[Objectives]Polysaccharide was extracted from Lactarius deliciosus by ultrasonic-assisted method to improve polysaccharide yield.[Methods]Five variables including extraction temperature,extraction time,ultrasonic powe...[Objectives]Polysaccharide was extracted from Lactarius deliciosus by ultrasonic-assisted method to improve polysaccharide yield.[Methods]Five variables including extraction temperature,extraction time,ultrasonic power,ultrasonic time and material-to-liquid ratio were selected for single factor experiments.The extraction process of L.deliciosus polysaccharide was optimized by response surface analysis,and its antioxidant activity was evaluated by measuring its total reduction ability and DPPH free radical scavenging capacity.[Results]The optimal extraction conditions were determined as follows:material-to-liquid ratio 1:35 g:ml,ultrasonic power 462 W,ultrasonic time 10 min,extraction time 110 min and extraction temperature 90℃.Under these conditions,the extraction rate of polysaccharide was(10.83±0.03)%.The antioxidant test results showed that when the mass concentration of L.deliciosus polysaccharide was 0.5 mg/ml,its absorbance and DPPH free radical scavenging rate reached their maximum values,which were 3.274%and 41.27%,respectively.The L.deliciosus polysaccharide had good antioxidant properties.[Conclusions]This study provides a theoretical basis for further development and utilization of L.deliciosus polysaccharide in the future.展开更多
Objective:To establish an optimized aqueous extraction process for polysaccharides from Physalis alkekengi L.peel and to preliminarily explore its in vitro anti-inflammatory activity against colorectal cancer SW620 ce...Objective:To establish an optimized aqueous extraction process for polysaccharides from Physalis alkekengi L.peel and to preliminarily explore its in vitro anti-inflammatory activity against colorectal cancer SW620 cells.Methods:A single-factor test combined with orthogonal test analysis was used to evaluate the effects of the material-to-liquid ratio,extraction temperature,and extraction time on the yield of polysaccharides from Physalis alkekengi L.peel.The antioxidant activity of the polysaccharides was assessed by analyzing their free radical scavenging ability in vitro,and the anti-inflammatory effect was evaluated using SW620 cells.Results:The optimal extraction conditions were a material-to-liquid ratio of m(g):V(mL)=1:30,an extraction temperature of 100℃,and an extraction time of 40 minutes,with a predicted polysaccharide yield of 25.7%.The polysaccharides from Physalis peruviana peel effectively scavenged DPPH,superoxide anion,and hydroxyl radicals.After treatment with Physalis peruviana polysaccharides,the levels of IL-1β,IL-18,and TNF-αin the cell culture medium were significantly reduced,and the phosphorylation level of P65 protein in SW620 cells was decreased.Conclusion:This extraction method is stable and reliable,and the prepared Physalis alkekengi L.polysaccharides exhibit significant in vitro antioxidant and anti-inflammatory activities.This study provides a theoretical basis for developing drugs for the prevention and treatment of colorectal cancer.展开更多

