In this work, we investigated serum level of Galactin-1 (Gal-1) and its tissue immunohistochemical expression in SOC patients at different stages trying to find out its significance as a diagnostic and prognostic marker. Patients and methods: The study included 95 females I-Control group: Twenty five healthy females;II-Patients group: Seventy females diagnosed as SOC at different stages;Stage I: 8 cases, Stage II: 12 cases, Stage III: 32 cases and Stage VI:18 cases. Serum Galectin-1 and CA-125 were measured by ELIZA and tissue Galectin-1 was assessed by immunohistochemistry. All patients were followed for up to 3 years after surgery. Results: Serum Gal-1 and CA-125 levels were significantly higehr in SOC patients compared to controls (p 0.001). We found a direct positive statistically significant correlation between serum Gal-1 and CA125 levels (p 0.001). Serum Gal-1 at cut off value > 135 ng/ml was superior to CA-125 a cut off value > 49 u/ml with sensitivity, specificity of 100%, vs 88.57, 96% for CA-125. Serum Gal-1 was significantly associated with tumor stage (p 0.001). Immunohistochemistry showed that patients with strong Gal-1 expression had higher serum level (p = 0.002). Stromal and tumor Gal-1 expression were significantly correlated with tumor grade (p 0.001) and stage (p = 0.001). Serum Gal-1, CA-125 and IHC Gal-1 expression were associated with poor survival (p 0.001, p = 0.009 and p = 0.002) respectively. Conclusion: Serum Gal-1 and its tissue IHC expression are useful diagnostic and prognostic markers for SOC patients.展开更多

The result showed that P16 gene was positive expression in HO-8910 cell of mother line,HO8910PM cell line and xenograft of highly mcatstasizing human ovarian carcinoma in nude mice. However, P16gene in the metastatic cell had a weaker expression. P16gene positive expression were also found in sl cases of 69cases (73.9%) in the ovarian epithelial carcinoma paramn embedded tissues. Comparative studies showed that the positive rate of P16 gene expression markedly reduced with the increase of pathologic grade and clinical stage,metastasis in the lymph node and decrease of 5-year survival (P<0.05, p<0.01).P16 gene is not only a controller of cytokerastic cycle, but also a key member of tumorigenic suppresser:its absence and expression degree are also correlated with the ovarian carcinoma genesis and development,especially with the metastasis of the ovarian cancer.展开更多

The Wnt/β-catenin pathway is involved in various normal cellular activities,including determination,proliferation,migration and differentiation in embryonic development and adult homeostasis. Deregulation or constitutive activation of the Wnt/β-catenin pathway may lead to cancer formation. Immunohistochemical expression of β-catenin in gynecologic tumor have been reported recently. In normal epithelia,immunoreactivity was strongly observed at the membrane,partially at cytoplasm,nuclear staining of β-catenin was rarely seen in normal cases; In ovarian carcinomas,β-catenin nuclear expression was found more commonly in endometrioid carcinomas,nuclear β-catenin staining seemed to be of prognostic importance; In endometrium carcinomas,β-catenin nuclear expression were more common in pure endometrioid tumors than in unendometrioid tumors,associated with favorable prognosis,the staining pattern was independent of the menopausal status; In synchronous primary cancers of the endometrium and ovary,activating mutations in β-catenin seemed to distinguish synchronous primary tumors from metastatic tumors.展开更多
