Molecular Characterization and Antibacterial Activity of a G-Type Lysozyme in Yellow Drum(Nibea albiflora)

Lysozyme(EC3.2.1.17)plays an important role in the immune response;as a nonspecific immune factor,it can resist causative agents.Lysozyme can be divided into c-type and g-type in fish.In a previous study,through genome-wide association analysis,the g-type lysozyme gene,which is named NaLyg in yellow drum(Nibea albiflora),was found to be a key candidate gene for disease resistance in response to Vibrio harveyi infection.The cDNA of NaLyg was 1025 bp,including four exons and three introns,and its open reading frame(ORF)had a full-length of 582 bp,encoding 193 amino acids.NaLyg was found to be conserved during evolution through bioinformatic analyses.The NaLyg protein possessed a sugar binding domain and three catalytic sites,including Glu71,Asp84 and Asp101.Quantitative qRT-PCR results confirmed that NaLyg gene mRNA was visibly increased after V.harveyi infection.The NaLyg protein purified by prokaryotic expression killed some gram-negative bacterial pathogens by inducing cell wall destruction,including V.harveyi,Aeromonas hydrophila and Edwardsiella tarda.Moreover,the NaLyg protein killed two gram-positive bacteria,Bacillus subtilis and Staphylococcus aureus.Taken together,the experimental results suggested that the NaLyg protein of N.albiflora played an important role in fighting bacterial infections. 展开更多

Starting from sissotrin (1), a natural isoflavonoid isolated from Trifolium baccarinii (Fabaceae), one new semisynthetic derivative, 6-nitrobiochanin A (1b) and two known derivatives, 8-nitrobiochanin A (1a) and 2",3",4",6"-tetraacetylsissotrin (1c) have been obtained after performing nitration and acetylation reactions. Their structures were assigned after interpretation of their spectrometric (HR-ESI-MS) and spectroscopic (NMR 1D and 2D) data and by comparison with those reported in the literature. The substrate as well as the semisynthetic derivatives were evaluated for their antibacterial activities against six strains. The results reveal that they are inactive or weakly active on the strains tested with the exception of 8-nitrobiochanin A (1a) which showed moderate activity (MIC = 62.5 μg∙mL<sup>−1</sup>) on Staphylococcus aureus ATCC 43300. 展开更多

Superwetting materials have drawn unprecedented attention in the treatment of oily wastewater due to their preferable anti-fouling property and selective oil/water separation.However,it is still a challenge to fabricate multifunctional and environmentally friendly materials,which can be stably applied to purify the actual complicated wastewater.Here,a Ag/Ag/α-Fe_(2)O_(3) heterostructure anchored copper mesh was intentionally synthesized using a facile two-step hydrothermal method.The resultant mesh with superhydrophilicity and underwater superoleophobicity was capable of separating various oil/water mixtures with superior separation efficiency and high permeationflux driven by gravity.Benefiting from the joint effects of the smaller band gap of Ag/α-Fe_(2)O_(3) heterojunction,inherent antibacterial capacity of Ag/α-Fe_(2)O_(3) and Ag nanoparticles,favorable conductive substrate,as well as the hierarchical structure with superwettability,such mesh presented remarkably enhanced degradation capability toward organic dyes under visible light irradiation and antibacterial activity against both Escherichia coli(E.coli)and Staphylococcus aureus(S.aureus)compared with the pure Ag/α-Fe_(2)O_(3) coated mesh.Impressively,the mesh exhibited bifunctional water purification performance,in which organic dyes were eliminated simultaneously from water during oil/water separation in onefiltration process.More importantly,this mesh behaved exceptional chemical resistance,mechanical stability and long-term reusability.Therefore,this material with multifunctional integration may hold promising potential for steady water purification in practice. 展开更多

Carboxymethyl starch/silver oxide nanocomposites(CMS-Ag_(2)O NCs)were successfully fabricated by modifying carboxymethyl starch(CMS)with Ag_(2)O obtained from an aqueous AgNO_(3)solution as silver source.Ag_(2)O nanoparticles(NPs)formed on the surface of CMS by ion exchange.Based on SEM images,the diameters of Ag_(2)O NPs were determined to be between 50 and 100 nm.From the XRD spectra of CMS-Ag_(2)O NCs,the new diffraction peaks appeared at 33.88°and 38.08°,which were attributed to the Ag_(2)O NPs.According to the XPS analysis,Ag 3d_(5/2)and Ag 3d_(3/2)peaks in CMS-Ag_(2)O NCs were fitted into two main peaks centered at 367.6 eV and 373.6 eV,which were attributed to Ag^(+).The antibacterial efficiencies of CMS-Ag_(2)O NCs against Escherichia coli,Staphylococcus aureus,Pseudoalteromonas tetraodonis,Micrococcus luteus,and Shewanella putrefaciens were determined to be 99.6%,99.7%,99.4%,99.5%,and 99.6%,respectively.The antibacterial efficiencies of CMS-Ag_(2)O NCs against the bacterial species were all greater than 99%.Therefore,these results indicated that CMS-Ag_(2)O NCs was highly effective as a bactericidal agent against multiple bacterial species.CMS-Ag_(2)O NCs can be further applied to antifouling coating. 展开更多

