【目的】南方根结线虫是威胁桃产业绿色发展的地下主要害虫,开发抗性分子标记,对抗性砧木分子育种具有重要意义。【方法】根据前人对桃砧木抗根结线虫的定位结果,在GDR网站peach genome V2.0查询定位区间的候选基因。在5个抗病、5个感... 【目的】南方根结线虫是威胁桃产业绿色发展的地下主要害虫,开发抗性分子标记,对抗性砧木分子育种具有重要意义。【方法】根据前人对桃砧木抗根结线虫的定位结果,在GDR网站peach genome V2.0查询定位区间的候选基因。在5个抗病、5个感病种质中扩增候选基因,并通过DNAMAN、IGV等软件对候选基因进行序列差异分析,开发抗南方根结线虫KASP分子标记,在抗性种质筑波3号与感性种质哈露红的杂交F2群体中对该分子标记进行验证,并与前人开发的SCAR和35 bp indel抗南方根结线虫分子标记的准确性进行比较。【结果】KASP标记结果将基因型划分为3种,分别为抗性纯合型(A),抗性杂合型(B),感性纯合型(C),A∶B∶C=42∶94∶64,与表型符合率为88.5%;SCAR标记检测结果划分为2种,分别为抗病型(A1)和感病型(C1),A1∶C1=135∶65,与表型符合率为87.0%;35 bp indel分子标记分为3种类型,分别为抗性纯合型(A2),抗性杂合型(B2),感性纯合型(C2),A2∶B2∶C2=1∶154∶45,与表型符合率为52.0%。【结论】本研究中开发的KASP标记提高了分子标记选择准确率,对抗南方根结线虫分子育种具有重要意义。 展开更多

【目的】深入探寻东北杏种质资源遗传多样性,筛选关键综合评价指标,提升育种效率,发掘优良东北杏无性系并高效利用。【方法】采用多样性分析、聚类分析、相关性分析和主成分分析,对69个东北杏无性系的45个果实表型性状(23个质量性状和2... 【目的】深入探寻东北杏种质资源遗传多样性,筛选关键综合评价指标,提升育种效率,发掘优良东北杏无性系并高效利用。【方法】采用多样性分析、聚类分析、相关性分析和主成分分析,对69个东北杏无性系的45个果实表型性状(23个质量性状和22个数量性状)进行研究。【结果】41个表型性状变异系数大于0.1,数量性状的平均Shannon-wiener多样性指数(1.91)高于质量性状(0.62),数量性状的多样性较质量性状更丰富。聚类分析将69个东北杏无性系分为3类,A类适用于薄壳、出仁率高的品种类群;B类可用于选育高产、大仁型品种,属优良仁用杏品种类群;C类可作为高出核率和出仁率的重要育种材料。果肉薄、果小的无性系出核率更高;核壳薄、单核质量小的无性系出仁率更高;单核质量大的无性系产核量更高。5个主成分的累计贡献率达84.42%,能够反映22个数量性状的基本信息。结合相关性分析和主成分分析,22个数量性状可简化为果肉厚度、果纵径、单果质量、单核质量、单仁质量、出核率、出仁率、产核量共8个关键评价指标,筛选出10个果实经济性状表现优异的无性系。【结论】东北杏无性系的果实性状表现出丰富变异,经多指标综合评价筛选出的803、777、783、720、721、725、719、711、731和784号无性系可用于东北杏良种选育及开发利用。 展开更多

A wide survey was conducted to study plant-parasitic nematodes(PPNs)associated with Prunus groves in Spain.This research aimed to determine the prevalence and distribution of PPNs in Prunus groves,as well as the influence of explanatory variables describing soil,climate and agricultural management in structuring the variation of PPNs community composition.A total of 218 sampling sites were surveyed and 84 PPN species belonging to 32 genera were identified based of an integrative taxonomic approach.PPN species considered as potential limiting factors in Prunus production,such as Meloidogyne arenaria,M.incognita,M.javanica,Pratylenchus penetrans and P.vulnus,were identified in this survey.Seven soil physico-chemical(C,Mg,N,Na,OM,P,pH and clay,loamy sand and sandy loam texture classes),four climate(Bio04,Bio05,Bio13 and Bio14)and four agricultural management variables(grove-use history less than 10 years,irrigation,apricot seedling rootstock,and Montclar rootstock)were identified as the most influential variables driving spatial patterns of PPNs communities.In particular,younger plantations showed higher values for species richness and diversity indices than groves cultivated for more than 20 years with Prunus spp.Our study increases the knowledge of the distribution and prevalence of PPNs associated with Prunus rhizosphere,as well as on the influence of explanatory variables driving the spatial structure PPNs communities,which has important implications for the successful design of sustainable management strategies in the future in this agricultural system. 展开更多

