The effects of f...Studies were performed to determine the extent of nuclear DNA degradation induced by iron, iron-ascorbate, or iron-bleomycin under aerobic conditions in a model system using isolated rat liver nuclei. The effects of five antioxidants (catalase, superoxide dismutase, dimethyl sulfoxide, glutathione and diallyl sulfide) on this oxidative nuclear damage were also investigated. At the 0.05 level for statistical significance, iron induced concentration-dependent DNA degradation, and this effect was enhanced by ascorbate and bleomycin. The antioxidants catalase, dimethyl sulfoxide, and diallyl sulfide significantly reduced the iron-ascorbate-induced DNA damage, whereas superoxide dismutase and dimethyl sulfoxide significantly reduced iron-bleomycin-induced damage. Glutathione significantly increased the iron-bleomycin-induced DNA damage. These results suggest that the reactive oxygen species generated by iron, iron-ascorbate, and iron-bleomycin are responsible for the DNA strand breaks in isolated rat liver nuclei.展开更多

Knockout(KO) of As3 mt was shown to reduce the capacity to methylate i As in mice. However, no data are available on the oxidation states of As species in tissues of these mice. Here, we compare the oxidation states of As species in tissues of male C57BL/6 As3mt-KO and wild-type(WT) mice exposed to arsenite(iA s~Ⅲ) in drinking water. WT mice were exposed to50 mg/L As and As3mt-KO mice that cannot tolerate 50 mg/L As were exposed to 0, 15, 20, 25 or30 mg/L As. iA s~Ⅲaccounted for 53% to 74% of total As in liver, pancreas, adipose, lung, heart, and kidney of As3mt-KO mice; tri- and pentavalent methylated arsenicals did not exceed 10% of total As. Tissues of WT mice retained iA s and methylated arsenicals: iA s~Ⅲ, MAs~Ⅲand DMAs~Ⅲ represented 55%‐68% of the total As in the liver, pancreas, and brain. High levels of methylated species, particularly MAs~Ⅲ, were found in the intestine of WT, but not As3mt-KO mice,suggesting that intestinal bacteria are not a major source of methylated As. Blood of WT mice contained significantly higher levels of As than blood of As3mt-KO mice. This study is the first to determine oxidation states of As species in tissues of As3mt-KO mice. To evaluate the potential environmental effects of engineered nano metals,it is important to determine the adverse effects of various nanomaterials on aquatic species.Adult tilapia (Oreochromis mossambicus) were mai...To evaluate the potential environmental effects of engineered nano metals,it is important to determine the adverse effects of various nanomaterials on aquatic species.Adult tilapia (Oreochromis mossambicus) were maintained in 10 L glass aquaria,and exposed to a graded series of synthesized silver nanoparticles (Ag-NPs) at 25,50 and 75 mg/L for eight days.The LC 50 value was 12.6 mg/L.Reduced activities of antioxidant enzymes and the contents of antioxidants were lowered in the gills and liver of fishes treated with AgNPs,which resulted in heavy accumulation of free radicals.Histopathological results imply that the balance between the oxidative and antioxidant system in the fish was broken down during Ag-NPs exposure.The principal concern related with the release of nanomaterials and their smaller particle may change the materials transport and potential toxicity to aquatic organisms compared to larger particles.展开更多
In this study,graphene oxide was covalently immobilized on silica-coated magnetite and then modified with 2-phenylethylamine to give a nanocomposite of type Fe3O4@SiO2@GO-PEA that can be applied to the magnetic solid-...In this study,graphene oxide was covalently immobilized on silica-coated magnetite and then modified with 2-phenylethylamine to give a nanocomposite of type Fe3O4@SiO2@GO-PEA that can be applied to the magnetic solid-phase extraction of polycyclic aromatic hydrocarbons(PAHs) from water samples.The resulting microspheres(Fe3O4@SiO2@GO-PEA) were characterized by Fourier transform-infrared spectroscopy(FT-IR),scanning electron microscopy(SEM),CHNS elemental analysis,and vibrating sample magnetometry(VSM) techniques.The adsorbent possesses the magnetic properties of Fe3O4 nanoparticles that allow them easily to be separated by an external magnetic field.They also have the high specific surface area of graphene oxide which improves adsorption capacity.Desorption conditions,extraction time,amount of adsorbent,salt concentration,and pH were investigated and optimized.Following desorption,the PAHs were quantified by gas chromatography with flame ionization detection(GC-FID).The limits of detection(at an S/N ratio of 3) were achieved from 0.005 to0.1 μg/L with regression coefficients(R2) higher than 0.9954.The relative standard deviations(RSDs) were below 5.8%(intraday) and 6.2%(inter-day),respectively.The method was successfully applied to the analysis of PAHs in environmental water samples where it showed recoveries in the range between 71.7%and 106.7%(with RSDs of 1.6%to 8.4%,for n = 3).The results indicated that the Fe3O4@SiO2@GO-PEA microspheres had a great promise to extraction of PAHs from different water samples.展开更多