And their antioxidative as well as glycosidase inhibitory activities ar...In the present study, ultrasonic extraction technique (UET) is used to improve the yield of polysaccharides from Lami- naria japonica (LJPs). And their antioxidative as well as glycosidase inhibitory activities are investigated. Box-Behnken design (BBD) combined with response surface methodology (RSM) is applied to optimize ultrasonic extraction for polysaccharides. The optimized conditions are obtained as extraction time at 54 min, ultrasonic power at 1050 W, extraction temperature at 80℃ and ratio of material to solvent at 1:50 (g mL-1). Under these optimal ultrasonic extraction conditions, an actual experimental yield (5.75% + 0.3%) is close to the predicted result (5.67%) with no significant difference (P〉0.05). Vitro antioxidative and glycosidase inhibitory activities tests indicate that the crude polysaccharides (LJP) and two major ethanol precipitated fractions (LJP1 and LJP2) are in a concentration-dependent manner. LJP2 (30%-60% ethanol precipitated polysaccharides) possesses the strongest α-glucosidase in- hibitory activity and moderate scavenging activity against hydroxyl radicals (66.09% ±2.19%, 3.0 mg mL-l). Also, the inhibitory activity against a-glucosidase (59.08% ± 3.79%, 5.0 mg mL-1) is close to that of acarbose (63.99% ± 3.27%, 5.0 mg mL-l). LJP 1 (30% ethanol precipitated polysaccharides) exhibits the strongest scavenging activity against hydroxyl radicals (99.80%q-0.00%, 3.0mg mL-1) and moderate a-glucosidase inhibitory activity (47.76%± 1.92%, 5.0 mgmL-1). LJP shows the most remarkable DPPH scav- enging activity (66.20%±0.11%, 5.0mgmL-1) but weakest a-glucosidase inhibitory activity (37.77%±1.30%, 5.0mgmL-1). How- ever, all these LJPs exert weak inhibitory effects against a-amylase. These results show that UET is an effective method for extract- ing bioactive polysaccharides from seaweed materials. LJP 1 and LJP2 can be developed as a potential ingredient in hypoglycemic agents or functional food for the management of diabetes. This study provides scientific evidence and advances in the preparation technology and a hypoglycemic activities evaluation method for seaweed polysaccharides, especially glycosidase inhibition in com- bination with an antioxidative activity evaluation method.展开更多

Single factor experiments were conducted to examine the effects of extracting time,temperature,the solid-liquid ratio and pH value on the extraction yield of polysaccharide from pumpkin. [Method] The best enzyme ratio and extraction conditions for complex enzymes extraction were determined through orthogonal tests. Scavenging ·OH and O-2 activities of pumpkin polysaccharides were also investigated by salicylic acid and improved self-oxidation of o-pheno methods respectively. [Results] The results showed that the biggest extraction yield of polysaccharide from pumpkin can be got when adding 1% cellulose enzyme,1.5% pectinase,1.0% papain and Na2HPO4-citric acid buffer solution (pH was 4.6),and oscillating for 30 min under water at 40 ℃ with the solid-liquid ratio of 1:30. In addition,pumpkin polysaccharides had a strong activity of eliminating ·OH,but very weak activity to scavenge O-2. Extraction temperature, extrac- tion duration, solution pH and solid to liquid ratio were selected as technical parameters which required to be optimized. With the extraction rate of seaweed poly- saccharides as an index, single-factor experiment and orthogonal experiment were carried out to determine the optimal extraction technique. Seaweed polysaceharides were extracted with the optimal technical parameters to analyze the biological activities. Savage method was used to process seaweed polysaceharide extract. In vitro biological activities of preliminarily purified seaweed polysaeeharides were investigated successively, including reducing ability, antioxidant ability and antibacterial activity. The results showed that there was a positive correlation between polysaccharide concentration and its reducing ability, antioxidant ability and antibacterial activity. Furthermore, there was also a correlation among the reducing ability, antioxidant ability and antibacterial activity of seaweed polysaecharides; the relation- ship between each two of these three indicators was similar to positive linear correlation.展开更多

[ ...[ Objective ] This study aimed to investigate the optimal extraction process of crude polysaccharides from wild Lactarius volemus Fr. in Yunnan Province and preliminarily analyzed its antioxidant activity in vitro. [ Method ] With water extraction and alcohol precipitation method, the optimal conditions for extracting crude polysaccharides from wild L. volemus Ft. were screened by single-factor and orthogonal experiments. The antioxidant activity of the extracted crude polysac- charides was determined with DPPH assay. [ Result ] The optimal conditions for pigment removal with activated carbon were: activated carbon amount of 20 g/L, water bath time of 40 min, water bath temperature of40 ℃ ; the optimal conditions for extracting crude polysaccharides from wild L. vo/emus Fr. with hot water ex- traction method were: hot water extraction time of 3 h, solid-liquid ratio of 1: 45, extraction frequency of twice. Under the optimized extraction conditions, the yield of crude polysaccharides was 21.33 mg/g. In addition, the antioxidant activity of 0. 665 mg/ml crude polysaccharides was 52.46% ; the amount of crude polysac- eharides was proportional to the antioxidant activity. [ Conclusion] Hot water extraction method can be used as a high-efficiency extraction technology of crude pol- ysaecharides from wild L. volemus Fr. with simple operation and low costs. Crude polysaccharides extracted from L. volemus Ft. exhibited certain antioxidant activi- ty in vitro.展开更多