Water management is an important practice that affects fruit size and quality.Effective implementation of irrigation scheduling requires knowledge of the appropriate indicators and thresholds,which are established manly based on the effects of water deficits on final fruit quality.Few studies have focused on the real-time effects of water status on fruit and shoot growth.To establish soil water potential (ψ_(soil)) thresholds to trigger irrigation of peach at pivotal fruit developmental stages,photogrammetry,^(13)C labelling,and other techniques were used in this study to investigate real-time changes in stem diameter,fruit projected area,net leaf photosynthetic rate (P_(n)),and allocation of photoassimilates to fruit under soil water potential conditions ranging from saturation to stress in 6-year-old Shimizu hakuto’peach.Stem growth,fruit growth,and P_n exhibited gradually decreasing sensitivity to water deficits during fruit developmental stages I,II,and III.Stem diameter growth was significantly inhibited whenψ_(soil)dropped to-8.5,-7.6,and-5.4 k Pa,respectively.Fruit growth rate was low,reaching zero when theψ_(soil)was-9.0 to-23.1,-14.9 to-21.4,and-16.5 to-23.3 k Pa,respectively,and P_ndecreased significantly when theψ_(soil)reached-24.2,-22.7,and-20.4 kPa,respectively.In addition,more photoassimilates were allocated to fruit under moderateψ_(soil)conditions (-10.1 to-17.0 k Pa) than under otherψ_(soil)values.Our results revealed threeψ_(soil)thresholds,-10.0,-15.0,and-15.0 kPa,suitable for triggering irrigation during stages I,II,and III,respectively.These thresholds can be helpful for controlling excessive tree vigor,maintaining rapid fruit growth and leaf photosynthesis,and promoting the allocation of more photoassimilates to fruit. 展开更多

Peach, an economically important model plant of the Rosaceae family, has been domesticated and cultivated in China for approximately5 000 years. The Hexi Corridor, an important corridor connecting east-central China and the Tarim Basin, is the starting point of the Silk Road that links China and the Eurasian region. As a globally distributed fruit tree, the spread of peach was accomplished through historical trade routes in the Hexi Corridor and the Tarim Basin. However, knowledge of peach genetic diversity in these regions remains limited. In this study,we examined the relationships and the spread history of domesticated peaches through sequencing and genomic analysis of 161 peach accessions collected from Northwest China, including 43 from the Hexi Corridor and 104 from the Tarim Basin. The results indicate that peach landraces in the Hexi Corridor and the Tarim Basin are derivatives of peaches from the east and south of China (ESC). Notably, the genetic diversity of accessions from both the Hexi Corridor and the Tarim Basin was lower than that of ESC accessions. Reduction of diversity (ROD) and linkage disequilibrium (LD) analyses detected a genetic bottleneck in peaches from these regions. Additionally, these peaches have undergone varying degrees of selection from natural environment. Moreover, genes responsive to biotic and abiotic stresses were under selection, which could be the result of the climate change of Northwest China after the Last Glacial Maximum (LGM). Our findings provide a better understanding of the genetic basis of peach migration in Northwest China. Furthermore, this study expands the available genomic data for peaches and provides critical information for future peach breeding programs. 展开更多

Bud dormancy facilitates the survival of meristems under harsh environmental conditions.To elucidate how molecular responses to chilling accumulation controlling dormancy in peach buds,chromatin immunoprecipitation sequencing to identify the H3K27me3 modifications and RNA sequencing of two peach cultivars with pronounced differences in chilling requirement were carried out,the results showed that genes associated with abscisic acid and gibberellic acid signal pathways play key roles in dormancy regulation.The results demonstrated that peach flower bud differentiation occurred continuously in both cultivars during chilling accumulation,which was correlated with the transcript abundance of key genes involved in phytohormone metabolism and flower bud development under adverse conditions.The more increased strength in high chillingrequirement cultivar along with the chilling accumulation at the genome-wide level.The function of the dormancy-associated MADS-box gene PpDAM6 was identified,which is involved in leaf bud break in peach and flower development in transgenic Nicotiana tabacum(NC89).In addition,PpDAM6 was positively regulated by PpCBF,and the genes of putative dormancy-related and associated with metabolic pathways were proposed.Taken together,these results constituted a theoretical basis for elucidating the regulation of peach bud dormancy transition. 展开更多

