A simple method to prepare of DNA template suitable for PCR amplification from filamentous fungi will be valuable for improving experimental efficiency.Here,a method was developed which just needed ultrasonic treatment of the mycelium at usual condition,and the produced solution could directly be used as DNA template for internal transcribed spacer(ITS)amplification successfully.The PCR could be improved by additional treatment of 60℃water baths,but was not centrifugation.When the template amount was 0.5-2μL and the ultrasonic time was 7-11 min,there was no distinctly influences on PCR.The method was commonly used for M.purpureus,I.cicadae,Lentinula sp.,Flammul sp.and Dictyophora sp.etc.to detect target sequences of ITS,hygromycin resistance gene(Hyg),CRISPR-associated protein 9(Cas9),Citrinin gene C(CtnC),Citrinin gene D(CtnD),large subunit rRNA gene(NL),and so on.The method could provide a simple,rapid,safe and economic approach to prepare the DNA template for large-scale PCR of the special filamentous fungi materials. 展开更多

Increased Effluent Dosage Effects on On-Site Wastewater Treatment Systems of Differing Architecture Type

Approximately 20% of homes nationwide use an on-site treatment system as a form of household wastewater management. However, approximately 10% to 20% of on-site treatment systems malfunction each year, many of which have either failed or exceeded the soil’s long-term acceptance rate (LTAR), causing environmental and human health risks. The objective of this field study was to evaluate the effects of soil condition (e.g., wet and dry) and product architecture type [i.e., chamber, gravel-less-pipe (GLP), polystyrene-aggregate, and pipe-and-aggregate] on in-product solution storage and biomat thickness in a profile-limited soil in northwest Arkansas under increased loading rates and to estimate the LTAR for each product. During Phase I of this study (March 13 to October 4, 2013), effluent loading rates were approximately doubled, while rates were approximately quadrupled during Phase II (October 8, 2013 to May 29, 2014), from the maximum allowable loading rate for each product. The pipe-and-tire-chip, 46-cm-wide trench pipe-and-gravel, and the 25-cm diameter GLP products had the greatest (p < 0.001), while the 31-cm-width and the 5.4-m-long chambers had the lowest (p < 0.001) in-product solution storage during wet-soil conditions of Phase I monitoring. The 25-cm diameter GLP product had the greatest (p < 0.001), while the 61-cm-width, 5.4-m-long chamber had the lowest (p < 0.001) in-product solution storage during Phase II. Results of this study indicate that some alternative products may be able to effectively handle effluent loading rates in excess of those currently allowed by the State of Arkansas. 替雷利珠单抗一线治疗晚期非小细胞肺癌的快速卫生技术评估

目的评价替雷利珠单抗(TIS)一线治疗晚期非小细胞肺癌(NSCLC)的有效性、安全性和经济性。方法计算机检索PubMed、Embase、Cochrane Library、CNKI、WanFang Data、SinoMed数据库和卫生技术评估(HTA)相关网站,搜集TIS一线治疗晚期NSCLC的... 目的评价替雷利珠单抗(TIS)一线治疗晚期非小细胞肺癌(NSCLC)的有效性、安全性和经济性。方法计算机检索PubMed、Embase、Cochrane Library、CNKI、WanFang Data、SinoMed数据库和卫生技术评估(HTA)相关网站,搜集TIS一线治疗晚期NSCLC的HTA报告、系统评价/Meta分析和药物经济学研究,检索时限均从建库至2024年4月30日。由2名研究者独立筛选文献、提取资料和质量评价,并采用定性描述方法进行快速卫生技术评估。结果共纳入9篇文献,包括系统评价/Meta分析7篇,药物经济学研究2篇。有效性方面,与化疗(CT)相比,TIS+CT可提高晚期NSCLC患者的无进展生存期(PFS)和客观缓解率(ORR),可提高任何程序性细胞死亡受体配体-1(PD-L1)表达情况、伴或不伴肝转移、年龄≥65岁或<65岁、有吸烟病史的晚期NSCLC患者的PFS;与CT相比,TIS+CT可提高晚期非鳞状NSCLC患者的PFS,可提高PD-L1≥50%的晚期非鳞状NSCLC患者的PFS;与CT相比,TIS+CT可提高PD-L1为1%~49%、PD-L1≥50%、男性、年龄≥65岁、有吸烟史、ECOG评分1分、ⅢB期和Ⅳ期晚期鳞状NSCLC患者的PFS。安全性方面,与卡瑞利珠单抗+CT和阿替利珠单抗+贝伐珠单抗+CT相比,TIS+CT可降低严重不良反应发生率。经济学方面,对于无表皮生长因子受体突变和渐变淋巴瘤激酶重排的非鳞状NSCLC,与CT相比,TIS+CT在中国具有一定的成本-效果优势。亚组分析结果显示,在PD-L1表达≥50%、肝转移和有吸烟史的非鳞状NSCLC患者中,一线TIS+CT方案的生存益处更大。结论TIS+CT一线治疗晚期NSCLC具有较好的有效性、安全性和经济性。 展开更多