This study optimized the alkali-extraction conditions and antioxidant activities of soluble polysaccharides from the body wall of Sipunculus nudus. The effects of solid-liquid ratio, extraction duration, extraction temperature, and alkali concentration on the yield of S. nudus polysaccharides(SNP) were examined, according to which the optimal combination of extraction parameters was obtained by an orthogonal test. The relative influencing importance of different extraction parameters on the yield of SNP followed the order as solid-liquid ratio > extraction temperature > alkali concentration > extraction duration. The highest extraction yield, 1.98%, was achieved under an optimal extraction condition: temperature 60℃, solid-liquid ratio 1:6 g mL^(-1), duration 5 h, and alkali(Na OH) mass fraction 6%. The in vitro antioxidant activities examination showed that extracted SNP under this optimized condition had strong power in reducing certain hydroxyl and superoxide radical scavenging abilities. The promising results showed that extracted SNP could be a potent natural antioxidant.展开更多

[Methods] The single factor orthogonal design was applied to explore the effects of ultrasonic time,solid-to-liquid ratio,extraction time and ethanol precipitation concentration on the extraction rate of crude polysaccharides from C. Capitata,to obtain the optimal extraction process; DPPH and ABTS^+ free radical scavenging ability was detected to evaluate its in vitro antioxidant activity.[Results]The optimal extraction process of C. Capitata crude polysaccharide was as follows: solid-to-liquid ratio was 40∶ 1,the extraction temperature was 50℃,the ultrasonic time was 80 min. Under such conditions,the extraction rate of C. Capitata crude polysaccharide was 10. 51%,and the purity was 56. 9%; C. Capitata crude polysaccharide has good scavenging ability to DPPH and ABTS^+ free radicals,and its IC_(50) values are 0. 468 and 0. 231 mg/mL,respectively. [Conclusions]The optimized process is simple and feasible,and the C. Capitata polysaccharide has good antioxidant activity in vitro. The antioxidant activity of their ethanolic extracts, as well as their effects on the oxidation of edible corn oil during accelerated ageing, was studied. Maceration of taxonomically identified commercial vegetable samples, previously peeled and thinly sliced, was carried out at ambient temperature, out of direct light, with occasional agitation and ultrasound, employing 95% ethyl alcohol as the extraction solvent, allowing them to stand for 7 days. The total polyphenolic contents were determined on the filtrated extracts using the Folin-Ciocalteau method. The antioxidant activity was evaluated on emulsions of ethanolic extracts of onion prepared in edible commercial corn oil, using sorbitan monooleate as emulsifying agent. The peroxide values were analyzed using the iodometric method;oxidation induction times were obtained from the peroxide evolution graphs, using the tangent method. Oil samples emulsified with ethanolic onion extracts showed an extension of the induction period. A 7 day ageing study at 45℃ was additionally performed to determine the conjugated dienes on pure commercial corn oil and its emulsions by visible spectrophotometry. The spectral analysis showed an increase of the measured absorbances in oil samples without additives and no change for the oils emulsified with onion extract. An increasing of diene values was observed for corn oil without additives during ageing;no changes in the value were observed in oils emulsified with onion extracts.展开更多