In this study,high performance liquid chromatography(HPLC)and RNA-seq transcriptome sequencing were used to study the changes in soluble sugar components and flavonoids in Prunus persica‘Jinxiangyu’at different developmental stages(20–90 d after flowering)and screen the key genes regulating the formation of soluble sugar and flavonoids in the fruits.The results showed that 60–85 d after flowering was the key stage of quality formation of Prunus persica‘Jinxiangyu’,and the content of soluble sugar,soluble solid,fructose,and sucrose in the fruit increased significantly during this period.The sugar content of ripe fruits was mainly fructose and sucrose.The content of kaempferol glycoside was low in the fruit.Quercetin glycoside content was higher in the young fruit stage and decreased with fruit maturity.There were no anthocyanin compounds in the fruit.The expression levels of genes involved in flavonoid metabolism(ANS,DFR,F3H,FLS,4CL1,etc.)were low in the fruit.A total of 181 differentially expressed genes were identified during fruit development to participate in five sugar metabolism pathways,among which the SDH gene had a higher expression level,which continuously rised in the later stage of fruit development.It mainly promoted the accumulation of fructose content in the later stage of fruit development.The expression levels of SPS1,SS,and SS1 genes were continuously up-regulated,which played a key role in sucrose regulation.The higher expression levels of SUS3 and INVA genes in the early stage of fruit development promoted the degradation of sucrose. 展开更多

This paper delves into and assesses the differences in biological traits and fruit nutritional composition of 3 varieties of Prunus humilis introduced from 2 different regions.Results showed that(i)the contents of protein,total sugar,and calcium of No.5 variety were higher those of the other 2 varieties,which was recognized as the most suitable variety for fresh fruit;(ii)the comparison of leaf areas index and fruit shape index of 3 varieties shows that the fruit shape index and individual fruit weight of the introduced varieties were higher than those of the control,the diameter of individual fruit of the introduced varieties higher than that of the control indicated that they had a higher fruit hardness,more dry matter accumulation and better fruit quality,and that the introduction region was more suitable for scale-up of P.humilis;(iii)there was a positive correlation with the number of fruiting branches and there was a significant negative correlation with individual fruit weight,while the leaf area had a significant negative correlation with the number of fruiting branches.The introduced varieties can develop normally in the 2 testing areas,notably,No.5 variety performed better than No.4 and No.6.Improving management is the prerequisite for maintaining proper number of fruiting branches and high yield. 展开更多

Cameroon is one of the important exporters of the barks. Despite the important measures adopted in Cameroon for sustaining its harvesting, some many chalenges still remain. The objective of this work is to refine the forest management parameters in relation to P. africana in the regions of Adamaoua and the South-West by developing a volume rate which makes it possible to estimate the production for a new stem. The work took place in two phases: in the South-West in 2010 and in Adamaoua in 2011. Data collection used the semi-direct method, while the cubing equation was deduced by the multiple linear regression method. Two models for volume estimation and three models for mass prediction were developed. The predictive parameters retained are diameter, height of the bole and thickness of the bark. Results show that the average mass of the dry bark for a given P. africana tree species is 27.55 ± 14.44 kg and this varies according to the site. The strong adjusted coefficient of determination (adjusted R2) observed illustrates the reliability of the proposed models. These models provide a reliable tool that can be adopted as a standard in Cameroon for P. africana. 展开更多

Polygalacturonase (PG,EC3.2.1.15) is the key cell wall hydrolase in fruit ripening. Polygalacturonase (PG,EC3.2.1.15) is the key cell wall hydrolase in fruit ripening. The identification and characterization of a full length cDNA (pMT18) encoding for PG from Feicheng peach (Prunus persica (L.) Batsch cv. Feicheng) is described. The pMT18 clone is 1188 bp in length, with an open reading frame of 393 amino acids. The homology and phylogenetic analyses indicate a remarkable similarity between peach PG and other ripening related PG. And seven consensus sequences have revealed in peach PG compared to the PG from other plants. However, the profound divergence with other PG and the unique structure features suggest that peach PG probably belongs to a new evolutionary class. In RT PCR analysis, pMT18 related RNA was undetectable in leaves, and was much abundant in ripe fruits. The ripening specific expression pattern of this cDNA will be useful in investigating the roles of PG in fruit ripening and developing a transgenic peach with the improved post harvesting quality in the future. 展开更多

