To address such issues, a new colony based label-free detection method working on the principles of elastic light scattering was introduced. In order to build libraries of scattering images for bacterial pathogens, it is pertinent to determine the effect of preparation and storage of the agar media on the scatter patterns. Scatter patterns of three Escherichia coli serovars (O26, O111 and O157) were studied and used in a model system, after growth on Sorbitol-MacConkey agar plates that were prepared and stored at different conditions in the laboratory. Quantitative image processing software was used to analyze variation in scatter patterns of the same serovar on media prepared under various standard laboratory conditions and to generate a cross-validation matrix for comparison. Based on the results, it was determined that attention should be given during preparation of media so that the agar plates are not air-dried more than 10 - 20 min after solidification at room temperature. The plates could be stored in sealed bags in cold room (4oC - 10oC) for up to a month before use. The findings of this study should provide guidelines in preparation, storage, and handling of media for generation of reproducible scatter patterns of bacterial colonies with the light scattering sensor for pathogen detection.展开更多

The UV-Visible spectroscopy result reveals that the band gap energy of ZnO/Nb2O5 nanoparticles to be 3.8 eV. The XRD results show that the crystallite size is to be 31.9 nm. The ICPMS data indicate the presence of 3,3461,328 counts of 93 Nb and 577,906,390 counts of 66 Zn. An improvement in the photocatalytic degradation of Indigocarmine dye (IC) in comparison to commercially available pure ZnO was observed. The photodegradation efficiency for ZnO/Nb2O5 and ZnO were found to be 97.4% and 52.1% respectively. The enhancement in photocatalytic activity of ZnO/ Nb2O5 was ascribed to the extended light absorption range and suppression of electron hole pair recombination upon Nb loading. The antibacterial activity of ZnO/Nb2O5 nanoparticles was investigated. These particles were shown to have an effective bactericide.展开更多

Objective Ubiquitin conjugate enzyme E2O(UBE2O)is a ubiquitin-conjugating enzyme that has been reported to be involved in tumorigenesis.This study investigated the role of UBE2O in hepatocellular carcinoma(HCC).Method...Objective Ubiquitin conjugate enzyme E2O(UBE2O)is a ubiquitin-conjugating enzyme that has been reported to be involved in tumorigenesis.This study investigated the role of UBE2O in hepatocellular carcinoma(HCC).Methods The expression of UBE2O was detected using qRT-PCR,Western blotting,and immunohistochemical staining.Cell proliferation and Transwell assays were used to detect proliferation,migration,and invasion of HCC cells,respectively.Bioinformatic analysis was performed to analyze the relationship between UBE2O and the clinical features,prognosis,and immune cell infiltration of HCC.Results UBE2O was significantly over-expressed in HCC tissues.High expression of UBE2O was associated with poor tumor grade and poor prognosis.Functional experiments showed that down-regulation of UBE2O inhibited HCC cell proliferation,migration,and invasion.Co-expression gene analysis and gene set enrichment analysis showed that UBE2O was associated with protein hydrolysis,cell cycle,and cancer-related pathways in HCC.The results of immune analysis revealed that the expression of UBE2O was positively correlated with the immune infiltration and expression of immune-related chemokines of HCC.Conclusions UBE2O is significantly correlated with the prognosis of HCC and may be a valuable prognostic biomarker for HCC.展开更多

The construction of E pH diagrams has turned to multi component systems. However, there are some limits in plotting such diagrams. There is only one diagram for one multi component system, which can not reflect the truth of the aqueous reaction. In the paper, a new computation method is proposed to construct E pH diagrams. Component activity term is used to determine the boundary of stable areas. For the multi component systems, different atom ratios of elements have been taken into account. M S H 2O system is chosen to study since it is of importance in metallurgical solution. Compared with conventional methods, the algorithm is simple and conforms to real conditions.展开更多

On an average 〉 5.0 log CFU/g of natural microbial populations and c...This study was conducted to isolate and identify antibiotic resistant bacteria in fresh tomato sold in three different local markets of Dhaka city. On an average 〉 5.0 log CFU/g of natural microbial populations and coliform bacteria was present in the samples. Prevalence of Salmonella spp., E. coli, Listeria spp., and Yersinia spp., were recorded through cultivation dependent assay of tomato samples of different markets. Almost all of the isolates were uniformly resistant to rifampicin, erythromycin, clindamycin and oxacillin (100%), vancomycin (93%), amoxicillin (87%), whereas 60% of the isolates were found to be resistant to ampicillin and cephalexin. Antibiotic sensitivity test of the presumptive E. coli and Salmonella spp. and corresponding plasmid profiling indicated a correlation of plasmid mediated multidrug resistance (MDR) of these environmental bacteria. Washing with sanitizing agents such as acidified sodium chlorite (ASC) and 200 ppm chlorinated water was able to reduce _〈 2.0 log CFU/g of natural microflora and coliform bacteria. In contrast, ASC was able to reduce 〈 2.0 log CFU/g of other food borne pathogens and, chlorinated water was able to reduce up to undetectable level. Therefore, washing of tomato with 200 ppm chlorinated water could be useful in reducing the pathogen populations on tomatoes.展开更多