快速反应团队工作模式对重症孕产妇抢救效率及救治结局的影响

目的:观察对重症孕产妇实施快速反应团队(RRT)工作模式的干预效果。方法:回顾性收集2021年10月-2022年10月开展常规护理模式的148例重症孕产妇临床资料纳入对照组,2022年11月-2023年11月开展RRT工作模式的152例重症孕产妇临床资料纳入... 目的:观察对重症孕产妇实施快速反应团队(RRT)工作模式的干预效果。方法:回顾性收集2021年10月-2022年10月开展常规护理模式的148例重症孕产妇临床资料纳入对照组,2022年11月-2023年11月开展RRT工作模式的152例重症孕产妇临床资料纳入观察组。比较两组抢救效率、救治结局、新生儿情况、重症孕产妇并发症。结果:观察组处理时间(22.6±4.0min)、抢救情况(10.5%)、剖宫产率(35.5%)、新生儿死亡率(0)、ICU入住率(2.0%)、住院时间(7.56±1.34d)、并发症总发生率(2.6%)均少于对照组(33.0±4.5min、18.9%、52.0%、4.1%、27.7%、10.86±3.33d、12.2%),预警情况(40.1%)、有效处理率(98.0%)、5minApagar评分(8.74±0.58分)、脐带血pH值(7.64±1.96)高于对照组(27.7%、81.8%、8.46±0.64分、4.96±1.34)(均P<0.05),两组孕产妇死亡率、新生儿1minApagar评分无差异(P>0.05)。结论:RRT工作模式可提高重症孕产妇的抢救效率,改善救治结局、新生儿情况。 展开更多

Heat treatment plays an important role in tailoring the mechanical properties of powder-metallurgy(PM)titanium alloys.However,only limited work about the rapid heat treatment(RHT)of PM titanium alloys has been reported.In this work,RHT was applied to PM Ti-5Al-5Mo-5V-1Cr-1Fe alloy after hot rolling to study the evolution of its mechanical properties and the influence of residual pores on its properties.Through hot rolling and annealing,a fine and uniformα+βstructure with few residual pores is ob-tained.During RHT,the primaryαdissolves gradually and completes in theβregion,and theβgrains then grow,resulting in the continuous decrease in elongation after aging.Moreover,the tensile strength first increases and then decreases with increasing RHT temperature,owing to the increase in volume fraction of secondaryαinα+βregion and theβgrain growth inβregion.In contrast to the RHT of cast-and-wrought titanium,the negative influence of residual pores lowers the RHT temperature for obtaining the highest tensile strength to a temperature below theβ-transus temperature.Despite the negative influence of the residual pores,retained primaryαand fineβgrains with fine secondaryαinside contribute to achieving a good strength/ductility balance(1570 MPa and 6.1%,respectively).Addi-tionally,although at higher cycles to failure,the negative influence of residual pores increases as it affects the crack initiation zone at the subsurface,the good resistance of secondaryαto fatigue crack propaga-tion still enhances the fatigue strength considerably(about 300 MPa).This work suggests a cost-effective strategy to produce titanium alloys with high performance. 展开更多

日的探讨基于脑计算机断层扫描灌注成像(CTP)的RAPID软件在行血管内治疗的前循环急性缺血性脑卒中患者预后预测中的应用价值,并探寻影响患者预后的指标。方法回顾性分析2017年1月至2018年2月于海军军医大学(第二军医大学)长海医院行血... 日的探讨基于脑计算机断层扫描灌注成像(CTP)的RAPID软件在行血管内治疗的前循环急性缺血性脑卒中患者预后预测中的应用价值,并探寻影响患者预后的指标。方法回顾性分析2017年1月至2018年2月于海军军医大学(第二军医大学)长海医院行血管内治疗,完成脑CTP检查及RAPID软件进行后期图像分析,且术后血管再通达改良脑梗死溶栓(mTICI)2b级及以上的前循环急性缺血性脑卒中患者的病历资料。根据术后3个月改良Rankin量表(mRS)评分,将患者分为预后良好组(mRS评分≤2分)和预后不良组(mRS评分>2分)。比较两组年龄、入院时美国国立卫生研究院卒中量表(NIHSS)评分等一般资料以及RAPID软件分析结果,包括达峰时间>4s、>6s、>8s、>10s的低灌注区体积(V_(Tmax>4s)、V_(Tmax>6s)、V_(Tmax>8s)、V_(Tmax>10s))和梗死核心区体积(V_(CBF<30%))、脑血流不匹配体积和不匹配指数(MMR)。采用多因素logistic回归分析筛选预测因素,并获得预测公式。采用受试者工作特征(ROC)曲线评价该预测公式的预测能力。结果符合入组标准的前循环急性缺血性脑卒中患者共137例,预后良好组92例,预后不良组45例。预后良好组患者的年龄、入院时NIHSS评分均低于预后不良组,差异均有统计学意义(t=-4.21、-4.06,P均<0.01)。预后良好组V_(Tmax>6s_、V_(Tmax>8s_、V_(Tmax>10s)、V_(CBF<30%)均小于预后不良组,MMR大于预后不良组,差异均有统计学意义(Z=-3.11、-3.17、-3.38、-4.52、-3.74,P均<0.01)。Logistic回归分析显示年龄、V_(CBF<30%)小是大血管闭塞所致前循环急性缺血性脑卒中血管内治疗患者预后良好的影响因素[比值比(OR)=0.904,95%置信区间(CI):0.860~0.950;OR=0.976,95%CI:0.964~0.988;P均<0.01]。预测公式为Logit(P)=8.454-0.024×V_(CBF<30%)-0.101×年龄。ROC曲线分析结果显示该预测公式的曲线下面积为0.786(95%CI:0.699~0.873,P<0.01)。结论 V_(CBF<30%)和年龄是前循环急性缺血性脑卒中血管内治疗患者预后的独立预测因素,年龄越小、V_(CBF<30%)越小预后良好的可能越大。基于CTP的RAPID软件可用于术前筛选适合血管内治疗的前循环急性缺血性脑卒中患者,值得临床推广。 展开更多