Analysis on Antibacterial, Detumescent and Antioxidant Activity of Huoshan Large-leaf Yellow Tea

[Objectives] To provide experimental basis for the effective development and utilization of Huoshan large-leaf yellow tea resources and the screening of safe and effective active ingredients of large-leaf yellow tea. [Methods] The active substances of Huoshan large-leaf yellow tea were extracted by hot-water extraction, and the freeze-dried powder of Huoshan large-leaf yellow tea was obtained by freeze drying. The antibacterial activity of the extract was preliminarily confirmed using the Oxford cup method, and its antimicrobial spectrum was analyzed using 14 strains. A xylene-induced mouse auricle swelling test was carried out to detect the swelling inhibition rate of the extract and analyze its in-vitro detumescent activity. Then, the antioxidant activity of the extract was identified through a DPPH free radical scavenging capacity test and a ferric reducing antioxidant power assay. [Results] The extract had significant inhibitory effects on various bacteria. The extract could effectively inhibit the growth of Escherichia coli, Staphylococcus aureus, Enterococcus hirae, Acinetobacter baumannii, Klebsiella pneumonia, Pseudomonas aeruginosa, Bacillus subtilis , and other strains. The diameter of the inhibition zone increased with the increase of sample concentration. The extract had a significant inhibitory effect on auricle swelling induced by xylene in mice. When the concentration of the drug reached 1.0 mg/mL, its inhibition rate on mouse auricle swelling reached 55.2% ( P <0.01), slightly lower than the swelling inhibition rate of the aspirin group (66.52%, P <0.01). The results of the antioxidant test showed that large-leaf yellow tea extract also had strong activity. Within the concentration range of 0.1-1.0 mg/mL, its DPPH radical scavenging rate increased with the increase of sample concentration. Within the concentration range of 0.1-1.0 mg/mL, its DPPH radical scavenging rate increased with the increase of sample concentration. When the concentration reached 1.0 mg/mL, the scavenging rate reached 69.75%. The Fe 3+ -reduction capacity of the extract also increased with the increase of sample concentration within the concentration range of 0.1-2.5 mg/mL. When the concentration was 2.5 mg/mL, the reducing power of the extract reached 1.43±0.04. However, its DPPH free radical scavenging rate and reducing power were slightly lower than the capacity of V C at the same concentration. [Conclusions] The extract of Huoshan large-leaf yellow tea obtained by hot-water extraction had strong activity in many aspects, including inhibiting the growth of various microbes, subsiding swelling in vitro and resisting oxidation. These experimental results provide certain guiding significance for the basic research of Huoshan large-leaf yellow tea extract, as well as experimental data support for the subsequent development of functional foods and drugs of Huoshan large-leaf yellow tea. 展开更多

Two new dihydropyrones,rhytismatones C(1)and D(2),and a known compound,penicillenol A1(3),were isolated from the co-culture broth of the deep-sea-derived fungus Penicillium crustosum PRB-2 and Suaeda salsa-derived endophytic fungus Peni-cillium citrinum HDN11-186.Their structures were elucidated through comprehensive analysis of nuclear magnetic resonance(NMR)spectra and mass spectra.The absolute configurations of new compounds were determined by calculating the electronic circular di-chroism(ECD)spectrum.UPLC-MS data showed that compounds 1–3 could only be detected in the media of co-culture,suggesting new biosynthetic pathways were activated in the co-cultured fungi.Compound 1 showed obvious antibacterial activities against Pro-teus sp.MMBC-1002 and Bacillus subtilis MMBC-1004 with minimum inhibitory concentration(MIC)both at 25μmolL^(-1). 展开更多