A comparison of sensitivity in extratropical circulation in the Northern Hemisphere (NH) and Southern Hemisphere (SH) is conducted through observational analyses and diagnostic linear model experiments for two typ...A comparison of sensitivity in extratropical circulation in the Northern Hemisphere (NH) and Southern Hemisphere (SH) is conducted through observational analyses and diagnostic linear model experiments for two types of El Ni(n)o events,the traditional El Ni(n)o with the strongest warmth in the eastern tropical Pacific (EP El Ni(n)o) and the El Ni(n)o Modoki with the strongest warmth in the central tropical Pacific (CP El Ni(n)o).It is shown that CP El Ni(n)o favors the occurrence of a negative-phase Northern Annular Mode (NAM),while EP El Ni(n)o favors that of the Pacific-North American (PNA) pattern.In SH,both EP and CP El Ni(n)o induce a negative phase Southern Annular Mode (SAM).However,the former has a greater amplitude,which is consistent with the stronger sea surface temperature (SST) warmth.The difference in the two types of El Ni(n)o events in NH may originate from the dependence of heating-induced extratropical response on the location of initial heating,which may be associated with activity of the stationary wave.In SH,the lack of sensitivity to the location of heating can be associated with weaker activity of the stationary wave therein.展开更多

This study examined the survival of acid-adapted E. coli O157 in kombucha during fermentation and refrigerated kombucha mixed with fruit juices. Acidic and non-acidic kombucha mixes were fermented at 25°C usin...This study examined the survival of acid-adapted E. coli O157 in kombucha during fermentation and refrigerated kombucha mixed with fruit juices. Acidic and non-acidic kombucha mixes were fermented at 25°C using a commercially available starter culture and inoculated with a 5-strain mixture of acid-adapted E. coli O157. There was >5-log reduction in the pathogen count for both starter mixes within 7 days of fermentation. For the kombucha-juice blends at refrigerated temperature, 14 ml of lemon, apple, orange, and mango juices were mixed with 186 ml kombucha separately. The treatments were inoculated with a 5-strain mixture of acid-adapted E. coli O157 and incubated at 5°C for 14 days. >5-log reduction in the pathogen count was observed in lemon, control, and mango juice blend after 1, 3, and 14 days, respectively. The total reduction in pathogen count in the apple and orange juice blend after 14 days was 4.43 and 4.12 log CFU/ml, respectively. The inability of the kombucha fruit blend to cause a 5-log reduction of E. coli O157 suggests the need for following strict hygienic and good sanitation practices during blending and bottling for home fermenters and an approved HACCP plan for foodservice operators to ensure product safety.展开更多

The theoretical noise was defined as a scattering signal from a rough produce surface while the signal was defined as a scattering signal from the increasing numbers of rod-shaped bacteria on the produce surface. In this research, we measured the surface topography of a tomato using BioAFM to provide the quantitative nature of the surface roughness which was, in turn, modeled with the discrete dipole approximation (DDA) for an accurate estimation of the background scattering signature. Then we included the DDA model of rod-shaped bacteria and calculated the combined elastic scattering signature in the upper hemispherical space with different polarizations, wavelengths, and incident angles. The total scattering cross-section (TSC) and partial scattering cross-section (PSC) were both computed on six predefined aperture locations. The results indicate that, upon proper selection of the wavelength and incident angle, it was possible to provide the minimum number of bacteria (~32) to provide a differentiable elastic scattering signal from the produce surface.展开更多

