"Synthetic"allopolyploids recreated by interspecific hybridization play an important role in providing novel genomic variation for crop improvement.Such synthetic allopolyploids often undergo rapid genomic s..."Synthetic"allopolyploids recreated by interspecific hybridization play an important role in providing novel genomic variation for crop improvement.Such synthetic allopolyploids often undergo rapid genomic structural variation(SV).However,how such SV arises,is inherited and fixed,and how it affects important traits,has rarely been comprehensively and quantitively studied in advanced generation synthetic lines.A better understanding of these processes will aid breeders in knowing how to best utilize synthetic allopolyploids in breeding programs.Here,we analyzed three genetic mapping populations(735 DH lines)derived from crosses between advanced synthetic and conventional Brassica napus(rapeseed)lines,using whole-genome sequencing to determine genome composition.We observed high tolerance of large structural variants,particularly toward the telomeres,and preferential selection for balanced homoeologous exchanges(duplication/deletion events between the A and C genomes resulting in retention of gene/chromosome dosage between homoeologous chromosome pairs),including stable events involving whole chromosomes("pseudoeuploidy").Given the experimental design(all three populations shared a common parent),we were able to observe that parental SV was regularly inherited,showed genetic hitchhiking effects on segregation,and was one of the major factors inducing adjacent novel and larger SV.Surprisingly,novel SV occurred at low frequencies with no significant impacts on observed fertility and yield-related traits in the advanced generation synthetic lines.However,incorporating genome-wide SV in linkage mapping explained significantly more genetic variance for traits.Our results provide a framework for detecting and understanding the occurrence and inheritance of genomic SV in breeding programs,and support the use of synthetic parents as an important source of novel trait variation.展开更多

Using reciprocal cross, self-cross, and backcross, w...To fully understand the tomato ovary locule formation of inheritance, two varieties, ovary multi-locule (MLK1) and few-locule (FL1), were used in these studies. Using reciprocal cross, self-cross, and backcross, we have constructed various groups to seek the genetic law and mechanisms of tomato locule formation, for modifying the tomato breeding theory. The parental and the resulting FI and F2 generations have been used to examine the heredity of the locule number. The results showed that few-locule was incompletely dominant. The data from backcross demonstrated a significant effect on the locule number, whereas, the reciprocal cross showed an insignificant effect. Not surprisingly, the locule number was controlled by nuclear genomes, not by exogenous substances. The model that inherited the locule number belonged to the additive-dominant model: additive effects played a very important role and were partially dominant. In addition, the results revealed that the locule number in tomato was mainly controlled by a single gene, whereas, it was modulated by a number of other genes. Finally, the general inheritability and narrow inheritability of the locule number were 69.44% and 52.98%, respectively.展开更多

Hook. and Cryptomeria fortunei Hooibrenk. The chloroplast trnL trnF region and one intra genic segment of the mitochondrial gene, Cox Ⅲ, were amplified from those of the parents and hybrids by PCR using gene specific primers. Cp and mtDNA polymorphisms of the amplified regions were detected between the parents after restriction digestions. Restriction fragment length polymorphism (RFLP) analysis revealed that all the F 1 individuals possessed Cox Ⅲ restriction fragment patterns (characteristic of the paternal parent Cryptomeria fortunei ) and the trnL trnF region (identical to the maternal parent Cunninghamia lanceolata ) showing that a different mode of inheritance for organelle DNA has occurred in the hybrids. Contents of cry1A insecticidal protein were significantly different among transgenic maize lines and various tissues of the same transgenic lines. High expression of cry1A protein occurred in green tissues, such as leaf and husk leaf, and low expression occurred in pith, tassel, ear pith, pollen and silk. The results also showed that the contents of cry1A insecticidal protein in leaves of transgenic maize increased with the advance of development and there was no significant difference in cry1A expression level among various generations of transgenic maize.展开更多

Presently, the ultrastructure of the cytoplasm of the egg cell and pollen tube—immediately before fertilization and during cytoplasmic transmission of male gametophyte—has been described for the same species. The fate of parental plastids and mitochondria in the proembryo has also been followed. The mature egg cell contains a large amount of mitochondria, but seems to lack normal plastids. Most plastids have transformed into large inclusions. Apart from the large inclusions, there are abundant small inclusions and other organelles in the egg cell. During fertilization, pollen tube penetrates into the egg cell at the micropylar end and thereafter the contents are released. Plastid and mitochondrion of male origin are lacking near the fusing sperm_egg nuclei. The second sperm nucleus—not involved in karyogamy—remains at a site near the receptive vacuole. This nucleus is surrounded by large amount of male cytoplasm containing mixed organelles from the sperm cell, tube cell, and egg cell. At the free nuclear proembryo stage, organelles of male and female origin are visible in the perinucleus_cytoplasmic zone. Most of the mitochondria have the same morphological features as those in the egg cell. Some of the mitochondria appear to have originated from the sperm and tube cells. Plastids are most likely of male gametophyte origin because they have similar appearance as those of the sperm and tube cell. Large inclusions in the egg cell become vacuole_like. Paternal plastids have been incorporated into the neocytoplasm of the proembryo. In the cellular proembryo, maternal mitochondria are more abundant. Plastids resembling those of the sperm and tube cell are still present. These cytological results clearly show that in P. tabulaeformis, plastids are inherited paternally and mitochondria bipaternally. The cytological mechanism of plastid and mitochondrion inheritance in gymnosperm is discussed.展开更多

