Orientin is a flavonoid monomer.In recent years,its importance as a source of pharmacological active substance is growing rapidly due to its properties such as anti-myocardial ischemia,anti-apoptosis,anti-radiation,an...Orientin is a flavonoid monomer.In recent years,its importance as a source of pharmacological active substance is growing rapidly due to its properties such as anti-myocardial ischemia,anti-apoptosis,anti-radiation,anti-tumor,and anti-aging.However,the neuroprotective effects of Orientin on stroke injury have not been comprehensively evaluated.The aim of the present study was thus to investigate the neuroprotective capacity and the potential mechanisms of Cyperus esculentus L.orientin(CLO)from Cyperus esculentus L.leaves against ischemia/reperfusion(I/R)injury using standard orientin as control.For in vitro studies,we treated HT22 cells with CoCl2 as an in vitro ischemic injury model.HT22 cells in the control group were treated with CoCl2.For in vivo studies,we used rat models of middle cerebral artery occlusion,and animals that received sham surgery were used as controls.We found that CLO protected CoCl2-induced HT22 cells against ischemia/reperfusion injury by lowering lipid peroxidation and reactive oxygen species formation as well as decreasing protein oxidation.However,CLO did not reduce the release of lactate dehydrogenase nor increase the activity of superoxide dismutase.Results showed that CLO could decrease neurological deficit score,attenuate brain water content,and reduce cerebral infarct volume,leading to neuroprotection during cerebral ischemia-reperfusion injury.Our studies indicate that CLO flavonoids can be taken as a natural antioxidant and bacteriostastic substance in food and pharmaceutical industry.The molecular mechanisms of CLO could be at least partially attributed to the antioxidant properties and subsequently inhibiting activation of casepase-3.All experimental procedures and protocols were approved on May 16,2016 by the Experimental Animal Ethics Committee of Xinjiang Medical University of China(approval No.IACUC20160516-57).展开更多

In this study, an aged mouse mo...Total flavonoids are the main pharmaceutical components of Trollius chinensis Bunge, and orientin and vitexin are the monomer components of total flavonoids in Trollius chinensis Bunge. In this study, an aged mouse model was established through intraperitoneal injection of D-galactose for 8 weeks, followed by treatment with 40, 20, or 10 mg/kg orientin, vitexin, or a positive control (vitamin E) via intragastric administration for an additional 8 weeks. Orientin, vitexin, and vitamin E improved the general medical status of the aging mice and significantly increased their brain weights. They also produced an obvious rise in total antioxidant capacity, superoxide dismutase, catalase, and glutathione peroxidase levels in the serum, and the levels of superoxide dismutase, catalase and glutathione peroxidase, Na+-K+-ATP enzyme, and Ca2+-Mg2+-ATP enzyme in the liver, brain and kidneys. In addition, they significantly reduced malondialdehyde levels in the liver, brain and kidney and lipofuscin levels in the brain. They also significantly improved the neuronal ultrastructure. The 40 mg/kg dose of orientin and vitexin had the same antioxidant capacity as vitamin E. The deionised water and radiofrequency electromagnetic radiations(RF-EMR) groups were administered with deionised water while orientin was administered to the RF-EMR + low dose of orientin group(10 mg/kg), RF-EMR + high dose of orientin group(20 mg/kg) and high dose of orientin group(20 mg/kg). All the groups except deionised water and high dose groups were exposed to 900 MHz radiofrequency-electromagnetic radiation for 28 consecutive days(1 h/day). Learning and memory was assessed via the step-down inhibitory avoidance task. Activities of lipid prexidation and antioxidant enzymes were measured using kits.Results: Radiofrequency electromagnetic radiation caused impairment in learning and memory and reduced activities of brain antioxidant enzymes, increased lipoperoxidation and corticosterone concentration as well as histopathological aberrations in the hippocampal tissues. Conversely, orientin alleviated learning and memory deficit, improved the activities of endogenous antioxidant enzymes and mitigated brain lipoperoxidation and neuronal degeneration in mice exposed to radiofrequency electromagnetic radiation. Conclusions: Orientin alleviates learning and memory impairment due to radiofrequency electromagnetic radiation in mice by improving antioxidant defence mechanism and may be considered as a promising therapeutic agent for improving the antioxidant system of people living in radiofrequency electromagnetic radiation-prone environment.展开更多
目的建立基于多种内参物的一测多评法测定金莲清热颗粒中芒果苷、荭草素-2″-O-β-L-半乳糖苷、荭草苷、藜芦酸、牡荆苷和哈巴俄苷6个成分的含量。方法以Agilent Eclipse Plus C_(18)为色谱柱,以乙腈-0.1%磷酸溶液为流动相(梯度洗脱),...目的建立基于多种内参物的一测多评法测定金莲清热颗粒中芒果苷、荭草素-2″-O-β-L-半乳糖苷、荭草苷、藜芦酸、牡荆苷和哈巴俄苷6个成分的含量。方法以Agilent Eclipse Plus C_(18)为色谱柱,以乙腈-0.1%磷酸溶液为流动相(梯度洗脱),流速为1mL/min,柱温为30℃,检测波长为270 nm。分别以荭草苷、牡荆苷、荭草素-2″-O-β-L-半乳糖苷为内参物,采用一测多评法建立另外5个待测成分与内参物的相对校正因子,然后计算21批金莲清热颗粒样品中6个成分的含量,并与外标法测定结果进行比较。结果一测多评法测得21批金莲清热颗粒样品中芒果苷、荭草素-2″-O-β-L-半乳糖苷、荭草苷、藜芦酸、牡荆苷和哈巴俄苷的含量范围分别为0.234~0.516、1.804~2.270、2.143~2.606、0.190~0.223、0.594~0.782、0.080~0.152 mg/g,外标法测定结果分别为0.235~0.523、1.798~2.265、2.137~2.599、0.190~0.224、0.597~0.786、0.077~0.151 mg/g,2种方法所测结果的差异百分比不大于4.00%。结论建立了基于多种内参物同时测定金莲清热颗粒中6种成分含量的一测多评法,该方法与外标法所得结果无明显差异,可用于金莲清热颗粒的质量控制。展开更多