Single-factor tests and orthogonal experimental design(L_9(3~3)) were used to obtain the optimal...An ultrasonic-assisted technique was employed to extract crude polysaccharide from Tricholoma matsutake fruiting bodies. Single-factor tests and orthogonal experimental design(L_9(3~3)) were used to obtain the optimal extraction conditions. Results showed that the optimal parameters were as follows: ultrasonic temperature, 40 °C; ultrasonic time, 50 min; water to raw material ratio, 25 ml/g; ultrasonic frequency, 45 k Hz; and ultrasonic power, 100 W. Three novel T. matsutake polysaccharide(TMP) fractions(TMP30, TMP60, and TMP80) were isolated and purified from TMP by stepwise alcohol precipitation. Their preliminary structural features were determined by highperformance anion-exchange chromatography with pulsed-amperometric detection(HPAEC-PAD) and Fourier transform infrared spectrophotometer(FT-IR) analyses. Furthermore, their in vitro antioxidant activity was investigated in terms of a reducing power assay and the scavenging rates of 2,2-diphenyl-1-picrylhydrazyl(DPPH) and hydroxyl radicals. The order of the various fractions based on their antioxidant activity was TMP80>TMP>TMP60>TMP30. These findings suggested that novel polysaccharide fractions from T. matsutake, especially TMP80, could be promising active macromolecules for biomedical use.展开更多

The effects of extraction temperature,time,ultrasonic power,solid-liquid ratio on polysaccharides yield were investigated by single-factor experiment and orthogonal experiment to obtain optimum extraction conditions; the antioxidant activities of lychee pericarp polysaccharides were evaluated by the total antioxidant activity. Optimum extraction process was under extraction temperature of 80 ℃,extraction time of 2. 5 h,ultrasonic power of 180 W,solid-liquid ratio of 1∶ 25 g / m L; the total antioxidant activity was increased with the increasing concentration of polysaccharides. The extraction process is simple and efficient. The extraction rate of lychee pericarp polysaccharides with antioxidant activities is high.
文摘[Objectives]This study was conducted to optimize the optimal process conditions for extracting polysaccharides from Rose laevigate Michx.with hot water.[Methods]The extraction rate of R.laevigate was used as an evaluation index,and extraction temperature,material-to-liquid ratio,extraction time and extraction times were selected as influencing factors.On the basis of single factor tests,L 9(34)orthogonal tests were carried out for optimization,and the content of polysaccharides was determined by the phenol-sulfuric acid method.The in vitro antioxidant activity of the extracted R.laevigate polysaccharides was studied.[Results]The effects of various influencing factors on the extraction rate of R.laevigate polysaccharides ranked as material-liquid ratio>extraction temperature>extraction times>extraction time.The optimal extraction process was extraction temperature 80℃,extraction time 2 h,and material-to-liquid ratio 1∶25,extraction times 3 times,under which the extraction rate was(9.55±0.12)%.R.laevigate polysaccharides had certain scavenging capacities of DPPH free radicals and hydroxyl free radicals,and its scavenging capacity of DPPH free radicals was better than that of hydroxyl free radicals.[Conclusions]This study provides a theoretical basis for the optimization of the extraction process of R.laevigate polysaccharides and the in-depth development and application of polysaccharides.

文摘[Objective] This paper aims to study a new method of extracting pumpkin polysaccharide from pumpkin. Single factor experiments were conducted to examine the effects of extracting time,temperature,the solid-liquid ratio and pH value on the extraction yield of polysaccharide from pumpkin. [Method] The best enzyme ratio and extraction conditions for complex enzymes extraction were determined through orthogonal tests. Scavenging ·OH and O-2 activities of pumpkin polysaccharides were also investigated by salicylic acid and improved self-oxidation of o-pheno methods respectively. [Results] The results showed that the biggest extraction yield of polysaccharide from pumpkin can be got when adding 1% cellulose enzyme,1.5% pectinase,1.0% papain and Na2HPO4-citric acid buffer solution (pH was 4.6),and oscillating for 30 min under water at 40 ℃ with the solid-liquid ratio of 1:30. In addition,pumpkin polysaccharides had a strong activity of eliminating ·OH,but very weak activity to scavenge O-2. [Conclusion] This study provided basic data for research and application of Pumpkin polysaccharide.