The results indicate that the sperms of Pinus tabulaeformis Carr. contain abundant plastids, mitochondria and organelle DNA. These data provide reliable cytological evidence of paternal plastid and mitochondrion inheritance in Pinus . The results are in line with the confirmed general rule of paternal plastid inheritance in Pinaceae. But whether mitochondria in sperm cells can be transmitted into the embryos is an issue needs further developmental studies. Another important finding is that contrary to earlier classification of the male gamete of Pinaceae into the male nuclei type, the results reveal that male gametes in Pinus tabulaeformis are actually cells. However, the sperm cells are only surrounded by plasma membranes without cell walls. The larger leading sperm cell in a pollen tube section is long in shape, with a large amount of cytoplasm; while the second sperm cell is smaller, round in shape and contains less cytoplasm. Whether this feature of the male gamete type could be considered as a representative characteristic of the family is discussed and further conclusions await more experimental evidences from studies on plants from different species.展开更多

For the grains of F1 generation, the performance of aroma was tested by chewing method, while for the plants of generations of F1, F2 and BC1 F1, the performance of aroma was tested by KOH solution method. The results showed that the non-aroma of restorer line is a dominant trait, but the aroma of GHS line is a recessive trait; the aroma genes of GHS, Yuexiangzhan and Xiangwanxian 17 were non-al elic; the aroma genes of GHS, Yuxiangyouzhan, Tianlongxiang 103, Xin-wanxiang and Chuangxiang 5 may have the same al elic loci. Among the combinations of GHS and non-aromatic restorer lines, the non-aromatic to aromatic ratios of the F2 plants and the BC1 F1 plants were 3∶1 and 1∶1, respectively, and the inheritance of aroma of GHS was control ed by a pair of recessive nuclear genes. In hybrid rice breeding, using GHS, Qiongxiang S has been bred out, and a series of aro-matic hybrid rice combinations have also been selected. Among the hybrid combi-nations, Qiongxiangliangyou 1, Qiongxiangliangyou 2 and other high-yielding and high-quality combinations have been approved by provincial government, and they have good application prospects.展开更多

文摘This study aims to further promote the inheritance and innovative development of intangible cultural heritage in Yunnan Province,promote the protection,inheritance,integration,and innovation of ethnic culture in Lijiang,strengthen the protection and inheritance of Baisha murals in Lijiang,and change the teaching of art design majors in vocational colleges in Yunnan.Given the lack of traditional Chinese culture and local ethnic culture,this article focuses on the teaching of art design majors in Yunnan vocational colleges.It explores the construction model of the cultural inheritance and innovation carrier of Lijiang Baisha murals to meet the spiritual and cultural needs of the local people,and efforts will be made to promote the high-quality development of the Baisha ethnic area in Lijiang.

文摘To fully understand the tomato ovary locule formation of inheritance, two varieties, ovary multi-locule (MLK1) and few-locule (FL1), were used in these studies. Using reciprocal cross, self-cross, and backcross, we have constructed various groups to seek the genetic law and mechanisms of tomato locule formation, for modifying the tomato breeding theory. The parental and the resulting FI and F2 generations have been used to examine the heredity of the locule number. The results showed that few-locule was incompletely dominant. The data from backcross demonstrated a significant effect on the locule number, whereas, the reciprocal cross showed an insignificant effect. Not surprisingly, the locule number was controlled by nuclear genomes, not by exogenous substances. The model that inherited the locule number belonged to the additive-dominant model: additive effects played a very important role and were partially dominant. In addition, the results revealed that the locule number in tomato was mainly controlled by a single gene, whereas, it was modulated by a number of other genes. Finally, the general inheritability and narrow inheritability of the locule number were 69.44% and 52.98%, respectively.