Chlorine is commonly used to sanitize processing equipment where Escherichia coli O157:H7 (Ec) may survive and contaminate food products. The objective of this study was to characterize the survival behavior of Ec with different stresses on RTE meats. A multi-strain cocktail of Ec was pre-treated with freezing shock for 15 - 20 h and/or chlorine (0, 25, and 50 ppm) for one hour, and then inoculated onto RTE meat surfaces to obtain about 3.0 log CFU/g. Samples were stored at three abuse temperatures (12℃, 18℃, and 24℃) and Ec was enumerated during the storage. The freezing shock impact was studied using the Ec cocktail stored in a freezer overnight followed by chlorine exposure for one hour. The lag phase and growth rate of Ec were estimated using DMFit (Combase, Baranyi’s model). Results indicated that Ec growth was suppressed by chlorine treatment. Freezing shock was found to have little impact in terms of lag time and growth rate. The lag phase of Ec after exposure to 0 ppm of chlorine (50.3 h) was shorter than that of Ec treated with 25 ppm (54.6 h) and 50 ppm (164.1 h) at 12℃. However, the lag phase decreased with an increase in temperature, e.g. at 25 ppm, lag times were 54.6, 51.1 and 48.9 h for 12℃, 18℃ and 24℃, respectively. Lag times increased with an increase in chlorine concentration. At 24℃, lag times were 15.8, 48.9, and 52.4 h for 0, 25, and 50 ppm, respectively. The growth rate increased with an increase in temperature for 0 and 25 ppm chlorine levels, but decreased at 50 ppm level. Growth rate and lag phase as a function of temperature and chlorine concentration can be described by polynomial models and modified Ratkowsky-type and Zwietering-type models. Results of this study will contribute to risk assessment of RTE meats.展开更多
Organically modified silica (ORMOSILS) thin films produced by sol-gel method were imprinted with two bacterial strains as whole cells in order to develop an easy, fast and specific probe to detect and specifically ide...Organically modified silica (ORMOSILS) thin films produced by sol-gel method were imprinted with two bacterial strains as whole cells in order to develop an easy, fast and specific probe to detect and specifically identify these micro-organisms when present in water samples. An important feature of the imprinting process was the molecular finger-prints left by these microorganisms alongside morphology, into imprinted film cavities. The films also showed high selectivity toward the imprinted template and were able to discriminate between two very close bacterial species (E. coli and S. typhimurium). In addition, several central physical parameters of the experimental water solution were examined (i.e., pH, ionic strength and the organic load exemplified by NaCl and TOC concentration, respectively). The method sensitivity to different bacterial concentrations was studied by confocal microscopy (CLSM) and quartz crystal microbalance (QCM) tools. Results showed that increased bacterial concentrations favor rapid adsorption onto imprinted sol-gel films with high affinity, while low pH, increased organic load and high ionic concentrations (i.e., seawater) interfere with bacteria re-adsorption, reducing detection capability. Under average drinking water chemical composition the method proved to be highly efficient.展开更多

文摘[Objectives]This study aimed to optimize the extraction process of total flavonoids in Cardamine tangutorum O.E.Schulz.[Methods]On the basis of single-factor experiments,the ultrasonic-enzymatic extraction process of total flavonoids in C.tangutorum O.E.Schulz was optimized by Box-Behnken design from aspects of ethanol volume fraction,extraction time,extraction temperature and enzyme addition,with yield of total flavonoids as the evaluation index.[Results]The optimal extraction process of total flavonoids in C.tangutorum O.E.Schulz was as follows:ethanol volume fraction of 70%,ultrasonic extraction time of 43 min,ultrasonic extraction temperature of 37℃and cellulose addition of 0.07 mg.Under the optimal conditions,the yield of total flavonoids from C.tangutorum O.E.Schulz reached 75.90 mg/g.[Conclusions]The optimized extraction process was reasonable and feasible,and it could provide a reference for the extraction of total flavonoids in C.tangutorum O.E.Schulz.This study provided a foundation for the quality control of food/medicinal materials and the development and research of big health products of C.tangutorum O.E.Schulz.

文摘A comparison of sensitivity in extratropical circulation in the Northern Hemisphere (NH) and Southern Hemisphere (SH) is conducted through observational analyses and diagnostic linear model experiments for two types of El Ni(n)o events,the traditional El Ni(n)o with the strongest warmth in the eastern tropical Pacific (EP El Ni(n)o) and the El Ni(n)o Modoki with the strongest warmth in the central tropical Pacific (CP El Ni(n)o).It is shown that CP El Ni(n)o favors the occurrence of a negative-phase Northern Annular Mode (NAM),while EP El Ni(n)o favors that of the Pacific-North American (PNA) pattern.In SH,both EP and CP El Ni(n)o induce a negative phase Southern Annular Mode (SAM).However,the former has a greater amplitude,which is consistent with the stronger sea surface temperature (SST) warmth.The difference in the two types of El Ni(n)o events in NH may originate from the dependence of heating-induced extratropical response on the location of initial heating,which may be associated with activity of the stationary wave.In SH,the lack of sensitivity to the location of heating can be associated with weaker activity of the stationary wave therein.