Developing and excavating new agrochemicals with highly active and safe is an important tactic for protecting crop health and food safety.In this paper,to discover the new bactericide candidates,we designed,prepared a new type of1,2,3,4-tetrahydro-β-carboline(THC)derivatives and evaluated the in vitro and in vivo bioactivities against the Xanthomonas oryzae pv.oryzae(Xoo),Xanthomonas axonopodis pv.citri(Xac),and Pseudomonas syringae pv.actinidiae(Psa).The in vitro bioassay results exhibited that most title molecules possessed good activity toward the three plant pathogenic bacteria,the compound A17 showed the most active against Xoo and Xac with EC50 values of 7.27 and 4.89 mg mL^(-1)respectively,and compound A8 exhibited the best inhibitory activity against Psa with EC50value of 4.87 mg mL^(-1).Pot experiments showed that compound A17 exhibited excellent in vivo antibacterial activities to manage rice bacterial leaf blight and citrus bacterial canker,with protective efficiencies of 52.67 and 79.79%at 200 mgmL^(-1),respectively.Meanwhile,compound A8 showed good control efficiency(84.31%)against kiwifruit bacterial canker at 200 mg mL^(-1).Antibacterial mechanism suggested that these compounds could interfere with the balance of the redox system,damage the cell membrane,and induce the apoptosis of Xoo cells.Taken together,our study revealed that tetrahydro-β-carboline derivatives could be a promising candidate model for novel broadspectrum bactericides. 展开更多

Natural products have long been a crucial source of,or provided inspiration for new agrochemical discovery.Naturally occurring 18β-glycyrrhetinic acid shows broad-spectrum bioactivities and is a potential skeleton for novel drug discovery.To extend the utility of 18β-glycyrrhetinic acid for agricultural uses,a series of novel 18β-glycyrrhetinic acid amide derivatives were prepared and evaluated for their antibacterial potency.Notably,compound 5k showed good antibacterial activity in vitro against Xanthomonas oryzae pv.oryzae(Xoo,EC50=3.64 mg L–1),and excellent protective activity(54.68%)against Xoo in vivo.Compound 5k induced excessive production and accumulation of reactive oxygen species in the tested pathogens,resulting in damaging the bacterial cell envelope.More interestingly,compound 5k could increase the activities of plant defense enzymes including catalase,superoxide dismutase,peroxidase,and phenylalanine ammonia lyase.Taken together,these enjoyable results suggested that designed compounds derived from 18β-glycyrrhetinic acid showed potential for controlling intractable plant bacterial diseases by disturbing the balance of the phytopathogen’s redox system and activating the plant defense system. 展开更多

[Objectives]To investigate the antioxidant,anti-inflammatory and antibacterial activities of different extracts(aqueous,ethanol,ethyl acetate and n-butanol extracts)of Miao medicine Polygonum capitatum.[Methods]Eleven batches of P.capitatum in Guizhou province were collected,and water,ethanol,ethyl acetate and n-butanol extracts were prepared by reflux extraction.Antioxidant activity was determined by radical scavenging capacity of 1,1 diphenyl-2-picyl hydrazine(DPPH),anti-inflammatory activity was screened by lipopolysaccharide(LPS)induced RAW264.7 cells to produce NO,and the minimum inhibitory concentration(MIC)was screened by broth microdilution method.[Results]When the concentration of ethyl acetate extract was 10 mg/L,the scavenging rate of DPPH ranged from 90%to 99%.The(MIC of the ethyl acetate extract against Staphylococcus aureus(SA),Pseudomonas aeruginosa(PA)and Escherichia coli(EC)was 0.18-0.65,0.13-0.82,and 0.15-0.78 g/L,respectively.In the anti-inflammatory activity,ethyl acetate extract inhibited NO production with inhibition rate of 70%.[Conclusions]The ethyl acetate extract and ethanol extract of Miao medicine P.capitatum have strong antioxidant,anti-inflammatory and antibacterial activities. 展开更多

Indeed,medicinal importance of honey has been documented in the world's oldest medical literatures,and since the ancient times,it has been known to possess antimicrobial property as well as wound-healing activity.The healing property of honey is due to the fact that it offers antibacterial activity,maintains a moist wound condition,and its high viscosity helps to provide a protective barrier to prevent infection.Its immunomodulatory property is relevant to wound repair too.The antimicrobial activity in most honeys is due to the enzymatic production of hydrogen peroxide.However,another kind of honey,called non-peroxide honey(viz.,manuka honey),displays significant antibacterial effects even when the hydrogen peroxide activity is blocked.Its mechanism may be related to the low pH level of honey and its high sugar content(high osmolality) that is enough to hinder the growth of microbes.The medical grade honeys have potent in vitro bactericidal activity against antibiotic-resistant bacteria causing several life-threatening infections to humans. But,there is a large variation in the antimicrobial activity of some natural honeys,which is due to spatial and temporal variation in sources of nectar.Thus,identification and characterization of the active principle(s) may provide valuable information on the quality and possible therapeutic potential of honeys(against several health disorders of humans),and hence we discussed the medicinal property of honeys with emphasis on their antibacterial activities. 展开更多