文摘[Objective] The aim was to explore the inheritance of fat and fatty acid compositions in peanut.[Method] Genetic and correlation analyses were performed using a mixed genetic model of major gene plus polygene with 215 F9 RILs and their parents ZHENG 8903 and YUHUA No.4.[Result] The optimal genetic model of the fat content was two linked complementary major gene plus polygene inheritance model.The heritability of major genes was 22.88%.Oleic,linoleic,and palmitic acid content were controlled by two major genes and the heritability was 75.11%,75.00% and 60.95%,respectively.Stearic and arachidic acid content were controlled by two major genes plus polygenes,the heritability of major genes was 26.43% and 33.17%,respectively.O/L ratio and behenic acid content were controlled by three major genes,the heritability was 70.65% and 30.70%,respectively.[Conclusion] The result in this study was that accumulating polygenes should be emphasized in the improvement of fat content,while utilization of major genes is enhanced in improvement of oleic and linoleic acids.
文摘The inheritance of mitochondrial (mt) DNA and chloroplast (cp) DNA was investigated in intergeneric hybrids from crossing between Cunninghamia lanceolata (Lamb.) Hook. and Cryptomeria fortunei Hooibrenk. The chloroplast trnL trnF region and one intra genic segment of the mitochondrial gene, Cox Ⅲ, were amplified from those of the parents and hybrids by PCR using gene specific primers. Cp and mtDNA polymorphisms of the amplified regions were detected between the parents after restriction digestions. Restriction fragment length polymorphism (RFLP) analysis revealed that all the F 1 individuals possessed Cox Ⅲ restriction fragment patterns (characteristic of the paternal parent Cryptomeria fortunei ) and the trnL trnF region (identical to the maternal parent Cunninghamia lanceolata ) showing that a different mode of inheritance for organelle DNA has occurred in the hybrids. Furthermore, the maternal inheritance of chloroplast DNA is reported here for the first time in coniferophyta.
文摘The inheritance of plastid DNA in Pharbitis was studied by the method of restriction fragment length polymorphisms (RFLP).Experimental results showed that plastid DNA from Pharbitis was paternally inherited in reciprocal crosses,P. nil ×P. limbata and P. limbata×P. nil hybrids.But,in the cross of P. limbata×P. nil,the possibility of biparental inheritance of plastid DNA could not be roled out in our preliminary experiment.Thus Pharbitis became the third genus among angiosperms characterized with male plastid transmission.The mechanisms of paternal plastids DNA inheritance in Pharbitis is unclear.The authors proposed that dilution,exclusion and/or degeneration of maternal plastid,including their DNA,after fertilization should be considered.
文摘[Objective] This study aimed to investigate the inheritance mechanism of tomato fruit firmness.[Method] Two tomato cultivars significantly different in fruit firmness were selected for investigation of the inheritance mechanism of tomato fruit firmness using combination analysis of six generations (P1,P2,F1,F2,B1 and B2).[Result] The results indicated that the heredity of tomato fruit firmness was consistent with the additive-dominant model controlled by one pair of major genes; the additive effect (d),dominant effect (h) and degree of dominance (h/d) of major genes were 17.37,-7.96 and-0.46,respectively,showing positive additive effect and incompletely dominant-negative effect; the hereditability of major gene effect in B1,B2 and F2 generation was 88.59%,45.81% and 85.62%,respectively.[Conclusion] The heredity of fruit firmness was controlled by one pair of major gene,showing significant additive effect and dominant effect.
文摘The inheritance of chloroplast DNA (cpDNA) in sweet potato (Ipomoea batatas Lain.) was analyzed using DNA restriction fingerprinting. The cpDNA fingerprints of hybrids from reciprocal crosses between Xushu18 and AB78-1 were found to be identical to those of their female parents, which reveals that cpDNA of sweet potato is maternally inherited in this intervarietal crossing. This maternal cpDNA transmission pattern does not accord with the putative one based on former cytological studies. The plastid inheritance in Convolvulaceae has been briefly reviewed in this study, and the utility of DNA restriction fingerprinting analysis in the study of plastid inheritance is also discussed.

文摘Cultural characteristics of the traditional architectural environment and places of traditional cultural activities in Hanjiaxiang Village,an immigrant village of the Ming Dynasty located in the present Yuangong Township,Chenggu County of Shaanxi Province,were studied to analyze influence of geographical environment,history and culture,and social environment on the village cultures.Specifically,cultural characteristics of traditional architectural environment in this village were elaborated from spatial layout,architectural structure,architectural style and village landscapes.In terms of places for traditional cultural activities,layouts and cultural characteristics of places for the clan's sacrifice and community's fire,traditional sacrifice activities,and local opera performances were introduced.On this basis,cultural inheritance means for the protective renovation and construction of overall village spaces,village buildings,village landscapes and cultural characteristics were proposed.