MADS box proteins play an important role in floral development. To find genes involved in the floral transition of Prunus species, cDNAs for two MADS box genes, PpMADS1 and PpMADSIO, were cloned using degenerate primers and 5'- and T-RACE based on the sequence database of P. persiea and P. duleis. The full length of PpMADS1 cDNA is 1,071 bp containing an open reading frame (ORF) of 717 bp and coding for a polypeptide of 238 amino acid residues. The full length of PpMADSIO cDNA is 937 bp containing an ORF of 633 bp and coding for a polypeptide of 210 amino acid residues. Sequence comparison revealed that PpMADS1 and PpMADSIO were highly homologous to genes API and PI in Arabidopsis, respectively. Phylogenetic analysis indicated that PpMADS1 belongs to the euAP1 clade of class A, and PpMADSIO is a member of GLO/PI clade of class B. RT-PCR analysis showed that PpMADS1 was expressed in sepal, petal, carpel, and fruit, which was slightly different from the expression pattern ofAPl; PpMADS10 was expressed in petal and stamen, which shared the same expression pattern as PI. Using selective mapping strategy, PpMADSI was assigned onto the Binl:50 on the G1 linkage group between the markers MCO44 and TSA2, and PpMADSIO onto the Bin1:73 on the same linkage group between the markers Lap- 1 and FGA8. Winter chill is essential for the growth and development of deciduous species. To understand the relationship between accumulated chilling hours during endodormancy and blooming and fruit shape development, we controlled chilling hours and investigated their effects on blooming date and fruit shape of peaches. The results showed that the number of days to full bloom date and the heat requirement for blooming were negatively correlated with accumulated chilling hours. Accumulated chilling hours were significantly negatively correlated with fruit shape index and fruit tip lengths, suggesting that the number of chilling hours affect the fruit shape development. Fewer accumulated chilling hours may be the major reason for longer fruit shape and protruding fruit tips. In conclusion, our results indicate specifically that decreased winter chilling hours can delay the bloom date and may lead to aberrant fruit shape development in peaches. Our study provides preliminary insights into the response of temperate fruit species to global climate change. 展开更多

Objective:To investigate the physicochemical properties of unripe peach-Pruuns persica cv.Mibaekdo(Mibaekdo) and Prunus persica cv.Nagasawa Hakuho(Nagasawa Hakuho) as an alternative to fowl supplement while Japanese apricot(Prunus mume cv.Backaha)(Backaha) was used as a control sample.Methods:The unripe fruits were analyzed for soluble solid(Brix).titratable acidity.pH,total polyphenol content,1,1-diphenyl-2-pierylhydrazyl(DPPH) radical scavenging activity,amygdalin content,free amino acid content,organic acid content,free sugar content,and α-amylase activities.Results:Total polyphenol content of unripe peach ranged between 137.27-151.64 μg/g whereas that of apricot was 160.73 μg/g.DPPH radical scavenging activities of Rackaha was the highest189.16%.) followed by Mibaekdo(85.03%) and Nagasawa Hakuho(41.30%).The highest amount of oxalic acid 1612.8 mg/100 g) was observed in Mibaekdo while that of Nagasawa llakuho and Backaha were(184.6±18.1) and(334.8±16.1) mg/100 g,respectively.Amygdalin contents of Mibaekdo.Nagasawa Hakuho and Backaha were 486.61,548.60 and 174.28 μg/g,respectively.Conclusions:The results suggest that the unripe fruit of peach has a significant biochemical potential of using as a food supplement with potential health benefit for human health. 展开更多

A pot experiment was conducted to investigate the effects of nitrogen content [Nl (no fertilizer), N2 (0.15 g.kg-l), and N3 (0.3 g.kg 1)] on the growth and the hydraulic characteristics of peach seedlings under different soil moisture conditions (Wl, W2 and W3, in which the soil water content was 45% to 55%, 60% to 70%, and 75% to 80% of the field water capacity, respectively) by using a specialized high pressure flow meter with a root chamber and a coupling, which was connected to plant organs. Leaf area and leaf hydraulic conductivity (KL) increased significantly in the seedlings because of increased soil moisture and N content. KL increased with leaf area. A linear correlation was documented between KL and leaf area. KL was higher in the morning and began to decline sharply after 16:00, at which KL declined after an initial increase. Soil moisture and N content enhanced shoot (Ks) and root (Kr) hydraulic conductivities, thereby improving the low soil moisture condition to a large extent. Ks and Kr of the seedlings were reduced by 32% and 27% respectively in N~, and by 14.7% and 9.4%, respectively in N2, and both in Wb compared with the control treatment. N3 had no significant effect on Ks and Kr under similar conditions. Linear negative correlations were observed between Kr and the excised root diameter as well as between Ks and the shoot stem diameter. The shoot-to-root ratio increased with in- crease in N content. The shoot-to-root ratio in N3 was increased by 14.37%, compared with N1 in W1 as well as by 12% and 4.39% in Wz and W3, respectively. Knowledge of the effects of soil moisture and N fertilizer on hydraulic characteristics and growth is important. Our results provide basic guidelines for the implementation of water-saving irrigation and fertilization management of nursery stock. 展开更多