文摘In this study,an orthogonal experiment was performed to explore the optimal extraction technique of water-soluble polysaccharides from pumpkin.Moreover,the polysaccharides' capabilities to scavenge ·OH and DPPH· and their gross reducing power was measured with Vc and BHT as references.In the orthogonal experiment,various factors including temperature,extraction time,solid-liquid ratio,were optimized.The results showed that the polysaccharides extracted from pumpkin are endowed with strong in vitro antioxidant properties.Our study provided references for the extraction of pumpkin polysaccharides and for their use as antioxidants.

文摘Dried seaweed powder was used as the raw material to extract polysaccharides by ethanol precipitation after water bath. Extraction temperature, extrac- tion duration, solution pH and solid to liquid ratio were selected as technical parameters which required to be optimized. With the extraction rate of seaweed poly- saccharides as an index, single-factor experiment and orthogonal experiment were carried out to determine the optimal extraction technique. Seaweed polysaceharides were extracted with the optimal technical parameters to analyze the biological activities. Savage method was used to process seaweed polysaceharide extract. In vitro biological activities of preliminarily purified seaweed polysaeeharides were investigated successively, including reducing ability, antioxidant ability and antibacterial activity. The results showed that there was a positive correlation between polysaccharide concentration and its reducing ability, antioxidant ability and antibacterial activity. Furthermore, there was also a correlation among the reducing ability, antioxidant ability and antibacterial activity of seaweed polysaecharides; the relation- ship between each two of these three indicators was similar to positive linear correlation.

文摘[ Objective ] This study aimed to investigate the optimal extraction process of crude polysaccharides from wild Lactarius volemus Fr. in Yunnan Province and preliminarily analyzed its antioxidant activity in vitro. [ Method ] With water extraction and alcohol precipitation method, the optimal conditions for extracting crude polysaccharides from wild L. volemus Ft. were screened by single-factor and orthogonal experiments. The antioxidant activity of the extracted crude polysac- charides was determined with DPPH assay. [ Result ] The optimal conditions for pigment removal with activated carbon were: activated carbon amount of 20 g/L, water bath time of 40 min, water bath temperature of40 ℃ ; the optimal conditions for extracting crude polysaccharides from wild L. vo/emus Fr. with hot water ex- traction method were: hot water extraction time of 3 h, solid-liquid ratio of 1: 45, extraction frequency of twice. Under the optimized extraction conditions, the yield of crude polysaccharides was 21.33 mg/g. In addition, the antioxidant activity of 0. 665 mg/ml crude polysaccharides was 52.46% ; the amount of crude polysac- eharides was proportional to the antioxidant activity. [ Conclusion] Hot water extraction method can be used as a high-efficiency extraction technology of crude pol- ysaecharides from wild L. volemus Fr. with simple operation and low costs. Crude polysaccharides extracted from L. volemus Ft. exhibited certain antioxidant activi- ty in vitro.

文摘Marine organisms constitute unlimited resource of bioactive substances due to their high biodiversity and represent a valuable source of new compounds. This study optimized the alkali-extraction conditions and antioxidant activities of soluble polysaccharides from the body wall of Sipunculus nudus. The effects of solid-liquid ratio, extraction duration, extraction temperature, and alkali concentration on the yield of S. nudus polysaccharides(SNP) were examined, according to which the optimal combination of extraction parameters was obtained by an orthogonal test. The relative influencing importance of different extraction parameters on the yield of SNP followed the order as solid-liquid ratio > extraction temperature > alkali concentration > extraction duration. The highest extraction yield, 1.98%, was achieved under an optimal extraction condition: temperature 60℃, solid-liquid ratio 1:6 g mL^(-1), duration 5 h, and alkali(Na OH) mass fraction 6%. The in vitro antioxidant activities examination showed that extracted SNP under this optimized condition had strong power in reducing certain hydroxyl and superoxide radical scavenging abilities. The promising results showed that extracted SNP could be a potent natural antioxidant.

