNAs) play important roles in response to various biotic and abiotic stresses. So far, systematic identification and characterization of lncRNAs have been reported in a few model plant species and major crops, but their roles in abiotic stress response have not yet been reported in common wild rice(Oryza rufipogon). Dongxiang wild rice(DXWR) possesses a high degree of drought resistance and has been well recognized as a precious genetic resource for drought resistant rice breeding. We presented the reference catalog of 1 655 novel lncRNA transcripts in DXWR using strand-specific RNA sequencing and bioinformatics approaches. Meanwhile, a total of 1092 lncRNAs were determined as differentially expressed lncRNAs under drought stress. Quantitative real-time PCR results exhibited a high concordance with RNA sequencing data, which confirmed that the expression patterns of lncRNAs based on RNA sequencing were highly reliable. Furthermore, 8 711 transcripts were predicted as target genes of the differentially expressed lncRNAs. Functional annotation analysis based on Gene Ontology and Kyoto Encyclopedia of Genes and Genomes databases showed that the target genes were significantly enriched in cellular and metabolic processes, cell part, binding and plant hormone signal transduction, as well as many other terms related to abiotic stress resistance. These results expanded our understanding of lncRNA biology and provided candidate regulators for genetic improvement of drought resistance in rice cultivars.展开更多
We evaluated the effects of rice black streak dwarf virus (RBSDV)-infested rice plants on the ecological parameters and its relevant defensive and detoxification enzymes of white-backed planthopper (WBPH) in labor...We evaluated the effects of rice black streak dwarf virus (RBSDV)-infested rice plants on the ecological parameters and its relevant defensive and detoxification enzymes of white-backed planthopper (WBPH) in laboratory for exploring the relationship between RBSDV and the non-vector planthopper. The results showed that nymph survival rate, female adult weight and fecundity, and egg hatchability of WBPH fed on RBSDV-infested rice plants did not markedly differ from those on healthy plants, whereas the female adult longevity and egg duration significantly shortened on diseased plants. Furthermore, significantly higher activities of defensive enzymes (dismutase, catalase and peroxidase) and detoxification enzymes (acetylcholinesterase, carboxylesterase and glutathione S-transferase) were found in WBPH adults fed on infected plants. Results implied that infestation by RBSDV increased the ecological fitness of non-vector planthopper population.展开更多

Field and pot culture experiments were conducted to explore the underlying mechanisms o...Grain filling, a crucial stage of grain yield formation in rice, is usually affected by the panicle nitrogen (N) fertilization. Field and pot culture experiments were conducted to explore the underlying mechanisms of N effect. Two rice cultivars with high lodging resistance were grown in the field and pot. Four panicle N fertilization treatments were conducted in 2006 and repeated in 2007. The result showed that medium level of panicle N fertilization treatment (NM) enhanced the accumulation and translocation of non-structural carbohydrate (NSC) in the stem and sheath. Compared with non-nitrogen treatment (NO), NM promoted the translocation of labeled ^13C from stem and sheath to grain. But, low level of panicle N fertilization treatment (NL) and high level of panicle N fertilization treatment (NH) showed the negative effect. The endosperm cell, grain length, and grain width of NM increased more quickly than that of NO from 4 to 10 d after anthesis. During the early period of grain filling, sucrose-phosphate synthase (EC 2.4.1.14, SPS) activity were significantly higher for the NM treatment than those of the NL and NH treatments. Sucrose synthase (EC 2.4.1.13, SuSase) activity in the grains was substantially enhanced by NM, with the duration of higher activity being longer than those of the other treatments. At maturing stage, NM significantly increased the filled grain number, the seed-setting rate, and the grain weight compared with NL and NH. The results suggest that NM have a positive effect on the activities of enzymes of physiological importance, thereby increasing the grain size and promoting grain filling.展开更多

The total amount of water used by rice unde...A field experiment was conducted to study water use efficiency and agronomic traits in rice cultivated in flooded soil and non-flooded soils with and without straw mulching. The total amount of water used by rice under flooded cultivation (FC) was 2.42 and 3.31 times as much as that by rice under the non-flooded cultivation with and without straw mulching, respectively. The average water seepage was 13 560 m^3/ha under the flooded cultivation, 4 750 m^3/ha under the non-flooded cultivation without straw mulching (ZM) and 4 680 m^3/ha under non-flooded cultivation with straw mulching (SM). The evapotranspiration in the SM treatment was only 38.2% and 63.6% of the FC treatment and ZM treatment, respectively. Compared with the ZM treatment, straw mulching significantly increased leaf area per plant, main root length, gross root length and root dry weight per plant of rice. The highest grain yield under the SM treatment (6 747 kg/ha) was close to the rice cultivated in flooded soil (6 811.5 kg / ha). However, the yield under the ZM treatment (4 716 kg/ha) was much lower than that under the FS treatment and SM treatment. There are serious reproductive barriers, mainly incrossability and hybrid sterility, in the interspecific hybridization of O. sativa with non-AA genome wild rice. These are the 'bottleneck' for transferring elite genes from wild rice to O. sativa. Combining traditional crossing method with biotechnique is a reliable way to overcome the reproductive barriers and to improve the utilizing efficiency of non-AA genome wild rice.展开更多

The mutant exhibits normal plant morphology but has apparently few tillers. The most striking change in nop is that its panicle differentiation is blocked, with masses of fluffy bract nodes generate from the positions where rachis branches normally develop in wild-type plants. Genetic analysis suggests that nop is controlled by a single recessive gene, which is temporarily named Nop(t). Based on its mutant phenotype, Nop(t) represents a key gene controlling the initiation of inflorescence differentiation, By using simple sequence repeat markers and sequence tagged site markers, Nop(t) gene was fine mapped in a 102-kb interval on the long arm of chromosome 6. These results will facilitate the positional cloning and functional studies of the gene.展开更多
In order to study the correlation between the cracking of rice (Oryza sativa L.) kernels and the molecular structure of the amylopectin in them, we attempted optical sum frequency generation (SFG) spectroscopy in the ...In order to study the correlation between the cracking of rice (Oryza sativa L.) kernels and the molecular structure of the amylopectin in them, we attempted optical sum frequency generation (SFG) spectroscopy in the C-H stretching vibration region for normal and cracked japonica non-glutinous rice kernels. The samples were Koshihikari and Nipponbare. In Nipponbare, the width of the SFG spectrum peak at 2915 cm<sup>- 1</sup> of the cracked rice kernels was broader than that of the normal ones, while for Koshihikari there was no clear difference. The width of the 2915 cm<sup>- 1</sup> peak is suggested to originate from the variety of the higher-order structure of the saccharide chains in amylopectin. Although this is a tentative result, this method is shown to have a potential of serving for preventing the cracking of the rice kernels.展开更多

The recessive gene controlling thermo-sensitive genetic male sterility (TGMS) is expressed when the plants are grown in conditions with higher or lower critical temperatures. To transfer tgms gene(s) control ing TGMS to Thai rice cultivars by backcross breeding method, a male sterile line was used as a donor parent while Thai rice cultivars ChaiNat 1, PathumThani 1, and SuphanBuri 1 were used as recurrent parents. The BC2F2 lines were developed from backcrossing and selfing. Moreover, the simple sequence repeat (SSR) markers were developed for identifying tgms gene and the linked marker was used for assisting selection in backcrossing. The identification lines were confirmed by pol en observation. The results showed the success of introgression of the tgms gene into Thai rice cultivars.