Prunus species include many important perennial fruit crops,such as peach,plum,apricot,and related wild species.Here,we report de novo genome assemblies for five species,including the cultivated species peach(Prunus persica),plum(Prunus salicina),and apricot(Prunus armeniaca),and the wild peach species Tibetan peach(Prunus mira)and Chinese wild peach(Prunus davidiana).The genomes ranged from 240 to 276 Mb in size,with contig N50 values of 2.27−8.30Mb and 25,333−27,826 protein-coding gene models.As the phylogenetic tree shows,plum diverged from its common ancestor with peach,wild peach species,and apricot~7 million years ago(MYA).We analyzed whole-genome resequencing data of 417 peach accessions,called 3,749,618 high-quality SNPs,577,154 small indels,31,800 deletions,duplications,and inversions,and 32,338 insertions,and performed a structural variant-based genome-wide association study(GWAS)of key agricultural traits.From our GWAS data,we identified a locus associated with a fruit shape corresponding to the OVATE transcription factor,where a large inversion event correlates with higher OVATE expression in flat-shaped accessions.Furthermore,a GWAS revealed a NAC transcription factor associated with fruit developmental timing that is linked to a tandem repeat variant and elevated NAC expression in early-ripening accessions.We also identified a locus encoding microRNA172d,where insertion of a transposable element into its promoter was found in double-flower accessions.Thus,our efforts have suggested roles for OVATE,a NAC transcription factor,and microRNA172d in fruit shape,fruit development period,and floral morphology,respectively,that can be connected to traits in other crops,thereby demonstrating the importance of parallel evolution in the diversification of several commercially important domesticated species.In general,these genomic resources will facilitate functional genomics,evolutionary research,and agronomic improvement of these five and other Prunus species.We believe that structural variant-based GWASs can also be used in other plants,animal species,and humans and be combined with deep sequencing GWASs to precisely identify candidate genes and genetic architecture components. 展开更多

As the preferred nitrogen(N)source,ammonium(NH_(4)^(+))contributes to plant growth and development and fruit quality.In plants,NH 4+uptake is facilitated by a family of NH_(4)^(+) transporters(AMT).However,the molecular mechanisms and functional characteristics of the AMT genes in peach have not been mentioned yet.In this present study,excess NH_(4)^(+) stress severely hindered shoot growth and root elongation,accompanied with reduced mineral accumulation,decreased leaf chlorophyll concentration,and stunned photosynthetic performance.In addition,we identified 14 putative AMT genes in peach(PpeAMT).Expression analysis showed that PpeAMT genes were differently expressed in peach leaves,stems and roots,and were distinctly regulated by external NH_(4)^(+) supplies.Putative cis-elements involved in abiotic stress adaption,Ca^(2+) response,light and circadian rhythms regulation,and seed development were observed in the promoters of the PpeAMT family genes.Phosphorylation analysis of residues within the C-terminal of PpeAMT proteins revealed many conserved phosphorylation residues in both the AMT1 and AMT2 subfamily members,which could potentially play roles in controlling the NH 4+transport activities.This study provides gene resources to study the biological function of AMT proteins in peach,and reveals molecular basis for NH_(4)^(+) uptake and N nutrition mechanisms of fruit trees. 展开更多

Because there are thousands of peach cultivars,cultivar classification is a critical step before starting a breeding project.Various molecular markers such as simple sequence repeats(SSRs)can be used.In this study,67 polymorphic primers produced 302 bands.Higher values for SI index(1.903)suggested higher genetic variability in the genotype under investigation.Mean values for observed alleles(Na),expected heterozygosity(He),effective alleles(Ne),Nei’s information index(h),and polymorphic information content(PIC)were 4.5,0.83,5.45,0.83,and 0.81,respectively.The dendrogram constructed based on Jaccard’s similarity coefficients outlined four distinct clusters in the entire germplasm.In addition,an analysis of molecular variance(AMOVA)showed that70.68%of the total variation was due to within-population variation,while 29.32%was due to variation among populations.According to this research,all primers were successfully used for the peach accessions.The EST-SSR markers should be useful in peach breeding programs and other research. 展开更多