文摘This study examined the survival of acid-adapted E. coli O157 in kombucha during fermentation and refrigerated kombucha mixed with fruit juices. Acidic and non-acidic kombucha mixes were fermented at 25°C using a commercially available starter culture and inoculated with a 5-strain mixture of acid-adapted E. coli O157. There was >5-log reduction in the pathogen count for both starter mixes within 7 days of fermentation. For the kombucha-juice blends at refrigerated temperature, 14 ml of lemon, apple, orange, and mango juices were mixed with 186 ml kombucha separately. The treatments were inoculated with a 5-strain mixture of acid-adapted E. coli O157 and incubated at 5°C for 14 days. >5-log reduction in the pathogen count was observed in lemon, control, and mango juice blend after 1, 3, and 14 days, respectively. The total reduction in pathogen count in the apple and orange juice blend after 14 days was 4.43 and 4.12 log CFU/ml, respectively. The inability of the kombucha fruit blend to cause a 5-log reduction of E. coli O157 suggests the need for following strict hygienic and good sanitation practices during blending and bottling for home fermenters and an approved HACCP plan for foodservice operators to ensure product safety.
文摘We investigated the presence and related signal-to-noise ratio (SNR) of rod-shaped bacteria on a produce surface using elastic scattering. The theoretical noise was defined as a scattering signal from a rough produce surface while the signal was defined as a scattering signal from the increasing numbers of rod-shaped bacteria on the produce surface. In this research, we measured the surface topography of a tomato using BioAFM to provide the quantitative nature of the surface roughness which was, in turn, modeled with the discrete dipole approximation (DDA) for an accurate estimation of the background scattering signature. Then we included the DDA model of rod-shaped bacteria and calculated the combined elastic scattering signature in the upper hemispherical space with different polarizations, wavelengths, and incident angles. The total scattering cross-section (TSC) and partial scattering cross-section (PSC) were both computed on six predefined aperture locations. The results indicate that, upon proper selection of the wavelength and incident angle, it was possible to provide the minimum number of bacteria (~32) to provide a differentiable elastic scattering signal from the produce surface.
文摘Foodborne pathogens continue to pose a potential food safety hazard in ready-to-eat (RTE) meat. Chlorine is commonly used to sanitize processing equipment where Escherichia coli O157:H7 (Ec) may survive and contaminate food products. The objective of this study was to characterize the survival behavior of Ec with different stresses on RTE meats. A multi-strain cocktail of Ec was pre-treated with freezing shock for 15 - 20 h and/or chlorine (0, 25, and 50 ppm) for one hour, and then inoculated onto RTE meat surfaces to obtain about 3.0 log CFU/g. Samples were stored at three abuse temperatures (12℃, 18℃, and 24℃) and Ec was enumerated during the storage. The freezing shock impact was studied using the Ec cocktail stored in a freezer overnight followed by chlorine exposure for one hour. The lag phase and growth rate of Ec were estimated using DMFit (Combase, Baranyi’s model). Results indicated that Ec growth was suppressed by chlorine treatment. Freezing shock was found to have little impact in terms of lag time and growth rate. The lag phase of Ec after exposure to 0 ppm of chlorine (50.3 h) was shorter than that of Ec treated with 25 ppm (54.6 h) and 50 ppm (164.1 h) at 12℃. However, the lag phase decreased with an increase in temperature, e.g. at 25 ppm, lag times were 54.6, 51.1 and 48.9 h for 12℃, 18℃ and 24℃, respectively. Lag times increased with an increase in chlorine concentration. At 24℃, lag times were 15.8, 48.9, and 52.4 h for 0, 25, and 50 ppm, respectively. The growth rate increased with an increase in temperature for 0 and 25 ppm chlorine levels, but decreased at 50 ppm level. Growth rate and lag phase as a function of temperature and chlorine concentration can be described by polynomial models and modified Ratkowsky-type and Zwietering-type models. Results of this study will contribute to risk assessment of RTE meats.
文摘Organically modified silica (ORMOSILS) thin films produced by sol-gel method were imprinted with two bacterial strains as whole cells in order to develop an easy, fast and specific probe to detect and specifically identify these micro-organisms when present in water samples. An important feature of the imprinting process was the molecular finger-prints left by these microorganisms alongside morphology, into imprinted film cavities. The films also showed high selectivity toward the imprinted template and were able to discriminate between two very close bacterial species (E. coli and S. typhimurium). In addition, several central physical parameters of the experimental water solution were examined (i.e., pH, ionic strength and the organic load exemplified by NaCl and TOC concentration, respectively). The method sensitivity to different bacterial concentrations was studied by confocal microscopy (CLSM) and quartz crystal microbalance (QCM) tools. Results showed that increased bacterial concentrations favor rapid adsorption onto imprinted sol-gel films with high affinity, while low pH, increased organic load and high ionic concentrations (i.e., seawater) interfere with bacteria re-adsorption, reducing detection capability. Under average drinking water chemical composition the method proved to be highly efficient.
