Editing Algorithms for Non-Linear Editing Systems Based on MPEG-2

MPEG-1/2-based non-linear editing systems appear to have a tendency to replace the M-JPEG systems, but in so doing it is difficult to realize the video, audio and the synchronization editing algorithms. Such editing algorithms are presented. Based on an analysis of the structure of the MPEG-1/2 stream, and using parameters of the video, audio and the synchronization information, the video, audio and synchronization editing algorithms are provided. The characters of the algorithms are efficient, the quality loss of frames is low because it only decodes and codes part of the data; the editing algorithm is fast through use of some index files; synchronization editing is realized using the synchronization information, such as PTS, ESCR and other parameters. 展开更多

Stochastic Models to Mitigate Sparse Sensor Attacks in Continuous-Time Non-Linear Cyber-Physical Systems

Cyber-Physical Systems are very vulnerable to sparse sensor attacks.But current protection mechanisms employ linear and deterministic models which cannot detect attacks precisely.Therefore,in this paper,we propose a new non-linear generalized model to describe Cyber-Physical Systems.This model includes unknown multivariable discrete and continuous-time functions and different multiplicative noises to represent the evolution of physical processes and randomeffects in the physical and computationalworlds.Besides,the digitalization stage in hardware devices is represented too.Attackers and most critical sparse sensor attacks are described through a stochastic process.The reconstruction and protectionmechanisms are based on aweighted stochasticmodel.Error probability in data samples is estimated through different indicators commonly employed in non-linear dynamics(such as the Fourier transform,first-return maps,or the probability density function).A decision algorithm calculates the final reconstructed value considering the previous error probability.An experimental validation based on simulation tools and real deployments is also carried out.Both,the new technology performance and scalability are studied.Results prove that the proposed solution protects Cyber-Physical Systems against up to 92%of attacks and perturbations,with a computational delay below 2.5 s.The proposed model shows a linear complexity,as recursive or iterative structures are not employed,just algebraic and probabilistic functions.In conclusion,the new model and reconstructionmechanism can protect successfully Cyber-Physical Systems against sparse sensor attacks,even in dense or pervasive deployments and scenarios. 展开更多

Although different genome editing technologies are available, the clustered regularly interspaced short palindromic repeats/Cas9 (CRISPR/ Cas9) system, which utilizes engineered endonucleases to generate a double-stranded DNA break (DSB) in the target DNA region and subsequently stimulates site-specific mutagenesis through DNA repair machineries, is emerging as a powerful genome editing tool for elucidating mecha- nisms of protection from plant viruses, plant disease resistance, and gene functions in basic and applied research. In this review, we provide an overview of recent advances in the CRISPR system associated genome editing in plants by focusing on application of this technology in model plants, crop plants, fruit plants, woody plants and grasses and discuss how genome editing associated with the CRISPR system can provide insights into genome modifications and functional genomics in plants. 展开更多

Secondly, by constructing appropriate Lyapunov function and using comparison theorem with an impulsive differential equation, we study that a positive periodic solution exists. CRISPR-Cas tools for mammalian genome editing typically rely on single Cas9 or Cas12a proteins.While type I CRISPR systems in Class I may offer greater specificity and versatility,they are not well-developed for genome editing.Here,we present an alternative type I-C CRISPR system from Desulfovibrio vulgaris(Dvu)for efficient and precise genome editing in mammalian cells and animals.We optimized the Dvu type I-C editing complex to generate precise deletions at multiple loci in various cell lines and pig primary fibroblast cells using a paired PAM-in crRNA strategy.These edited pig cells can serve as donors for generating transgenic cloned piglets.The Dvu type I-C editor also enabled precise large fragment replacements with homology-directed repair.Additionally,we adapted the Dvu-Cascade effector for cytosine and adenine base editing,developing Dvu-CBE and Dvu-ABE systems.These systems efficiently induced C-to-T and A-to-G substitutions in human genes without double-strand breaks.Off-target analysis confirmed the high specificity of the Dvu type I-C editor.Our findings demonstrate the Dvu type I-C editor′s potential for diverse mammalian genome editing applications,including deletions,fragment replacement,and base editing,with high efficiency and specificity for biomedicine and agriculture. 展开更多