文摘We investigated the inheritance and expression of cry1A gene in transgenic maize ( Zea mays L.) by Southern blotting analysis and enzyme_linked immunosorbent assays (ELISA). The results showed that cry1A had been transmitted to progeny of transgenic maize as a single gene. Contents of cry1A insecticidal protein were significantly different among transgenic maize lines and various tissues of the same transgenic lines. High expression of cry1A protein occurred in green tissues, such as leaf and husk leaf, and low expression occurred in pith, tassel, ear pith, pollen and silk. The results also showed that the contents of cry1A insecticidal protein in leaves of transgenic maize increased with the advance of development and there was no significant difference in cry1A expression level among various generations of transgenic maize.
文摘In an earlier report the ultrastructure and nucleoid organelles of male gamete in Pinus tabulaeformis Carr. have been described. Presently, the ultrastructure of the cytoplasm of the egg cell and pollen tube—immediately before fertilization and during cytoplasmic transmission of male gametophyte—has been described for the same species. The fate of parental plastids and mitochondria in the proembryo has also been followed. The mature egg cell contains a large amount of mitochondria, but seems to lack normal plastids. Most plastids have transformed into large inclusions. Apart from the large inclusions, there are abundant small inclusions and other organelles in the egg cell. During fertilization, pollen tube penetrates into the egg cell at the micropylar end and thereafter the contents are released. Plastid and mitochondrion of male origin are lacking near the fusing sperm_egg nuclei. The second sperm nucleus—not involved in karyogamy—remains at a site near the receptive vacuole. This nucleus is surrounded by large amount of male cytoplasm containing mixed organelles from the sperm cell, tube cell, and egg cell. At the free nuclear proembryo stage, organelles of male and female origin are visible in the perinucleus_cytoplasmic zone. Most of the mitochondria have the same morphological features as those in the egg cell. Some of the mitochondria appear to have originated from the sperm and tube cells. Plastids are most likely of male gametophyte origin because they have similar appearance as those of the sperm and tube cell. Large inclusions in the egg cell become vacuole_like. Paternal plastids have been incorporated into the neocytoplasm of the proembryo. In the cellular proembryo, maternal mitochondria are more abundant. Plastids resembling those of the sperm and tube cell are still present. These cytological results clearly show that in P. tabulaeformis, plastids are inherited paternally and mitochondria bipaternally. The cytological mechanism of plastid and mitochondrion inheritance in gymnosperm is discussed.
文摘The cytological mechanism of plastid and mitochondrion inheritance in Pinus is an interesting research topic with only a limited number of published articles. The results indicate that the sperms of Pinus tabulaeformis Carr. contain abundant plastids, mitochondria and organelle DNA. These data provide reliable cytological evidence of paternal plastid and mitochondrion inheritance in Pinus . The results are in line with the confirmed general rule of paternal plastid inheritance in Pinaceae. But whether mitochondria in sperm cells can be transmitted into the embryos is an issue needs further developmental studies. Another important finding is that contrary to earlier classification of the male gamete of Pinaceae into the male nuclei type, the results reveal that male gametes in Pinus tabulaeformis are actually cells. However, the sperm cells are only surrounded by plasma membranes without cell walls. The larger leading sperm cell in a pollen tube section is long in shape, with a large amount of cytoplasm; while the second sperm cell is smaller, round in shape and contains less cytoplasm. Whether this feature of the male gamete type could be considered as a representative characteristic of the family is discussed and further conclusions await more experimental evidences from studies on plants from different species.

文摘The inheritance of aroma of GHS was studied by crossing GHS with non-aromatic lines 9311, Xianhui 207, Yuefengzhan, Taihuangzhan, Yuexiangzhan and aromatic lines Yuxiangyouzhan, Xiangwanxian 17, Tianlongxiang 103, Xinwanxiang and Chuangxiang 5. For the grains of F1 generation, the performance of aroma was tested by chewing method, while for the plants of generations of F1, F2 and BC1 F1, the performance of aroma was tested by KOH solution method. The results showed that the non-aroma of restorer line is a dominant trait, but the aroma of GHS line is a recessive trait; the aroma genes of GHS, Yuexiangzhan and Xiangwanxian 17 were non-al elic; the aroma genes of GHS, Yuxiangyouzhan, Tianlongxiang 103, Xin-wanxiang and Chuangxiang 5 may have the same al elic loci. Among the combinations of GHS and non-aromatic restorer lines, the non-aromatic to aromatic ratios of the F2 plants and the BC1 F1 plants were 3∶1 and 1∶1, respectively, and the inheritance of aroma of GHS was control ed by a pair of recessive nuclear genes. In hybrid rice breeding, using GHS, Qiongxiang S has been bred out, and a series of aro-matic hybrid rice combinations have also been selected. Among the hybrid combi-nations, Qiongxiangliangyou 1, Qiongxiangliangyou 2 and other high-yielding and high-quality combinations have been approved by provincial government, and they have good application prospects.