Antibacterial activity of zinc oxide nanoparticles(Zn O-NPs) has received significant interest worldwide particularly by the implementation of nanotechnology to synthesize particles in the nanometer region. Many microorganisms exist in the range from hundreds of nanometers to tens of micrometers. Zn O-NPs exhibit attractive antibacterial properties due to increased specific surface area as the reduced particle size leading to enhanced particle surface reactivity. Zn O is a bio-safe material that possesses photo-oxidizing and photocatalysis impacts on chemical and biological species. This review covered Zn O-NPs antibacterial activity including testing methods, impact of UV illumination, Zn O particle properties(size, concentration, morphology, and defects), particle surface modification, and minimum inhibitory concentration. Particular emphasize was given to bactericidal and bacteriostatic mechanisms with focus on generation of reactive oxygen species(ROS) including hydrogen peroxide(H2O2), OH-(hydroxyl radicals), and O2-2(peroxide). ROS has been a major factor for several mechanisms including cell wall damage due to Zn O-localized interaction, enhanced membrane permeability, internalization of NPs due to loss of proton motive force and uptake of toxic dissolved zinc ions.These have led to mitochondria weakness, intracellular outflow, and release in gene expression of oxidative stress which caused eventual cell growth inhibition and cell death. In some cases, enhanced antibacterial activity can be attributed to surface defects on Zn O abrasive surface texture. One functional application of the Zn O antibacterial bioactivity was discussed in food packaging industry where Zn O-NPs are used as an antibacterial agent toward foodborne diseases. Proper incorporation of Zn O-NPs into packaging materials can cause interaction with foodborne pathogens, thereby releasing NPs onto food surface where they come in contact with bad bacteria and cause the bacterial death and/or inhibition. 展开更多

Methods: The essential oil of leaf of F. koreana was extracted by using hydrodistillation process and the volatile components investigated with the help of gas chromatography coupled with mass spectrometry. The antibacterial study was carried out with the help of agar disc dif usion method, MIC, MBC and viable count. The mode of action was determined with help of potassium ion l ux, cellular material release and scanning electron microscopy. The antioxidant activity was determined with the help of 2, 3-diphenyl-2-picrylhydrazyl method, nitric oxide scavenging activity and superoxide anion radical scavenging assay. Results: Total ten compounds were identii ed as trans-phytol(42.73%), cis-3-hexenol(12.95%), 毬-linalool(10.68%), trans-2-hexenal(8.86%), trans-2-hexenol(8.86%), myrcenol(3.86%), 4-vinylphenyl acetate(3.86%),(4Z)-4,6-heptadien-1-ol(3.18%), lemonol(2.73%) and benzeneacetaldehyde(2.27%) by gas chromatography coupled with mass spectrometry. The antibacterial study was demonstrated that leaf essential oil of F. koreana act against foodborne and other pathogenic bacteria. The mode of action revealed that this essential oil acted on the cytoplasmic membrane, resulting in loss of integrity and increased permeability. In addition, leaf essential oil of F. koreana was shown to be rich in linalool, which contributes to improved antioxidant activity. Objective:To find the effectiveness of essential oil of lemongrass for the treatment of pathogenic organisms.Methods:Lemongrass oil was investigated for activity against Staphylococcus aureus (S.aureus),Bacillus cereus(B.cereus),Bacillus subtilis(B.subtilis),Escherichia coli(E.coli), Klebsiella pneumoniae(K.pneumoniae) and Pseudomonas aeruginosa(P.aeruginosa),using Agar Diffusion Method and Broth Dilution Method.Minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) were determined by the Broth Dilution Method. The antibiotic susceptibility test against the test organisms was performed by Disc Diffusion Method.Results:Lemongrass was found effective against all the test organisms except P. aeruginosa.Gram positive organisms were found more sensitive to lemon grass oil as compared to gram negative organisms.The test organisms were found inhibited by Lemon grass oil at lower concentrations in Broth Dilution Method as compared to Agar Diffusion Method.Conclusions: The tested organisms,particularly gram-negative organisms had shown high resistance towards different antibiotics whereas they were found to be inhibited by lemongrass oil even at lower concentration.Thus lemongrass oil is effective against drug resistant organisms.It can be suggested that use of lemongrass oil would be helpful in the treatment of infections caused by multidrug resistant organisms. 展开更多