香蕉枯萎病菌内源报告基因Foc4carS的鉴定及其应用

香蕉枯萎病是由尖孢镰刀菌古巴转化型(Fusarium oxysporum f. sp. cubense, Foc)引起的香蕉毁灭性土传病害,其中4号生理小种(Foc4)能感染几乎所有的香蕉品系,危害最严重。carS基因通过调控下游car结构基因参与调控镰刀菌类胡萝卜素的生... 香蕉枯萎病是由尖孢镰刀菌古巴转化型(Fusarium oxysporum f. sp. cubense, Foc)引起的香蕉毁灭性土传病害,其中4号生理小种(Foc4)能感染几乎所有的香蕉品系,危害最严重。carS基因通过调控下游car结构基因参与调控镰刀菌类胡萝卜素的生物合成,本研究克隆鉴定了Foc4carS基因(FOIG_05085),Foc4carS蛋白具有典型的RING-finger蛋白结构域。利用分割标记法(Split-marker PCR)获得Foc4carS基因的融合片段,同时构建含有Foc4carS基因sgRNA591序列的pUC-fFuCas9-HTBNLS-hph-Foc4carS基因编辑载体,通过PEG介导的原生质体转化获得该基因的敲除突变体、回补突变体以及基因编辑敲除体,并对敲除和回补突变体的生物学特性和致病力进行分析。结果显示:ΔFoc4carS突变体的菌落直径、产孢量和致病力等生物学表型与野生菌株Foc4无显著差异,而ΔFoc4carS突变体菌落颜色呈深橙色,Foc4carS基因的缺失影响了次生代谢产物类胡萝卜素的生物合成;基因编辑的ΔFoc4carS(HDR)突变体不论是再生筛选板还是继代后的PDA平板,其菌落均出现典型的深橙色,表明Foc4carS可作为内源报告基因,在香蕉枯萎菌Foc4中进行基因质粒型CRISPR/Cas9编辑可行。 展开更多

碱性/螺旋-环-螺旋(basic/helix-loop-helix,bHLH)转录因子在植物的生长发育、次生代谢、信号转导、逆境胁迫等方面起重要的调控作用。棉花GhbHLH71基因c DNA全长996 bp,编码331个氨基酸残基,蛋白序列中含有保守的bHLH结构域,属于bHLH... 碱性/螺旋-环-螺旋(basic/helix-loop-helix,bHLH)转录因子在植物的生长发育、次生代谢、信号转导、逆境胁迫等方面起重要的调控作用。棉花GhbHLH71基因c DNA全长996 bp,编码331个氨基酸残基,蛋白序列中含有保守的bHLH结构域,属于bHLH转录因子家族成员。实时荧光定量分析结果表明,GhbHLH71基因在棉花纤维快速伸长期3~12 DPA (days post anthesis,DPA)相对高表达,暗示其主要在棉花纤维发育过程中发挥作用。构建该基因的CRISPR/Cas9基因编辑载体进行了棉花遗传转化,T_0代再生株经过Cas9基因的PCR检测以及靶位点突变情况检测分析,获得6个T_0代基因编辑突变体。T_1代不同突变株系在棉花生长发育期的表型性状与对照相比无明显差异,但成熟纤维的长度与对照相比皆明显变短。T_2代突变株系可以稳定遗传T_1代株系纤维变短的表型。其中4#和8#株系连续2代的纤维长度与对照相比缩短比率皆达20%以上,表明GhbHLH71基因的突变主要影响了棉花纤维细胞的伸长。本研究为深入了解棉花中bHLH转录因子的生物学功能以及棉花纤维发育的分子机制提供了一定的参考。 展开更多

[目的]强化经常性卫生费用核算数据政策分析,为基本公共卫生服务项目后续开展提供依据。[方法]以天津市2016-2019年基层医疗卫生机构预防服务费用核算结果为基础,分析基层医疗卫生机构基本公共卫生服务项目开展情况和存在问题。[结果]2016-2019年,天津市基层医疗卫生机构预防服务费用呈逐年增长趋势,由2016年的14.25亿元增加至2019年的18.04亿元。预防服务费用主要流向预防接种项目、慢性病管理和老年人健康管理项目;预防服务费用筹资来源仍以政府筹资方案为主,但家庭卫生支出占比呈快速增长趋势,主要是由于二类疫苗接种费用的快速增长所致。[结论]天津市基层医疗卫生机构预防服务费用流向与其基本公共卫生工作重点一致,但基层医疗卫生机构传染病防控投入程度与其职能不匹配。基层医疗卫生机构二类疫苗接种服务家庭卫生支出负担过重。 展开更多

病原微生物是影响家禽健康的重要因素之一,严重阻碍了家禽养殖业的快速发展,因而开发快速高效的病原检测方法对于家禽疫病的有效预防和控制具有十分重要的意义。CRISPR-Cas12a是一门新兴的基因编辑工具,具有高度的特异性、敏感性以及操... 病原微生物是影响家禽健康的重要因素之一,严重阻碍了家禽养殖业的快速发展,因而开发快速高效的病原检测方法对于家禽疫病的有效预防和控制具有十分重要的意义。CRISPR-Cas12a是一门新兴的基因编辑工具,具有高度的特异性、敏感性以及操作简便等优点,通过与聚合酶链式反应(PCR)、等温核酸扩增(LAMP)、重组酶聚合酶扩增(RPA)、重组酶介导等温扩增(RAA)等方法相结合,可大大提高检测效率及准确性,通过使用荧光信号、侧向层析试纸条等技术,还可使检测结果可视化,在病原检测中发挥了重大作用。本文综述了CRISPR-Cas12a系统作用机制及其结合多种核酸扩增技术在家禽病原检测中的应用研究进展,以期为家禽疫病的诊断和防控提供参考,为未来开发更有效的检测平台奠定基础。 展开更多