文摘[Objectives]To optimize the ultrasonic-assisted extraction process of polysaccharides from Cyathula Capitata( Wall.) Moq. and evaluate its antioxidant capacity in vitro. [Methods] The single factor orthogonal design was applied to explore the effects of ultrasonic time,solid-to-liquid ratio,extraction time and ethanol precipitation concentration on the extraction rate of crude polysaccharides from C. Capitata,to obtain the optimal extraction process; DPPH and ABTS^+ free radical scavenging ability was detected to evaluate its in vitro antioxidant activity.[Results]The optimal extraction process of C. Capitata crude polysaccharide was as follows: solid-to-liquid ratio was 40∶ 1,the extraction temperature was 50℃,the ultrasonic time was 80 min. Under such conditions,the extraction rate of C. Capitata crude polysaccharide was 10. 51%,and the purity was 56. 9%; C. Capitata crude polysaccharide has good scavenging ability to DPPH and ABTS^+ free radicals,and its IC_(50) values are 0. 468 and 0. 231 mg/mL,respectively. [Conclusions]The optimized process is simple and feasible,and the C. Capitata polysaccharide has good antioxidant activity in vitro. This experiment provides a basis for the further development and utilization of the C. Capitata polysaccharide.
文摘The content of polyphenolic in commercial onions has been determined. The antioxidant activity of their ethanolic extracts, as well as their effects on the oxidation of edible corn oil during accelerated ageing, was studied. Maceration of taxonomically identified commercial vegetable samples, previously peeled and thinly sliced, was carried out at ambient temperature, out of direct light, with occasional agitation and ultrasound, employing 95% ethyl alcohol as the extraction solvent, allowing them to stand for 7 days. The total polyphenolic contents were determined on the filtrated extracts using the Folin-Ciocalteau method. The antioxidant activity was evaluated on emulsions of ethanolic extracts of onion prepared in edible commercial corn oil, using sorbitan monooleate as emulsifying agent. The peroxide values were analyzed using the iodometric method;oxidation induction times were obtained from the peroxide evolution graphs, using the tangent method. Oil samples emulsified with ethanolic onion extracts showed an extension of the induction period. A 7 day ageing study at 45℃ was additionally performed to determine the conjugated dienes on pure commercial corn oil and its emulsions by visible spectrophotometry. The spectral analysis showed an increase of the measured absorbances in oil samples without additives and no change for the oils emulsified with onion extract. An increasing of diene values was observed for corn oil without additives during ageing;no changes in the value were observed in oils emulsified with onion extracts.

文摘An ultrasonic-assisted technique was employed to extract crude polysaccharide from Tricholoma matsutake fruiting bodies. Single-factor tests and orthogonal experimental design(L_9(3~3)) were used to obtain the optimal extraction conditions. Results showed that the optimal parameters were as follows: ultrasonic temperature, 40 °C; ultrasonic time, 50 min; water to raw material ratio, 25 ml/g; ultrasonic frequency, 45 k Hz; and ultrasonic power, 100 W. Three novel T. matsutake polysaccharide(TMP) fractions(TMP30, TMP60, and TMP80) were isolated and purified from TMP by stepwise alcohol precipitation. Their preliminary structural features were determined by highperformance anion-exchange chromatography with pulsed-amperometric detection(HPAEC-PAD) and Fourier transform infrared spectrophotometer(FT-IR) analyses. Furthermore, their in vitro antioxidant activity was investigated in terms of a reducing power assay and the scavenging rates of 2,2-diphenyl-1-picrylhydrazyl(DPPH) and hydroxyl radicals. The order of the various fractions based on their antioxidant activity was TMP80>TMP>TMP60>TMP30. These findings suggested that novel polysaccharide fractions from T. matsutake, especially TMP80, could be promising active macromolecules for biomedical use.