Objective:To evaluate the antibacterial activity of the methanolic extract of mango(Mangifera indica L) seed kernel.Methods:Chokanan mango seed kernel and seed kernels from assorted mango varieties were collected,cleaned,dried and powered.Crude methanolic extracts of mango seed kernel were analyzed for the phytochemical constituents.The free radical scavenging activity was determined by 2,2-Diphenyl-1-picrylhydrazyl(DPPH) assay.Antibacterial activity was evaluated by disc diffusion assay with three medically important bacterial pathogens such as methicillin resistant Staphylococcus aureus(S.aureus)(MRSA),Escherichia coli(E.coli) and Vibrio vulnificus(V.vulnificus).Results:Qualitative phytochemical analysis indicated the presence of important phytochemical compounds such as glycosides,saponins,flavanoids, tannins and alkaloids.There was no significant difference in the phytochemical content between the single and assorted mango seed kernels.However,the free radical scavenging study indicated that the assorted mango kernels showed slighdy higher activity than the single species(P【0.05). The crude methanolic extract of mango seed kernel at a concentration of 100 mg/mL is found to have potential antimicrobial activity against MRSA and E.coli compared to V.vulnificus. Study on the antibacterial activity also indicated that there was no significant difference in the antibacterial activity of the single and assorted mango seed kernel extracts.Conclusions:The present study conclusively demonstrates the free radical scavenging activity and antibacterial activities of mango seed kernel.In addition,the results also indicated that there is no significant difference in the phytochemical content and biological activity of mango kernels from single and assorted mango varieties. 展开更多

Objective:To evaluate the antimicrobial efficacy of berberine,a plant alkaloid.Methods:Five multi-drug resistant(MDR) STEC/EPEC and five MDR ETEC isolates from yaks with haemorrhagic diarrhoea were selected for the study.Antibacterial activity of berberine was evaluated by broth dilution and disc diffusion methods.The binding kinetics of berberine to DNA and protein was also enumerated.Results:For both categories of enterovirulent Escherichia coli(E.roli) isolates, berberine displayed the antibaclerial effect in a dose dependent manner.The MIC<sub>50</sub> of berberine chloride for STEC/EPEC isolates varied from 2.07μM to 3.6μM with a mean of(2.95±0.33)μM where as for ETEC strains it varied from 1.75 to 1.96μM with a mean of(1.87±0.03)μM. Berberine bind more tightly with double helix DNA with Bmax and Kd of(24.68±2.62) and(357.8±57.8),respectively.Berberine reacted with protein in comparatively loose manner with Bmax and Kd of(18.9±3.83) and 【286.2±113.6),respectively.Conclusions:The results indicate clearly that berberine may serve as a good antibacterial against multi drug resistant E.coli. 展开更多

Objective:To search for antimicrobial agents among natural products.Methods:Ethanolic extracts of 4 plant species,including Beta vulgaris L.(Chenopodiaceae),Amaranthus graecizans (A.graecizans) L.(Amaranthaceae),Rumex obtusifolius(R.obtusifolius) L.and Polygonum patulum(P.patulum) M.B.(Polygonaceae),were evaluated for antibacterial activity using agar disc diffusion method against some gram-positive and gram-negative bacteria[Pseudomonas aeruginosa(P.aeruginosa),Listeria monocytogenes(L.monocytogenes).Staphylococcus epidermidis(S.epidermidis),Staphylococcus aureus(S.aureus),Klebsiella pneumoniae(K. pneumoniae),Salmonella typhi(S.typhi),Bacillus cereus(B.cereus).Bacillus anthracis(B. anthracis),Escherichia coli(E.coli) and Streptococcus pyogenes(Str.pyogenes)].These extracts were obtained from aerial parts of the used plants.Results:The majority of these extracts had inhibitory effect at different concentrations(0.05 g/mL,0.10 g/mL,0.20 g/mL and 0.40 g/mL) against above mentioned bacteria.E.coli was the most resistant strain.The highest inhibitory zone was showed by ethanolic extract of P.patulum against Str.pyogenes(28 mm) and followed by ethanolic extract of B.vulgaris against S.epidermidis(23 mm).The extract of A.graecizans didn’t show inhibitory activity except at 0.40 g/mL against B.cereus.The minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) values of R.obtusifolius extract that was measured against Str.pyogenes were equal(MIC=MBC=5.00mg/mL).Conclusion: The findings of this study could also be as new source for antibiotics discovery and infection treatment. 展开更多