Estrogen rapid effects:a window of opportunity for the aging brain?

Estrogen produces several beneficial effects in healthy neurological tissues and exhibits cardioprotective effects.Hormone therapy has been widely used to treat menopausal estrogen deficiency for more than 80 years.Despite high initial expectations of cardioprotective effects,there has been substantial distrust following important randomized clinical trials,such as the Women’s Health Initiative.Subsequently,the timing of treatment in relation to the onset of menopause came under consideration and led to the proposal of the timing hypothesis,that early initial treatment is important,and benefits are lost as the timing since menopause becomes prolonged.Subsequent analyses of the Women’s Health Initiative data,together with more recent data from randomized and observational trials,consistently show reductions in coronary heart disease and mortality in younger menopausal women.Regarding cognitive function,the timing hypothesis is consistent with observations from basic and animal studies.There is some clinical evidence to support the benefits of hormonal therapy in this context,though skepticism remains due to the paucity of clinical trials of substantial length in younger menopausal women.It is likely that the effects of estrogens on cognitive performance are due to rapid mechanisms,including mechanisms that influence Ca2+homeostasis dynamics,provide protection in a hostile environment and reduce inflammatory signals from neural tissues.In the future,inflammatory profiles accounting for early signs of pathological inflammation might help identify the‘window of opportunity’to use estrogen therapy for successful cognitive protection. 展开更多

Pathogenic microorganisms can cause infectious diseases in the human body and cause harm to humans.Under the continuous changes of human life style,sudden infectious diseases and sudden pathogenic microbial contamination in food have brought great challenges to human health.Traditional pathogenic microorganism detection is mainly carried out in laboratories with low speed,and requires experienced operators,which greatly affects the detection efficiency,and real-time monitoring cannot be achieved in the face of epidemics or food contamination.In response to these new situations,new on-site rapid detection methods have become a must,but to achieve the purpose of on-site rapid and immediate detection,higher requirements have been put forward to sensitivity and specificity.Due to their special photoelectric properties and adsorption properties,nanomaterials have gradually been applied in on-site rapid detection methods such as biosensors,and some substantial progress has been made.In this study,we introduced the actual application of nanomaterials in testing and their applications in on-site rapid testing,and made some prospects for the development of on-site rapid testing methods. 展开更多

This paper evaluated two different wastewater primary treatments of wastewater from the“Universidad Autonoma Metropolitana,Azcapotzalco Campus”,a conventional one integrated for coagulation,flocculation,sedimentation and filtration versus one alternative integrated for in-line coagulation to precondition to rapid filtration.The wastewater was from the Azcapotzalco Campus in Mexico City,and in both cases,we characterized the wastewater,measured:pH,conductivity,temperature,total dissolved solids,dissolved oxygen,ORP(Oxide Reduction Potential),turbidity,and COD(Chemical Oxygen Demand).Conventional treatment was slightly better treatment,but the alternative treatment represents saving in equipment,reactive,energy and time,so it was considered a viable technical and economic wastewater primary treatment. 展开更多

Effects of induction heat treatment on mechanical properties of TiAl-based alloy

The effects of rapid heating cyclic heat treatment on mechanical properties of a TiAl based alloy (Ti 33Al 3Cr) were studied by means of an induction heating machine. The results show that: 1) fine fully lamellar microstructure with colony size of about 50 μm and lamellar spacing of about 0.12 μm can be obtained; 2) the compression mechanical properties can be improved to a large extent and the best comprehensive compression mechanical properties can reach the yield stress 745 MPa, the large flow stress 1 672 MPa and the compression ratio 19.4%; and 3) the compression fracture at room temperature after induction heat treatment and aging is still typical cleavage fracture. 展开更多