Objective:To determine the antimicrobial activity of rosemary(Rosmarinus of ficinalis L.) and to investigate the synergistic effects of this extract combined with ceforuxime against methicillinresistant Staphylococcus aureus(MRSA).Methods:The inhibitory and bactericidal activities of rosemary ethanol extract,alone and in combination with cefuroxime,were studied.Results:The minimum inhibitory concentrations(MICs) of the ethanol extract of rosemary were in the range of 0.39-3.13 mg/mL.The minimum bactericidal concentrations(MBCs) were usually equal to or double that MICs.The antimicrobial activity of combinations of the ethanol extract of rosemary and cefuroxime indicated their synergistic effects against all MRSAs.Conclusions:The present work clearly demonstrates that rosemary has a key role in the elevation of susceptibility toβ-lactams. 展开更多

Objective:To assess the antibacterial activity of eight Iranian plant extracts against Staphylococcus aureus(S.aureus) strains which were isolated from infected patients.Methods: The studied strains were isolated from urine,stool,blood and wound of infected patients and identified by biochemical tests.In further,the antibacterial activity of 8 ethanolic local plant extracts including Querqus brantii(Q.brantii),Ziziphus spina-christi(Z.spina-christi),Peganum harmala(P.harmala),Oliveira decumbens(O.decumbens),Galium tricornutum(G.tricornutum), Vitex pseudo negundo(Vi.pseudo negundo),Salvia officinalis(S.officinalis),Vaccaria pyramidata (V.pyramidata) were then evaluated using agar disc diffusion method.Results:A total of 9 isolates were isolated and identified as S.aureus.Antibacterial profile of the strains showed that the strains were resistant to methicillin and cefixime.The highest antibacterial activity against the studied strains were belong to Q.brantii,P.harmala,Z.spina-christi and O.decumbens vent extracts with 11-40 mm,15-28 mm,8-26 mm and 10-20 mm of diameters,respectively. Intermediate antibacterial activity was exhibited by G.tricornutum and Vi.pseudo negundo against some of the studied strains with 7-20 mm and 7-13 mm of diameters,respectively. Noteworthy,out of 9 studied strains;6 and 2 of them were resistant to the G.tricornutum and Vi. pseudo negundo extracts,respectively.The S.officinalis and Va.pyramidata,however,showed no antibacterial activity against the studied strains.Conclusions:The Q.brantii,P.harmala,Z. spina-christi and O.decumbens extracts were shown to possess strong antibacterial activity against methicillin and cefixime resistant S.aureus strains and can be considered as the promising natural antibiotics for treating the studied strains. 展开更多

According to our results the maximum antibacterial activity was observed in ethanolic extract when compared to other extract. So, the ethanolic extract was studied for antioxidant activity which exhibited high antiradical activity against DPPH, hydroxyl, and nitric oxide radicals. In conclusion, green hull of J. regia showed strong reducing power activity and total antioxidant capacity. Objective:To isolate novel actinomycetes and to evaluate their antibacterial activity.Methods:Three soil samples were collected from Vengodu(village)in Kanchipuram district,Tamil Nadu,India.Actinomycetes were isolated using serial dilution and plating method on actinomycetes isolation agar.Results:Totally 35 isolates were obtained on the basis of colony characteristics on actinomycetes isolation agar.All the isolates were screened for antibacterial activity by cross streak method.Medium and optimization of day were done for the potent strains using Nathan's agar well diffusion method.Isolation of bioactive compounds from significant active isolates was done by using different media.The most active isolate VAS 10 was identified as Actinobacterium Loyola PBT VAS 10(accession No.JF501398)using 16s rRNA sequence method.The hexane,ethyl acetate,dichloromethane and butanol extracts of VAS 10 were tested against bacteria.The maximum antibacterial activity was observed in dichloromethane and ethyl acetate;maximum zones of inhibition were observed against Enterococcus durans.The rRNA secondary structure and the restriction sites of Actinobacterium Loyola VAS 10 were predicted using Genebee and NEBCutter online tools respectively.Conclusions:The present study showed that among the isolated actinomycetes,Actinobacterium Loyola PBT VAS 10(accession No.JF501398)showed good antibacterial activity against the tested bacteria. 展开更多
